Quality Characteristics of Noodles Containing Shoots and Leaves of Aralia elata and Kalopanax pictus (참두릅과 개두릅을 첨가한 국수의 제조특성)

  • The objective of this study was to evaluate the quality characteristics of noodles made with a dried powder of the shoots and leaves from $Aralia$ $elata$and $Kalopanax$ $pictus$. Ash, protein, and dietary fiber contents of $A.$ $elata$ and $K.$ $pictus$ were higher than those of wheat flour. The protein and fat contents of $K.$ $pictus$ powder were higher than those of the $A.$ $elata$ powder, but ash and dietary fiber contents were less than those of $A.$ $elata$. When the amount of $A.$ $elata$ powder increased, brightness (L) values of dough decreased, and redness (a) and yellowness (b) values of dough increased. The weight and volume of the cooked noodles also increased, as did the turbidity of the soup. Addition of $A.$ $elata$ and $K.$ $pictus$ powders in noodles reduced the cutting forces needed to cut the cooked noodles. The sensory evaluation indicates that noodles with 5% $A.$ $elata$ and $K.$ $pictus$ powder added were the most preferable.

  • 염증은 물리화학적 자극이나 세균 감염과 같은 외부 자극에 대응하기 위한 생체조직의 방어 반응의 하 나이며 조직이나 장기의 손상을 회복시키는 기전으로서 매우 중요한 역할을 하지만 염증반응이 과도해 질 경우 궤양성 대장염, 기관지염, 천식, 대장암 등을 유발하는 원인으로 작용하기 때문에 염증반응을 조 절하는 항염증제 개발은 매우 중요시되고 있다. 국내 염증개선 관련 시장은 현재 약 3조원 규모이며 지속 적인 성장세에 있지만 천연소재의 기능성 중심으로 출시된 제품 중 다수의 제품에서 주로 수입 원료를 사용하고 있어 국내 자생 및 재배하는 천연 소재들의 효능에 대한 검증을 통해 수입 원료 의존도를 낮추 기 위한 연구가 요구되고 있다. 따라서 본 연구에서는 순창에서 재배되는 참두릅 (Aralia elata Seem)과 도라지 (Platycodon grandiflorum)를 이용하여 천연 염증개선 소재화를 목적으로 기능성 증진을 위한 추 출조건 선정과정과 대식세포에 대한 세포독성 및 항염증 효능을 확인하여 천연소재를 이용한 염증개선 소재화를 시도하고자 한다. 본 연구는 전북 순창에서 재배되는 참두릅과 도라지의 추출조건을 선정하기 위해 용매, 온도 및 시간별 추출물의 total polyphenol 함량 평가를 통하여 최적 추출조건 선정을 진행하 였으며, 선정된 추출조건에서 추출된 추출물의 항산화 활성을 측정하기 위하여 DPPH & ABTS radical scavenging activity 및 total flavonoids 함량을 확인하여 항산화 효능을 평가하였다. 또한 대식세포인 Raw 264.7을 사용하여 MTT assay, Nitric oxide (NO) 생성 억제 효능을 확인하여 세포독성 및 항염증 활 성을 평가하였다. 실험결과, Total polyphenol 함량 분석을 통해 최적 추출조건이 선정된 두릅 (주정 40%, 25℃, 3 h), 도라지 (주정 60%, 25℃, 1 h)의 추출물을 이용하여 DPPH & ABTS radical scavenging activity 및 total flavonoids 함량을 분석한 결과, 도라지보다 두릅에서 더 높은 항산화 활성을 나타내었다. 대식세포를 활용한 두릅과 도라지의 세포독성을 측정한 결과, 100 ug/mL 이내의 농도에서 독성활성이 나타내지 않음을 확인되었으며, 항염증 활성을 측정한 결과 100 ug/mL에서 두릅 추출물이 도라지 추출 물보다 약 33% 이상 NO 생성억제 활성이 높게 나타내었다.

