Alteration of Carbohydrate Metabolism in Rice Seedlings under Low Temperature (저온 처리한 벼 유식물에서 탄수화물 대사의 변화)

  • The contents of reducing sugar, sucrose, starch and fructose-2,6-bisphosphate (F-Z,$6-P_2$) in relation to the activities of amylase, invertase and fructose-1,6-bisphosphatase (FBPase) were investigated from the leaves of rice (Oryza sativa L. cv. Samjin) seedlings grown at $4^{\circ}C$ for 3 days_ In the seedlings, the contents of reducing sugar and sucrose were increased, but soluble and insoluble starch were declined. Under this condition, amylase activity was increased. but acid invertase activity was declined and alkaline invertase activity was not changed. Cytosolic and stromal FBPase activities were increased. But F-2,$6-P_2$ content was declined. It seemed that the increase of reducing sugar content might be due to the increased activity of amylase and the increase of sucrose content might be related to the increased activity of cytosolic FBPase, reduced content of F-Z,$6-P_2$ and reduced rate of hydrolysis of sucrose during the cold treatment. These results suggested that the changes in carbohydrate rnetabolim of rice seedlings under low temperature reflect one of the protection mechanism to the low temperature during the cold treatment.atment.

  • This study was carried out for the analysis of chemical composition and antioxidative activity of lotus leaf. The lotus leaf contained 63.8% of crude carbohydrate, 16.9% of crude protein, 1.0% crude fat, and 9.3% of crude ash including high amount of calcium (2.2%). The antioxidative effect of several solvents extracts of lotus leaf was investigated. Among them 70% ethanol extract showed relatively higher extraction yield and higher total phenol content as well as the highest electron-donating ability using 1,1-diphenyl-2-picryl hydroxyl.

Carbohydrates Analyses of Korean Yam(Dioscorea) Tubers (한국산 마의 당질 분석)

  • Yam(Dioscorea) tubers were used as one of the dietary starches in Korea, China and Japan because of its characteristic properties. Yam tubers were powdered after freeze drying the sliced tubers to investigate carbohydrates. Chemical properties such as proximate components and carbohydrates were studied using three varieties of Korean yams, namely. D. batatas, D. aimadoimo and D. japonica. The proximate components of yam tubers showed that the major components of Korean yam tubers were carbohydrates and moisture. The main components of free sugars in yams were identified as fructose, glucose, sucrose and maltose. In addition to these four sugars, there were two unidentifiable peaks whose areas are too big to ignore. These results were obviously different from other reported data of free sugars in yams. The main components of total sugars were mannose and glucose.

  • Glucose, trehalose, and glycogen content were measured in haemolymph and the body during larval development of the oriental corn borer, Ostrinia furnacalis ($Guen\'{e}e$). Glucose content varied in haemolymph, but in the body it gradually decreased at all larval stages. As the larva proceeds growth, the initial high level of trehalose increased further in haemolymph whereas the low level trehalose gradually decreased in the body. Glycogen levels in haemolymph rose gradually during larval growth whereas those in the body increased sharply at the two latter instar larval stages. It is apparent that the existence of a homeostasis in carbohydrate levels between haemolymph ana the body.

  • Carborhydrate contents were compared between diapausing and non-diapausing pupae of the fall webworm, Hyphantria cunea DRURY. Glycogen content in the whole body of diapausing pupae kept at $-5{\pm}1^{\circ}C$ was less than that of those left at room temperature, while the amounts of trehalose and sorbitol showed the reverse trend. The osmotic pressure of haemolymph was higher in diapausing pupae than that in non-diapausing pupae. The mean oxygen consumption rate of diapausing pupae was $4{\sim}6.7$ times less than that of non-diapausing, normal pupae.

  • We investigated effects of a topical treatment of methoprene(0.5-5.0$\mul$/egg), a juvenLle horm mane analogue, on embryo development in the gypsy moth, Lymantri$\alpha$ dispar. Methoprene lowered egg hatching rate, and also reduced the mean wet weights of hatched 1st instar larvae w with the most effect shown at the highest concentration. The differences in protein(p < 0.01) and carbohydrate(p < 0.05) contents between control and methoprene(5$\mul$/ egg) treatment g groups were observed during embryo development.

