Effect of Planting Density and a Silicate Fertilizer on Strength of Stem in Chrysanthemum 'Baekma' (국화 'Baekma' 줄기경도에 미치는 재식밀도와 규산질비료 엽면시비 효과)

  • The research was carried out a solution to prevent the breakage of stem resulted from stem cavity in the Dendranthema grandiflorum Ramat. 'Baekma'. Stem cavity occurred in the lower parts of stem for about 5 cm long as the stem grew to 20 cm, and the breakage increased to about 60 cm as the stem grew to 80 cm. Plant height, stem diameter, and flower stalk length were the highest in the planting density of $11cm{\times}11cm$. The light interception by plants increased as the planting density decreased (47, 99 and 143%). The strength and hardness values were the greatest, $567kg{\cdot}cm^{-2}$ and $1,339kg{\cdot}cm^{-2}$ in the planting density of $11cm{\times}11cm$. Plant height and plant weight increased in the foliage fertilization of one time, but the plants in the control flowered earlier. The strength and hardness increased in the foliage fertilization treatments and the greatest in the treatment of one time application of $60mg{\cdot}L^{-1}$ Silicate fertilization. The Si content of stems increased as the fertilization concentration and treatment time increased. The Si content in $30mg{\cdot}L^{-1}$ silicate treated twice was two times as high as that of the control.

Enhancement of Stem Firmness in Standard Chrysanthemum 'Baekma' by Foliar Spray of Liquid Calcium Compounds (액상 칼슘 화합물 엽면살포에 의한 스탠다드 국화 '백마'의 줄기 경도 강화)

  • This study was conducted to enhance the stem firmness of standard chrysanthemum 'Baekma' bred in Korea for commercial quality improvement and inhibition of stem breaking during transportation through foliar spray with calcium agents. Calcium agent screening 'Baekma' was examined using $CaCl_2{\cdot}2H_2O$, $Ca(NO_3)_2{\cdot}4H_2O$, and OS-Ca (natural liquid calcium compounds extracted from oyster shell) depending on each concentration (0, 0.001, 0.01, 0.1, and 1.0%, respectively). All calcium agents sprayed with 1.0% caused chemical injury such as stem bending or leaf burn. OS-Ca also showed more sensitive response to chemical injury than the other calcium agents because OS-Ca was absorbed very well by 'Baekma' leaves. Maximum stem firmness measured during the final harvest was greater in OS-Ca than in the other calcium agents. Especially, maximum stem firmness was greatest in 0.01% OS-Ca. However, elastic strength and maximum bending stress were greater in 0.001% OS-Ca than in the others. Thus, OS-Ca ranged from 0.005 to 0.05%, which did not show any chemical injury, was finally selected as the first candidate for hardening the stem of 'Baekma'. The next experiment using OS-Ca was conducted with the concentrations of 0, 0.005, 0.01, and 0.05%, respectively. From the results, 0.05% OS-Ca showed better plant growth and parameters such as plant height, stem diameter (upper and middle part), the number of leaves, and dry weights of each part than the other concentrations of OS-Ca and control. As for stem firmness depending on OS-Ca concentration, the Ca content within stem, maximum firmness, elastic strength, and maximum bending stress of stem in 'Baekma' sprayed with 0.05% OS-Ca showed the highest values among all the treatments and it turned out to be very high level of significance between control and OS-Ca treatments. However, the area and percentage of the inside cavity within horizontal stem section in 'Baekma' did not show any significance between any treatments including control. Thus, stem firmness of 'Baekma' did not show any correlation with the inside cavity area of stem. In conclusion, we recommend foliar sprays with 0.05% OS-Ca at vegetative growth stage to enhance stem firmness of 'Baekma' during transportation.

