Some Observations on the Fine Structure of Korean Bat Livers (한국산 박쥐 간장의 미세구조적 고찰)

  • To understand the fine structure of Korean bat livers , the authors investigated the liver of four Korean bats; Rhinolephus ferrumequinum korai, Myotis macrodactylus, Myotis dauhentonii ussrinesis, and Murina leucogaster intermedia by transmission electron microscopy. The hepatocytes of Rhinolophus ferrumequinum korai had large-sized mitochondria and many peroxisomes. In the Myotis macrodactylus, juctional completes, especially desmosomes, were well developed. The Myotis daubentonii ussrinesis had many glycogen particles in the cytoplasm. Also, the space of Disse and sinusoidal spare was filled with amorphous materials. In the Murina leucogaster intemedia, basement membrane was prominent in the sinusoid, and no Kupffer and Ito cells were observed These results suggest the characteristic differences in the liver ultrastructure among Korean bats.

Fine Structural Study of Pollen Wall Development at Late Stage of Microsporogenesis in Panax ginseng (인삼의 화분벽 발달에 관한 미세구조적 연구)

  • The ontogeny of pollen wall in Panax ginseng was studied with transmission and scanning electron microscopy from early tetrad stage until pollen maturity. Initial indication of exine development is undulation of plasma membrane for the preparation of bacular mound. The first recognizable structure of the pollen wall is the cellulosic primexine which is formed outside of the plasma membrane while microspore tetrads are still surrounded by callose wall. As development proceeds, foot-layer and baculum differentiation, callose dissolution and exine formation were progressed. During this process, sporopollenin is deposited into the exine, and then endexine development was followed. The intine, innermost pollen wall layer, is developing form hypertrophic Golgi vesicles. The thickness of exine is very even on all along the pollen wall, but intine thickness of apertural region is thicker than that of nonapertural region. Mature pollen of ginseng is $20{\mu}m$ in size, tricolpate and shows fine reticulate sculpturing.

Preparation and Characteristics of Te Fine Particles Doped SiO2 Glass Thin Films by Sol-gel Method (졸-겔법에 의한 Te 미립자 분산 SiO2 유리 박막의 제조와 특성)

  • Nanocomposite of Te doped $SiO_2$ films was prepared for the new functional materials like non-linear optic materials, selective absorption and transmission films. The effects of particle size and morphology with different hydrolysis conditions on the properties were examined with TGA/DTA. XRD. UV-spectrometer, SPM, SEM and EDS. It was found that Te/$SiO_2$ films showed high absorption peak at 550nm visible region by plasma resonance of Te fine particles. The Rm surface roughness of the films was about 2.5 nm and the size of Te particles was 5~10nm.

Ultrastructural Changes of Hair Treated with Bleaching Agent (탈색된 머리카락의 미세구조적 변화)

  • This study applied a bleaching agent. which is commonly used in the beauty salons, to the hair of normal adult women, collected the hair immediately and 10 days and 20 days from the bleaching, were investigated the degree of degradation of the hair by using scanning and transmission electron microscopes. The surface of hair just after bleaching was observed to be similar to that of normal hair, showing no split or damage of scale. In the hair of 10 days after bleaching, however, the scale came off. From this time, scale on the cuticular layer of hair began to be separated. In 10 days from bleaching, the scale on the cuticular layer was separated from hair and some cytoplasm of cuticular cells was broken into pieces or fell off. The cell remains made the surface coarse and uneven. At this period, damaged scales had a sharp end. In the hair of 20 days after bleaching, scale fell off from the whole surface of the hair and the surface looked rough. On the bleached hair, many vacuoles were formed in the endocuticle of cuticular cells. As a result, deformation caused by the formation of vacuoles in cuticles broke up the cuticular cells.

Preparation and Evaluation of Microcapsule/Emulsions via the Electroatatic Interations of Polysaccharide and Protein (식물 유래 다당류/단백질 기반 마이크로캡슐/에멀젼 제조 및 평가)

  • A novel microcapsule/emulsions for cosmetics was studied. Our present studies demonstrate that the biopolymer-stabilized microemulsion composed of polysaccharide and protein can encapsulate and stabilize remarkably coenzyme-Q10 (Q10). Polysaccharide and protein complex were incorporated in the microcapsule in order to reinforce the physical strength of the microspheres. We compared the long-term stability of the activity of Q10 in biopolymer-stabilized microemulsion. There was no noticeable negative effect on the activity of Q10. Optical microscope (OM) and transmission electron microscope (TEM) showed that microcapsules were spherical and had a smooth surface. Consequently, the polysaccharide/protein emulsion produced in this study may be beneficial in improving the emulsion stability and the protection capability of labile substances.

