Isolation and Identification of Xylanase Secreting Yeast (Xylanase를 분비하는 효모 균주의 분리 및 성질)

  • Among the new yeast strains which were isolated from soils by incubating in the xylan containing minimal medium at 3$0^{\circ}C$, one strain(XB-33) was finally selected by the results of extracellular xylanase production test. The characteristics of XB-33 was almost consistent with those of the Cryptococcus ater. The formation of xylanase activity was induced by xylan and repressed by xylose or glucose. The xylanase was partially purified from the culture supernatant with DEAE-Sephadex A5O chromatography. The enzyme had a pH optimum for activity at 5.0 and its stability range was pH 5-7. The temperature optimum was at 5$0^{\circ}C$, but the enzyme activity was greatly lost by heating at 7$0^{\circ}C$ for 60 minutes. The hydrolysis products from xylan by crude enzyme detected by TLC, were xylose and n series of higher oligosaccharides. The Km value of xylanase was 20 (mg/ml).

Study on Anti-estrogenic Activity of DEHP as an Endocrine Disruption Chemical (내분비 교란성 DEHP의 항-에스트로젠 활성에 관한 연구)

  • Di-2-ethylhexyl phthalate (DEHP), is a widely used plasticizer known to be a suspected endocrine disrupter, but its exact effects on aquatic organisms are not yet known. When Japanese medaka (Oryzias latipes) were exposed from the time of hatching to 3 months of age to an aqueous DEHP solution at nominal concentrations of 1, 10, and 50 $\mu\textrm{g}$/l, DEHP treated female fish showed distinct reproductive effect. And the midge (Chironomus riparius.). an aquatic invertebrate, was exposed to DEHP to evaluate the effects on reproductive processes via sediment toxicity. The test endpoints included emergence, sex ratio, fecundity, and the viability of F1 offspring egg ropes. The result implied that the normal developmental and/or reproductive processes in C. riparius had been disrupted when exposed to DEHP, the effect also being displayed in the next generation. In summary, DEHP hinders the development of reproductive organs in the female Japanese medaka and C. riparius.

Studies on the Mineralogical Characteristics of Apple Orchard Soils (사과원토양(園土壤)의 광물학적특성(鑛物學的特性)에 관(關)한 연구(硏究))

  • The mineralogical studies of the eleven sub-soil samples derived from granite, granodiorite, diorite and arkose sandstone, taken from apple orchards in the province of Kyungsangbukdo, Korea are made to investigate the relationships between the mineral weathering, soil forming processes and mineralogical composition. The fine sand fraction (less than 0.2mm) and the clay fraction (less than 2 micron) are dispersed with the shaker after hydrogen peroxide treatment for the removal of organic matter, and separated from each suspension by gravity sedimentation. The fine sand are observed by mineral microscope and the clay are observed by X-ray diffraction patterns, differential thermal analysis curves and infrared spectrum. The outline of the results are as follows. 1. The primary minerals ; Quartz, changed-feldspar, plagioclase, alkali-feldspar are dominant in almost all samples, and some samples contain an appreciable amount of hornblende, biotite, muscovite and plant opal. There are also those samples which contain very small quantity of pyroxene group, tourmaline, epidote, cyanite, magnetite, volcanic glass and zircon. They are mainly derived from weathering products of granite, granodiorite, diorite, arkose or its mixtures. 2. All samples contain expanding or nonexpanding $14{\AA}$ minerals, illite and kaolin minerals, and some samples contain chlorite, cristobalite, gibbsite, and those primary minerals as quartz and feldspar, but the quantities vary according to the parent matrials. 3. Non-expanding $14{\AA}$ minerals may be dioctahadral vermiculite which sandwiches gibbsite layer or chlorite in between layer lattices. 4. As for clay minerals, montmorillonite was principal component in the samples derived from weathering products of arkose sandstone and tertiary. Minerals which are derived from weathering products of arkose have kaolin minerals and vermiculite as their principal component, and minerals derived from weathering products of acidic rock group are generally classified into two groups, the kaolin mineral group, and the kaolin minerals and vermiculite group.

  • A laboratory experiment was conducted to find out the denitrification rate upon the levels of nitrogen and source of organic matter in submerged sandy and sandy loam soil. The results obtained were sumarized as follows; 1. Evolution of nitrous oxide was increased at 1st and 10 days after incubation. And dinitrogen was increased at 1st and 30 days after incubation. Applications of green manure was enhanced the evolution of nitrous oxide ($N_2O$) and dinitrogen ($N_2$). 2. The cumulative denitrification rates at 50 days was high in Gyuam sandy loam soil (O-M: 1.52%) than that of Hamchang sandy soil (O-M: 3.81%). On the other hand, the cumulative emission of dinitrogen was high in Gyuam sandy loam soil while nitrous oxide was high in Hamchang sandy soil. The total mount of denitrification rate was high in order of green manure > rice straw > compost > control soil. 3. Increases of fertilizer nitrogen was enhanced the rate of emission of dinitrogen and nitrous oxide during the incubation time. 4. According to Michaelis-Menten kinetic equation, denitrification rates and reaction efficiency were remarkably increased by application of readily decomposable organic matter with in higher organic matter content of soil. 5. The negative relationship was observed between the evolution of dinitrogen and carbon ($CO_2+CH_4$) while the nitrous oxide with carbon was positive. 6. Under the this experiment conditions 1 mg of carbon was required for production of 4 mg N as $N_2O$ and 3 mg of N as $N_2$, respectively.

