Skin Transcriptome Profiling of the Blass Bloched Rockfish (Sebastes pachycephalus) with Different Body Color Patterns (체색 패턴이 다른 개볼락(Sebastes pachycephalus) 피부 전사체 프로파일링)

  • The body color pattern in fish is a distinctive feature for species identification. The blass bloched rockfish Sebastes pachycephalus is a commercially important marine fish species, distributed in the central and southern parts of Korea and south Hokkaido of Japan. It has a morphological feature divided into four subspecies according to with or lacking distinct spots on the body surface, and to the location of markings on the body surface. However, the genetic basis of body color pattern of S. pachycephalus is still unknown. Thus we analyzed the transcriptome of S. pachycephalus skin samples using RNA-seq analysis to investigate functional genes related to body color patterns. The experimental skin samples were prepared by classified into 'Wild type' (lacking distinct spots and markings) and 'Color type' (with distinct spots and marking). Two skin sample transcriptomes were compared pairwise and the results revealed that were 164 differentially expressed unigenes in the skin samples of 'Wild type' and 'Color type'. Gene Ontology analysis of 164 differentially expressed unigenes showed that these genes were included in the functional group of molecular function (2 genes), biological process (46 genes), and cellular component (6 genes). There were several genes that body color type skin specific expression and the genes were CTL (Galactose-specific lectin nattectin), CUL1 (Cullin-1), CMAS (N-acylneuraminate cytidylyltransferase), NMRK2 (Nicotinamide riboside kinase 2), ALOXE3 (Hydroperoxide isomerase ALOXE3), SLC4A7 (sodium bicarbonate cotransporter 3). Our study is the first attempt to search for functional genes involved in the formation of body color patterns in S. pachycephalus. The differentially expressed unigenes obtained in this study can be used as candidate genes for functional gene study related to body coloration of fish.

Spatial and Temporal Variation of Macroinvertebrates according to Physical Factors in Gongji Stream Area (공지천 수계에서 물리적인 요인에 따른 저서성 대형무척추동물 군집의 시.공간적인 변동)

  • Our study is purposed to understand effect on spatio-temporal variability of macroinvertebrate community 11 sampling times at 4 sites between two streams (Shinchon stream and Gongji stream in Chuncheun City) from May 2011 to October 2013. In this study, the possible physical factors on spatio-temporal variability of macroinvertebrate community were discussed. After stream improvement project, the effects of anthropogenic disturbance in study sites appeared as increased water temperature more than $30^{\circ}C$ and the difference of water temperature between Shinchon stream and Gongji stream was by maximum $9^{\circ}C$ on 2012. The monsoon rainfall decreased number of species, individuals and biodiversity index of macroinvertebrate community, particularly, in Shincheon stream compare to Gongji stream. Dominant species of macroinvertebrate taxa was caddisfly in Shincheon stream and Diptera in Gongji stream but on August 2013, it was dominated by Diptera, Chironomidae spp. in all study sites. The spatio-temporal variability of macroinvertebrate community in the streams may be useful as bio-indicator influencing anthropogenic factors such as soil erosion (landslide or cultivation) or monsoon rainfall.

Characterization of Tobacco mosaic virus Isolated fromSolanum tuberosum ‘Chubak’ in Korea (감자 '추백' 에 발생한 Tobacco mosaic virus 의 특성)

  • An isolate of Tobacco mosaic virus (TMV) was isolated from potato cultivar ‘Chubak’ showing vein clearing and mild mosaic at Namhae in Korea. This isolate, TMV-St, was differentiated from other tobamoviruses based on biological properties, serological relationships and nucleotide sequence analyses of coat protein genes. TMV-St caused typical symptoms on four indicator plants as compared to the tobamovirus of TMV-U1, Pepper mild mottle virus (PMMoV), and Tomato mosaic virus (ToMV), which caused economic losses in Solanaceous vegetables, tomato, pepper, and eggplant. Remarkably, the TMV-St induced distinctly different symptom of systemic chlorotic spots on Chenophodium murale. On C. murale, Gomphorena globosa, and Nic-otiana rustica, the four viruses were classed by the virulence of systemic or local infections. In serological test TMV-St antiserum showed a precipitation line with each other tabamovirus. The CP gene of TMV-St contain 477 nucleotides, and the nucleotides sequence was the most similar to that of TMV-U1.

