Effect of medium pH on the extracellular production of red pigments using Monascus purpureus (Monascus purpureus 에 의한 세포외 적색색소 생산성 증가에 대한 배지내 pH 조절의 영향)

  • The Effect of pH red pigment production by Monascus purpureus ATCC 16365 has been studied in pH-controlled batch fermenter culture. A maximum of yellow and red pigments were detected using UV-Vis spectrophotometer at 385nm and 495nm, respectively. Fungal growth and pigment production were favoured at low pH(pH 4.0-5.5). Especially extracellular formation rate of orange to yellow pigment was decreased compared with that of orange to red pigment at pH 7.0. In addition, the enhancement of ratio of extracellular to intracellular pigment and the red pigment production in pH 7.0-controlled batch fermenter was observed. However, the pH 7.0-controlled batch cultures depressed the total production of pigments. The pH change from 4.0 to 7.0 during batch fermenter cultivations sharply increased both red pigment production and the extracellular composition.

  • This study describes extinguishment mechanism of the purely buoyant diffusion flame using the water spray. Experiments are systematically carried out for the oul pool fire with the six different atomizing nozzles. From the measurement of burning rate which represents the combustion intensity of fire, it is observed that the water spray is able to act to enhance fire rather than to extinguish fire. The air entertainment due to the water spray is visualized to understand this phenomenon, acting to enhance fire. In order to observe effects of droplet size on fire extinguishment, and amount of water which reaches the flame base, fuel surface, and mean diameter of droplets are measured. When water droplets are too small, they do not reach the flame base because they can the water spray having too small doplets is ineffective for extinguishment of the oil fire.

  • The effects of temperature and of $Na_2O$ and $SiO_2$ contents on the crystallization of zeolite A were studied, by examining crystallization curves and particle size distributions of final products at various crystallization conditions. Crystallization process could be simulated adopting the assumptions of constant linear growth rate and equilibrium between amorphous solid phase and soluble species. Rate constants were determined by comparing the simulated crystallization curves with experimental data. Rate constant for linear growth increased with temperature and crystallization rate at different mole ratio of $Na_2O/H_2O$ correlated reasonably well with increase of soluble species. The rate constant of crystallization did not increase with increase in mole ratio of $Na_2O/H_2O$, but the rate of nuclei formation and the fraction of soluble species were enhanced. The rate constants for linear growth of zeolite A were determined as $0.07{\sim}0.24{\mu}m{\cdot}min^{-1}$ at these experimental conditions Apparent activation energy was estimated as $49kJ{\cdot}mol^{-1}$.

Confirmation of Non-Siderophore Antifugal Substance and Cellulase from Bacillus lichemiformis Kll Containing Antagonistic Ability and Plant Growth Promoting Activity (생물방제능과 식물성장촉진능을 동시에 가지는 Bacillus licheniformis K11의 non-siderophore 항진균 물질 및 cellulase의 생산조건 확인)

  • Bacillus lichemiformis Kll, a plant growth promoting rhizobacterium was reported as a producer of auxin, siderophore, as well as antifungal cellulase under some culture conditions. In vitro test, B. licheniformis Kll represented excellent antagonistic ability against Fusarium oxyspoum (KACC 40037), and showed broad spectrum against other phytopathogenic fungi. B. licheniformis Kll had cellulolytic activity toward not only carboxymethyl-cellulose (CMC) but also insoluble cellulose, such as fungal cell wall cellulose, filter paper (Whatman No. 1), and Avicel. In addition, we confirmed antifungal substance production by butanol-extract methods. The strain produced optimally the antifungal substance when it was cultivated at pH 9.0, 30${\circ}$C for 4 days on nutrient medium. The biological control mechanisms of B. lichemiformis Kll were caused by antifungal substance, cellulase and siderophore against phytopathogenic fungi.

Degradation of Amaranth by Microorganisms (미생물(微生物)에 의한 Amaranth의 분해(分解))

  • Fundamental study was carried out to elucidate the mechanisms of biological degradation of dyestuff in environments. A few bacterial strains which were capable of degrading amarnath were obtained from soil through an extensive screening program and identified by microbiolological properties. Conditions for bacterial growth and amaranth degradation were characterized and optimized, and the degradation products were identified. The results were as follows. 1. The most active strain A12-1 to be capable of degradation of amaranth was identified as Pseudomonas sp. 2. Optimal conditions for growth of the strain A12-1 were:$35^{\circ}C$ and pH 7.5, and growth was markedly increaesd by aeration. 3. Degradation of amaranth by the strain was accessed under similiar conditions for growth, however significantly inhibited when the culture was aerated. 4. Both bacterial growth and amaranth degradation were gradually decreased with increased concentration of amaranth in the culture. 5. Reaction of the crude enzyme from the strain A12-1 was optimal at $35^{\circ}C$ and pH 7.5 for degrading amaranth. 6. Sodium naphthionate and R-amino salt were found to be the products of amaranth degradation by the strain A12-1.

