Production of Fungal Lipids -V. Effects of Vitamins, Metabolic Intermediates and Mineral Salts on the Growth and Lipid Accumulation of Mucor plumbeus- (곰팡이 유지(油脂) 생산(生産)에 관(關)한 연구(硏究) -제 5 보 : 비타민류(類), 대사중간생성물(代謝中間生成物) 및 미량원소(微量元素)가 Mucor plumbeus의 균체(菌體) 및 지방질(脂肪質) 생산(生産)에 미치는 영향(影響)-)

  • Effects of vitamins, metabolic intermediates and several inorganic mineral salts on the biomass and lipid accumulation of Mucor plumbeus were investigated after 15 days of incubation at $37^{\circ}C$ under static culture condition. The optimum concentrations of various vitamins were ${\gamma}/l$ for biotin, and 0.01 g/l for nicotinic acid, pyridoxine, thiamine and riboflavin. Among them pyridoxine was the most stimulatory. The maximum felt weight and lipid content per 50ml medium were $2.82{\pm}0.14\;g$ and 62.8%. Triglyceride content of neutral lipid produced under this condition was 64.9%. The major fatty acids were oleic acid (50.0%), linoleic (23.8%) and palmitic acid (13.9%). Malonic acid was considered not to be desirable even though it stimulated the biomass and lipid accumulation because triglyceride content was lowered considerably comparing with control. $MgSO_4{\cdot}7H_2O$ was the most stimulatory among the various magnesium salts and its optimum concentration was 5 g/l. Mucor plumbeus did not require $NaH_2PO_4$ for the stimulation of felt and lipid production. However, the addition of $MnCl_2$ at the concentration of 2 g/l was stimulatory to show $2.76{\pm}0.28\;g$ of felt/50 ml and 56.4% lipid content, and 73.9% triglyceride in the neutral lipid.

  • Prunus tomentosa Thunberg seed was investigated to evaluate its possibility for use as food resources of fats and proteins. The seed contained 40.38% of crude fat and 26.59% of crude protein. The lipid fractions obtained by silicic acid column chromatography were mainly composed of 95.49% of neutral lipids, whereas compound lipids were only 4.51%. Among the neutral lipid components by thin-layer chromatography, triglycerides were 89.86%, sterols, monoglycerides, sterol esters, free fatty acids and diglycerides were 4.14%, 2.98%, 1.77%, 1.07%, and 0.18%, respectively. Oleic acid (65.06-66.05%) and linoleic acid (26.56-28.40%) were the main fatty acids in the total lipid, neutral lipid and triglyceride fractions. In the glycolipid and phospholipid fractions, predominant fatty acids were oleic acid (40.55-51.46%), linoleic acid (20.26-30.89%) and palmitic acid (17.64-21.43%). The extractability of salt soluble protein of seed was 60%, and recovery rate of main protein fraction separated by Sephadex G-200 was about 46.5%. The electrophoretic analysis showed 7 bands in seed protein.

An Analysis of Vegetation Structure and Vegetation-Environment Relationships with DCCA in Forest Community of Ullung Island (울릉도 산림군락의 구조 및 DCCA에 의한 식생과 환경과의 상관관계 분석)

