Ethnobotany of Hallyeo-Haesang National Park and Its Adjacent Regions, South Korea (한려해상국립공원 일대의 민속식물)

  • The ethnobotanical study was conducted to record and conserve the traditional knowledge of botanical taxa in Hallyeo-Haesang National Park area, S. Korea from 2012 to 2013. According to the survey results, derived from 1,430 sheets of 183 residents at 70 places, the ethnobotanical plants in Hallyeo-Haesang National Park area consisted of a total 247 taxa; 82 families, 197 genera, 224 species, 3 subspecies, 19 varieties and 1 form. The analysis of usage for 247 taxa showed that the edible use was the highest with 166 taxa, followed by medicinal with 138 taxa, craft with 27 taxa, material with 16 taxa and ritual with 9 taxa respectively. The leaf of plant was the most useful part, followed by stem and fruit plant. Quantitative analysis of the ethnobotanical plants in Hallyeo-Haesang National Park area was performed by the basic values of FC, NU and UR, and indices of CI, RFC, RI and CV. As a result, The CI places Artemisia princeps in the first position, followed by Pueraria lobata, Taraxacum platycarpum. The RFC place A. princeps in first position, followed by Achyranthes japonica, Aster scaber. The RI places A. princeps in the first position, followed by P. lobata, Pinus densiflora. The CV places A. princeps in the first position, followed by P. lobata, A. japonica. Farfugium japonicum had been used as specific ethnobotanical plant in this area, for arthralgia, postpartum arthrodynia, circulatory disturbance.

  • Alkali gelatinization of rice, corn, arrow root and ginger root starches at various sodium hydroxide concentrations was investigated. Critical concentrations of alkali for starch gelatinization ranged from 2.33 to 3.17 meq NaOH per gram of starch. Ginger root starch was most resistant to alkali gelatinization and arrow root starch was least stable to alkali.

Specific Marker Gene Analyses for DNA Polymorphism of the Blood Cell in Korea Native Brindled Cattle (칡한우 혈액에서 DNA 다양성 분석을 통한 표지 유전자 탐색)

  • This study was conducted to detect the specific expressing genes by using RAPD-PCR and RFLP method in the Korea Native Brindled Cattle, Korean Native cow and Holstein cattle. And then, the specific marker gene was investigated by the analysis of the genes for detection significance according to the expressing pattern. We found the specific expression gene by the RAPD-PCR analysis in Korea Native Brindled Cattle. It was detected the differences of the species in the colour and external section. The Korea Native Brindled Cattle were vary low compare to the Korean Native cow and Holstein cattle by analysis result of polymorphism and distribution. And there were a found the specific marker gene by sequencing in the R9B gene fragment of Korea Native Brindled Cattle. And the sequencing result of the R9B was different between Korean Native cow and Holstein cattle. Thus, this gene can be apply as the specific marker gene in the Korea Native Brindled Cattle.

혈액형지배 유전자에 의한 칡소의 유전적 특성

  • 혈액형을 지배하는 유전자는 진화에 대하여 중립적인 작용을 하고 있어서 집단의 유전적 구조의 특성 파악, 계통분류학 등에 많이 응용되고 있다. 본 연구는 칡소에 대한 유전학적 특성을 구명하고자 혈액형 분석기술을 응용하여 실시하였다. 공시동물은 (주)한경게놈텍 목장에서 사육중인 외모적으로 칡소의 특징을 보이는 25두를 이용하였다. 혈액은 경정맥에서 헤파린 처리된 진공 채혈관에 무균적으로 채취하여 혈장, 백혈구 및 적혈구로 원심분리한 후 냉동 혹은 냉장 보관하여 각 실험에 이용하였다. 적혈구 항원형의 검출은 2% 적혈구 부유액과 축산기술연구소에서 생산된 항혈청 11종을 이용하여 용혈반응으로 실시하였고, 혈액단백·효소를 지배하고 있는 6개의 유전자 좌위에 대하여 전분 혹은 포리아크릴 아미드겔 전기영동으로 다형 검출을 실시하였다. 용혈반응으로 검출한 적혈구 항원형의 반응양상은 검사한 11종의 항체에 대하여 6종은 50%이상의 개체에서 양성반응을 보였다. 이와 같은 결과는 일반 한우에서 보이는 양성반응율보다는 높은 것으로 판단되어진다. 전기영동법으로 분석한 6개의 혈액단백·효소 지배 유전자 좌위 중 ALB좌위을 제외한 5개 유전자 좌위에서 다형이 관찰되었다. HB, AMY-1, GC 및 PTF-2 유전자 좌위는 2개의 대립유전자가 관찰되었고, TF 유전자 좌위는 4개의 대립유전자가 관찰되었다. 표 1에서 같이 칡소에서 관찰된 각 유전자 좌위의 대립유전자 빈도의 구성은 일반적인 한우와는 상이한 결과를 보였으나 평균 이형접합도는 칡소가 0.438, 일반한우가 0.442로 계산되어 유전적 변이성은 유사한 것으로 추정되었다. 이상의 결과로 본 연구에서 분석한 칡소는 다른 한우집단과는 상이한 유전적 구조를 가지고 있으나, 유전적 다형성은 비교적 높은 것으로 시사되었다. 보다 정확하고 많은 량의 유전정보 수집을 위하여 Microsatellite DNA 및 모색 관련 유전자를 분석할 필요성이 있을 것으로 사료된다.(Table Omitted)