Desmutagenic Activity of Heated Mountain Herb Juices (산채류(山菜類) 가열즙(加熱汁)의 돌연변이 억제 작용에 관(關)한 연구(硏究))

  • Potential mutagenicity of ten heated edible mountain herbs were examined with spore recassay, Ames test and DNA breaking test. Samples of edible mountain herbs were prepared with water extraction at $100^{\circ}C$ for 20 minutes. With the rec-assay, no significant mutagengic activity could be obtained from all of the samples, but among the eight of metal ions added to sample solution, $Pb^{2+}$ to R. crispus heated juice, $Zn^{2+}$ to L. fischeri and S. bracycarpa heated juice increased mutagenic activity of the samples. With the Ames test and DNA breaking test, all of the samples did not show mutagenicity. However, breaking action was activated on heated L. fischeri, P. japonicus. A. triphylla and A. tataricus juices in the presence of 25mM $Cu^{2+}$. But heated A. elata, H. aurantiaca, A. triphylla, S. bracycarpa and A. scaber juices were inactivated in the presence of 25mM $Fe^{2+}$. Desmutagenic activities against benzo$({\alpha})$pyrene significantly increased as increasing concentration of the heated edible mountain herb juices.

Inhibitory Activity of Aralia elata Leaves on Protein Tyrosine Phosphatase 1B and α-Glucosidase (참두릅 잎의 Protein Tyrosine Phosphatase 1B와 α-Glucosidase 저해 활성)

  • Anti-diabetic potential of the leaves of A. elata through the inhibitory activity on PTP1B and ${\alpha}$-glucosidase has not been reported. In this study, the EtOAc fraction of methanolic extract from the leaves of A. elata showed potent inhibitory activity against the PTP1B and ${\alpha}$-glucosidase with $IC_{50}$ value of $96.29{\pm}0.3$ and $264.71{\pm}14.87{\mu}g/mL$, respectively. Three known triterpenoids, oleanolic acid, oleanolic acid-28-O-${\beta}$-D-glucopyranoside and oleanolic acid-3-O-${\beta}$-D-glucopyranoside were isolated from the most active EtOAc fraction. We determined the chemical structure of these triterpenoids through comparisons of published nuclear magnetic resonance (NMR) spectroscopic data. Furthermore, we screened these triterpenoids for their ability to inhibit PTP1B and ${\alpha}$-glucosidase over a range of concentrations ($12.5-50{\mu}M$). All three terpenoids significantly inhibited PTP1B in a concentration dependent manner and oleanolic acid effectively inhibited ${\alpha}$-glucosidase. In addition, these compounds revealed potent inhibitory activity with negative binding energies toward PTP1B, showing high affinity and tight binding capacity in the molecular docking studies. Therefore, the results of the present study clearly demonstrate that A. elata leaves and its triterpenoid constituents might be beneficial in the prevention or treatment of diabetic disease.

Analysis of Vegetation-Environment Telationships of Main Wild Vegetables on Short-term Income Forest Products, in Korea (단기소득임산물 자생지 주요 산채류 식생과 환경의 상관관계 분석)

  • This study was conducted in 2016-2017 to provide the basic ecological data needed to establish environmental conditions for the cultivation of wild vegetables. It used TWINSPAN to classify the vegetation structure of natural habitats of wild vegetable nationwide and DCCA ordination to analyze the correlation between the by community structure and environmental factors. We performed TWINSPAN on 100 taxa with high importance values in 91 plots of major habitats of wild vegetables. The vegetation was classified into Cirsium setidens and Synurus deltoides group, Ligularia fischeri and Hemerocallis fulva group, Adenophora divaricata var. manshurica group, Platycodon grandiflorum and Aster scaber group, Aralia elata and Pteridium aquilinum group, and Pimpinella brachycarpa and Osmunda japonica group communities. We then performed DCCA ordination of 11 communities classified by TWINSPAN and 11 environmental factors. The results showed that the altitude had the strongest correlation with the vegetation. The Cirsium setidens, Synurus deltoids, and Lifularia fischeri communities were distributed in areas with similar environmental factors such as high altitude, gentle slope, and nutrient. The Aralia elata and Osmunda japonica communities were distributed in the location environment with low altitude, pH, O.M, T-N, $Ca^{2+}$, and C.E.C. The Hemerocallis fulva community was distributed in the location environment with moderate northeastern and northwestern slope, low altitude and pH, and high $P_2O_5$, whereas the Adenophora divaricata var. manshurica community was distributed in the location environment with gentle southeastern and southwestern slope, high altitude and pH, and low $P_2O_5$, which was the opposite tendency of the location environment from Hemerocallis fulva community. The Platycodon grandiflorum community was distributed in the location environment with gentle southwestern slope, low altitude, pH, O.M, T-N, $P_2O_5$, $Ca^{2+}$, and C.E.C., and high $Mg^{2+}$. The Pteridium aquilinum community was distributed in the location environment with southwestern slope, low altitude, O.M, T-N, C.E.C, $P_2O_5$, $Ca^{2+}$, and $K^+$. The Aster scaber and Pimpinella brachycarpa communities were widely distributed in many plots with various location environments.

