Stability of the enzyme-modified starch-based hydrogel model premix with curcumin during in vitro digestion (효소변형 전분기반 하이드로젤 모델 프리믹스 내 탑재된 커큐민의 소화과정 중 안정성)

  • In this study, the effect of enzyme-modified starch used in the preparation of filled hydrogel powder loaded with curcumin (FHP) on redispersibility, thermal and UV stability, and curcumin retention during in vitro digestion was investigated. FHP maintained stability without layer separation when redispersed and showed more stability against UVB than the emulsion powder (EMP). There was no significant difference in the chemical stability of curcumin between rice starch-based filled hydrogel powder (RS-FHP) and enzyme-modified starch-based filled hydrogel powder (GS-FHP). However, the gel matrix of GS-FHP maintained greater stability of lipid droplets in the stomach compared to RS-FHP, thereby improving the retention rate of curcumin after in vitro digestion. GS-FHP could be used as a novel material for developing premixes that require stable formulation and maintenance of functional substances, as it can increase the dispersion stability and retention rate of functional substances after digestion.

Oral Bioavailability for Sub-micron Particle Curcumin (TheracurminCR-033P) and Absorption Rate by Product Composition in Healthy Human Volunteers (서브마이크론 입자 형태 커큐민의 생체 흡수율 및 제품 조성에 따른 흡수율 동등성 연구)

  • Curcumin have various health-beneficial properties in numerous studies. However, its bioavailability is low due to its limited intestinal uptake and rapid metabolism. This study aimed to evaluate the pharmacokinetics of newly developed sub-micron particle curcumin with increased water dispersibility (Theracurmin(R) CR-033P). Plasma curcumin levels were measured at 0, 1, 2, 4, 8 h after Theracurmin(R) CR-033P intake using high-performance liquid chromatography. For analyzing pharmacokinetics of Theracurmin(R) CR-033P, eighteen healthy subjects were recruited and received Theracurmin(R) CR-033P at a single oral dose containing curcumin 30 mg. $C_{max}$ was 28.14 ng/ml, and the area under the curve for 8 h was estimated to be 104.36 ng/ml. Based on the area under the plasma concentration (AUC), the bioavailability of sub-micron particle curcumin was higher 22-, 35-, 28-fold than native curcumin in men, women, and all subjects, respectively. For comparing by formulation, seven healthy subjects were recruited and received two type of treatment: (1) existing dosage form 300 mg (contained curcumin 30 mg) ${\times}$ 3 capsule, (2) high dosage form 300 mg (contained curcumin 90 mg) ${\times}$ 1 capsule + placebo 300 mg ${\times}$ 2 capsule. In the cross-over study, there was no significant differences in $C_{max}$ and AUC of plasma curcumin. In conclusion, submicron particle curcumin with increased water dispersibility significantly improved its oral bioavailability and women absorbed curcumin more effectively than men. Different formulation of Theracurmin(R) CR-033P has shown equivalent to the reference in terms of pharmacokinetics.

  • Rapid increase of diabetic population is a major health concern in Korea. In a trial to develop food components which can prevent and/or cure diabetes, we investigated the anti-diabetic activity of curcumin in high fat diet (HFD)-induced type 2 diabetes mellitus (T2DM) animal model. C57BL/6 mice were divided into three groups: normal diet (ND), high fat diet (HFD), and curcumin (CUR, HFD+0.02% curcumin). Mice were fed each diet for 16 weeks. CUR significantly reduced body weight gain, the levels of plasma glucose, insulin, total-cholesterol (T-C), and LDL-C, whereas increased HDL-C compared to those of HFD group. Notably, insulin signaling pathway was activated by CUR. This suggests that CUR improves obesity-associated T2DM by overcoming insulin resistance in part.

Colorimetric Sensing Behavior of Curcumin Printed on Paper and Cotton Fabric (커큐민으로 프린팅된 종이와 면직물의 유해가스에 대한 색채 감지 거동)

  • Increasing the needs for eco-friendly natural materials, much attention on natural dyes has been attracted. Curcumin, one of abundant natural dyes available in nature, is an eco-friendly molecule found in turmeric. In this study, the colorimetric sensing behavior characteristics of ammonia and hydrogen chloride gases was analyzed using curcumin ink-printed paper and cotton fabric by inkjet printing method. The fabricated paper and fabric were utilized as a colorimetric sensor that can track food spoilage. The color changes of the samples printed on the paper and cotton upon exposure to the toxic gases, the reversibility of the color changes, and the water resistance were investigated. Both samples showed reversible reactions with NH3 and HCl, and small amount of ammonia produced by food spoilage was successfully sensed with naked eyes, confirming its capability to warn food spoilage in our daily life.

