Effects of Concentrations of Glucose and Maltose on the Growth of Bacillus amyloliquefaciens (B. amyloliquefaciens 세포 성장에 미치는 포도당과 맥아당 농도의 영향에 관한 연구)

  • Cell growth and production of ${\alpha}$-amylase, acetic acid and lactic acid were investigated in Bacillus amyloliquefaciens(ATCC 23350) flask culture with various carbon sources. Maximum dry cell density increased with increase in initial maltose concentration. Maximum dry cell density was the highest(1.4g/$\ell$) at 10g/$\ell$ of initial glucose concentration. With 10g/$\ell$ of initial glucose concentration, maximum specific cell growth rate was obtained. Among the various carbon sources maximum ${\alpha}$-amylase production was obtained with 149 unit/ml at 20g/$\ell$ of initial maltose concentration. With 5g/$\ell$ of initial maltose concentration, maximum ${\alpha}$-amylase production rate was obtained. By increasing carbon source concentration, acetic acid formation decreased. Acetic acid formation was higher in glucose than in maltose. By increasing carbon source concentration, lactic acid formation increased. Lactic acid formation was higher in maltose than in glucose.

  • 다공성 물질이 충전된 밀폐용기 내에서 수용성 이원혼합용액($H_2O{+}NaCl$)이 수평한 상부전열면으로 부터 동결될 때 혼합용액의 초기농도, 액체의 과열 및 다공성물질의 입자직경 크기가 온도와 농도분포에 미치는 영향을 실험하였으며, 동결이 진행됨에 따라 이동하는 고액상 혼합영역의 계명위치를 측정하였다. 다공성물질은 평균직경이 2.85mm, 6mm인 구형의 유리구슬을 이용하였다. 수용성 혼합용액의 초기농도는 공융농도도 이하로 하였으며 상부 전열면은 공융온도 이하로, 하부전열면은 액상선온도 이상으로 유지하여 동결 실험한 결과 상부 전열면으로 부터 고체 영역, 고액상혼합영역, 액체영역으로 구분되었다. 액체의 초기농도가 5%인 경우 과냉현상이 관찰되었으나 10%, 15%인 경우 액체온도는 액상선 온도보다 더 높았다. 용액의 초기농도를 감소시킬수록 고체와 고액상혼합영역의 범위는 증대되었으며 고액상혼합영역과 고체영역의 계면은 더욱 강해진 자연대류에 의하여 2차원성이 증가된 형상을 보였다. 용액의 자연대류는 다공성물질의 직경이 클수록 증가되었으며 계면에서의 제융해현상은 관찰되지 않았다.

  • This study was carried out to evaluate the effects of initial concentration and moisture content on removal of phenol in soil. Texture of soil used was sandy loam. Air was supplied at the rate of $31{\ell}/m^3$(soil)/min. Initial phenol concentrations of contaminated-soils were about 700mg/kg and 1,200mg/kg(as dry weight basis), respectively. Low phenol concentration (about 700mg/kg) was degraded more rapidly than high concentration (about 1,200mg/kg). After phenol concentration of 1,200mg/kg was decreased to about 700mg/kg, its degradation was similar to the case of initial phenol concentration of about 700mg/kg. Phenol degradationrate rate of 15% soil moisture content was higher than that of 20%.

  • The growth of recombinant E. coli and formation of the by-products were investigated. Glucose was consumed in approximately 8hours of cultivation when initial glucose concentration was 5g/1. When higher initial glucose concentrations were employed, it took approximately 16 to 20hours for the glucose to be consumed. When maltose or glucose was used as a carbon source, lactic acid and acetic acid were main by-products, while propionic acid and methanol production was not significant. Much by- products were formed at the higher initial carbon source concentrations. Higher concentration of lactic acid was observed when glucose was used, while higher concentration of acetic acid was observed when maltose was used. Lactic acid production affected the cell yield while acetic acid production affected the growth rate.