Effects of Pueraria flos and radix Water-extracts on levels of Several Serum Biomarkers in Ethanol-treated Rats (갈화와 갈근 열수추출물들이 에탄올 투여 흰쥐의 혈청성분에 미치는 영향)

  • The present study was investigated effect of each water extract from Pueraria flos (PF) and Pueraria radix (PR) on serum several biomarkers in ethanol-treated rats. Male Sprague-Dawley rats were randomly divided into six groups: Normal (None-treated group); Ethanol (only ethanol-treated group); EPF I (ethanol-treated, supplemented group); EPR (ethanol-treated, PF II-supplemented group); EPR I (ethanol-treated, PR I-supplemented group) ; EPR (ethanoltreated, PR II-supplemented grou). Five groups of male Sprague-Dawley rats were orally administered 25% ethanol (5 g/kg body weight/day) and sacrified 5 weeks post treatment. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and ${\gamma}-glutamyl$ transpeptidase activities were significiantly lowered by feeding of PF or PR than those of only ethanol-treated group. Whereas serum glucoseand liver glycogen contents were significantly decreased (p<0.05) by ethanol administration and increased decreased (p<0.05) by PF or PR supplement. This results indicate that Pueraria flos and radix water extract supplement improves alcoholic disorder.

The Properties of Plant Fiber and Polyester Blended Nonwoven Fabrics (식물성 섬유와 폴리에스테르 섬유의 혼합 부직포 제조 및 특성 -어저귀, 칡섬유를 중심으로-)

  • Nonwoven fabrics have been widely used in various fields that include household, industrial, agricultural, medical goods, especially in the automobile industry. In this study, eco-friendly fiber materials were developed and investigated as a substitute material for polyester fibers in nonwovens. To make plant fiber bundles, stems of Indian mallow (IM), and Kuzu vine (KV) were retted; in addition, the non-cellulose component was partially removed. Plant fiber bundles and polyester fibers (P) were blended and needle punched to produce nonwovens. Five kinds of nonwovens were manufactured: P100 (Polyester 100%), IM10 (IM 10% and Polyester 90%), IM20 (IM 20% and Polyester 80%), KV10 (KV 10% and Polyester 90%), and KV20 (KV 20% and Polyester 80%). The color values, surface appearance, tensile strength, elongation, tear strength, abrasion strength, flexstiffness, moisture regain, water or oil absorbency, and static electricity of manufactured nonwovens are investigated. As the blended ratios of IM or KV increased, the brightness of nonwovens decreased compared to that of polyester 100%. Tensile strength, tear strength, abrasion strength, and flexstiffness of IM10 as well as KV10 were higher than those of P100, IM20, and KV20, resulting from the influence of the structure and properties of nonwoven fibers. Moisture regain and water or oil absorbency increased, while static electricity decreased in proportion to the amount of plant fibers. IM or KV and polyester blended nonwovens showed improved properties over P100 that could be substituted for P100 as a novel material for textiles.

Effect of Electric Purse Conditions on the Fusion and Development Embryos Produced by Ear Cell Nuclear Transfer in Brindle Coated Hanwoo (Korean Cattle) (칡소의 귀세포를 이용한 핵이식에서 전기융합조건이 융합 및 배발달에 미치는 영향)

  • This study was conducted to investigate the effects of embryo development by fusion condition on the nuclear transfer with brindle coated cow's ear cells. Ear cells were transferred into an enucleated oocyte and fused with cytoplasm in the fusion condition with 1.9kv/cm, 2.0kv/cm, 2.1kv/cm each 10 and 20ug duration Nuclear transfer embryo were activeted with a combination of 5ug/ml and 1.9mM 6-DMAP (4min, 4h). Fusion rate was 51∼68% range among fusion condition (1.9, 2.0, 2.1kv/cm; 10, 20us). But, cytoplasm lysis rate was increased by higher electric condition (0∼51.8% range). Each parameter's cleavage and blastocyst formation rate were 1.9kv/cm for 10us (75.8 and 19.5%), 20us (69.8 and 48.6%), 2.0kv/cm for 10us (76.9 and 20.0%), 20 us (68.5 and 40.9%), 2.1kv/cm for 10us (70.5 and 44.2%), 20 us (68.5 and 27.0%). We compared the effectiveness of cloning for between brindle coated cow's ear cells and Hanwoo fetal fibroblast cells. There was no significant differences in the fusion rate and developmental rate to the blastocyst stage. After transfer of blastocysts derived from nuclear transfer embryos, pregnancy rates of the Hanwoo fetal fibroblast cells and brindle coated cow's ear cells were checked pregnant on day 60 as assessed by ultrasonography, 40% (2/5) and 15.8% (3/19), respectively. This studies conclude that brindle coated cow's ear cells have the developmental potentiality to term by nuclear transfer. These results demonstrate that the increased the field strength was to be profitable for development of blastocyst or reduce of cytoplasm's damage than increasing the pulse duration.