Total Folate Contents of 15 Edible Plants Consumed in Korea Using Trienzyme Extraction Method (국내 소비되는 봄나물의 Trienzyme 추출법을 적용한 엽산 함량 분석)

  • Trienzyme digestion (AOAC Official Method 2004.05) procedure using protease, ${\alpha}$-amylase, and chicken pancreas conjugase was evaluated to determine its usefulness in the microbiological quantitation of total folate in foods. Folate values obtained by alkali hydrolysis (Korean Food Standards Codex) were compared to those obtained by the trienzyme method for four certified reference materials (CRM) representing diverse matrixes. Trienzyme treatment increased measurable folate from most CRM compared to levels found after alkali hydrolysis. The largest increases were observed with CRM 487 (pig liver, 5.8-fold) and CRM 121 (whole meal flour, 3.1-fold) after trienzyme digestion. Using trienzyme digestion method, total folate contents of raw and blanched edible plants were determined. Eleutherococcus senticosus ($146.9{\mu}g/100g$) showed the highest total folate content, followed by Aster glehni F. Schmidt ($142.8{\mu}g/100g$) and Ledebouriella seseloides H. Wolff ($140.4{\mu}g/100g$) on a wet weight basis. Blanching of samples resulted in an insignificant decrease in folate content for five samples and 11~63% reduction for nine samples. Our finding suggests that trienzyme digestion method is accurate for the determination of food folate in leafy vegetables.

A Survey on the Usage of Wild Grasses (산채류의 이용실태에 대한 조사)

  • This survey investigated on the degree of acknowledgment, intake frequency and using methods of wild grasses of the residents in Kyeonggi, Kangwon, Chunnam, Chungbook, Kyeongnam & Cheju area. The results were as follows: 1. An answer that impression of wild grasses is 'nature food' was the highest(42.6%). It was founded that interest of nature food has been increasing. 2. It has been shown that the most common method for elimination of astringent taste is to blanch and then wash several times with water.(62.9%) 3. The most common obtaining routes was traditional market. It has been shown that rate of gathering in the fields is higher in rural community and the group of over 60 years old than that of urban community and the other groups. 4. In rural area, people(28.7%) ate wild grasses more frequently than in urban area. Family with the old and the group of over 40 years old eat wild grass often. 5. More than 95% of answerers were familiar with the names of wild grasses, codonopsis lanceolata, chinese bellflower, braken, mugwort, wild rocambole and edible shoots of a fatsia & Korean lettuce are known to them with over 88.6%, 85.1% respectively. Frequently eaten wild grasses were braken, chinese bellflower, wild tocambole, codonopsis lanceolata, mugwort and korean lettuce orderly. But in Chunnam & Kyeongnam, they were braken, chinese bellflower, mugwort, wild rocambole and Korean lettuce orderly. 6. Wild grasses eaten with rice were total 25 species, and mugwort had the highest usage rate with 41.9%. Mugwort was used for $D'{\breve{o}}k$(rice cake)(77.8%), T'wigim $J{\breve{o}}n$(deep fat fried dish swallow fat fried dish)(50.1%) and liquor(4.6%). In the cooking of Guk(soup), Chigae(stew), Sengch'ae(raw vegetable) and kinds of wild grasses for Namul(cooked seasonal vegetable) & Bokkum(saute) are the most various with 58 species, especially braken was most commonly used for saute. Sedum & Korean lettuce were the common ingredients of Kimch and codonopsis lanceolata was used in liquer & Jangachis(pickle) with 40%, 46% each. Chinese bellflower was used with the most variable cooking method. 7. 43 species of wild grasses were stored by blanching and drying, and braken had the highest rate(32.3%) then aster scaber, flowering fern, Pimpinella brachycarpa, mugwort and ligularia orderly.
