Arylsulfatase catalyzes the release of SO$\sub$4//sup2/- from sulfate esters of simple phenols. Arylsolfatase occurs widely in animal tissues and in microorganisms including soil bacteria. Its widespread distribution suggests that it has a rather fundamental function and environmental meaning. It has been shown previously that arylsulfatase of Klebsiella was purified and characterized. A condition of arylsulfatase synthesis was tested with several strains of Serratia. Serratia marcescens could not utilize some sugars, such as xylose, rhamnose, glucosamine and arabinose hut glucose and mannitol as a sole carbon source. However, arylsulfatase synthesis was repressed by glucose but not by mannitol. The enzyme synthesis was repressed ob inorganic sulfate and methionine, and this repression was relieved by addition of tyramine. Arylsulfatase of S. marcescen was purified by fractionation with ammonium sulfate and followed by chromatographies on DEAE-Cellulose CM-Cellulose, and DEAE-Sephadex A-25. The molecular weight of arylsulfatase was determined to be 46,000 by SDS-Gel electrophoresis and 49,000 by Sephadex G-100 column chromatography. The enzyme showed some different properties with that of K. aerogenes. The activity was maximum at pH 6.8. The Km and Vmax values for p-nitrophenyl sulfate were 2.5${\times}$10$\^$-4/ M and 20 nmoles/min/mg protein, respectively. The enzyme showed high activities toward phenyl sulfate, ο-and p-nitro phenyl sulfates, and p-nitrocatechol sulfate. The inhibition of enzyme was strongly affected by hydroxylamine, inorganic fluoride, sulfide and phosphate, but by inorganic sulfate. Like Klebsiella arylsulfatase, tyramine, octopamine, and dopamine gave signifcant inhibitory effect.

In this experiment, persulfate, a strong oxidant for ISCO (In-Situ Chemical Oxidation) was used to degraded RDX in artificial ground water at ambient temperature. Results of RDX degradation by persulfate in a batch reactor showed that the oxidation reaction was pseudo first order with estimated Ea (activation energy) of $1.14{\times}10^2kJ/mol$ and the rate was increased with the increase of reaction temperature. The oxidation of RDX by persulfate increased slightly with the increase of initial solution pH from 4 to 8. The RDX oxidation rate increased 13 times at pH 10 compared with that at pH 4, however, alkaline hydrolysis was found to be the main reaction of RDX degradation rather than oxidation. The study also showed that the oxidation rate of RDX by persulfate was linearly dependent upon the molar ratios of persulfate to RDX from 5 : 1 up to 100 : 1, with a proportion constant of $4{\times}10^{-4}$ ($min^{-1}$/molar ratio) at $70^{\circ}C$. While NOM (Natural Organic Matter) exerted negative effects on the oxidation rate of RDX by persulfate, with a proportion constant of $1.21{\times}10^{-4}$ ($min^{-1}{\cdot}L/mg-NOM$) at $70^{\circ}C$ and persulfate/NOM molar ratio of 10/1. The decrease in RDX oxidation rate was linearly dependent upon the added NOM concentration. However, the estimated activation energy in the presence of 20 mg-NOM/L was within 3.3% error compared to that without NOM, which implies the addition of NOM does not alter intrinsic oxidation reaction.
This study was carried out to induce plant regeneration via multiple shoot formation from sucker explants of Rubus fruticosus L. ${\times}$ R. parvifolius L. To induce adventitious shoots, the sucker explants were sterilized in 1.2% sodium hypochlorite (NaOCl) solution, and then were cultured on the full and 1/2 MS solid medium supplemented with BA (0.1, 0.5, 1.0, $2.0mg{\cdot}L^{-1}$). After 4 weeks of culture, the highest frequency (83.3%) of shoot formation from sucker explants was obtained from the full MS medium with $1.0mg{\cdot}L^{-1}$ BA. The highest shoot number (3.7) per explant was obtained from the full MS medium with $0.5mg{\cdot}L^{-1}$ BA. After 12 weeks of culture, the number of shoots (15.4) per explant was increased. The highest frequency (95%) of root formation was obtained from the 1/2 MS medium, when the explant with shoot were cultured on the full, 1/2 and 1/4 MS medium. The survival rate of the plantlets after transfer to plastic pots containing sand, soil, and vermiculite (1.1:1, vol.) was 95%. The results indicate that multiple shoot procedure can be applied for an efficient mass propagation of Rubus fruticosus L. ${\times}$ R. parvifolius L.