  • This study was undertaken to investigate the origin of suspended matter to induce turbidity water in Gachang dam in view of environmental geology. During the period from May to August 2004, field works and sampling were carried out three times at the dam and along its streams, and chemical and mineralogical analyses such as ICP, IC, particle size analyzer, XRD and SEM were made on water, soil and suspended matter in water. Electrical conductivity (EC), turbidity, the contents of cation and anion increase from upstream toward the dam mostly due to the geological factors such as weathring of the rocks causing the increase of the total ion content. Vermiculite, illite, kaolinite, quartz, feldspar and iron hydroxide are commonly found in suspended matters in water and soils. Finer particles (d10) in soil increase slightly toward downstream and the vermiculite content is highest in the dam water. Since geological differences are not significant, mineralogy are similar in suspended matters and soils. Clay mineral compositions present in suspended matters were alsmost the same as those in soils, indicating the origin of soils by weathering of host rocks and being transported to the dam by stream water.

  • Aspergillus sp. (MK-24) producing a biological active substance that inhibited the venom proteinase activity was isolated from soil. The substance also inhibited the activity of trypsin and coagulation of blood, but did not inhibit papain, $\alpha$-chymotrypsin and pepsin. The substance was partially purified from culture filtrate by precipitaion with acetone, and by chromatography of DEAE-Sepadex A-50 column and Amberlite IRC-50 ion exchange. The inhibitory substance was stable in the wide pH range from 2.0 to 12.0 at 37$^{\circ}C$, but not stable at $65^{\circ}C$ in the alkaline pH. Only 12% of the activity was decreased by the heat treatment at 10$0^{\circ}C$ for two hours. The inhibition on venom proteinase (Agkistrodon bromohoffi brevicaudus) was a mixed type. The inhibitory activity depended on the preincubation time and completely depressed by cupric, zinc and cobalt ions. The inhibition on the venom proteinase was appeared strongly on casein but not on ovalbumin or hemoglobin as a substrate.

  • This study reports on the genesis and mineralogical characteristics of the clay minerals in the soils derived from the five major parent rocks of granite, granite-gneiss, limestone, shale, and basalt in Korea. The investigation on the mineralogical aspects of primary and secondary minerals of the rocks and coarse fractions in the soils have been already reported. In this report, the identification of clay minerals in the soil clay fractions was done through the analyses of chemical, X-ray diffraction, and thermal methods. The studies showed clearly that much of the clay minerals was evolved by the weathering of primary minerals and some were further developed by the transformation of secondary minerals. Cation exchange capacity(CEC) of the clay fractions increased with higher amotunts of vermiculite, chlorite, and illite, however, decreased with higher hydroxy octahedral sheet within the interlayer spaces of vermiculite even if dominant clay with vermiculite. Feldspars in the granite and granite-gneiss might be completely transformed to kaolin mineral, Illite, chlolrite, and vermiculite formed by the alteration of micas, amphibole, augite, and primary chlorile seem to be subsequently transformed to the mixed layer minerals such as illite/vermiculite, illite/chlorite, and chlorite/vermiculite. These weathering products may be ultimately transformed into kaolin minerals. The smectite minerals in the clay fractions of the soils developed on the limestone are considerably present and they seem to be formed directly by the precipitation from high Mg solution and/or by the transformation of vermiculite from micas and chlorite in the parent materials. Abundant presence of illite in the soil clays developed on the shale is considered to have inherited from the fine particles and more resistant hydrous muscovite. The weathering sequences of the hydrous muscovite were as follows according to the degree of soil development ; hydrous muscovite ${\rightarrow}$ illite/vermiculite mixed layer(Inceptisols, Daegu series) and hydrous muscovite ${\rightarrow}$ illite/vermiculite mixed layer ${\rightarrow}$ vermiculite ${\rightarrow}$ kaolin mineral(Alfisols, Buyeo series). The plagioclase in the basalt might be mostly weathered to kaolin minerais. The augite in the basalt is likely to be transformed through progressive stage of weathering, augite ${\rightarrow}$ chlorite ${\rightarrow}$ chlorote/vermiculite mixed layer ${\rightarrow}$ vermiculite ${\rightarrow}$ kaolin. Another weathering sequence of augite could be expected, augite ${\rightarrow}$ chlorite ${\rightarrow}$ illite by the presence of illite and illite/vermiculite mixed layer in the clay fractions. Vermiculite and gibbsite were quantified from thermogravimetry(TG) and kaolin minerals, from both TG and differerential thermal analysis (DTA). Vermiculite in Jangseong series from the limestone was the dominant clay mineral of 21.7 percent and had a range in the order of 9.2 percent in Buyeo series to 5.4 percent in Daegu series from the shale. The rest soils ranged from 8.8 to 28.3 percent. Kaolin minerals were the dominant clay mineral of 32.7 percent in Asan series from the granite-gneiss and Gueom series of 32.0 percent from the basalt. The soils from the limestone ranged from 9.4 to 14.9 percent. The rest soils ranged from 8.9 to 28.6 percent. Gibbsite were 3.9 and 2.3 percent for Weoljeong and Chahang series from the granite, respectively. In Asan and Cheongsan series from the giranite-gneiss were 1.4 and 4.5 percent, respectively, and 3.6 percent in Jangpa series from the basalt.

