플라즈마 처리에 의한 원뿔형 다중벽 탄소나노튜브 다발의 형성기전

  • 플라즈마 처리를 통하여 수직 합성된 다중벽 탄소나노튜브가 원뿔형 다발이 될 수 있으며 원뿔형 탄소나노튜브 다발은 기존의 구조적, 기계적 성질의 향상과 더불어 향상된 전계방출 능력을 가질 것으로 기대되어 이를 X-선원, 전계방출디스플레이(FED), 유기발광다이오드(OLED) 백라이트 등의 전자빔 원으로 적용하기 위한 연구가 진행되고 있다. 원뿔형 탄소나노튜브 다발의 형상 제어를 통하여 전계방출특성을 향상시킬 수 있으며 이를 위해 원뿔형 탄소나노튜브 다발이 생성되는 메커니즘과 조사되는 플라즈마의 역할에 대해서 이해하는 것이 중요하다. 본 연구에서는 플라즈마 생성부와 조사부를 분리한 유도결합형 플라즈마 원을 사용하여 입사되는 이온의 에너지, 조사량, 입자 종을 독립적으로 제어하였고 이를 통하여 원뿔형 탄소나노튜브 다발이 형성되는 메커니즘과 플라즈마의 역할을 밝혀내었다. 알곤 및 수소 플라즈마 처리에서는 원뿔형 탄소나노튜브 다발이 형성되지 않았으나 질소 및 산소 플라즈마 처리에서는 원뿔형 탄소나노튜브 다발이 형성되었다. 특히 산소 플라즈마 처리가 원뿔형 탄소나노튜브 다발 형성에 효과적이었다. 원뿔형 탄소나노튜브 다발의 형성 메커니즘은 탄소나노튜브의 분극과 쉬스 전기장의 상호작용을 이용한 모델을 사용하여 설명하였다. 질소 및 산소 플라즈마 처리에서는 탄소나노튜브 끝단에 생성되는 C-N, C-O 결합에 의해 향상된 유도 쌍극자와 쉬스 전기장에 의해 탄소나노튜브 끝단이 모여 원뿔형 탄소나노튜브 다발이 생성됨을 밝혀내었다. 산소 플라즈마 처리에서 입사되는 이온의 에너지 조절에 의한 쉬스 전기장 조절과 조사량 조절을 독립적으로 수행하여 원뿔형 탄소나노튜브 다발의 직경 및 높이가 쉬스 전기장 및 조사량에 따라 조절 가능함을 보였다. 이로부터 입사되는 이온의 입자 종, 쉬스 전기장 및 조사량 조절 등의 플라즈마 인자 조절을 통하여 원뿔형 탄소나노튜브 다발의 형상 제어가 가능함을 보였다. 탄소나노튜브의 형상 제어와 더불어 세슘 입자 삽입을 통한 탄소나노튜브의 일함수 감소를 통하여 향상된 전계 방출 특성을 갖는 탄소나노튜브 팁의 제조 가능성을 확인하였다.

Growth and Production of Insecticidal Crystal Proteins of Bacillus thuringiensis as Affected by Carbon Sources (Bacillus thuringiensis 생장과 살충성 결정단백질 생성에 대한 탄소원의 영향)

  • Effects of 14 carbohydrates supplied as carbon sources on cell growth and sporulation of, and the production of insecticidal crystal proteins by Bacillus thuringiensis strains were investigated in liquid cultures. Strains grew well in media containing any one of the 14 carbohydrates supplied, reaching maximum cell densities of $10^7{\sim}10^8\;cells/ml$ in 16.7 to 22 hours after inoculation depending on the strain. Spores first appeared in 16.7 to 24.7 hours after inoculation, and 80% sporulation was reached in 28 to 51.3 hours after inoculation depending on the strain. No change in pH of media was observed after cell multiplication. The production of total protein was highest when supplied with sucrose and was lowest with starch. More insecticidal crystal proteins were produced when supplied with glucose, lactose, maltose, or sucrose. The amount of insecticidal crystal proteins produced by the strains was proportional to that of the total protein. The relative amount of individual insecticidal crystal protein species produced by B.t. kurstaki and B.t. israelensis was not influenced by the carbohydrates supplied.

Effects of Carbon, Nitrogen Sources and pH on Direct Somatic Embryogenesis in Liquid Culture of Rehmannia glutinosa Lib. (지황의 액체배양에서 탄소원.질소원 및 pH가 직접 체세포배 형성에 미치는 영향)

  • Basic informations for direct somatic embryo formation in Rehmannja glutinosa Lib. were obtained in 500ml erlenmyer flask. The ratio of ammonium to nitrate nitrogen of 825(mg/l) : 1900(mg/l) was proper condition for somatic embryo formation from stem and petiole explants and 3% sucrose was the most effective carbon source. Full strength MS medium with 2mg/l BA was better than LS medium for somatic embryogenesis. The initial pH 5.7 of medium(full strength MS with 2.0mg/l BA and 0.1mg/l NAA) was good for embryo production. Potassium ion was taken up rapidly within 2 weeks. while $Ca^{++}$ and $Mg^{++}$ ion contents were almost constant during culture period. Sucrose hydrolysis occurred throughout the culture, while glucose and fructose were absorbed simultaneously from the third week of culture.