  • 본연구는 울릉도의 성인봉과 태하령 지역의 산림 식생을 대상으로 199년 7-8월에 식생조사와 토양조사에 의한 너도밤나무 군락의 임분구조 및 DCCA ordination을 이용하여 분석한 결과는 다음과 같다. 1. 울릉도 산림의 중요치를 각 군락별로 살펴보면 너도밤나무-섬조릿대 군락에서 중요치가 높은 종은 너도밤나무, 우산고로쇠, 마가목, 섬단풍, 섬벚나무 등의 순으로 너도 밤나무-일색고사리 군락은 우산고로쇠 너도밤나무, 마가목 층층나무, 등수국 등의 순으로 나타났다 그리고 너도밤나무-큰두루미꽃 군락에서 중요치가 높은 종은 너도밤나무 우산고로쇠 등수국 마가목 음나무등의 순으로 솔송나무-섬잣나무 군락은 섬잣나무, 너도밤나무, 솔송나무, 회솔나무, 섬피나무 등의 순으로 나타났다. 2. DCCA ordination에 의하면 산림군락과 환경요인과의 상관관계는 다음과 같다 너도밤나무-섬조릿대 군락은 해발고가 높고 네 군락 중 토양수분이나 전절소 유기물 등이 많은 지역에 분포하고 있었다. 너도밤나무-일색고사리 군락은 해발고가 다른 군집보다 높고 토양수분이나 전질소, 유기물 등이 많아 너도밤나무-섬조릿대 군락과 매우 유사한 입지환경을 가진 지역이나 토성 중 clay 가 많이 함유된 지역에 분포하고 있었다. 너도밤나무-큰두루미꽃 군락은 해발고가 네 군락 중 중간지역에 분포하고 있으며 토양수분이나 유기물, 전질소 등도 중간인 지역에 분포하고 있었다. 솔송나무-섬잣나무 군락은 해발고가 낮고 토양수분이나 전질소, 유기물이 적고 sand가 많이 함유된 토양에 분포하고 있었다. 3. 울릉도 산림군락으 Shannon의 종다양도 지수는 0.5455~0.8801으로 비교적 낮은 수치를 나타내고 있다. 또한 너도밤나무 군락에서 분포하고 있는 주요 종의 조서열 중요치 곡선을 보면 전체의 기울기가 완만하여 너도밤나무 군락은 안정적이라 할 수 있다.단 생산성 향상을 위한 세포의 고농도 배양에는 조사한 여러 배양 시스템 중에 가장 효율적인 시스템임올 알 수 있었다 하지만 이 시스템 에서 포도당을 낮은 level로 유지할 수 있었으나, 초산의 과도한 축적으로 항체 생산성의 향상은 예상에 비해 크지 않았다. 81%), C18 0(12.38%), C18: 1(25.93%), C22:6(9.95%)이며 결합지방질(結合脂肪質)은 C14 : 0(11.60%), C16 : 0(18.94%), C16: 1(10.42%). C18 : 1(10.89%), C22 : 6(23.44%)이었다. 총필수지방산(總必須脂肪酸) 함량(含量)은 극성지방질(極成脂肪質)$(20.14{\sim}31.12%)$이 비극성지방질(非極成脂肪質)$(6.97{\sim}11.13%)$보다 훨씬 높았고, 결합지방질(結合脂肪質)이 유리지방질(遊離脂肪質)보다 높았으며 부위별(部位別)로는 피부(皮部)$(15.18{\sim}15.41%)$가 육질부(肉質部)$(6.97{\sim}11.13%)$보다 높았다. 또${\omega}3$고도부포화지방산(高度不飽和脂肪酸) 함량(含量)은 육질부(肉質部)$(15.15{\sim}28.32%)$가 피부(皮部)$(6.77{\sim}18.18%)$나 내장부(內臟部)$(8.35{\sim}9.74%)$보다 높았으며, 육질부(肉質部)에서는 극성지방질(極成脂肪質)$(26.28{\sim}34.18%)$이 비극성지방질(非極成脂肪質)$(15.15{\sim}28.32%)$보다 높았다.veral world-wide prediction models. Comparison of Lipid Components in Wild and Cultured Bastard (천연 및 양식산 넙치의 지방질성분)

