Enzymatic Synthesis of Polyphenol Glycosides by Amylosucrase (재조합 아밀로수크라아제를 이용한 효율적인 폴리페놀 배당체의 합성)

  • The capability of synthesizing polyphenol glycosides was examined using recombinant amylosucrase from the hyperthermophilic bacterium Deinococcus geothermalis. Based on the action mode of amylosucrase, sucrose and twenty-one polyphenols were used as a donor and acceptors respectively. The transglycosylation reaction by amylosucrase produced one or two major polyphenol glycosides depending on the type of polyphenols used. The synthesized polyphenol glycosides were detected by thin-layer chromatography. The structures of the newly synthesized polyphenol glycosides were predicted based on the transglycosylation mechanism of the enzyme. According to the acceptability of the polyphenols, the structural characteristics of polyphenol as an efficient acceptor were evaluated. The results indicate that amylosucrase is an efficient catalyst for the enzymatic synthesis of polyphenol glycosides, which have high potentials in food, cosmetics, and pharmaceutical industries.

Volatile Flavor Components in Watermelon(Citrullus vulgaris S.) and Oriental Melon(Cucumis melo L.) (국내산 수박(Citrullus vulgaris S.) 과 참외(Cucumis melo L.) 의 휘발성 향기성분)

  • Volatile flavor components of watermelon (Citrullus vulgaris S.) and oriental melon (Cucumis melo L.) obtained by simultaneous steam distillation and extraction apparatus were separated by gas chromatography (GC) and gas chromatography-mass spectrometry (GC/MS). Thirty seven and fifty five volatile flavor components were identified in watermelon and oriental melon, respectively. (Z)-3-Nonen-1-ol, (Z,Z)-3,6-nonadien-1-ol, (E,Z)-2,6-nonadienal and (E)-2-nonenal containing unsaturated nine carbon atoms were the characteristic flavor components of watermelon. $C_{9}-Unsaturated$ esters including (Z)-3-nonenyl acetate, (Z)-6-nonenyl acetate, (Z,Z)-3,6-nonadienyl acetate and thioester were important components in the flavor profile of oriental melon.

  • In this study, mixed catalytic char gasification of Indonesia low-rank coal Kideco was investigated under nitrogen atmosphere and isothermal conditions at a fixed reactor. The effects of the temperature were investigated at various temperature (700, 750, 800, $850^{\circ}C$). The effects of blend ratio of catalysts ($K_2CO_3$, Ni) were investigated with different blend ratios (1:9, 3:7, 5:5, 7:3 and 9:1). The sample was prepared by mixing with $K_2CO_3$ physically and by ionexchange method with Ni. The data from thermogravimetric analyzer and gas chromatography were applied to four gassolid reaction kinetic models including shrinking core model, volumetric reaction model, random pore model and modified volumetric reaction model.

  • A new gas-chromatographic method for determining carbon and hydrogen in organic compounds has been developed. After sample combustion was performed in a regular analytical combustion tube with an internal oxidant (a mixture of silver oxide and manganese dioxide) under a helium flow, the water produced was converted to acetylene by passing through a calcium carbide tube. The carbon dioxide and acetylene were trapped by a molecular sieve 5A column at room temperature. The trapped gases were released under programmed temperature raise up to $340^{\circ}C$ and the released gases were passed through a silica gel column. The adsorption of $CO_2$ and $C_2H_2$ in the molecular sieve 5A trapping column were found to be quantitative and the silica gel column showed an excellent resolution of $CO_2$ and $C_2H_2$ for analytical purpose. The analytical results for various known compounds based on the out-put of the thermal conductivity cell calibrated for the amounts of carbon and hydrogen contents in benzoic acid, showed average errors ${\pm}0.5%$ and ${\pm}0.33%$ for carbon and hydrogen, respectively.

  • From water extract of red ginseng, two new-type glucosides, $A:\;C_{12}H_{16}O_8\;and\;B:\;C_9H_{16}\;O_7,$ were isolated by fractionation with highly porous polymer ollowed by chromatography on silica gel in yields of 0.04 and $0.16\%,$ respectivley. The structures of A and B were elucidated by means of NMR and MS. Neither glucosides were detected in the extract of white ginseng and the mechanism of formation of A and B during the process of steaming will be discussed. The isolation and identification of saponins of corms (head of the root) of red ginseng are also reported.