Analysis of Microbial Communities in Aquatic Sediment Microbial Fuel Cells Injected with Glucose (포도당을 주입한 수중퇴적물을 이용한 연료전지시스템에 있어서 미생물군집 분석)

  • The purpose of this research was to optimize electric current production of sediment microbial fuel cells by injecting glucose and to investigate its impact on microbial communities involved. It was shown that injection of proper concentration of glucose could increase electric current generated from sediment microbial fuel cells. When 1,000 mg/L of glucose, as opposed to higher concentrations, was injected, electric current increased up to 3 times. This increase is mainly attributed to the mutual relationship between fermenting bacteria and exoelectrogenic bacteria. Here the organic acids generated by fermenting bacteria could be utilized by exoelectrogenic bacteria, removing feedback inhibition caused by the organic acids. When glucose was injected, the population of Clostridium increased as to ferment injected glucose. Glucose fermentation can have either a positive or negative effect on electric current generation. When exoelectrogenic bacteria may readily utilize the end-product, electric current could increase. However, when the end-product was not readily removed, then detrimental chemical reactions (pH decrease, methane generation, organic acids accumulation) occurred: exoelctrogenic bacteria population declined and non-microbial fuel cell related microorganisms prospered. By injecting a proper concentration of glucose, a mutual relationship between fermenting bacteria, such as Clostridium, and exoelectrogenic bacteria, such as Geobacter, should be fulfilled in order to increase electricity production in mixed cultures of microorganisms collected from the aquatic sediments.

  • Sand and silt size fractions of soils which were derived from five major rocks of granite, granite-geniss, limestone, shale, and basalt in Korea were studied. Determination of the mineralogical and chemical composition of rock-forming mineral breakdown which is accompanied by the formation of secondary minerals. The chemical composition of the fraction was largely changed with the content of weatherable and resistant soil minerals such as ferromagenesian minerals, carbonates, and guartz. In the sand fractions of the soils from the granite and granite-gneiss, chlorite-vermiculite mixed layers seem to be an intermediate weathering product prior to the weathering state of the formation of vermiculite from chlorite. Kaolin minerals in the silt fractions of the soils from the granite-gneiss are considered to be formed by the pseudomorphic transformation of plagioclase. In the sand and silt fractions of the soils derived from the limestone, large amount of calcite and dolomite seems to have been inherited from the parent rocks. The primary chloritc, micas, and feldspars are considered to be formed from the weathering remains after leaching of carbonate minerals during the soil formation. In the residual soils(Gueom series) developed from the basalt, quartz and micas were coexisted with plagioclase and augite inherited from the parent rock.

  • This study was conducted to find out the behavior of anaerobic microorganisms and anaerobic decomposition products of rice straw in the strict anaerobic condition. The number of methanogenic bacteria were more isolated than cellulolytic bacteria from the digester decaying rice straw during the entire incubation time. The activity of anaerobic microorganisms, such as methanogens and cellulolytics, were high the early incubation time in the treatment of rice straw with urea, but without urea was low at that time and increased moderately after 10 days incubation. Volatile fatty acid as intermediate anaerobic decomposition products had a longer retention time and higher accumulation rate in the treatment of rice straw without urea than with urea, and predominant fatty acid was propionic acid. Gas generation rate as final products were very intimate relationship with the activity of methanogenic bacteria. Average Eh value was -250mV during the incubation time and $CH_4$ : $CO_2$ percent ratio was about 60~65 : 35~40 in this Eh value. Decomposition rate of rice straw calculated from $CH_4$ and $CO_2$ gas wars 45.6% for 50 days in the treatment of rice straw with urea, and 36.8% without urea.