  • This study tried to analyze the factors of corporate growth potential of CJ E&M, a representative company in the content industry. First, in order to analyze CJ E&M's growth Engine, Michael Porter's diamond model was used to review key factors, and then, based on the 4P model, directions for future corporate growth were suggested. As a Result, Factors behind corporate growth included "Media content of various genres" and "recruitment of star-class human resources", in terms of Production Conditions, and "Gratification Chinese market demand" in terms of Demand Conditions. In addition, "Korean wave industry aiming at K-Culture" in terms of Related Industry and "Two track strategy: global·glocal strategy" and "Media commerce strategy" in terms of business environment: strategy, structure, and competition was able to analyze. For the direction of development, there are "various products through collaboration of affiliated companies" in terms of Product and "TVING Benchmarked Netflix" in terms of Price. In addition, "global expansion through OTT platform TVING" in terms of place and "challenge marketing utilizing the Tiktok platform" in terms of promotion must be carried out.

A Study on the Strength Properties of Mortar Under Various Types and Contents of Accelerators for Freezing Resistance (내한촉진제의 종류 및 혼입양 변화에 따른 모르터의 강도특성에 관한 연구)

  • When fresh concrete is exposed to low temperature, the concrete may suffer frost damage due to freezing at early ages and strength development may be delayed. One of the solution methods for resolve these problems is to reduce freezing temperature of concrete by the use of chemical admixture called Accelerators for freezing resistance. In this study, we investigate the effect on strength development of cement mortar using accelerators for freezing resistance with the variance curing condition. As the result of this study, the mortar using accelerators for freezing resistance show that continuously strength development in curing condition of -5$^{\circ}C$. And compressive strength under the variance temperature condition was higher than constant temperature condition in same maturity.

토끼의 수종 점막을 통한 [D-알라$^2$-메치오닌엔케팔린아미드의 투과 증진

  • 효소억제제와 투과촉진제를 첨가하지 않은 경우 비강, 직장 및 질점막을 통한 YAGH니 투과정도를 검토한 결과 donor측 잔존률이 경시적으로 급격히 감소하여 24시간후각각 7.04, 4.21 및 51.34% 잔존하였다. 또한 receptor측으로의 투과량는 비강점막의 경우 8시간대에 3.9%의 투과율을 기점으로 다시 분해되었으며 직장 점막에서는 최대 0.72% 로 거의 검출되지 않았다. 질점막의 경우에는 24시간대에 1.8%의 투과에 머물렀다. 한편 혼합효소억제제로 TM가 EDTA를 양측 셀에 첨가한 조건에서는 첨가되지 않은 경우와 비교하여 donor측에서의 YAGFM이 서서히 감소됨과 동시에 receptor cell로의 투과량이 다소증대되었다. 더욱 양측 셀에 혼합효소억제제를 넣고 donor cell에 투과촉진제를 첨가한 경우 점막투과성을 현저히 증대시켰으며 LPC를 사용한 경우 효소억제제만을 사용한 경우와 비교하여 비강에서 3.6배, 직장에서 3.2배 및 4.1배 투과 flux를 증가시켰다.

  • The buildings constructed with steel structure is coated with certified fire resistive material to resist from fire. Coating materials lose their initial performances as time passes, so they need some maintenance. This study is covers standardization of the methods for assessing the durability of SFRM. In Korea, more than 90 percent of SFRM are used indoors. So This study is to decide proper test method through investigation and examination of effect factor to performance of fire-resistant structure and applicable test method.

난치성 질환의 병태모델실험

  • 산소라디칼-질병모델을 보다 간편한 방법으로 만들어 생화학적 발병 기전을 추구하므로서 신약창출에 필요한 기초자료를 제시하고자 본 실험을 실시하였다. 미세혈관의 혈류이상은 과잉의 활성산소를 생산하게되며 이로 인해 여러 종류의 난치성 질환 및 염증을 수반하는 많은 질병이 발병하게 된다. 일반적으로 흔히 이용되어지는 허혈-재관류모델이 아닌 환경독성물질을 활용한 병태모델을 만들어 이 모델조건에서 활성산소생성과 밀접한 관계를 갖는 잔틴 옥시데이즈(Xanthine Oxidase)의 변화와 과산화지질생성정도를 연관지어 검토하였다. 수은, 구리, 몰리브덴등의 금속이온들은 잔틴 디하이드로저네이즈(Xanthine dehydrogenase)로 부터 옥시데이즈(Oxidase)형으로의 전환을 촉진시켰으며 막독성의 지표로 이용되어지는 과산화지질의 생성도 현저히 증가시켰다. 또한 알데하이드류의 첨가실험에서도 잔틴산화효소의 형전환이 촉진되었으며 첨가한 알데하이드의 탄소수와 수소수 그리고 탄소와 탄소의 결합상태와도 밀접한 관계가 있음이 관찰되었다.
