Isolation of Cadmium-tolerant Bacteria and Accumulation of Cadmium into the Bacterial Cell (카드뮴의 내성균분리 및 균체내 축적)

  • A strain of Enterobacter cloacae isolated from soil showed the evidence of growth in medium containing 1,500 ppm of Cd$\^$++/ though its growth was inhibited at high cadmium concentrations. This strain accumulated 59% of cadmium into the cells during incubation in medium containing 0.5 ppm of cadmium and its uptake was nearly proportional to the dry weight of cells, the average being 7.8 mg cadmium per g dry cells. It was also found that 60-70% of cadmium accumulated in cells were distributed in the cell wall fraction.

  • An intracellular accumulation of cadmium by the intact cell of an extremely cadmium tolerant yeast, Hansenula anomala B-7, was investigated in the presence of Triton X-100. The uptake of cadmium by the intact cell was efficiently enhanced up to approximately 40% or more by 0.1% of Triton X-100 and Aerosol OT, respectively. The Michaelis constant, Km, done by Lineweaver-Burk plot of accumulation velocity of cadmium vs. cadmium concentration was calculated to be 0.247mM. The optimal conditions of pH and the temperature for the effective cadmium uptake were from neutrality to alkali and 4$0^{\circ}C$, respectively. The accumulation of cadmium was increased approximately 3 times under the shaking incubation, with no correlation to shaking rate. By zinc the cadmium accumulation was decreased.

  • As a pary of investgation on effective accumulation of cadmium in yeast cells, effect of surfactant was studied on intracellular accumulation of cadmium in extremely cadmium-tolerant yeast, Hansenula anomala B-7. Cadmium accumulation was enhanced up to approximately 40% by the addition of non-ionic surfactant, Triton X-100 and its optimal concentration was found to be 0.1-0.2%. Furthermore, optimum conditions for intracellular accumulation of cadmium were at $40^{\circ}C$ and initial pH 6.0 for 48 hours under shaking culture.

  • The cultural conditions and the intra cellular accumulation of cadmium was studied using a cadmium tolerant yeast strain B-7 which had been isolated from activated sludge collected from a zinc mining area. The organism was able to grow in a medium containing 3,000 $\mu\textrm{g}$/$m\ell$ of cadmium-ion. (C $d^{++}$) Optimum conditions for the growth of the organisms were 20~22$^{\circ}C$ and pH 5.0~8.0 under aerobic condition. The maximum cadmium accumulation was observed when the organism was grown at pH 6.0. The growth of B-7 was not affected by the addition of a silicone-based antifoamer, which stimulated the intra cellular accumulation of cadmium. The intra cellular cadmium accumulation started after the cell ceased to grow. One gram of cells accumulated 34.17mg of cadmium when the organism was grown in a medium containing 500 $\mu\textrm{g}$/$m\ell$ of cadmium and 0.2%, v/v silicone-based antifoamer at 28$^{\circ}C$ for 48 hours with shaking. About 73 % of the accumulated heavy metal by the organism was found in the cytoplasm.m.

  • A mechanistic study by which Cadmium-tolerant P.Putida C1 accumulates high conc of Cd in its cell body was performed. Approximately 57% Cd accumulated was distributed on the cell wall and the other 43% portion was in cytoplasm. 84% Cd of the Cd in the cell wall fractions present in the polyphosphate-polysaccharide fractions, but most of Cd in the cytoplasm fraction was in protein and nucleic acid. Cadmium affected the protein synthesis in P. Putida. The intracellular protein content was decreased by cadmium addition, but the soluble protein precipitated by ammonium sulfate($30{\sim}75%$ satruation) was increased as compared to that from the cells grwon without cadmium. Furthermore, in the cells grown with of cadmium, high-molecular-weight soluble protein was increased, with of cadmium, high-molecular-weight soluble protein was increased, compared with the cells grown without cadmium, but low-molecular-weight soluble protein was decreased. These results indicate that Cd inhibited the intracellular protein biosynthesis but enhance biosynthesis of the high-molecular-weight soluble protein precipitate by ammonium sulfate($30{\sim}75%$ saturation).

The Cadmium Biosorption Mechanism in Gram Negative Bacteria, Serratia marcescens (Gram 음성 세균인 Serratia marcescens에 의한 카드뮴 흡착 기작)

