A Suppressive Component on Phagocytosis of Murine Peritoneal Macrophage in Aurantii immaturi pericarpium (청피에 함유된 복강 마크로파지의 탐식작용 억제 성분)

  • The phagocytic activity of murine peritoneal macrophage, was determined by lucigenin chemiluminescence and engulfment of fluorescein-conjugated E. coli particle. The acti vity-guided fractionation upon the methylenechloride fraction of Aurantii immaturi pericarpium led to the isolation of a flavonoid, isosinensetin, as a suppressive component of phagocytosis. Isosinensetin suppressed the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles and enhanced the production of nitric oxide in murine peritoneal macrophage.

Yields and Fruit Quality of Different Aged Walnut Trees (Juglans sinensis Dode) Derived from Seedlings in Gimcheon (김천지역 실생묘 유래 호두나무의 수령별 수확량 및 과실 품질)

  • This study was conducted on cultivation farms in the Gimcheon area, which is the main production area of walnuts, in order to compare and analyze the yield and fruit quality of walnut trees (Juglans sinensis Dode) derived from seedlings. The official trees were classified into nine classes from 10 to 60 years old, and a total of 135 trees were selected, with 15 trees selected for each grade. The growth and fruit characteristics for each age group were then examined. The average tree height was 10.3 m, and the average crown area was 63.7 ㎡. The average number of fruit settings per tree was 573.1, with the largest number of walnuts per tree at 35 years (974.0 walnuts), and the lowest walnuts per tree at 10 years (124.7 walnuts). In addition, the number of fruit settings per bearing mother branch in 35 year old trees was the highest at 3.1. The average weight for each fruit type was found to be 58.7 ± 4.0 g of walnuts in green peel, 10.5 ± 0.5 g of dried shell walnuts, and 4.9 ± 0.2 g of walnuts, respectively. The average yield by fruit type was 32.5 ± 13.8 kg of walnuts in green peel, 5.9 ± 2.5 kg of dried shell walnut, and 2.8 ± 1.2 kg of walnuts. The yield was positively correlated with the crown area, the number of fruits setting per bearing mother branch, and the number of fruits setting per 1 ㎥. The formula used to estimate the yield per tree by age was calculated as y = 8993.9 ln (tree age-9) 8230.3, R2 = 0.8531 (walnut in green peel) and y = 1647.1 ln (tree age-9) 1442.1, R2 = 0.8527 (dried shell walnut).

  • Purpose: Livedo vasculitis is recurrent painful ulceration of the feet, ankles and legs characterized by purpuric papules and plaques that undergo superficial necrosis and healing with residual white atrophic scars (atrophie blanche). The typical histopathologic findings of livedo vasculitis are characterized by endothelial proliferation and hyaline degeneration along with thrombosis of dermal vessels. Standard therapeutic strategies for treatment of livedo vasculitis are usually on the basis of rheologic, anti-inflammatory or immnosuppressive treatments, a aspirin, dipyridamole, glucocorticosteroids, pentoxyfylline, or high-dose intravenous immunoglobulin are often ineffective or partially effective. Methods: We report a case of 24-year-old male patient with livedo vasculitis on the ankles and dorsal surfaces of both feet. Results: The lesion that had been unresponsive to medical treatment were successfully healed with complete debridement and skin grafting without recurrences. Conclusion: Surgical treatment can be one of the therapeutic choice in Livedo vasculitis.

  • Citri Reticulatae Viride Pericarpium(CRVP) is being used to regulate the flow of Q(氣) However, the mechanism of it's pharmacological actions is not well understood. The purpose of this research was to investigate effects of CRVP contractil response of isolated on abdominal and femoral artery in rabbits and renal artery in pigs. 1. Abdominal artery was relaxed by CRVP in a dose-dependent manner. 2. Femoral artery was relaxed by CRVP in a dose-dependent manner 3. Pretreatment with methylene blue and indomethacin did not inhibited CRVP induced relax in abdominal artery and femoral artery. 4. Renal artery was relaxed by $H_{2}O$ fraction in a dose-dependent manner, 5. Pretreatment with regitine inhibited $H_{2}O$ fraction(CRVP) induced relax in renal artery in a dose-dependent manner. 6. Renal artery was not relaxed by hexane fraction(CRVP) in a dose-dependent manner. 7. Pretreatment with regitine$(10^{-7}M)$ was relaxed by methylene chloride(MC)(CRVP) and $H_{2}O$ fraction in a $0.5{\mu}g/m{\ell}$.