  • Immature apple fruits of cultivars Fuji and Miller were completely resistant to Macrophoma sp. until 10 July. When inoculated on 31 July, apples became susceptible to this fungus, irrespective of cultivar. This fungus grew better on Fuji than Miller. Lesion enlargement on apple at $25^{\circ}C$ was remarkably pronounced relative to $20^{\circ}C$. In both cultivars, the amount of total soluble carbohydrates or reducing sugars in apple fruit flesh was gradually increased as apples became mature. The healthy apples of cultivar Fuji had a higher amount of carbohydrates than those of Miller. The infected apples contained more soluble carbohydrates in comparison to the healthy ones. In healthy apples, there were no consistent differences between Fuji and Miller in amount of total soluble amino acids. With increasing age of apples, the amount of soluble amino acids declined in the fruit flesh of both cultivars. Levels of carbohydrates and amino acids in apple fruits were discussed in association with the immature fruit resistance to Macrophoma sp.

Effects of Elevated CO2 Concentration on Leaf Phenology of Quercus acutissima (이산화탄소 농도 증가가 상수리나무 잎의 계절현상에 미치는 영향)

  • Effects of elevated $CO_2$ on leaf phenology of Quercus acutissima were examined using open-top chambers, which had ambient and elevated $CO_2$ concentrations (ambient ${\times}1.4$, ambient ${\times}1.8$). To analyze the effect of chamber, non-treatment block was established near outside of the chambers. In 2013, budburst, leaf unfolding, coloring, and shedding were surveyed, and spring phenology was surveyed in 2014. Thermal sum (base temperature $+5^{\circ}C$) of each phenological event occurred was recorded. In addition, bud samples were collected and analyzed for carbohydrate contents in March 2014. Elevated $CO_2$ concentration advanced budburst and leaf unfolding, and delayed shedding in 2013. However, in 2014, the temperature of the spring season was high, and there was no significant effect of elevated $CO_2$ concentration on spring phenology. Carbohydrates content, such as starch, total non-structural carbohydrate and total soluble sugar, were significantly increased in response to elevated $CO_2$ concentration. It has been proposed that elevated $CO_2$ concentration could extend the growing season of temperate species with increased possibility of frost damage due to early bud opening and leaf unfolding. However, our analysis showed that the increased carbohydrate concentration in bud under elevated $CO_2$ would reduce the possibility of early spring frost damage by acting as cryoprotectant.

Properties of Porphyran and Hemicellulose extracted with Different extract Solutions and Enzymatic Pretreatments from Porphyra (추출 용매 및 효소 전처리 방법에 따른 포피란과 헤미셀루로오즈의 특성)

  • Laver, Porphyra, is distinctive for its high content of proteins and polysaccharides such as porphyran and hemicellulose. The chemical properties of the polysaccharides extracted with different extraction methods such as hot water, dilute acid(pH 4.0) or alkali solution(2N NaOH) were examined to investigate the suitable extraction conditions for porphyran and hemicellulose from laver. For porphyran extraction, dilute acid solution was more preferable to hot water and alkali solution because of its higher 3,6-anhydrogalactose content and lower protein content. However, alkali solution was more suitable to extract the hemicellulose because of higher mannose content indicating the extraction of mannan. To decrease contamination of the polysaccharides with protein, the dried lavers were pretreated with enzymes (Protamex, Flavourzyme, Alcalase, Viscozyme) before hot water extraction. All enzyme pretreatments increased the yield of polysaccharides by compared with control (enzyme unpretreated) and Flavourzyme pretreatment was most effective to decrease protein contamination in the polysaccharide. All viscosities of porphyran solutions pretreated by enzymes were lower compared to the control porphyran solution and showed pseudoplastic behavior with yield stress. In case of alkali extraction of residues obtained after enzyme hydrolysis and hot water extraction, protease pretreatment increased the mannose contents in the polysaccharide while the xylose content was increased by Viscozyme pretreatment.

Seasonal Changes of Biochemical Composition in Cultured Bivalves (양식패류의 생화학적 체성분의 계절적 변화)

  • Seasonal changes in biochemical composition of Crassostrea giggas and Scapharca broughtonii in the Kosong bay and Kangjin bay, southern Korea were studied over 12 months in relation to their habitat. In Crassostrea giggas, protein was high during summer in all experimental station, but lipid and carbohydrate were high during winter, and no clear difference was between experimental station. Glycogen content was sharply decreased during summer, and energy content was low but carbohydrate was high during summer. In Scapharca broughtonii, protein was low, but carbohydrate was high during summer. Lipid was high from February to March, glycogen content was low from June to September. Energy content was low during winter and no clear difference biochemical composition was between experimental station of Scapharca broughtonii.