The effect of flavonoids on the TREK-1 channel (TREK-1 채널에 대한 플라보노이드의 효과)

  • TREK-1 channel is a member of the two-pore domain potassium (K2P) channel family that is regulated by intracellular pH, membrane stretch, polyunsaturated fatty acids, temperature, and some neuroprotectant agents. TREK-1 channel can influence neuronal excitability by regulating leakage of potassium ions and resting membrane potential. TREK-1 channel has been shown to be overexpressed in prostate cancer cells. Although the importance of these properties, relatively little is known about flavonoid effects in the regulations of TREK-1 channel. The purpose of the study was to screening of flavonoids as the TREK-1 channel modulator using one of electrophysiological techniques such as excised inside-out patch configuration. We demonstrated blocking effect on TREK-1 channel by flavonoids such as epigallocatechin-3-gallate (EGCG), curcumin and quercetin in CHO cells transiently expressing TREK-1 channel. The inhibition of TREK-1 channel by quercetin and curcumin was reversible, whereas EGCG was little reversible. Quercetin, EGCG and curcumin decreased the relative channel activity to 73%, 91% and 94%, respectively. The half-inhibitory concentration (IC50) of curcumin, quercetin and EGCG was $1.04{\pm}0.19\;{\mu}M$, $1.13{\pm}0.26\;{\mu}M$ and $13.5{\pm}2.20\;{\mu}M$ in CHO cells expressing TREK-1 channel, respectively. These results indicate that flavonoids might regulate TREK-1 and this regulation might be one of the pharmacological actions of flavonoid in nervous systems and cancer cells.

Research regarding curcumin and soybean extract's influence on anti-inflammatory and acne and skin-repairing effect on RAW 264.7 macrophages (커큐민과 대두 추출물의 RAW 264.7 대식세포에서의 항염증 효과 및 여드름 피부 개선효과에 관한 연구)

  • By studying bioactivity in the aspect of skin care via curcumin, the main component of turmeric, and isoflavones, the soybean extract, this research tried to prove the possibilities as cosmetic ingredients. This research identified the curcumin and soybean extract's anti-inflammatory and toxic effects on skin cells through cell experiment, and using HPLC, the moisture, oiliness, and changes in erythema were measured when the soybean extract was applied to the skin. The result showed that curcumin had the least toxin on RAW 264.7 cell and the anti-inflammatory effect was identified. There were statistically significant changes on the moisture, oiliness and erythema when cosmetics containing curcumin and soybean extract were used. Therefore, it showed the possibilities of the utilization of curcumin and soybean extract to be used as cosmetics for acne skin.

Wound Healing Effect of Curcumin Gel for Transdermal Delivery (경피전달을 위한 커큐민 젤의 창상치유효과)

  • The aim of this study was to investigate the effect of trasdermal delivery of the curcumin gel on healing of the rats' dorsum wounds. Carbopol 934 and propylene glycol were used to prepare gels containing 1% curcumin. Curcumin gel was evaluated for various properties such as antioxidant, cell viability, anti-inflammatory, in vivo wound healing. The free radical scavenging activity using 1,1-diphenyl-2-picryl hydrazyl (DPPH) was 50% at 12.5 ppm concentration. Inhibition of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells by curcumin gel. In the curcumin gel-treated group, the re-epithelialization in wounds was significantly increased compared to the control group throughout the experimental period. These results suggested that curcumin may be helpful for the promotion of wound healing.

Analysis of the Degradation Products of Turmeric using GC-MS (GC-MS법을 이용한 울금의 퇴화물 분석)

  • Degradation products of the dye extracted from turmeric and the turmeric dyed textiles were examined by using GC-MS after 100 oven (OV) and $H_2O_2/UV/O_2$(PER) treatments for up to 28 days. Throughout the OV degradation times, 2-propenoic acid, 3-(2-hydroxyphenyl)- was found consistently, while isovanillin, and vanillic acid were newly detected. In 28 day PER degradation sample, feruloylmethane, 2-propenoic acid, 3-(2-hydroxyphenyl)-, benzoic acid, and vanillic acid were detected as well as isovanillin. Feruloylrnethane, and 2-propenoic acid, 3-(2-hydroxyphenyl)- were detected from the degraded fabric samples. With the absence of curcuminoids in the GC-MS result, the decreasing pattern of 2-propenoic acid, 3-(2-hydrokyphenyl)- reflect the degradation of curcuminoids in turmeric extraction with the progression of OV degradation times. It is suggested that isovanillin, feruloylmethane, 2-propenoic acid,3-(2-hydroxyphenyl)-, and vanillic acid are the probable fingerprint products for determining the turmeric dye from the badly faded archaeological textiles.