Reduction of Stem Cavity and Improvement of Flower Quality in Chrysanthemum 'Baekma' by Hydroponic Culture (양액재배에 의한 국화 '백마'의 줄기공동 경감과 절화품질 향상)

  • This study was conducted to investigate effect of ionic strength and feeding times of a nutrient solution on reduction of stem cavity size and improvement of flower quality in chrysanthemum 'Baekma'. A nutrient solution was applied with different strengths at three stages, namely, transplanting, budding, and flowering. The solution EC was adjusted as 1.61.82.0, 1.81.81.8, 1.82.01.8, and $2.02.02.0dS{\cdot}m^{-1}$ in four treatments. Feeding frequency per a day were 4 times for 12 min., 8 times for 6 min., 12 times for 4 min., and 18 times for 2.7 min. each. Cut flower length as affected by different strengths of a nutrient solution was the greatest in the plot of EC $2.02.02.0dS{\cdot}m^{-1}$. However number of leaves, stem diameter, and leaf size were greater in EC $1.82.01.8dS{\cdot}m^{-1}$ than in other treatments. Also, petal number of petals was the greatest and stem cavity size was the smallest in the plot of EC $1.81.81.8dS{\cdot}m^{-1}$. Plant height, number of leaves, stem diameter, leaf size were greater in the plot with 12 times feed ing per a day. number of petal was most in the plot with 8 times feeding per day, while stem cavity size was the smallest in the plot with 12 times feed ing per a day. Therefore, the better plant growth, the smaller stem cavity size.

  • Transformation system of rolC gene, dwarf gene in Lycium chinenese Mill. established by using system. Pin-punctured leaves induced numerous adventious buds in abaxial side when cultured on 3/2 MS medium containing 2.0 mg/L zeatin. Survival rate and shoot regeneration frequency of leaf explants decreased as kanamycin sulfate level increased. Shoot buds were not regenerated on 3/2 MS medium containing 10 mg/L kanamycin sulfate and 2.0 mg/L zeaein. Of the level tested, 10 mg/L of kanamycin sulfate was optimum in selection of kanamycin sulfate resistant plant. Co-culture time of bacteria and leaf explants was affected at the frequency of shoot regeneration and survival of leaf explants. Leaf explants co-cultivated during above 48hr severely decreased survival rate and shooting rate. Best result on survival rate and shooting rate were obtained when exposed for 24 h. 80 explants of 105 leaf explants survived on 3/2 MS medium containing 2.0 mg/L zeatin and 10 mg/L kanamycin sulfate, and 15 shoots was regenerated on the same medium. To select kanamycin sulfate resistant plant, regenerate as cultured on 3/2 MS medium containing 10 mg/L kanamycin sulfate, and obtained 5 kanamycin resistant plants. Southern blot analysis conformed that the rolC gene was incorporated into the genomic DNA of kanamycin resistant plants.

Analysis of Global Gene Expression Profile of Human Adipose Tissue Derived Mesenchymal Stem Cell Cultured with Cancer Cells (암세포주와 공동 배양된 인간 지방 조직 유래 중간엽 줄기 세포의 유전자 발현 분석)

  • Mesenchymal stem cells (MSC) are multipotent and can be isolated from diverse human tissues including bone marrow, fat, placenta, dental pulp, synovium, tonsil, and the thymus. They function as regulators of tissue homeostasis. Because of their various advantages such as plasticity, easy isolation and manipulation, chemotaxis to cancer, and immune regulatory function, MSCs have been considered to be a potent cell source for regenerative medicine, cancer treatment and other cell based therapy such as GVHD. However, relating to its supportive feature for surrounding cell and tissue, it has been frequently reported that MSCs accelerate tumor growth by modulating cancer microenvironment through promoting angiogenesis, secreting growth factors, and suppressing anti-tumorigenic immune reaction. Thus, clinical application of MSCs has been limited. To understand the underlying mechanism which modulates MSCs to function as tumor supportive cells, we co-cultured human adipose tissue derived mesenchymal stem cells (ASC) with cancer cell lines H460 and U87MG. Then, expression data of ASCs co-cultured with cancer cells and cultured alone were obtained via microarray. Comparative expression analysis was carried out using DAVID (Database for Annotation, Visualization and Integrated Discovery) and PANTHER (Protein ANalysis THrough Evolutionary Relationships) in divers aspects including biological process, molecular function, cellular component, protein class, disease, tissue expression, and signal pathway. We found that cancer cells alter the expression profile of MSCs to cancer associated fibroblast like cells by modulating its energy metabolism, stemness, cell structure components, and paracrine effect in a variety of levels. These findings will improve the clinical efficacy and safety of MSCs based cell therapy.