  • 비점오염원은 강우시 토지이용형태에 따라 유출특성이 다양하게 나타나며, 특히 강우 초기에 고농도의 유출수가 수계로 유입하게 된다. 또한 강우량, 강우강도, 선행건기일수 등은 비점오염물질의 농도에 영향을 미치며, 특히 선행건기일수가 길고 강우강도가 강한 강우사상일 경우 고농도의 오염물질이 배출되는 것으로 나타나고 있다(안태웅 등, 2012). 이에 본 연구에서는 3년간(2010년 ~ 2012년)의 모니터링을 통해 고랭지 밭에서 발생하는 비점오염물질의 초기세척효과를 분석하고자 하였다. 초기세척효과의 판단은 Geiger(1987)이 제시한 누적오염부하량과 누적유출량과의 그래프를 비교하여 기울기가 45도 기울기보다 상향하여 분포하였을 경우 양분선과 최대로 벌어진 부분의 시간까지에서 초기세척효과가 발생한다고 판단하였다. 연구결과, 고랭지 밭 지역의 경우 뚜렷한 초기세척효과 현상이 나타나지 않았다. 하지만 CODCr의 경우 3차 강우사상에서 초기세척 효과가 나타난 것으로 분석되었다. 3차 강우사상은 선행건기일수가 0.9일로 매우 짧고 발생강우량이 27.3 mm로 연구기간 동안 가장 적은 강우량이 관측되었지만, 유출발생시점 전후 3시간 동안 강우강도가 4.0 ~ 6.2 mm/hr로 높게 나타났기 때문에 유출초기에 고농도의 오염물질이 발생된 것으로 판단된다. 그동안 선행된 초기세척효과에 대한 연구를 살펴본 결과 이병수 등(2008)은 경안천 유역 농업지역에 대해 초기세척효과를 분석한 결과 일부 강우에서는 초기세척현상이 나타나지만 대부분의 강우에서는 초기세척현상이 나타나지 않아 농촌지역의 경우 초기세척현상의 접근방법은 적절치 않다고 보고하고 있다. 정성민 등(2009)은 고랭지 농업지역에서 SS와 TP는 뚜렷한 초기세척현상이 나타나는 반면, TN과 DOC는 경향을 보이지 않는다고 하였다. 하지만 윤영삼 등(2011)은 강우시 포도밭에서 초기세척효과를 분석한 결과 모든 수질항목에서 약한 초기세척효과가 나타났고 특히 SS의 경우 초기세척효과가 강하게 나타났다고 보고하고 있다. 이처럼 투수지역에서의 초기세척효과에 대한 판단은 연구자에 따라 차이를 보이는데 이러한 이유는 투수지역의 토양의 특성, 강우조건, 지형적 특징 등이 영향을 미치는 것으로 판단된다.

Improvement of Alcohol Productivity by Means of Repeated Batch Fermentation (반복적인 회분식 발효공정을 이용한 에탄올 생산성의 향상)

  • The functional relationship between the initial cell concentration and the ethanol productivity was investigated in the repeated batch fermentation of Sacharomyces cerevisiae ATCC 24858. The repeated batch fermentations were performed in the range of 60 to $150g/\ell$ of initial sugar concentration and 17.5g/$\ell$ to $53.1g/\ell$ of initial cell concentration. The time of one batch fermentation was 1 or 2 hours and the batch fermentation was repeated ten times in every repeated formentaction. The functional relationship showed that the productivity increased non-linearly according to the increase of initial cell concentration regardless of initial sugar concentration. When the initial concentration of sugar was $60g/\ell$ and that of biomass was $34.5g/\ell$, the fermentation was completed within one hour and its ethanol productivity was $26.7g/\ell$.hr, the latter including the times of cell separation, pouring the new substrate into a flask and sampling. When the initial sugar concentration was $120g/\ell$ and the initial cell concentration $50.3g/\ell$, the fermentation was also finished within one hour and its productivity was $45.8g/\ell$$.$hr, The maximum ethanol productivity for eight different repealed fermentations in this work was $53g/\ell$.hr.

