Use of Plant Materials for Decontamination of Waste Water Polluted with 2,4-Dichlorophenol (2,4-Dichlorophenol로 오염된 폐수의 정화를 위한 식물체의 이용)

  • This study was performed to estimate the possibility of use of plant materials as catalytic agents fur the decontamination of waste waters contaminated with organic pollutants by using 2,4-dichlorophenol(2,4-DCP) as a model pollutant. Plant materials containing high peroxidase activity were selected as catalysts for the removal of 2,4-DCP. Peroxidase activity, which plant materials were containing, was measured, and the greatest peroxidase activity was observed in shepherd's purse, followed by turnip, sweet potato, Chinese cabbage and white radish. The peroxidase activity in shepherd's purse was four times higher than that of horseradish purchased in U.S.A. Using shepherd' s purse and turnip, it was investigated the effect of various factors on the decontamination of 2,4-DCP through oxidative coupling. The removal of 2,4-DCP was extremely fast, and a maximal removal could be achieved within 3 min for shepherd' s purse and 15min for turnip. The pH range was from 3.0 to 8.0 and the amount of $H_2O_2$ added was 9mM when maximal removal was achieved(over 90%). No increasing removal of 2,4-DCP was observed due to increasing the amount of $H_2O_2$ added (over 9mM). The initial concentration affected the transformation of 2,4-DCP incubated with plant materials. When turnip was used as catalytic agent, it was observed decreasing transformation of 2,4-DCP due to increasing initial concentration.

  • Isopropyl alcohol extracted filefish protein concentrate (FPC) and NaOH hydrolyzed filefish protein isolate (FPI) were used for the investigation on the effect of processing conditions on the molecular distribution and functional properties. The molecular distribution of FPC on polyacrylamide gel showed a resemblance to that of fish muscle, but that of alkaline hydrolyzed FPI showed the severe degradation of protein. The content of several amino acids in FPI were lower than those of FPC. The pepsin digestibility of the FPC dried at high temperature was relatively high. FPC didn't exhibit a significant difference in nitrogen solubility at the pH range of 3.0-9.0, while FPI showed a wide difference with the pH change. FPI was more suspensible and rehydrated in water than FPC. Although the aeration capacity of FPI was very low, foam viscosity was higher than that of FPC. In contrast with aeration capacity, FPI presented higher emulsion capacity and lower emulsion viscosity than FPC. The size of fat globule in the emulsion of FPC was larger than that of FPI. In general, most functional properties decreased with the increment in drying temperature, except water holding capacity.

  • To improve the mechanical properties of $Al_2$O$_3$/ZrO$_2$composites, the homogeneous dispersion of ultra low size ZrO$_2$ particles in $Al_2$O$_3$ceramics have been controlled by coprecipitation method. In case of mechanical mixing of ZrO$_2$ powders with $Al_2$O$_3$, homogeneous dispersion and controlling the ZrO$_2$ size were relatively difficult due to high sintering temperature. So nanosized Zr hydroxide was coprecipitated from ZrOCl$_2$/Y(NO$_3$)$_3$ solution with commercial sub-micron sized $\alpha$-alumina (Sumitomo : AES-11(0.4 ${\mu}{\textrm}{m}$)) and high purity ultra low sized $\alpha$-alumina (Taimei Chemical (0.22 ${\mu}{\textrm}{m}$)) for low temperature sintering. By this partial coprecipitation method, relatively low sized ZrO$_2$ dispersion in $Al_2$O$_3$/ZrO$_2$ composites was achieved at 150$0^{\circ}C$-1$600^{\circ}C$ of sintering temperature range and their mechanical properties were measured.

Characterization of Endoglucanase (F-I-III) Purified from Trichoderma sp. C-4 (새로운 섬유소분해 균주 Trichoderma sp. C-4에서 분리한 Endoglucanase (F-I-III)에 대한 연구)

  • One of the endoglucanases, F-I-III, was purified from the culture filtrate of T. sp. C-4 through procedures including chromatography on Sephacryl S-200, DEAE-Sepharose A-50, and Chromatofocusing on Mono-P (FPLC). The molecular weight of the enzyme was determined to be about 56,000 Da by SDS-PAGE, and pI of 4.9 by analytical isoelectric focusing. F-I-III showed the highest enzyme activity at $55^{\circ}C$, and the pH optimum of the enzyme was 5.0. There was no loss of activity when the enzyme was incubated at $50^{\circ}C$ for 24 hours. The specific activity of the enzyme F-I-III toward the CMC was 315.4 U/mg. The Km value for $PNPG_2$ of F-I-III was 2.69 mM. N-terminal sequence of F-I-III was analyzed to be QPGTSTPEVHPKKLTTYK. It showed $95\%$ of homology to that of EGI from T. reesei. The presence of some metal ions (1 mM) had only a little effect on CMCase activity. The treatment of the reducing agents resulted in the increase of endoglucanase activity.