  • Changes in activity of gibberellin C20-hydroxylase which converts $[^{14}C]GA_{12}\;to\;GA_{l5}$ were studied during seed maturation using the partially purified enzyme preparations of two cultivars, Kentucky Wonder (normal) and Masterpiece (dwarf) of Phaseolus vulgaris. The preparations obtained by methanol precipitation and hydrophobic interaction chromatography efficiently converted; $GA_{l2}\;to\;GA_{4},\;via\;GA_{15},\;GA_{24}\;and\;GA_{9};\;GA_{20}\;to\;GA_{1},\;GA_{5}\;and\;GA_{6}$. The activities of C20-hydroxylase which converts $GA_{l2}\;to\;GA_{l5}$ were almost the same in both cultivars. The C20-hydroxylase activity per protein reached a maximum at the very early immature seed stage, followed by a subsequent rapid decrease during seed maturation, whereas the enzyme specific activity per seed reached a maximum at 21 days after flowering, and showed a similar fiuctuation to that of the 313-hydroxylase which converts $GA_{20}\;to\;GA_{l}$ during seed maturation.ration.

Preparation and Characterization of Unsaturated Poly(3-hydroxyalkanoate) Nanoparticles (불포화 폴리히드록시알칸오에이트 나노입자의 제조 및 특성)

  • Nanoparticles with unsaturated poly(hydroxyalkanoate)s (UPHAs) biosynthesized with Pseudo-monas oleovorans were prepared by spontaneous emulsification solvent diffusion method. The influence of nanoparticle formation was investigated with various experimental parameters such as sonication conditions, sol-vent, surfactant and polymer contents, etc. The physical and chemical properties of UPHAS and its nanoparticles were characterized using $^1$H- and $\^$13/C-nuclear magnetic resonance spectroscopies, attenuated total reflection infrared spectroscopy, differential scanning calorimetry and gel permeation chromatography. The morphology of particles was observed using scanning electron microscope and the size and distribution of nanoparticles were measured with electrophoretic light scattering spectrophotometer. The mean diameter of particles decreased with increasing sonication amplitude and time. The addition of ethanol into UPHAS chloroform solution decreased the particle size presumably due to increased solvent diffusion into water phase. The particle size increased with increased the concentration of UPHAS solution. Under the 2-4% poly(vinyl alcohol) (PVA) aqueous solution the minimum mean diameter of particles was shown. The higher degree of hydrolysis and degree of polymerization of PVA increased the mean diameter of particles.

Conservation and Analysis of Inner Materials of a Stoneware Bottle from Incheon Ongjingun Yeongheungdo Shipwreck (인천 옹진군 영흥도선 출수 도기병의 보존처리와 내용물 분석)

  • A stoneware bottle was recovered from the Incheon Yeongheungdo Shipwreck in 2013. Primary observations showed that this object had a sealed opening and was covered with a foreign material. After it was cleaned via mechanical and chemical methods, wave patterns were observed on the outer surface of the bottle and a yellowish-brown transparent material was found within. In this paper, the process of conserving the stoneware bottle and the analysis conducted on the unknown material found within is explained. The conservation process included steps such as cleaning, desalination and restoration. After of the missing area located at the rim, the original shape of the bottle was made clear. In addition, the unknown inner material was analyzed using FT-IR and GC-MS. Results showed that the material is similar to golden lacquer. It is speculated that the Yeongheungdo Ship had wrecked during the Unified Silla period while carrying a bottle loaded with golden lacquer.

Production of Red Bean Starch Granule with Cellulase from Fusarium moniliforme (Fusarium moniliforme이 생산하는 셀룰라아제를 이용한 소두전분제조(小豆澱粉製造))

  • Two fractions of ${\beta}-glucanase$(CMCase), two fractions of filter paper degradation enzyme (FPase) and one ${\beta}-glucanase$ fraction were partially purified from Fusarium moniliforme and applied to recovery process of red bean starch. Red bean were incubated with the fractions of CMCase and FPase at $50^{\circ}C$ for 2 hours and the starch granules are separated. Maximal sedimentation rate of red bean starch granules was obtained with treatment of the mixture solution of 0.004 units/ml of FPase and 0.3 units/ml of CMCase. In the enzyme treated process percent recovery of red bean starch granule increased about 7% and suspended solid in waste water was reduced about 40%, compared with those of control. The results indicated that red bean cell treated with cellulase fractions absorbed water more rapidly and specific gravity of starch granule increased.

Analysis of Trace Levels of Halonitromethanes (HNM) in Water using Headspace-SPME and GC-ECD (Headspace-SPME와 GC-ECD를 이용한 수중의 미량 Halonitromethane (HNM)류 분석)

  • Halonitromethanes (HNMs) are one of the most toxic groups of disinfection by-products. Recently, various studies have been fulfilled. An automated headspace-solid phase microextraction (SPME) gas chromatography/electron capture detector (GC-ECD) technique was developed for routine analysis of 9 HNMs in water samples. The optimization of the method is discussed. The limits of detection (LOD) and limits of quantification (LOQ) range from 90 ng/L to 260 ng/L and from 270 ng/L to 840 ng/L for 9 HNMs, respectively. Matrix effects in tap water and sea water were investigated and it was shown that the method is suitable for the analysis of trace levels of HNMs, in a wide range of waters. The method developed in the present study has the advantage of being rapid, simple and sensitive.