Extension of Storage Time of Chicon using Nanoparticle of Machilus thunbergii Extracts (후박추출물의 나노입자화를 통한 치콘의 저장기간 연장)

  • Machilus thunbergii has been showed relation to antimicrobial activity on minimal inhibitoty concentration (MIC) and colony forming inhibitory activity (CFIA) test, so that can be used to food preservatives for green vegetable. Nanoparticles has been made of edible materials. 80% of the nanoparticles has been characterized by image analyser and electron microscopy, showing in the range under 300 nm diameter. The sprayed nanoparticles remained on the surface of chicon even after washing by dilution water, then activate biological activities for storage of chicon with storing and releasement system of extracts. Chicon treated nanoparticle has been kept fresh condition about 2 months longer than 3 weeks of the non-treated control. It can be tell that treatment with nanoparticle of M. thunbergii extracts extends storage time of chicon possibly by inhibition of ethylene production through efficiency control on cell breathing.

  • 최근, nanorod나 nanowire와 같은 1차원의 나노구조가 나노디바이스로 각광을 받고 있다. [1] 특히 InN는 3족 질화물 반도체 중 가장 작은 밴드갭 에너지와 뛰어난 수송 특성을 가지고 있어 나노디바이스로의 응용에 적합한 물질이다. [2] 그러나 InN는 큰 평형증기압을 가지므로 쉽게 인듐과 질소로 분해되는 특성이 있어 나노구조로의 성장이 쉽지 않음이 알려져 있다. [3] 최근 연구결과에 따르면, InN 나노구조는 금속 catalyst를 사용한 방법이나, 기판 위 패턴을 이용하여 성장하는 방법, 염소를 사용한 방법이 널리 쓰이고 있다. [4,5,6] 그러나 이 방법들은 의도치 않은 불순물의 원인이 되거나 다른 추가적인 과정을 필요로 한다는 문제점도 일부 가지고 있다. 본 연구에서는 catalyst-free 유기 금속 화학 증착법 (MOCVD)를 이용하여 $Al_2O_3$ (0001)면 위에 InN nanostructure를 성장하였다. InN nanostructure 성장 시 트리메틸인듐(TMIn)과 암모니아($NH_3$) 를 전구체로 사용하였으며, 캐리어 가스로는 질소를 사용하였다. 또한 모든 샘플의 성장시간은 60분으로 고정하였으나, 성장 시 온도의 의존성을 보기 위해 $680-710^{\circ}C$ 의 온도범위에서 성장을 진행하였다. 그 결과 InN는 본 실험에서 적용된 성장온도범위 내에서 온도가 증가함에 따라 초기에는 columnar구조로 성장된 박막의 형태에서 wall이 배열된 형태로 변화하며 결국 $710^{\circ}C$ 의 온도에서 nanorod로 성장하게 된다. 성장된 InN의 나노구조는 X-선 회절 측정법, 주사 전자 현미경 그리고 투과 전자 현미경을 이용하여 각각의 구조적 특성을 분석하였다. X-선 회절 측정법과 주사 전자 현미경을 통한 분석결과에서는 이들 nanorods가 대부분 c 방향으로 수직하게 정렬되어 있음을 확인 할 수 있었다. 또한, $690^{\circ}C$ 에서 60분간 성장된 InN의 wall 구조의 두께는 200 nm, 길이는 $2-2.5\;{\mu}m$로 관찰되었으며, $710^{\circ}C$에서 60분간 성장된 InN nanorod의 지름은 150 nm, 길이는 $3\;{\mu}m$ 정도로 관찰되었다. 이를 통하여 볼 때 성장 온도가 InN의 나노구조 형성 시 표면의 모폴로지변화에 중요한 변수로 작용함을 알 수 있다. 본 발표에서는 이러한 표면 형상 및 구조 변화가 성장온도에 따른 관계성을 가짐을 InN의 분해와 성장의 경쟁적인 관계에 의해 논의할 것이다.

Studies on the Visceral Ganglion and Right Parietal Ganglion in the African Giant Snail, Achatina fulica II. Ultrastructural Method (아프리카 왕달팽이 (Achatina fulica) 내장신경절 및 우 체벽신경절에 관한 연구 II. 미세구조적 방법)