  • In case of high contents of rare earths in the LiCl-KCl salt, it is not easy to recover U and TRU metals as a usable resource form from LiCl-KCl eutectic salts generated from the pyroprocessing of spent nuclear fuel. In this study, a conversion of $UCl_3$ into an oxide form using $K_2CO_3$ and an electrodeposition of $NdCl_3$ into a metal form in $LiCl-KCl-UCl_3-NdCl_3$ system were conducted to resolve the problem. Before conducting the conversion, experimental conditions for the conversion were determined by performing a thermodynamic equilibrium calculation. In this study, almost all of $UCl_3$ disappeared in the LiCl-KCl salt when the injection of $K_2CO_3$ reached theoretical equivalent for the conversion, and then $NdCl_3$ was effectively electrodeposited as a metal form using liquid zinc cathode. After that, the LiCl-KCl salt became transparent, and uranium oxides were precipitated to the bottom of the LiCl-KCl salt. These results will be utilized in designing a process to separate U and rare earths in LiCl-KCl salt.

Alkaline Pilot Processing for Recovery of Fish Muscle Protein and Properties of Recovered Protein (어육 단백질 회수를 위한 알칼리 Pilot 공정과 회수 단백질의 특성)

  • Optimum operation condition for pilot scale of alkaline processing for fish muscle was investigated by measuring protein solubility, yield, texture, and water-holding capacity. Recovered protein yield was 33.2% for whole fish and 61.8% for minced muscle. Optimum homogenized speed and time, using industrial scale homogenizer, were 3,000 rpm and 5 min, respectively. Limited centrifugal force of continuous cylinder type was 4,000 rpm for recovering soluble protein, and 2,000 rpm for recovering precipitated proteins. The pH control agents such as citric acid, sodium phosphate and calcium oxide decreased the breaking force and deformation of recovered protein gel. The breaking force and deformation of the recovered proteins were high compared to conventional surimi. The breaking force and deformation were decreased by addition of salt, starch and bovine plasma proteins. Whiteness of recovered protein gel was lower than that of conventional surimi. Alkaline processing greatly decreased nitrogen content and chemical oxygen demand in waste water. The results suggest that alkaline processing has a potential as industrial production for recovering the proteins from fish muscle.

항만개발에 의한 조석조류 및 수질변화 예측을 위한 시뮬레이션 모델 연구

  • 항만개발 사업은 매립이나 준설과 같은 해안선과 해저지형을 변화시키는 공사가 이루어지며 이는 조석조류 현상의 변화를 유발하게 된다. 특히 해안지형이 복잡한 항만이나 하구에서는 변화정도가 크게 나타나며 방파제나 선박접안 시설과 같은 항내 구조물을 축조 할 경우에도 조석조류 변화가 야기된다. 조석조류 현상의 변화는 해양 수질과 생물상에 영 향을 줄뿐만 아니라 인접한 육지에 이르기까지 크다란 피해를 유발할 수가 있다. 창조시 해 수를 육지로 범람하게 하거나 퇴적이나 침식조건을 변화시킬 수도 있으며 오염물질이 외해 로 확산되는 과정을 방해하고 해양생물의 서식조건을 악화시켜 부근 해역의 어업에 피해를 유발할 수도 있다. 또한 해안 공사 과정에 해수로 유입되는 토사나 항만 이용시에 배출되는 폐수 등이 항내 수질과 생태계를 변화시킬 수 있다. 따라서 조석조류와 수질변화를 사전에 예측하고 이를 최소화하는 방안이 강구되어야 한다. 조석조류와 수질 변화를 사전에 예측하 는 방법으로 축소모형 실험과 컴퓨터 시뮬레이션이 사용된다. 축소모형 실험은 현장을 재현 하는데 많은 경비와 시간이 소모되며 예측에 한계가 있다. 반면에 컴퓨터 시뮬레이션은 대 상해역의 지형과 조류 그리고 수질특성에 적합하게 모델을 개발할 경우 예측력이 매우 높기 때문에 현재 가장 널리 이용되고 있다. 본 연구는 유한 요소법에 기초하여 미국 공병단 (United States Army Corps of Engineers)에서 개발한 TABS-2 모델을 이용하여 온산항에 서 시행되는 부두건설에 따른 조석조류 및 수질변화를 시뮬레이션하였다. 측정된 자료로 모 델 계수를 보정하고 검증하였으며 항내 매립과 준설이 이루어질 때 나타나는 조위와 조속변 화를 예측하였다. 매립과 준설공사시 유출되는 토사가 조석조류현상에 따라 이동 확산되는 범위와 농도분포를 예측하였다.ophobic components)은 75~80%, 친수성 물질(hydrophilic components)은 30~33%정도의 분포를 보였고, 전염소 및 전오존 공정을 거친 처리수에서는 각각 62.2-62.8%, 43.9~49.0% 및 50~ 55%, 40~57% 정도의 분포를 보였다. 그리고 웅집-침전을 거친 처리수에서는 그 분포가 77~82%, 24-48%였다. 전주리 공정을 통하여 소수성 물질(byoghobic components)의 분포가 감 소하는 것을 볼 때 전염소 및 전오존 처리가 용존유기물의 응집에는 오히려 역 효과를 나타내는 것으로 판단된다. 것으로 판단된다.여 황토 2g에 대하여 Hieltijes and Lijklema 방법에 의해 Adsorbed-p, Nonapatite inorganic-P(NAI-P), Apatite-p, Organic-P로 구분하여 분석하고, 총인(Total Phosphorus)을 Standard Methods에 따라 Persulfate digestion후 0.45 m membrane 여지 여과하여 여액에 대해 PO3-4-P의 농도를 Ascorbic Acid 법으로 측정한 결과, NAI-P가 가장 큰 비율을 차지하였고, 부원료로 첨가된 금속 양이온 중 Fe3-이온이 흡착에 기여하는 정도가 가장 큰 것으로 평가되었다.다당류 T-AS의 보체 활성화 기작은 classical과 alternative complement pathway의 양 경로를 통해 활성화 되었다. T-AS 분획은 mouse내의 특정 혈청단백을 증가시켰으며, 항체 생성능의 증가가 관찰되어 effect T 세포의 활성화가 나타나고 있음을 알 수 있었다. T-AS는 생체내 투여시에 대식세포의 탐식능이 증진되었으며, 대식세포 기능 저해제에 의한 대식세포의 기능 저해 현상이 회복되었다. 이와 같은 결과들로부터, T-AS의 항암 활성은 활성