  • Since the Hwang scandal, the South Korean state has expressed often-conflicting interests of encouraging stem cell research and the IVF industry to save the country and introducing the ethical regulation in conformity with "Global Standard." As the tightening ethical regulation of stem cell research has enervated the field of human Embryonic stem cell(hESC) research, somatic stem cells (re-)emerged as an alternative savior that could rescue the future of research communities, bio-industry, practicing doctors, patients and the nation itself from the crisis. The recent literature on Korean biotechnology, however, mainly focus on hESC and relatively little attention has been given to the rapidly growing field of research on somatic stem cells like hematopoietic stem cells(HSCs) or Adipose derived stem cells(ASCs). While the hESC therapy is often regarded as experimental and ethically controversial, the HSCs or Mesenchymal stem cell(MSC) therapies have already made their ways into people's everyday life through market without much public discussion. Many ordinary people in South Korea are familiar with the story of patients who survived leukemia with the HSCs treatment; the number of doctors who are actively marketing the ASCs therapies is on the rapid increase; the concept of cosmetic products made from ASCs is gaining popularity among consumers. In this context, this article argues that the current ethical debates solely focusing on hESC or on the state policy and research regulation are too limiting to fully illuminate the politics of stem cell technologies in South Korea.

Co-author network for convergent research pattern analysis in stem cell sector (줄기세포분야 융합연구형태 분석을 위한 공저자 네트워크)

  • This study was carried out to confirm a convergent research pattern and researchers' role in stem cell sector by social network analysis. Articles were extracted from 1996 to 2012 in PubMed, 515 authors of 270 embryonic stem cell and induced pluripotent stem cell articles and 1,515 authors of 580 adult stem cell and mesenchymal stem cell articles. Degree(D) and betweenness(B) centrality was measured and co-author network was generated for researcher's role. As a result, Core researcher and Intermediary researcher was identified in co-author network. Core researcher had high D. centrality, otherwise high B. centrality or not. Intermediary researcher for convergent research had high B. centrality and low D. centrality. Conclusively, co-author network will be used as objective data not only to find core researchers in subject area for improving achievement but also to select experts for research project evaluation.