  • Serratia marcescens, an enterobacterium of gram-negative bacteria, is characterized by resistance of the admium. Cadmium sensitive PM strain did not grow in the medium at cadmium concentration of 50 ppm. PA strain was induced to accommodate to cadmium by cultivating the mother strain (PC strain) in the medium with 50 ppm cadmium. As compared with PC and PM strains, PA strain revealed the excellent growth in cadmium media and accumulated four to five times higher cadmium concentration in cell than other strains. PA strain accumulated 23% of cadmium in cells when cultured in medium treated with 100 ppm cadmium and this cadmium was more accumulated in cytosol fractions than membrane fractions. Analysis by TEM indicated that cadmium was concentrated as a form of granule in cytosol. In protein patterns of cell after the treatment of cadmium, two inducible proteins (28 KDa and 64 KDa) and one reducible protein (45 KDa) were detected by SDS-PAGE. By Atomic Absorption Spectrophotometer, the amounts of cadmium attached to inducible proteins of 28 KDa and 64 KDa were 318.28 ㎍ and 325.37 ㎍ per gram of protein, respectively. It is assumed that these inducible proteins play an important role in the mechanism of cadmium accumulation in cells. A plasmid of 23Kbp was found in S. marcescens. The ability of resistance to cadmium in plasmid was confirmed by curing experiments.

  • 서울주변 78지역의 토양과 하수를 수집하여 200ppm의 카드뮴을 함유한 배지에서 생장하는 142주의 세균류를 분리하였다 이들중 1,500ppm의 카드뮴함유 배지에서 증식하는 8주를 선발하고 그 중 1주에 대하여 100 및 500ppm의 카드뮴 함유배지에서의 생장곡선을 얻었다. 또한 0.5, 1, 10및 100ppm의 카드뮴함유 배지에서 배양시간에 따른 균체내 카드뮴의 축적을을 조사한바 0.5ppm에서 최고 59%이였다.(중략)

  • Accumulation, elimination and subcellular distribution of heavy metals in Littorina brevicula exposed to cadmium and zinc separately and concurrently were investigated. When the winkles had been exposed to 400 ㎍/L CdCl₂ and 3000 ㎍/l ZnSO₄ separately for 90 days, each of the metal body burden in the whole sofl parts increased in proportion to time of exposure until 70 days. But it didn't increase after 70 days. But when the winkles had been exposed to cadmium and zinc simultaneously, cadmium body burden decreased but zinc body burden increased as compared to the winkles exposed to each of the metal. We also found that cadmium accumulated in the winkles was not depurated for 42 days, but zinc accumulated in them was depurated. Especially, zinc was depurated faster when they had been exposed to mixture of cadmium and zinc. After the winkles had been exposed to cadmium and zinc separately for 70 days, about 60% cadmium of the total body burden was associated with the soluble fraction, while about 75% zinc of the total body burden was associated with insoluble fraction. And these trends of metal partitioning did not alter when the winkles had been exposed to metal mixture. After the soluble fraction applied to gel-filtration chromatography column, the distribution patterns of cadmium and zinc associated with proteins or ligands were different each other. Most of cadmium (>90%) in the soluble fraction was bound to MBP-1 (Metal-binding protein-1), about 6.5 kDa), while zinc was distributed evenly to HMW (High molecular weight fraction, >60 kDa), MBP-1, MBP-2 (about 5 kDa), LMW (Low molecular weight fraction, <1 kDa).

  • Of the cadmium-tolerant 162 bacterial strains isolated from soils, river waters or active sludges of waste-water disposal plants in the Gyeongnam province a strain C1, which showed considerably higher growth rate in the agar plate containing 2000 ppm than any other strains isolated, was identified as a Pseudomonas putida or its similar strain when analyzed by taxonomical characteristics. Optimum pH and temperature for the growth of the P, putida were 7.0 and $30^{\circ}C$, respectively. This strain was resistant to antibiotics(ampicillin, chloramphenicol and streptomycin), and heavy metals(lithium, cupper, lead and zinc). This strain utilized salicylate, naphthalene or xylene as a sole carbon source. The rate of cadmium accumulation in P. putida cell was enhanced at low concentration of Cd in the growth media. The maximum cadmium absorption by this strain grown in 1 and l0ppm of Cd was respectively 78% and 60% 24 hrs after culture, but in 100 ppm Cd, 40% 48 hrs after culture. Addition of a non-ionic surfactant Triton X-100(0.1%) to the medium enhanced the accumulation of cadmium in the P. putida up to approximately 37%.

A Study of the Potential for Phytoremediation of Cd Contaminated Soil (카드뮴 오염 토양에 Phytoremediation의 적용 가능성 연구)

  • Phytoremediation of soil contaminated with cadmium was studied using Indian mallow (Abutilon avicennae) in columns packed with 80 mg Cd/kg soil. At 90 days after transplat, root biomass of the exposed plants was 4 times more inhibited compared to the control. Also, shoot length of the exposed plants was 3 times more inhibited than that of control plants. Accumulation of cadmium into tissues was in the order roots> stems> leaves during the 50 days, but the order was roots> stems> seeds> leaves during the 90 days after transplant. Regardless of cadmium contaminations, microbial activities were significantly greater in soil with plants than without plants. In soil column, cadmium was not transferred toward the lower part. Uptake of Cd by plant tissues was about 3.5% of the initial bioavailable cadmium for leaves, stems, and roots during the 90 days after transplant.