  • This study was carried out to select the appropriate medium(especially, carbon and nitrogen source ) for somatic embryogenesis in order to develop the rapid mass production system in suspension culture of chuanxiang Hort. Suitable medium for somatic embryo formation was MS medium. The half strength MS medium was effective for somatic embryo development. Sucrose was the most effective carbon source for somatic embryo formation, however, production of somatic embryos was reduced at higher concentration of sucrose. Effects of suger was the same as sucrose. Somatic embryo formation was higher as the decrease of $NH_{4}NO_3$, and optimum ratio of $KNO_3\;:\;NH_4NO_3$ was 825 : 238mg /1. Regenerated plant was obtained in MS basal medium and survival late of plantlet was 60-70% after transplanted directly to the vermiculite.

Effects of Carbon and Nitrogen Sources on the Shoot Formation in bioreator culture of Scrophularia buergeriana Miquel (현삼에서 탄소원과 질소원의 종류와 농도가 기내 식물체 분화에 미치는 영향)

  • To determine the proper carbon and nitrogen sources and their proper levels for mass micro propagation of Scrophularia buergeriana Miquel, tonic and curing cough experiment were applied and a method for mass cultivation by using bioreactors (2.5 L) was expinined. Proper ratio of $NH_4NO_3\;:\;$KNO_3$ was 413 mg/L : 1900 mg/L for multiple shoot production. Sucrose was more effective than glucose or fractose as carbon source and 3% concentration was good for shoot formation. Total nitrogen was not detected after six weeks both in 500 ml flask and bioreactor culture. Sucrose was decreased sharply after two weeks and there was no sucrose left after three weeks both in 500 ml flask and bioreactor culture. The stirrer in bioreactor caused shear stress to shoots severely. The sphere type bioreactor was better than the cylinder type and removal of inner loop in sphere type was more effective to avoid shear stress.

  • VOCs 배출원은 크게 인위적인 배출원과 자연적인 배출원으로 나눌 수 있다. 식물은 자연 발생적 VOCs(NVOCs)의 가장 주요한 배출원이며, 대기 중으로 유입되는 반응성이 강한 탄소의 주요한 배출원이다. 이소프린, terpene, 알콜, 카르보닐화합물, 에스테르 등은 식물에서 발생되는 대표적인 탄화수소화합물들이다. 이러한 물질들은 에어로졸의 형성 등 전지구적 대기화학반응, 광역적 광화학산화제 (특히 오존)의 형성, 탄소순환의 수지균형 비도시지역의 산성침적에 기여하는 유기산의 생성 등에 주요한 영향을 끼친다. (중략)

  • High quality diamond films of the silicon on diamond (SOD) structure are deposited using CO and $H_2$ gas mixture in microwave plasma chemical vapor deposition (CVD), a SOD structure is fabricated using low pressure CVD polysilicon on diamond/ Si(100) substrate. The crystalline structure of the diamond films which composed of { 111} and {100} planes. were changed from octahedral one to cubo-octahedron one as the CO/$H_2$ ratios are increased. The high quality diamond films without amorphous carbon and non-diamond elements were deposited at the CO/$H_2$ flow rate of 0.18. and the main phase of the diamond films shows (111) plane. The diamond/Si(lOO) structure shows that the interface is flat without voids. The measured dielectric constant. leakage current and breakdown field were $5.31\times10^{-9}A/cm^2$ and $9\times{10^7}{\Omega}cm$ respectively.

  • Culture conditions affecting mycelial growth and sporulation of P. theae, SP2, SP3 and P. longiseta which causing leaf blight on oriental persimmon leaf blight have been investigated. The optimum temperature for the mycelial growth and sporulation on potato dextrose agar was $25{\sim}30^{\circ}C$ in all the fungi, but was inhibited and finally arrested at 10 and $30^{\circ}C$. The optimum pH for mycelial growth and sporulation were ranged at pH 4.5~5.0 and 5.0~6.0 in all the fungi. Lenonian agar, potato sucrose agar and oatmeal medium were good culture media for the mycelial growth and sporulation of all the fungi. The effective sources of nitrogen and carbon for the mycelial growth were tryptone, glycine, starch, dextrose, galactose and lactose. Glutamic acid, peptone and tryptone were good nitrogen sources for sporulation of the fungi. Sucrose, starch and galactose were also good carbon sources for sporulation. Generally, vitamins had no effect on mycelial growth and sporulation. The pH of the potato dextrose broth inoculated with SP2, SP3 and P. theae and P. longiseta was changed from 7.0 to 4.5~4.7 and 5.0~5.4 after incubating for 10 days, respectively. But, the initial pH of the medium adjusted to 5.0 was lowered to 4.5~4.7 after incubating for 10 days.

  • The optimal synthetic medium, temperature and pH range for the mycelial growth of Cordyceps scarabaeicola were PDA, $25^{\circ}C$, and pH $8.0{\sim}8.5$, respectively. The effects of carbon sources, nitrogen sources and mineral salts on mycelial growth were superior to lactose, ammonium phosphate and $CaCl_2$, respectively. Liquid spawn culture was selected out BM(brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%) medium.

Effects of Concentrations of Glucose and Maltose on the Growth of Recombinant E. coli (재조합 대장균의 세포성장에 대한 포도당과 맥아당 농도의 영향)

  • The growth of recombinant E. coli and formation of the by-products were investigated. Glucose was consumed in approximately 8hours of cultivation when initial glucose concentration was 5g/1. When higher initial glucose concentrations were employed, it took approximately 16 to 20hours for the glucose to be consumed. When maltose or glucose was used as a carbon source, lactic acid and acetic acid were main by-products, while propionic acid and methanol production was not significant. Much by- products were formed at the higher initial carbon source concentrations. Higher concentration of lactic acid was observed when glucose was used, while higher concentration of acetic acid was observed when maltose was used. Lactic acid production affected the cell yield while acetic acid production affected the growth rate.