Fabrication and Characterization of UV-curable Conductive Transparent Film with Polyaniline Nanofibers (폴리아닐린 나노섬유를 이용한 광경화형 전도성 투명필름의 제조 및 특성)

  • Conductive polyaniline (PANI) nanofibers in UV-curable resin were used for a transparent conductive film. The emeraldine-salt PANI (ES-PANI) nanofibers were prepared by chemical oxidation polymerization of aniline, which could be changed into emeraldine-base PANI by dedoping. EB-PANI nanofibers as a precursor for conductive fillers were thereby transformed into re-dpoed PANI (rES-PANI) by dodecylbenzenesulfonic acid in the UV-curable resin solution. rES-PANI nanofibers have high conductivity and long-term stability in the solution without a defect of nanostructure. The resulting conductive resin solution was proved to be highly stable where no precipitation of rES-PANI fillers was observed over a period of 3 months. The transparent film was spin-casted on a poly(methyl methacrylate) sheet of thickness ca. $5{\mu}m$. A surface resistance of $6.5{\times}10^8{\Omega}/sq$ and transmittance at 550 nm of 91.1% were obtained for the film prepared with a concentration of 1.4 wt% rES-PANI nanofibers in the solution. This transformation process of rES-PANI from ES-PANI by dedoping-redoping can be an alternative method for the preparation of an antistatic protection film with controllable surface resistance and optical transparencies with the PANI concentration in UV-curable solution.

  • As a second part of the study entitled "Field Measurement and Analysis of Fluvial Sediment in the Cheongmi-Strea", this study concerns mineralogical and contaminative characteristics of sediment particles collected at Wonbu-Gyo and Hanpyeong-Gyo in the Cheongmi-Stream. The mineralogical analysis of the bed sediment collected reveals that, in general , quarz is the most abundant mineral found in sands with feldspars and rock fragments in the next, which confirms what is known in the literature. The shape factor of sand particles is about 0.7, which also agrees to what is known in the foreign literature. The analysis also reveals that the clay particles collected are composed mainly of illite, kaolinate, and chlorite. The analysis of contamination reveals that there is no substantial difference between the contents of organic material and heavy metals in the surface water and those in the pore water beneath the river bed. It is because the sampling for this analysis was conducted right after the September flood during which the fresh top soil from the upstream watershed replaced the old bed sediment and pore water that would probably be more contaminated. The contents of geavy metals in the bed sediment of sand particles do not exceed those in the soils adjacent to the river. For fine sediment such as clays, however, the contents of heavy metals, especially of mercury and zinc, far exceed those in the soils adjacent to the river. These fine sediments are transported downstream in the form of wash load and deposited in part on flood plains, which could be a new source of contaminants.taminants.

  • This experiment has been carried out to study the changes of caseins is Gouda-type cheese during ripening by the use of DEAE-cellulose column and polyacrylamide gel electrophoresis. The results obtained were as follow: 1. The amount of water soluble nitrogenous compounds in the cheese ripened for 2 and 4 months was 19.03% and 30.61% for total nitrogen in the cheese, respectively. On the other hand, the amount of 5% NaCl soluble nitrogenous compounds in the cheese increased to 41.13% for 2 months ripening, but it decreased to 22.61% at 4th month. 2. By DEAE-cellulose column, various nitrogenous compounds of Gouda-type cheese were fractionated into 5 major and several minor peaks. The proportion of some fraction areas to the whole increased with the ripening period and the others decreased. 3. Electrophoretic patterns of various nitrogenous compounds in a 4-month-old cheese showed 5 and 8 bands, repectively. 4. Sixteen amino acids were identified in soluble and precipitable compounds at 12% TCA in a 4-month-old cheese.