Spatial Analysis of Landscape Structure Changes Caused by the US Conservation Reserve Program in the Central High Plains (미중부지역 농지보전 프로그램에 의한 경관구조 변화분석)

  • The U.S. Conservation Reserve Program (CRP) resulted in the conversion of approximately 14.8 million ha(36.5 million acres) of cropland to grassland, woodland, and other conservation uses throughout the U.S. between 1986 and 1992. One of the major results of CRP has been the addition of millions of hectares of potential wildlife habitat. primarily as grassland. In this study, we examined regional changes in landscape structure caused by the introduction of CRP. Utilizing multi-seasonal Landsat Thematic Mapper imagery, we produced maps of cropland and grassland for the pre- and post- CRP enrollment periods for a six-county region in southwest Kansas. We then applied post-classification differencing to identify regions of cropland that had been converted to CRP. Using the FRAGSTATS spatial pattern analysis program, we calculated a variety of spatial statistics to analyze changes in landscape structure due to CRP. The major impact of CRP in the six-county study area has been the reversal of an overall trend of grassland habitat fragmentation. From the standpoint of potential wildlife habitat, the introduction of CRP has greatly increased the number of patches, mean patch size, and the interior or core area of grassland patches. In addition, CRP has increased connectivity and aggregation between grassland patches, potentially important factors for species of conservation interest, particularly those that require larger expanses of unbroken habitat. Finally, the distance between neighboring patches of grassland has decreased, reducing travel distance between patches. Clearly, the introduction of CRP has substantially modified the spatial structure of the southwest Kansas landscape, with important implications for wildlife habitat.

Influence of Humic or Fulvic Acid on Phytotoxicity of Bentazone (Bentazone의 약해(藥害)에 미치는 부식산(腐植酸) 및 훌브산(酸)의 영향(影響))

  • This research was conducted to assess the influence of humic or fulvic acid on Bentazone phytotoxicity using a bioassay with hydroponically grown cabbage (Brassica campestris subsp. napus var. pekinensis Makino). Concentrations of Bentazone in the water culture media were ranged from 0 to 32 ${\mu}M $ and those of the organic ligands were 1.0mM as a soluble carbon. Media were prepared in a complete factorial combination with pHs of 4.5, 6.5 and 8.5. The phytotoxicity indices on growth rate and dry weight decrement were employed to evaluate the effects of organic ligands on the Bentazone phytotoxicity. Humic or fulvic acid without Bentazone treatment enhanced the growth of cabbage and this effect was evident at low pH of 4.5. Bentazone led to chlorosis and necrosis on cabbage leaves resulting in the decreases of dry and fresh weights and growth rate. This phytotoxic effect was increased with Bentazone concentration and evident at low pH. At pH 4.5, dry weight was decreased about 63% with 8${\mu}$M of Bentazone treatment. Effective concentration of Bentazone causing 50% decreases in fresh weight as compared to the control was estimated to be 21${\mu}$M. Presence of organic ligand reduced the phytotoxicity of Bentazone to cabbage significantly by increasing yields and growth rates as compared to the treatment of Bentazone alone. At pH 4.5, fulvic acid reduced phytotoxicity of Bentazone upto 46%, and this efficiency of fulvic acid was better than that of humic acid under the same condition.

  • The ultimate research goal of the current study was a development of hydrolyzed whey protein powder (7%-GNANA) manufactured with normal content of sialic acid, a marker compound, that is naturally occurring at 7% concentration in GMP obtained from the milk protein. GMP is a safe food, used worldwide in infant and baby foods, etc. The test substance was prepared using (7% sialic acid containing) GMP as a raw material, and then using alcalase, an enzyme approved as a food additive, after separation of sialic acid with 100% efficiency and 7%-GNANA (containing 7% sialic acid and protein; product name: HELICOBACTROL-7) provided by MEDINUTROL Inc. (Korea). Bacterial reverse mutation (Ames) test was conducted in accordance with GLP Guideline using the test substance specified above. To identify its mutagenic potential against microorganisms, histidine auxotrophic strains of Salmonella Typhimurium, TA98, TA100, TA1535, and TA1537, and tryptophan auxotrophic strain of Escherichia coli, WP2uvrA, were used. The bacterial reverse mutation (Ames) test was performed by dividing the test substances into five different concentration groups (0, 61.7, 185, 556, 1,670, $5,000{\mu}g/plate$). Results of this experiment did not reveal repetitive increase of colony generating values or positive criteria for reverse mutagenicity for any concentration of test substances in any of the five strains, regardless of the presence of a metabolic activation system, and no dose-dependency was identified. In conclusion, the safety of 7%-GNANA test substance was verified by bacterial reverse mutation test conducted before registration of 7%-GNANA as a food additive.

Morphological and Genetic Species Identification in the Chironomus Larvae (Diptera: Chironomidae) Found in Domestic Tap Water Purification Plants (국내 수돗물 정수장에서 발견된 깔따구 유충(파리목: 깔따구과)의 유전적-형태적 종 동정 연구)

  • The Chironomus(Diptera: Chironomidae) is a freshwater benthic invertebrate that is an important indicator organism used for environmental pollution and water quality monitoring. In this study, we performed morphological classification and genetic species identification using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA for an accurate species classification of Chironomus larvae found in tap water purification plants in Incheon, Korea. Twenty larvae in six water purification plants consist of four species, including twelve Chironomus kiiensis, six Chironomus flaviplumus, one Chironomus dorsalis, and one Polypedilum yongsanensis (not included Genus Chironomus). Morphological characteristics of each larvae were identified based on the head capsule, the mentum, the mandible, the antenna, and the claw. Based on the COI sequences of 21 individuals of 17 Chironomus species registered in NCBI Genbank, phylogenetic analysis indicated that the 20 individuals investigated in this study consist of the same clade with corresponding species of the high homology (99~100%) including C. kiiensis, C. flaviplumus, C. dorsalis, and P. yongsanensis. These results will be used as main classification indicator for monitoring freshwater ecosystems by providing integrated morphological and genetic information for the species identification of Korean Chironomus.

