출토 인골의 유전자분석-나주 복암리 3호분 옹관 인골을 중심으로 (The genetic analysis on ancient human bone in jar coffin excavated from Naju Bokam-ni 3rd tumulus)

  • We have analyzed the allele and genotype frequencies from 10 fractions of ancient human skeleton in 3 pieces of Jar coffin excavated from Naju Bokamni3rd tumulus by PCR amplification, high resolution polyacrylamide gelelectorphoresis and silver staining. We could isolate human genomic DNA from 3 bone fractions but the rest of them could not be used as materials due to being decayed. We could detect sex determination as male and 3 genotypes of STR system, HUMTHO1, HUMTPOX and HUMC5F1PO from the bone fraction of left side in Jar coffin 3 and see the slightly reaction suggesting the sex as male from the bone fraction of the left side in Jar coffin 2 and female from the right side in Jar coffin 3.We have also analyzed the genotype frequencies of mitochondria from the bone fractions of the left side and the right side in jar coffin 3, respectively. From the result of indetifiying at nucletide position between 16018 and 16378of the base of hyper variable region(HV1) in the control region, We can presume that the both bones have the same maternal inheritance.

경산 임당동 및 사천 늑도 출토 인골의 유전자 분석 (Genetic Analysis of Ancient Human Bones Excavated in Sacheon Nuk-do and Gyeongsan Yimdang-dong, Korea)

  • We investigated the nucleotide substitution and insertion polymorphism of the hypervariable region Ⅰ and Ⅱ in mt DNA by sequencing ancient DNA from 51 ancient bones and teeth excavated at Nuk-do and Yimdang-dong in Korea. It revealed 35 sequence types from the ancient Korean. Of these, different sequences were 34 sequences. There were 19 and 38 base substitutions in HVI and HVⅡ, respectively. Some substitutions were characteristic of East Asian populations as compared with data reported on Caucacianpopulations,16051, 16150, 16172, 16223 in region I and 73, 263 in region II were noted as polymorphic sites, respectively. These were distributed evenly along the control region, though the frequency of each site was variable. Nucleotide substitution rather than insertion and deletion was the prevalent pattern of variation. Insertion of cytosine between312 and 315 in region HVⅡ were detected up to 98% in 51 ancient bone samples. This sequence data represents a phylogenetic tree using NTI DNA Suite computer program. The phylogenetic tree showed that mt DNA sequences of Nuk-do bones were relative to west Siberian and Indonesian. The usefulness of mt DNA sequencing in ancient Korean population excavated atarchaeological sites is based on biological and historical evidence for origin and migration of ancient Korean.

조선 시대 인골로부터 분리한 미생물의 유전학적 특성연구 - 김포 장기지구 토광묘 출토 인골을 중심으로 (Genetic Characterization of microorganism from Human Remains in the Joseon Period)

  • Preservation of artifacts that are excavated from archeological sites is closely related to soil environment. Biological remains are especially influenced by degradation activity of microorganism from soil environment. In this study a preserved human bone in archaeological tomb, Tou-kwang-myo from Joseon Period was analyzed to characterize bacteria groups by molecular genetic tools using 16S rDNA sequences. 117 clones were identified and classified 9 phylogenetic groups : ${\alpha}$-, ${\beta}$-, ${\gamma}$-, ${\delta}$-Proteobacteria, Sphingobacteria, Clostridia, Actinobacteridae, Nitrospiraceae, and Gemmatimonadetes according to homologous 16S rDNA sequences submitted in NCBI. ${\gamma}$-Proteobacteria group appears the highest ratio in bones (about 35%) while about 19.6% belong to the Actinobacteria group. The results may contribute to study on the effect of microorganisms on the human remains with burial method.

  • In this study human bones and teeth, excavated from the Myeongam-risite in Asan, Chungcheongnam-do Province, have been analysed by nuclear DNA typing and mitochondrial DNA sequencing methods. Twenty-one samples of long bones and twenty-seven samples of teeth from twenty-one individuals were collected and analysed. Among these thirteenteeth were successfully subjected to nuclear DNA extraction, quantification, and PCR(Polymerase Chain Reaction) amplification. Silver STR III (D16S539, D7S820, D13S317) multiplex PCR method was used in this study for a short tandem repeat (STR) analysis. Mitochondrial DNAs of tooth samples were also amplified and sequenced by a DNA sequencer. These analyses show that a sample from Burial no. 29 and one from Burial no. 38(right) possessed the same maternal inheritance. This may suggest that the Myeongam-ri cemetery was used by a kin group for a relatively long period of time.
