  • 인공면역시스템을 이용한 침입탐지시스템 개발을 위해 적용한 동적클론선택(Dynamic Clonal Selection) 알고리즘과 그의 문제점을 소개하고 개선된 동적클론선택 알고리즘을 제안한다. 개선된 동적클론선택 알고리즘은 정상행위를 비정상행위로 판단하는 기억 탐지 자들을 제거함으로써 기존에 동적클론선택 알고리즘이 안고 있던 오류를 감소시키는 방안을 제시한다.

  • 인공면역시스템을 이용한 침입탐지시스템 개발을 위해 적용한 동적 클론 선택(Dynamic Clonal Selection) 알고리즘과 그의 문제점을 소개하고 보다 개선된 동적 클론 선택 알고리즘을 제안한다. 이전 연구에서 침입탐지시스템이 흔히 접하게 되는 상황, 즉 과거 안정적으로 관찰되었던 정상행위가 합법적인 요인들로 인하여 갑작스러운 변화를 보일 경우 과거 생성되었던 기억탐지자가 정상행위를 비정상행위로 오류 판단하는 것을 막기 위하여 인간면역시스템의 체세포 돌연변이 (somatic hypermutation)를 이용하여 유전자 라이브러리를 진화시키는 방법을 첨가한 동적 클론 선택 알고리즘을 소개한다.

Fuzzy-Neural Networks by Means of Advanced Clonal Selection of Immune Algorithm and Its Application to Traffic Route Choice (면역 알고리즘의 개선된 클론선택에 의한 퍼지 뉴로 네트워크와 교통경로선택으로의 응용)

  • In this paper, an optimal design method of clonal selection based Fuzzy-Neural Networks (FNN) model for complex and nonlinear systems is presented. The FNNs use the simplified inference as fuzzy inference method and Error Back Propagation Algorithm as learning rule. Also Advanced Clonal Selection (ACS) is proposed to find the parameters such as parameters of membership functions, learning rates and momentum coefficients. The proposed method is based on an Immune Algorithm (IA) using biological Immune System and The performance is improved by control of differentiation rate. Through that procedure, the antibodies are producted variously and the parameter of FNN are optimized by selecting method of antibody with the best affinity against antigens such as object function and limitation condition. To evaluate the performance of the proposed method, we use the time series data for gas furnace and traffic route choice process.

  • This paper proposes a novel optimum design method for the PID controller of magnetic levitation-based Rail-Guided Vehicle(RGV) by a genetic algorithm using clone selection method and a new performance index function with performances of both time and frequency domain. Generally, since an attraction type levitation system is intrinsically unstable and requires a delicate controller that is designed considering overshoot and settling time, it is difficult to completely satisfy the desired performance through the methods designed by conventional performance indexes. In the paper, the conventional performance indexes are analyzed and then a new performance index for Maglev-based RGV is proposed. Also, an advanced genetic algorithm which is designed using clonal selection algorithm for performance improvement is proposed. To verify the proposed algorithm and the performance index, we compare the proposed method with a simple genetic algorithm and particle swarm optimization. The simulation results show that the proposed method is more effective than conventional optimization methods.

  • 인공면역시스템에서 중요한 특징중의 하나는 지속적으로 변화하는 환경에서 자기(self)의 유동적인 패턴을 동적으로 학습하고 비자기(non-self)에 대한 새로운 패턴을 예측하는데 있다. 본 논문은 자기적 용(self-adaptation)의 인공면역체계 특성을 기반으로하여 설계된 dynamics(동적 클론선택 알고리즘, dynamic clonal selection algorithm)의 역할을 논한다. 시스템의 세가지 중요한 변수인 자기내성 기간(Tolerisation Period). 연역 반응 임계값(activation threshold). 수명(life span)에 따라 변화하는 dynamics의 성능을 네트워크 침입에서 흔히 발견되는 시나리오를 모의실험하여 평가한다

  • DNA 클로닝과 조작기술의 발전은 어떤 유전자의 특정한 위치에 선택적으로 돌연변이를 도입할 수 있는 site-specific mutagenesis 기술을 창출해 내었다. 이 기술로 DAN 염기의 치환, 결실, 삽입등을 클론된 유전자에 직접 도입할 수가 있게 되어 생체의 유전자 조작이나 유전자의 산물인 단백질의 구조와 기능을 의도적으로 변화시키는 protein engineering에 광범위하게 이용되고 있다. Protein engineering은 주로 단백질의 촉매 및 생리활성의 증가, 효소의 특성및 기질 특이성의 변화, 단백질 구조의 안정화 및 내염성 증가, 분자량의 감소, 효소및 생리활성 단백질의 구조의 안정화및 내열성 증가 등에 활용되고 있으며 산업적 유용성이 큰새로운 단백질의 창조에도 기여할 것으로 기대를 모으고 있다. Site-specific mutagenesis 기술로 현재 가장 널리 이용되는 것이 in vitro상에서 수행하는 oligonucleotide-directed site specific mutagenesis이다. 이 방법은 생화학적으로 합성한 특정한 염기서열을 가진 oligonucleotide들을 일종의 mutagen으로 사용하거나 효소적 DNA 합성을 위한 primer로 사용하여 클론된 DNA의 염기서열을 선택적으로 개조하거나 혹은 다른 조작을 하는 것이다. 여기서는 돌연변이율을 높이는 여러가지 개량된 방법들이 나왔으며 그중의 몇가지를 소개하였다.