Fresh-cut사과 전처리에 따른 물성학적 품질 특성

  • 편의와 fresh-like quality를 선호하는 소비 경향에 맞추어 가공한 fresh-cut사과의 선도 연장을 위한 저해제를 탐색하였다. 실험에 사용된 사과는 박피하여 속심을 제거한 사과를 8조각으로 절단하여 저해제에 3분간 침지한 후 물기를 제거하고 각각 LDPE 포장지에 포장하여 실온에서 저장하면서 colorimeter와 rheometer를 이용하여 색도와 물성을 측정하였다. 저해제로는 1% ascorbic acid, citric acid, NaCl, MgCl$_2$, CaCl$_2$, KCl, CaSO$_4$, MgSO$_4$, $Na_2$SO$_4$, oxalic acid, calcium lactate, calcium citrate를 사용하였으며 천연저해제로는 건칡, 맥문동, 목향, Rhubarb, 당귀, 오약, 시체, 지각, 천문동, 청피, 계피, 구기자, 당귀, 진피, 감초, 녹차, 후추 등을 열수추출하여 사용하였다. 물리적인 변화는 시간이 경과함에 따라 감소하는 추세를 보였으며 일부 건조로 인해 texture의 증가를 나타내기도 했다. 처리에 따른 물리적인 변화로는 CaCl$_2$, NaSO$_4$, Cl, 시금치를 처리하였을 때 그 변화가 가장 작았으나 시금치의 경우 시금치로 인한 사과의 색변화로 관능적으로 좋지 않았다. 한약재는 목향, 당귀, 지각, 계피를 처리하였을 때 물리적인 변화가 가장 작았으나 시금치와 마찬가지로 처리 초기에 한약재 자체의 향과 색으로 인하여 관능적으로 좋지 못한 결과를 나타냈다. 갈변 정도를 나타내는 ΔE의 변화는 KCl과 CaCl$_2$, CaSO$_4$를 처리한 구에서 변화가 가장 작았으며 한약재를 처리하였을 경우 당귀, 천문동을 처리한 구에서 효과를 보였다. oxalic acid는 갈변저해에는 효과적이었으나 시간이 경과함에 따라 수분의 용출이 증가하였으며 물성의 변화도 컸다. 또한 ZnCl$_2$를 처리한 구에서는 이취 발생과 수분 용출이 컸으며 색의 변화도 컸다.

Antidepressant Effects of Citri Reticulatae Viride Pericarpium in the Forced Swimming Test (청피(靑皮)가 강제수영부하에 따른 우울행동에 미치는 영향)

  • Objectives: Investigation of the antidepressant effects of Citri Reticulatae Viride Pericarpium(CR) Methods: In order to investigate the antidepressant effects of Citri Reticulatae Viride Pericarpium(CR), we performed the forced swimming test. Also the expression of CRF, HSP70 and c-fos was measured with immunohistochemical method at PVN. Results: 1. The duration of immobility in the forced swimming test was significantly decreased in the CR 100mg/kg, 400mg/kg groups. 2. In the Control group, CRF expression was significantly increased in the PVN. Also, these CRF increase were significantly reduced in the CR 100mg/kg and 400mg/kg treated group. 3. HSP70 expression was significantly decreased at PVN in the CR 100mg/kg and 400mg/kg treated group. 4. C-fos expression was significantly decreased at PVN in the CR 100mg/kg and 400mg/kg treated group. Conclusions: According to the results, it can be considered that Citri Reticulatae Viride Pericarpium has antidepressant effect.