  • The lipid components of cultured bastard(Paralichthys olivaceus) were analyzed and compared with those of wild one and flounder(Pleuronichthys cornutus). Total lipid(TL) content of dorsal muscle in wild and cultured bastard, flounder was 2.0%, 1.6%, and 1.9%, respectively. Wild bastard and flounder were higher in neutral lipid(NL) and glycolipid(GL) contents, while lower in phospholipid(PL) content than those of cultured one. The NL was mainly consisted of triglyceride(70.7-72.6%), and PL was mainly occupied by phosphatidylcholine(53.6-58.3%), phosphatidyl-ethanolamine(25.9-29.5%). In fatty acid composition of TL, cultured bastard showed higher content in polyenes such as docosahexaenoic acid. and lower in monoenes such as palmitoleic, oleic acid than those of wild one. Flounder was higher in Percentage of arachidonic, eicosapentaenoic acid compard with bastard. The major fatty acids of these fishes were palmitic, docosahexaenoic, oleic, eicosapentaenoic and palmitoleic acid in order. These fatty acid composition of NL, GL and PL fractions were not significantly different among wild and cultured bastard, flounder.

Changes in Lipid Components of Pollack During Sun-Drying (명태 천일건조 중 지방질성분의 변화)

  • Changes in lipid components of pollack meat during sun-drying and effects of NaCl on lipid oxidation were examined. TBA values and peroxide values of sun dried pollack(SD), salted and sun dried pollack (SS) were 0.142 and 14.8 meq/kg, 0.226 and 20.0 meq/kg after sun-drying, respectively. Raw pollack contained 6.12% total lipid consisted of 2.42% neutral lipid(NL) and 3.70% phospholipid(PL) as dry basis, and there were $47{\sim}65%$ decrease in PL content during sun-drying. The NL class of raw pollack mainly consisted of triglyceride(TG), sterol(ST)+diglyceride(DG), hydrocarbon(HC)+sterol ester(SE), and main components in PL class were phosphatidylcholine(PC), phosphatidylethanolamlne(PE) and phosphatidylserine(PS). The contents of TG, ST+DG, PC and PE decreased, while those of free fatty acid, HC+SE, sphingomyelin and lysophosphatidylcholine increased markedly during sun-drying. The major fatty acids of TL in raw pollack, PD and SD samples were generally 22:6, 16:0, 20:5, 18:1 and 18:3; 20:5 decreased markedly during sun-drying, while saturates and monoenes such as 16:0, 18:0 and 18:1 increased slightly. And remaining ratios of polyunsaturated fatty acids of TL, NL and PL in SD and SS samples were 81.1%, 92.5%. 73.3%, and 74.6%, 74.1%, 45.4%, respectively. The results of changes in lipid components during sun-drying showed that sodium chloride catalyzed the lipid oxidation of pollack meat during drying processing.

  • The optimum conditions for growth and lipid production of Rhodotorula marina IFO 0879 were investigated. The optimum temperature and pH of cultivation was $30^{\circ}C$ and pH 6.0-7.0, respectively. During shaking of the culture for 8 days at $30^{\circ}C$, the maximum cell biomass of Rh. marina was 9.82g per liter of the medium, and the lipid content obtained was 35.4(w/w) of the dry cell biomass. Lactose and glucose were the most effective carbon sources for the lipid production. Ammonium sulfate was found to be the most effective nitrogen in culture medium the growth of the yeast was retarded, whereas its growth was favored at high concentrations with decreased lipid yield. When lacose was added during fermentation, in the initial stage cell biomass and lipid production were lower than those of the control, but in the later stage the trend were reversed. The major fatty acids of yeast lipid were palmitic acid(20.3%), oleic acid(46.6%) and linoleic acid(16.2%)

  • The cultural conditions of Mucor plumbeus FRI 0007 were investigated for the maximum production of felt and lipid. It was found that the lower the pH and the higher the incubation temperature, the higher accumulation of the felt and lipid. Shake culture rendered higher lipid accumulation and lower felt accumulation than static culture. Maximum production of felt and lipid content were 47.8 g/$\ell$ and 50.73%, respectively, when the organisms were static-cultured at a temperature of 37$^{\circ}C$ and pH of 3.5 for 25 days latroscan thinchrographic analysis showed that the higher amount of triglyceride was obtained when static-cultured at a low pH. Fatty acid composition of the microbial lipid was affected by the incubation temperature, types of nitrogen source and speed of agitation: lower degree of saturation was observed as the incubation temperature decreased and the speed of agitation increased. Fatty acids of monoglyceride and diglyceride were mainly palmitic and oleic acids and those of triglycerides were mainly palmitic, oleic acids.