EagBI is a type II restriction endonuclease from Enterobacter agglomerans strain CBNU45 isolated from soil. EagBI was partially purified by DEAE-cellulose, phosphocellulose P11 and hydroxylapatite column chromatography. EagBI recognizes and cleaves the sequence 5'-CGAT${\downarrow}$CG-3' and generates 2-base 3'-protruding cohesive ends. The optimal reaction conditions of EagBI are 10 mM Tris-HCl (pH 7.8), 6-10 mM $MgCl_2$, at 37 ${\circ}C$. The enzyme is maximally active in the absence of NaCl, able to cleave both $dam^-$ and $dam^+$ DNAs, and sensitive to heat treatment (at 65 ${\circ}C$ for 10 min). Therefore, although EagBI is an isoschizomer of PvuI, it is more useful than PvuI in respect of the NaCl requirement and heat-stability.
This experiment was conducted to investigate the effect of chitosan on the growth and yield of fall-planted potatoes. The research was carried out in fall-planted potatoes from August 2000 to Aprial 2001 at the height of 250m above sea level in Jeju Island. Soaking for 30 min. of seed potatoes in 200 times diluted solution of 3% liquid chitosan shortened days to emergence by 4 to 6 days. Stem length, number of stems and number of stolons per plant were 56.5 cm, 4.3, and 19.0, respectively, when seed potatoes were soaked for 30 min. in 200 times diluted solution of 3% liquid chitossan. Total potato yield in this treatment reached 2,963 kg per 10a and was significantly greater than in other treatments. Yield of marketable tubers (greater than 30g) per 10a for treatment of seed potatoes soaking in 200 times diluted solution of 3% liquid chitosan and for treatment of foliage spray with 200 times diluted solution of 3% liquid chitosan plus chitosna power (10 kg per 10a) miximg with soil was 2,761 kg and 2,628 kg, respectively. Contents of Mg, Fe and B were the greatest in the treatment of 30 min. soaking of seed potatoes in 200 times diluted solution of 3% liquid chitosanm. The increased contents of these elements are considered to have caused yield increase as a result of increased chlorophyll content for photosynthesis.
It is one of the important subjects in fall crop production of potatoes in korea to find the proper direct planting method as a substitute for transplanting method by sprouting inducement at seed-bed. From this point the author has undertaken this experiments to examine the effects of several handling methods of seed tuber pieces at planting on sprouting of fall grown potatoes at Ansung Agricultural College from August to september in 1969. The results of experiments induced sprouting with Gibberellin treatment using the variety Irish Cobbler were as follows; 1. Sun light exposure treatment to cut surface of potato seed pieces resulted in rotting of seed pieces and delay of sprouting and bud growth. Exposure more than three hours induced severe rotting(more than fifty percent) of seed pieces (Table I). 2. When soil moisture are abundant and the duration of sun light. exposure is short, even direct planting method of treated seed pieces with Gibberellin resulted in good potato stands (Table 2). 3. The direct planting method, as the rotting of seed pieces are avoidable are applicable in fall crop production of potatoes us a substitute for transplanting method because the sprouting and bud growth are promoted more in case of direct planting than transplanting (Table 3). 4. When the humidity in the bottle was low no healing effects was found by holding the seed pieces in large closed bottle under the trees for three days before planting (Table 2). 5. If duration of sunlight exposure is short, no difference of potato stands between clear day planting and cloudy day plant ing (Table 3). 6. As long as soil moisture deficiency. was not induced the irrigation at planting shows no effects to protect the seed piece rotting after planting (Table 3).