A Study on the Degradation Characteristics of 1,4-dioxane at Different Initial $H_2O_2$ Concentration with Advanced Oxidation Process using Ozone and Hydrogen Peroxide ($O_3/H_2O_2$를 이용한 고급산화공정에서 초기 $H_2O_2$ 농도에 따른 1,4-dioxane의 제거 특성 연구)

  • Advanced oxidation process involving $O_3/H_2O_2$ was used to eliminate 1,4-dioxane and to enhance the biodegradability of dioxane-contaminated water. Oxidation process was carried out in a bubble column reactor under different pH and $H_2O_2$ concentrations. The removal efficiencies of 1,4-dioxane were investigated at hydrogen peroxide concentration between 40 and 120 mg/L. At the same pH, removal efficiencies of 1,4-dioxane increased with increasing initial $H_2O_2$ concentration. There was a linear relationship between initial concentration of $H_2O_2$ and the amount of consumed $O_3$. It was observed that the high $H_2O_2$ concentration accelerated the generation of hydroperoxy ions(${HO_2}^-$) and hydroxyl radicals($OH{\cdot}$). Hydrogen peroxide enhanced the decomposition of 1,4-dioxane and the biodegradability of the solution.

The effect of gap between ingot and crucible on the distribution of initial melt concentration in Bridgman crystal growth (Bridgman 결정성장시 장입 주괴와 도가니 사이의 틈이 용액이 초기농도에 미치는 영향)

  • Dilute Al-Cu and Al-Mg alloys were solidified unidirectionally upward by Bridgman method. It is necessary that solute concentration of initial melt is uniform to be able to control the concentration of crystal. When solute concentration is not uniform, it can cause unusual macro-segregation in grown solid. A non-steady state solidification was observed where the solute concentration in the grown solid decreased with the progress of solidification, when a dilute Al-Cu melt with positive axial temeprature gradient was solidified. This was caused by leaking out of Cu-rich melt into the gap between ingot and crucible during melt-down and its sedimentation after complete melting. In the case of Al-Mg alloy, the solute concentration has a minimum in the middle of grown specimen because Mg-rich melt flowed down the gap between ingot and crucible and floated after complete melting. Uniform initial melt concentration can be achieved by the homogenization of the ingot or by the absence of the gap between ingot and crucible.

Extractive Ethanol Fermentation Characteristics of K.fragilis in an Aqueous Two Phase System (수성이상계에서 K.fragilis의 에탄올 추출발효 특성에 관한 연구)

  • Fermentation characteristics of Kluyveromyces fragilis CBS 1555 with Jerusalem artichoke juice, in extractive ethanol fermentation in aqueous two phase systems composed of polyethylene glycol 20000 (PEG) and crude dextran(Dx), were investigated as a function of initial sugar concentrations, concentrations of ethanol formed, or fermentation time. Both specific ethanol production rate increased with decrease in concentrations of PEG and Dx in two-phase systems. Without being related to the compositions of aqueous two-phase system, maximum specific cell growth rate and maximum specific ethanol production rate were showed in the initial sugar concentration fo $80g/\ell$ and $120g/\ell$, respectively. The inhibition effects of ethanol on specific cell growth rate and specific ethanol production rate decreased with decrease in PEG concentration and in the range of 2.5 to 5% Dx. Specific cell growth rate and specific ethanol production rate was fitted as an exponential function and a hyperbolic function, respectively, of the concentrations of ethanol formed. Overall ethanol productivity increased with increase in initial sugar concentrations, and also the required time for the maximum productivity was so. Ethanol production rate by the elapsed fermentation time showed the maximum value in the initial sugar concentration of $160g/\ell$.

Effects of Inoculum Density and Basal Media on Cell Growth and Taxol Production in Taxus Cell Suspension Cultures (주목 세포배양에서 초기 접종농도와 기본배지가 세포증식과 Taxol 생산에 미치는 영향)

  • Optimum inoculum concentration for the production of taxol was determined in Taxus brevifolia and Taxus cuspidata cell suspension cultures. By fresh weight, 2.5, 5, 7.5, 10 g/flask of cells were inoculated and cell growth as well as taxol production were examined. In both Taxus cell cultures, the higher the inoculum concentration, the shorter the length of the lag period. The optimum inoculum concentration for taxol production was found to be 5 g/flask. To produce taxol in large quantity, utilization of proper medium was thought to be important. In case of using a production medium with 6% sucrose, taxol production was noticed. Its level reached the maximum at the 9th day of culture and decreased afterwards. However, taxol was not detected from cell cultures in growth medium.