An Assessment of Heavy Metal Contamination in the Onsan Bay Using Biological Indicators (생물지표종을 이용한 온산만의 중금속 오염도 평가)

  • Concentrations of cadmium, copper and zinc were determined in three molluscan species including Sacculosiphonaria japonica, Reishia clavigera and Crassostrea gigas to assess heavy metal contamination in the Onsan Bay. The range of cadmium concentrations were 3.83∼39.67 ㎍/g d.w. in S. japonica, 7.47∼103.47 ㎍/g d.w. in R. clavigera, 1.32∼92.17 ㎍/g d.w. in C. gigas. Concentration of cadmium at contaminated site in C. gigas was 70 folds higher than that of reference site. The range of copper concentrations were 22.92∼1287.56 ㎍/g d.w. in S. japonica, 37.53∼656.97 ㎍/g d.w. in R. clavigera, 161.50∼3208.52 ㎍/g d.w. in C. gigas. The concentration of copper at contaminated site in S. japonica was 56 folds higher than that of reference site. The range of zinc concentrations were 93.48∼377.40 ㎍/g d.w. in S. japonica, 423.65∼1075.57 ㎍/g w. in R. clavigera, 2018.13∼22275.00 ㎍/g d.w. in C. gigas. The difference of zinc concentrations between reference site and contaminated site was relatively small than those of cadmium and copper in all the species tested. The largest difference was 11 folds in C. gigas. In the present work, we confirmed that all of these species, S. japonica, R. clavigera and C. gigas, could be used as biological indicators to monitor heavy metal contamination.

A Study of Urinary Ascorbic Acid as a Noninvasive Biomarker for Cadmium-Intoxication in Rats (카드뮴 중독 흰쥐에 대한 Noninvasive Biomarker로서의 요중 Ascorbic Acid에 관한 연구)

  • This study was planned to evaluate the urinary ascorbic acid as a new biological marker for the intoxication of cadmium, which could possibly be driven by its increased utilization and environmental pollution. In order to meet this goal, we have peformed measurement of urinary ascorbic acid concentration, histopathological examination of the kidney, and biochemical test for the liver function using cadmium-intoxicated rats by oral administration. The average concentrations of urinary ascorbic acid in the $CdCl_2$-treated rats were 214.0 mg/dl for 100 ppm group and 254.3 mg/dl for 200 ppm group during experimental period of 50 days. These levels are 24 and 28 times higher than one in the control group (9.0 mg/dl), respectively. Ultrastructural study showed the eosinophilic hyaline cast and focal effacement, fusion in the renal tubules, as well as loss of foot processes on the glomerular epithelial cells. These results suggested that cadmium may be responsible for renal glomerular injury. The blood levels of AST, ALT and LDH in the treated groups (199 IU/I, 88 IU/I, 1190 U/I for the 100 ppm group and 270 IU/I, 226 IU/I, 760 U/I for the 200 ppm group) were higher than ones in the control group(143 IU/I, 50 IU/I, 334 U/I). These results indicated the cadmium induced the damage of liver function. In conclusion, the administration of cadmium showed a remarkable increase of urinary ascorbic acid with renal and hepatic damage. Therefore, it is expected that measurement of urinary ascorbic acid would be an powerful method as a noninvasive biomarker for cadmium intoxication.

Distribution of spontaneously growing mushrooms in the Wolchulsan National Park (월출산국립공원의 자생버섯 분포상)

  • Mushrooms growing in the Wolchulsan National Park were surveyed from April to October 2018. As a result of this survey, 2 divisions, 9 classes, 24 orders, 71 families, 177 genera, and 407 species were found. Among them, 3 classes, 16 orders, 55 families, 150 genera, and 370 species belonged to Basidiomycota, while 6 classes, 8 orders, 16 families, 27 genera, and 37 species belonged to Ascomycota. The species richness was the highest in September at the altitudes between ~100 and 200 m. Among the investigated sites, the most diverse species occurred in the Gyeongpodae area. In this survey we found, Macrolepiota procera, Suillus bovinus, and Amanita pantherina, all of which have been known as climate-sensitive biological indicator species (CBIS). Based on the DNA sequence analyses, Craterellus parvogriseus was confirmed as a new record for Korea.