Ecological Evolution by Competitive Exclusion / An Experimental Approach with Cellular Slime Mold , Polysphondylium pallidum (경쟁배타에 의한 생태적 진화: 세포성 점균 Polysphondylium pallidum에 대한 실험적 접근)

  • Intraspecific clonal interactions have important influences on a population structure of the cellular slime mold (CSM). This study was to investigate whether or not evolutionary change in a population could be induced by clonal competition, and to elucidate how various clones in a population evolve in a homogeneous environment of laboratory culture. The characteristic clones of Polysphondylium pallidum which had different resource consumption rates (RCR) and mating types I and II were selected for study. Investigation was conducted for 4 experimental time interval $(T_0-T_4)$; one experimental time interval took almost 10-14 days from inoculation to havest of fruiting bodies. Two sets of 50 clones were cultured from 50 clones at To, and RCR variations of the population were compared between $(T_0\;and\;T_4)$ for each set of clones. Each clone of the CSM had a diverse resource consumption rate, or growth rate, in a homogeneous and limited Cerophyl agar plate despite the passage of 48-56 generations from the beginning of the experiment. Diverse clones with different growth rate could coexist in one site of the homogeneous agar plate as well as heterogeneous soil microenvironment. When there was high clonal diversity of RCR, a clone in a population had high chances to encounter other clones with resultant increased clonal competition. In one set, 26 of 37 clones of mating type I were changed to mating type Il for the 4 experimental time intervals, which indicated that the rate of competitive exclusion among clones during total experiment from $(T_0\;to\;T_4)$ was 0.703. In another set, 31 of 37 clones of mating type I were changed to mating type II , having the rate of competitive exclusion 0.838. The frequency of each of mat~ng types changed by 0.93-1.29% in each successive generation. The competitive exclusion among clones occurred by 1.26-1.75% when approximately $2.6{\times}10^8$ bacterial cells were provided as food and thereafter one generation of myxamoebae of CSM elapsed at room temperature. This finding implicated that in the vegetative state of P, pallidurn there was 1.26-1.75% probabil~ty of evolutionary change per generation changing from one clone to another clone.

Molecular Cloning and Identification of Novel Genes, Gomsin, Characteristically Expressed in Snailfish, Liparis tanakae (꼼치에서 특징적으로 발현되는 새로운 유전자 곰신의 분리 및 동정)

  • In order to obtain the specific genes of snailfish a subtracted cDNA library was constructed, and analysed by sequencing and GenBank search. Among them C90-171 clone was turned out to be genes showing low homology and nonredundant genes. This novel clone was named Gomsin(C90-171). Gomsin was shown to be intensely expressed in the epithelial cells, some mesenchymal cells, and sheaths of muscle bundles in the result of immunohistochemistry. In the cross reaction assay of Gomsin antibody against various human tissues, the Gomsin was strongly expressed in the ductal and acinar cells of salivary glands, which was similar to the expression patterns of proline-rich proteins(PRPs) of human. The antibody raised against the Gomsin was clearly cross-reacted with human salivary PRPs and also recombinant proteins of human PRPs in the Western blot and immunoprecipitation analysis. Contrast to the salivary PRPs, the Gomsin was not easily degraded in the mixed saliva, but rapidly attacked on the cultured keratocytes in vitro. The simulated protein structure of Gomsin was similar to the whorled pattern of PRPs, even though the amino acid sequence of Gomsin was quite different from those of PRPs. These data suggest that the Gomsin is a characteristic matrix protein in the skin and body of snailfish, which is also utilized for the tissue protection in the similar way to the PRPs of human muco-secretory organs.

Molecular Monitoring of Plankton Diversity in the Seonakdong River and Along the Coast of Namhae (분자 모니터링을 이용한 서낙동강과 남해 연안 플랑크톤 군집 분석)

  • The biodiversity of eukaryotic plankton has commonly been used to evaluate the status of aquatic ecosystems. Therefore, an accurate and rapid method for species identification is needed to reveal the biodiversity of environmental water samples. To date, molecular methods have provided a great deal of information that has enabled identification of the hidden biodiversity in environmental samples. In this study, we utilized environmental polymerase chain reaction (PCR) and constructed the 18S nuclear ribosomal RNA clone library from environmental water samples in order to develop more efficient methods for species identification. For the molecular analysis, water samples were collected from the Seonakdong River (Gimhae Bridge) and the coast of Namhae,(Namhaedo). Colony PCR and restriction fragment length polymorphism of PCR (PCR-RFLP) were then adopted to isolate unique clones from the 18S rDNA clone library. Restriction fragment length polymorphism pattern analysis of the Gimhae Bridge sample revealed 44 unique clones from a total of 60 randomly selected clones, while analysis of the Namhae sample revealed 27 unique clones from 150 clones selected at random. A BLAST search and subsequent phylogenetic analysis conducted using the sequences of these clones revealed hidden biodiversity containing a wide range of taxonomic groups (Heterokontophyta (7), Ciliophora (23), Dinophyta (1), Chytridiomycota (1), Rotifera (1) and Arthropoda (11) in the Gimhae Bridge samples Ciliophora (4), Dinophyta (3), Cryptophyta (1), Arthropoda (19) in the Namhae samples). Therefore, the molecular monitoring method developed here can provide additional information regarding the biodiversity and community structure of eukaryotic plankton in environmental samples and helps construct a useful database of biodiversity for aquatic ecosystems.

  • 전장 cDNA 클론을 시퀀싱하는 것은 선택적 스플라이싱 형태를 비롯한 정확한 유전자 구조를 정의하는데 유용하게 사용될 수 있으며, 유전자 및 단백질의 생물학적 기능연구에 중요한 자원을 제공한다. 포괄적이며 비 중복적인 cDNA의 생산은 인간 유전체 연구의 중요한 목표이다. 본 연구에서 제공하는 인간 cDNA 라이브러리 분석 파이프라인은 전장 cDNA를 분석하는 자동화 도구로 여러 연구자들에게 활용 될 수 있을 것으로 사료된다.