  • 생명공학의 시대로 일컬어 지고 있는 오늘날, 재생의학 분야에서는 난치성 질환 치료를 목적으로 활발한 연구가 진행되고 있다. 특히, 줄기세포를 이용한 세포 대체치료 관련 연구는 최근 국내 황우석 박사의 체세포 핵이식 배아세포주 확립에 이르기 까지 괄목할 만한 발전을 보여 주고 있다. 이와 관련해 생명보험산업에 적잖은 파장이 예상되며, 생명보험사 내부적으로 기존에 판매된 상품의 사차손 관리와 함께 급속도로 발전하는 줄기세포 연구에 직접적으로 대응하는 상품개발, 언더라이팅, 지급 심사 등 보험사 내외에서의 전방위적인 변화가 필요하다는 문제 제기가 있다. 줄기세포란 조직 분화 과정에서 볼 수 있는 세포이며 근육 뼈 뇌 피부 등 신체의 어떤 기관으로도 전환할 수 있는 만능세포로서, 간 폐 심장 등 구체적 장기를 형성하기 이전에 분화를 멈출 배아 단계의 세포를 말한다. 한편, 성체줄기세포는 조직이나 기관의 분화된 세포들 사이에서 발견되는 미분화 세포로써, 자기 스스로 증식할 수 있으며, 조직이나 기관의 특수한 기능을 가지고 있는 세포로 분화할 수 있는 능력을 가진 신체줄기세포를 말한다. 배아줄기세포와 생체줄기세포를 통한 장기이식 등 난치병 정복은 윤리적, 사회적으로 많은 논란이 예상되며, 기술적으로도 해결해야 할 문제점들이 산적해 있기 때문에 아직은 요원한 것이 사실이다. 현재 유럽 대부분의 나라와 미국에서는 인간 배아의 복제가 금지되어 있으며, 일본 정부는 연구용 배아 복제를 제한적으로 허용하고 있다. 하지만, 우리 나라의 경우 2005년 1월에 '생명윤리 및 안전에 관한 법'이 발효되었지만 정부는 관련 부작용에 대한 깊은 고찰 없이 전폭적인 지원들 약속하고 있는 실정이다. 줄기세포 연구의 발달로 인해 인류가 난치병 치료의 첫 장을 열었다고 하더라도 그 영향이 당장 보험사에 미친다고 할 수는 없다. 왜냐하면 앞으로 이러한 신기술이 실제 의료행위에 적용되기 위해서는 여러 단계의 안정화 작업과 임상시험이 필요한데 이러한 작업이 기술적으로 어렵고 그 시간도 만만치 않게 걸리기 때문이다. 또한, 보험사의 보장은 크게 사망/수술/입원/암/기타보장으로 구별할 수 있는데, 줄기세포 연구의 발달과 관련이 있는 보장이 제한되어 있어 보험사에 미치는 영향이 당장 우려할 만한 수준이라 할 수 없다. 하지만 만약 치료용 줄기세포 배양으로 인한 장기 기관의 이식이나 손상세포의 대체 등과 같은 의학신기술의 예상 외로 급격하게 발전한다면 보험사의 Risk 관리에 상당한 저해요인으로 작용할 것으로 판단된다. 특히 진단 입원 수술로 대표되는 생존보장에 대한 사차 Risk 및 사차손의 급증이나 역선택 증가는 보험사의 경영수지 악화를 유발하여 보험산업 전반에 위험으로 작용할 수도 있다. 따라서, 장기적인 안목으로 업계 공동의 대응이 필요하고, 각 사에서도 상품개발, 언더라이팅, 지급심사 간의 긴밀한 협조가 요구된다. 생명보험산업의 Risk 관리는 기존의 시장환경에 영향을 받는 비차, 이차중심에서 보험회사가 어느 정도 관리를 통해 적정규모를 유지할 수 있는 사차로 그 중심축이 이동하고 있다. 보험산업이 계속 활력을 갖고 성장하기 위해서는 체계적인 Risk나 관리가 핵심일 것이며, 보험사의 사차 Risk의 중요성이 더욱 커져 가고 있는 현실에서 거시적으로 의학신기술 발달 등 위험요인에 대해 미리 분석하고 이에 대한 대비책 마련이 필요할 것으로 판단된다.

Selection of Herbicide Resistant Potatoes Transformed with Phosphinothricin Acetyltransferase Gene (Phosphinothricin Acetyltransferase 유전자 도입에 의한 제초제 저항성 감자의 선발)

  • This experiment was conducted to introduce PAT (phosphinothricin acetyltransferase, non-selective herbicide bialaphos resistant gene) gene into potato (Solanum tuberosum. cv. Desiree). Optimal shoot regeneration from leaf discs and stem segments was obtained in MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L BA, and the frequency of shoot regeneration was 54% in left discs and 46% in stem segments. In this condition, leaf discs and stem segments of potato were co-cultivated with A. tumefaciens MP90 which contained binary vector with GUS: :NPTII gene and PAT gene. Transgenic shoots were regenerated from leaf and stem-derived calli on selection medium with 100mg/L kanamycin. The 100${\mu}M$ acetosyringone treatment during the co-cultivation highly enhanced(4 times than the control) the shoot regeneration on selection medium. When the putative transgenic plants were transferred to medium with 10mg/L basta, all of them were survived. After PCR. GUS test, and Southern blot analysis of the survived plant, we confirmed that the gene was stably integrated into the potato genome and expressed. After the transgenic plants were transplanted in soil, and the transgenic plants were sprayed with the herbicide basta (300ml/10a), the transgenic plants remained green but control plants were died.