We investigated natural background and enrichment characteristics and predicted geochemical disaster for stream sediments in the Hamyang-Sancheong area. Stream sediments samples were collected 95 ea in study area. The stream sediments were well known that had not possibility of contamination effect and represented drainage basins. We got the major and hazardous elements concentrations by XRF, ICP-AES and NAA analysis methods. Acid decomposition for the ICP-AES has been used $HClO_4$ and HF with $200^{\circ}C$ heating at 1st and after that $HClO_4$ HF and HCl with $200^{\circ}C$ heating at 2nd stage. We could know the characteristics that concentration of Cu and Co decreased when concentration of $SiO_2$ increased in correlation analysis. The enrichment factor of the stream sediments was below 2 in study area. This result indicated that study area belonged to moderate enrichment. The stream sediments of Hamyang area were enriched in order of Pb>Th>Cr>V>Co>Cu and those of Sancheong area were enriched in order of Pb>Th>Cr>Co>V>Cu. The enrichment factor(E.F.) of the Pb, Cr, Co and V was similar between Hamyang and Sancheong area. The enrichment factor of the Th was higher in Hamyang area and that of the Cu was higher in Sancheong area. The enrichment factor of the Pb was highly enriched in all study area than earth crust mean. But we could know that study area was not exposed to the pollution of the Pb through the tolerable level.
Bensulfuron concentrations of $10^{-7}$, $10^{-6}$, $10^{-5}$ and $10^{-4}M$ were applied to agar medium on susceptible (cv. KH 17854 and cv. IR 1846) and tolerant (cv. Chinsurah Boro II and IR 14252) rice cultivars were grown for microscopic inspection. Susceptible cultivars showed the decrease in shoot and root growth at the concentration of $10^{-5}M$ while ones showed no difference. Such a tendency was also observed from microscopic inspection in the elongation zone of shoot meristematic tissue. Seedlings grown in soil for 10 days were transfered to distilled water containing only bensulfuron solutions. There were significant differences between cultivars in terms of supression of shoot meristematic activity and swelling of cell volume. Observations of those cells made it clear that especially susceptible cultivars showed the irregular cell layering, vacuolation, cell swelling and partial damage in membrane of shoot tissue. The major response of root tips of susceptible cultivars showed the disorganization of cortex, rupture and contraction of membrane, inhibition of cell division, swelling and emergence of lateral root while tolerant ones showed no such responses.
Birnessite is one of the dominant Mn (oxyhydr)oxide phases commonly found in soil and deep ocean environments. It typically occurs as nano-sized and poorly crystalline aggregates in the natural environment. It is well known that birnessite participates in a wide variety of bio/geochemical reactions as a reactive mineral phase with structural defects, cation vacancies, and mixed valences of structural Mn. These various bio/geochemical reactions control not only the fate and transport of inorganic and organic substances in the environment, but also the formation of diverse Mn (oxyhydr)oxides through birnessite transformation. This review assessed and discussed about the phase transformation of birnessite under a wide range of environmental conditions and about the potential geochemical factors controlling the corresponding reactions in the literature. Birnessite transformation to other types of Mn (oxyhydr)oxides were affected by dissolved Mn(II), dissolved oxygen, solution pH, and co-existing cation (i.e., $Mg^{2+}$). However, there still have been many issues to be unraveled on the complex bio/geochemical processes involved in the phase transformation of birnessite. Future work on the detail mechanisms of birnessite transformation should be further investigated.

An oleaginous fungus was isolated from soil and identified as Mortierella sp. (M-12) for producing arachidonic acid (AA). Cell disruption methods, extraction methods, and particle sizes of freeze-dried biomass were tested to achieve maximum extraction of total lipids and AA. M-12 grown in glucose yeast media at $25^{\circ}C$ for 7 days contained 35.5% total lipid, and 47% of the total lipid was AA. Lipid extraction yield from wet biomass was shown to be similar to that in a dry state. Maximum lipid extraction was achieved using a mixture of chloroform and methanol (2:1) as an extraction solvent. Different mechanical cell disruption methods did not affect lipid extraction yields. The smaller the particle size of the biomass, the better the lipid extraction yield was observed. Particle size of biomass was shown to more strongly affect lipid extraction than extraction time. The highest AA content was observed in the class of neutral lipids.