  • Five kinds of neurosecretory cells (type-A, B, C, D and E) and neuropiles surrounding them were observed in the visceral ganglion and the right parietal ganglion of the African giant snail, Achatina fulica, by transmission electron microscopy. Type-A cells (diameter, $35{\mu}m$) are the most popular cells in the cortex of the two ganglions, which are of triangular or irregular forms. In their cytoplasm, there are found large granules of 1 fm in diameters and small round granules of about $0.1{\mu}m$ in diameters. Small granules are classified into the ones of high electron density and the others of middle electron density. Type-B cells (diameter, $19\times12{\mu}m$) are evenly distributed over various portions of cortex and medulla of the two ganglions. They are similar to type-A cells in shapes. The cytoplasm of type-B cells is crowded with high electron dense granules of about $0.1{\mu}m$. Round granules of about $0.7{\mu}m$ in diameters are also found but rarely. Type-C cells are the smallest cells whose sizes are about $8\times6{\mu}m$. Each of them contains a large nucleus of about $6\times5{\mu}m$. Its cytoplasm is full of electron dense granules of about $0.23{\mu}m$, each of which is artually an assembly of tiny granules of about $0.03{\mu}m$. Type-D cells are middle-size cells of about $28\times20{\mu}m$, which take ellipsoidal or irregular forms. They are found in the cortex more than in the medulla. Their cytoplasm looks dark due to the high electron density and, in it, two kinds of round granules whose sizes are $1.6{\mu}m$fm and $0.6{\mu}m$, respectively, are observed. Type-E cells are large cells of about $100\times50{\mu}m$, which are rarely found in the upper and middle portions of the two ganglions. The nucleus of the cell, which is very large $(70\times30{\mu}m)$ for the cytoplasm, contains electron dense round granules of diverse sizes (diameters, $1\sim0.2{\mu}m$). The surface of the cell protrudes filopodia of various forms and phagocytizes decrepit cells. Neuropiles are surrounding the neurosecretory cells. In nerve fibers, synaptic vesicles are observed, which are classified into six classes according to their electron densities , sizes and shapes.

Effect of filtrates of Serpulina(Treponema) hyodysenteriae on the epithelium in colonic loops in swine (Serpulina(Treponema)hyodysenteriae 배양여과액의 돼지 공장 계제 점막에 미치는 영향)

  • The cytotoxic effects of S hyodysenteriae strain B 204 on the mucosal surface were studied in surgically prepared ligated colonic loops in two male convenitonal mixed-breed pigs. In each one of four loops was inoculated with either sterile trypticase soy broth(TSB) of Serpulina, filtrate of Serpulina TSB culture. washed Serpulina cells or whole culture of Serpulina. Mucosal tissues were examined by transmission and scanning electron microscopy 24 and 48 hours after inoculation(p.i.). The filtrate did not induce any significant effect on the mucosal surface. The washed cells produced early lesions similar to those caused by the whole culture. These observations suggest that cytotoxins of the culture do not play a significant role in invasion of the epithelium in this experimental infection. The possible role of toxins associated with the organism at the site of interaction with the epithelial cells was not elucidated.

MORPHOLOGICAL CHARACTERISTICS OF ODONTOBLAST IN NFI-C KNOCK/OUT MICE (Nuclear Factor I-C 결손 생쥐에서 상아모세포의 형태학적 특징)

  • NFI-C null mice demonstrated aberrant odontoblast differentiation and thus abnormal dentin formation while other tissues/organs in the body, including ameloblasts, appear to be unaffected and normal. However little is known about the mechanism of NFI-C function in odontoblast differentiation and dentin formation. Odontoblasts are tall, highly polarized cells that are responsible for formation and maintenance of the predentin and dentin. An indication of their polarity is the acquisition of specialized intercellular junctions. As preodontoblasts differentiate into odontoblasts, they are Joined and attached at the apical end by well developed terminal webs of cytoskeletal actins, and associated tight as well as adherent njunctions. In this study, in order to investigate if disruption of the NFI-C gene interferes with formation of a specific or other structural proteins of the intercellular junctions, we examined morphological characteristic of the aberrant odontoblast in NFI-C null mice using light and electron microscope. In addition, we determined the expression of major structural proteins of intercellular junctions, ZO-1 and occludin, during the differentiation of odontoblasts using immunohitochemistry. The results were as follows : 1. In light microscopy, abnormal odontoblasts of incisors of the NFI-C null mice were round in shape, lost their polarity, and trapped in osteodentin-like mineralized tissue. Mutant molars have relatively normal crowns, but short and abnormal differentiating adontoblasts in root formation area. 2. Electron microscopy of abnormal odontoblasts revealed the dissociation of the round osteoblast-like cells, the loss of their cellular polarity, and the absence of an intercellular junctional complex known as the tight junctions. 3. A mutant incisor showed labeling for ZO-1 at the proximal and distal ends of secreting ameloblasts, while staining for ZO-1 was not observed in the abnormal odontoblasts. 4. A normal incisor showed immunoreactivity for occludin in the differentiating odontoblasts. However, staining for occludin was not observed in the abnormal odontoblasts of mutant incisor. These results suggest that NFI-C gene causes dissociation of odontoblast and thus abberant odontoblast differentiation and abnormal dentin formation by interfering with the formation of intercellular junctions.