Changes of Lipids in Raw and Processed Adlay Powder during Storage (율무가루 저장중 지방질 조성의 변화)

  • Raw adlay powder(RAP) and processed adlay powder(PAP) were prepared and the changes of lipids in RAP and PAP during storage at $5^{\circ}C$ and $35^{\circ}C$ for six months were studied. The typical patterns of lipid oxidation were observed during the storage; the values of acid, peroxide and carbonyl in the lipids were increased and the iodine value was decreased. These changes were especially significant(P<0.05) at higher temperature($35^{\circ}C$) and were more notable in the lipids from RAP than those from PAP during storage. The contents of neutral lipid, glycolipid and phospholipid were changed appreciably at $35^{\circ}C$, however, the changes were not significant(P<0.05) at $5^{\circ}C$. Further more, triglyceride in neutral lipid, digalactosyl diglyceride in glycolipid and phosphatidyl choline in phospholipid showed respectively a tendency of decrease during storage as compared with the other contents of lipid class.

Changes in the Concentration of Tocopherol and Carotenoid in Pinenut Oil during Autoxidation (잣 지방질의 자동산화에 따른 토코페롤 및 카로티노이드의 변화)

  • The changes in the concentration of toropherol and carotenoid in pinenut oil at various stage of refining are studied during autoxidation. The oxidative stability of pinenut oil decreased with the refining degree increased. The tocopheol content of total crude oil was 55.51mg/100g oil and $\alpha$, ${\gamma}$and $\beta$-tocopherol in the crude oil were 25.48mg, 23.94mg and 6.99mg in order, respectively. The amount of $\delta$-tocopherol was trace. The concentrations of $\alpha$, ${\gamma}$ and $\beta$-tocopherol in pinenut oil after degumming or alkaline refining were 23. 85mg, 19.79mg and 6.12mg or 24.08mg, 24.04mg and 5.33mg, respectively. The content of f-carotene was 0.63% of total unsaponifiable materials and that of lycopene was trace. The concentrations of carotenoids and toropherols in pinenut oil decreased while autoxidation progressed. Degrees of destruction of carotenoids and toropherols. were significant at first stage of oxidation. Decrease in $\alpha$-toropherol was found to be faster than that in $\alpha$, ${\gamma}$-tocopherol during oxidation.

Comparison of Physicochemical Characteristics of Alkali Refined and Esterified Rice Bran Oils (알칼리 정제(精製)와 에스테르화에 의한 미강유의 이화학적(理化學的) 특성(特性)의 비교(比較))

  • The alkali refined rice bran oil (ARBO) and the esterified rice bran oil (ERBO) with glycerol were analyzed for their physicochemical charateristics and the compositions. Specific gravity, refractive index and yellowness of ERBO was higher than ARBO but smoking point was 60-$80^{\circ}C$ higher than ERBO. The free fatty acid content was 0.05% a ARBO and 0.88-1.36% ERBO. The wax was not detected in ARBO but the soft and hard waxes were detected in ERBO. The lipids were composed of 98-99% neutral lipid, 0.2-0.5% glycolipid and 0.1-0.5% of phospholipid in all samples. The triglyceride content of neutral lipid was 96.3% ARBO and 93.0-94.1% ERBO, and its monoglyceride content was 0.11% ARBO and 0.39-0.69% ERBO. The major fatty acid composition of samples were 41-42% oleic, 36-40% linoleic and 17-18% of palmitic acid.