Effects of Citri Reticulatae Viride Pericarpium on 4-Hydroxynonenal-Induced Inflammation in PC12 Cells (청피의 항염증효과)

  • Objectives: The purpose of this study was to observe the effects of Citri Reticulatae Viride Pericarpium (CP) on 4-Hydroxynonenal (4-HNE)-induced inflammation in PC12 cells. Methods: 4-HNE was treated in PC12 cell to cause inflammatory response, and then treated with CP water extract at 25, 50, and $100{\mu}g/ml$. The phosphorylation of Jun N-terminal kinase (JNK) and the expression of $NF-{\kappa}B$ in PC12 cells were determined by Western blot, respectively. Results: The phosphorylation of JNK was significantly decreased in 4-HNE-stimulated PC12 cell by the treatment of CP extract at $25{\mu}g/ml$. The 4-HNE-induced expression of nuclear factor kappa-light-chain-enhancer of activated B cells ($NF-{\kappa}B$) p65 in nuclear of the cells was significantly decreased in PC12 cell by treatment with CP extract at 25, 50, and $100{\mu}g/ml$. Conclusions: These results suggest that CP water extract has an anti-inflammatory activity through suppressing the JNK and $NF-{\kappa}B$ activation.

Effect of BA and GA on Embryo Germination from Ovule Culture in Intergeneric Hybrids between Brassica and Raphanus (배추와 무의 속간 잡종육성을 위한 배주배양시 배 발아에 미치는 BA와 GA의 효과)

  • Intergeneric crosses over 34 cross combinations between genus Brassica and Raphanus were made. Ovules taken out of crossed were cultured on MS media supplemented with 1.0 mg/L GA and BA. Germination of embryo from ovule culture was not influenced by BA and GA in medium but by parental characters in its cross combination. Use of Raphanus sativus cv. Daibyosobudore and Jungkukcheongpi showed low embryo germination when they were used as male part. Cross combination with Brassica juncea as male parent showed slightly increased germination compared to other cross. These results indicated that embryo germination in ovule culture was not much influenced by casein hydrolysate, malt extract, BA, kinetin and glutamine in the medium, but parents in combination were key factor for increasing embryo germination.

  • This study was performed to certify the inhibitory effect of aqueous extracts from sixty-seven medicinal plants on the activity of xanthine oxidase in vitro. Among the sixty-seven medicinal plants, Scutellaria baicalensis Georgi, Citrus aurantium L., Polygonum multiflorum Thunb., Pueraria thunbergiana (Sieb. et Zucc.) Benth., Citrus unshiu Marcor., Rubus coreanus Miquel, Camellia sinensis L., and Poncirus trifoliata Raf. were regarded as effective anti-gout sources. The active substances of P. multiflorum root extract were very stable at pH 2.0 and high temperatures. Xanthine oxidase activity was proportionally inhibited when concentrations of P. multiflorum extract increased. The aqueous extract from P. multiflorum root at a concentration of 2.0 mg/0.1 mL inhibited xanthine oxidase by 73.8%.

Effects of Citri Reticulatae Viride Pericarpium on the Changes of Tyrosine Hydroxylase and ACTH in the Forced Swimming Test (청피가 FST에서 TH와 ACTH변화에 미치는 영향)

  • Objectives : The investigation of the antidepressant effects of Citri Reticulatae Viride Pericarpium (CR) Methods : we performed the forced swimming test. Also the expression of Tyrosine Hydroxylase (TH) was measured with immunohistochemical method at the Ventral Tegmental area (VTA), Locus coeruleus (LC). The Adrenocorticotropic Hormone (ACTH) level was measured in plasma. Results: 1. The duration of immobility in the forced swimming test was significantly decreased in the CR 100 mg/kg, CR 400 mg/kg groups, comparing with the control group (p < 0.05, p < 0.001). 2. TH expressions in the VTA, LC were significantly reduced in the CR 100mg/kg and CR 400mg/kg treated group, comparing with the control group (p < 0.05, p < 0.001). 3. ACTH expression in plama was significantly reduced in the CR 100 mg/kg treated group, comparing with the control group (p < 0.05). Conclusions : According to the above results, it can be considered that Citri Reticulatae Viride Pericarpium has antidepressant effect through the reduction of TH expression at VTA, LC and ACTH level in plasma.
