  • The factors affecting direct somatic embryogenesis from different parts of explant in liquid culture of Scrophularia buergeriana were investigated. Direct somatic embryogenesis was dependent on the explant tissues and stem was the most efficient explant. Rapid shoot development occurred on stem after 3-week culture but roots were not developed yet. Plantlets were not formed through somatic embryogenesis after 3-week culture of petiole. Though direct somatic embryo was not observed from leaf segment culture for 3 weeks, normal plantlets were developed after 8-week culture. BA played the main role for somatic embryogenesis in liquid culture and adding of either IAA or NAA caused rather adverse effects. Culture of stem segments in MS liquid medium with BA at 0.5 mg/ l or 0.1 mg/ l was proved to be the most efficient method for producing plantlets through direct somatic embryos.

  • This research was conducted to estimate the chlorophyll contents of chrysanthemum leaves using SPAD-502 chlorophyll meter under different photoperiod and light intensity. Measurements were done at every third leaf intervals starting from the top of the stem to the bottom at harvest time. SPAD value was highest at 10 hours of photoperiods, followed by 13 and 16 hours of photoperiods. In particular, under short day condition, SPAD value was highest in the young leaves below the flower bud and decreased down the leaf profile. Under long day condition, SPAD value reached maximum at leaves between $15-27^{th}$ from the top and decreased to the minimum immediately below the flower bud (youngest leaf). These results may indicate that the younger leaves become strong source of supporting flowers in reproductive stage, while the younger leaves serve as sinks and older leaves support their development in vegetative stage. Changes of SPAD value at different levels of irradiance showed that highest SPAD value were observed at high irradiance and decreased with decreasing irradiance.

  • When the leaf and stem tissues of Rehmannia glutinosa were cultured on MS medium with plant growth regulators, shoots were regenerated on MS medium with TDZ(0.01 to $2.0{\mu}M$), and root initiation was better on MS medium treated with NAA(0.01 to 2.0mg/$\iota$). On $B_5$ medium, shoot regeneration was better on medium with TDZ than those with Quincrac, NAA, and 2.4-D. The addition of Quincrac, NAA, and 2.4-D inhibited shoot regeneration on MS medium, but promoted shoot regeneration on $B_5$ medium. Shoot regeneration and growth was better on MS medium with the combination treatments of 2.4-D 0.01mg/$\iota$ and TDZ 0.01, 0.1 and $2{\mu}M$ compared with other treatments. Also shoot regeneration and growth on B_5 medium showed the similar results to that on MS medium.

  • Effects of the simulated acid rain on the growth response and the structural features were studied with the 3 month old seedlings of G. biloba and P. thunbergii treated with acid rain of pH 5.6, 4.0, 3.2, and 2.4. The diameter and area of tracheid cells in the transectioned stem of G. biloba decreased with acidity of simulated acid rain. The wall thickness of tracheid cells was the thinest at pH 2.4, but there was no different at other levels of pH. Increasing of the acidity, the height of tracheid cells were reduced steadily. The diameter and area of tracheid cells of the transectioned root reduced with decreasing pH of acid rain, but those at pH 3.2 were larger than those at control. The wall thickness and height of tracheid cells of root were gradually decreased with acidity of acid rain. The size variation of the fusiform cambial initials in the stem of G. biloba sections tangentially showed a shortening tendency with treatment of acid rain. The length of ray initials was the shortest at pH 2.4 and reduced with decreasing pH of acid rain. The diameter, area, wall thickness, and height of the tracheid cells in P. thunbergii stem and root decreased with decreasing pH of acid rain. The areas of the pith, cortex, and xylem in P. thunbergii treated with acid rain decreased, but the cortex and pith areas increased significantly after exposure to acid rain of pH 3.2 compared with control.

  • Local heating system providing hot air locally to growing parts including shoot apex and flower cluster which were temperature-sensitive organs of cherry tomato was developed to reduce energy consumption for greenhouse heating without decline of crop growth. Growing part following local heating system was composed of double duct distributer which connected inner and outer ducts with hot air heater and winder which moved ducts up and down following growing parts with plant growth. Growing part local heating system was compared with conventional bottom duct heating system with respect to distributions of air and leaf surface temperatures according to height, growth characteristics and energy consumption. By growing part local heating, air temperature around growing part was maintained $0.9{\sim}2.0^{\circ}C$ higher than that of lower part of crop and leaf surface temperature was also stratified according to height. Investigations on crop growth characteristics and crop yield showed no statistically significant difference except for plant height between bottom duct heating and growing part local heating. As a result, the growing part local heating system consumed 23.7% less heating energy than the bottom duct heating system without decrease of crop yield.

  • Willow plants are representative biomaterials used in river restoration and main target trees in stream managements. In order to understand the changes in the growth of Salix subfragilis and S. chaenomeloides with their stand ages, we investigated the density, height and basal area of stems and biomass at their different aged stands of the riparian zone of the sand-bed stream, the Nakdong River, Korea. We also developed allometric equations for estimating the biomass of these two species by establishing the relationship between diameter at breadth height and tree height with above-ground biomass. The stem density showed a sharp decrease for 3 years after germination for S. subfragilis and 6 years for S. chaenomeloides, resulting in strong self-thinning. The stem height of the two species increased to 7.5 m in 15 years for S. subfragilis, and to 14 m in 13 years for S. chaenomeloides. Aboveground biomass also increased rapidly at the early stage of growth. The biomass increased to 17 ton DM/ha in 13 years for S. subfragilis and to 1,110 ton DM / ha in 13 years for S. chaenomeloides. It is expected that the allometric equations of two Salix species derived from this study will be applied to the objectively estimating the biomass of willow plants for the management of floodplain trees in streams.
