Bambusae Calulis in Liquamen (Jukryuk) and Zingiberis Rhizoma Juice's (Saengkang- juice's) Effect on Ischemic Damage Secondary to MCA Occlusion in Mice (죽력과 생강즙이 중대뇌동맥 폐쇄에 의한 뇌허혈 손상에 미치는 영향)

  • Objective : The purpose of this study was to investigate the effect of Jukryuk and Saengkang-juices on cerebral vascular ischemia (CVI) of the middle cerebral artery (MCA). Method : By admiuistration Jukryuk and Saengkang-juices, we compared treated groups with untreated groups, in view of five points as follows: 1) cerebral damage; 2) damaged area of ischemia; 3) cerebral edema; 4) the number of neuronal cells adjacent to the areas damaged by ischemia; and 5) the number of neuronal cells adjacent to the areas damaged by ischemia Results : In this experiment, the effect of Jukryuk and Saengkang-juices was determined by inducing cerebral vascular ischemia after occluding the middle cerebral artery (MCA) in mice, and making observations and comparisons such as alterations in damaged areas and neuronal cellular changes in the brain. Conclusions : According to the above results, Jukryuk and Saengkang-juices can protect the cerebral vascular ischemia.

Effects of Bambusae Caulis in Liquamen on the Stress Proteins Induced by Heating in Endothelial Cells (혈관내피세포에 열 충격 부과시 죽력이 stress proteins의 발현에 미치는 영향)

  • We have previously observed that Bambusae Caul is in Liquamen (BCL) stimulates the adipose conversion of 3T3-L1 cells and molecular chaperones were involved in the process of the assembly and replacement of laminin subunits in Bovine aortic endothelial cells(BAEC). Endothelial cells are exposed to continuous shear stress due to the blood flow. Heat shock protens(hsp) are a well-known stress response protein, namely, stress proteins. To investigate effects of BCL on the stress proteins induced by heating in endothelial cells, we have analyzed synthetic amounts of stress proteins in sodium dodecyl sulfate gel electrophoresis under reducing conditions. Under the condition of heating stress, BCL inhibited the synthesis of stress proteins in endothelial cells. These results suggest that BCL may have an important role for expression of stress proteins induced by heating in endothelial cells.

Effects of Mixture of Yukgunja-tang and Bambusae Caulis in Liquamen on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (육군자탕(六君子湯)과 죽력(竹瀝) 혼합물이 국소 뇌혈류량 및 평균혈압에 미치는 효과)

  • The study was designed to investigate the effects of Mixture of Yukgunja-tang and Bambusae Caulis in Liquamen (YTBCL) on the change of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats, and further to determine the mechanism of action of YTBCL. The results in rats were as follows ; YTBCL 25 ${\mu}l$ significantly decreased rCBF and MABP compared with basal condition. YTBCL 100 ${\mu}l$ significantly increased rCBF compared with basal condition, but decreased MABP compared with basal condition. YTBCL 50 ${\mu}l$ significantly increased rCBF compared with basal condition, but MABP was somewhat decreased compared with basal condition. The VTBCL 50 ${\mu}l$-induced increase in rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g$/kg, i.p.), an inhibitor of guanylate cyclase and indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. The YTBCL 50 ${\mu}l$-induced decreased MABP significantly increased by pretreatment with methylene blue but was inhibited by indomethacin. This results were suggested that the mechanism of YTBCL was mediated Dy guanylate cyclase.

Revaluation of manufacturing process of Bambusae Caulis in Liquamen Based on Traditional Medical Texts (죽력(竹瀝)의 올바른 제법 고찰)

  • Objective : Bambusae Caulis in Liquamen has been used by East Asian doctors who approved efficient clinical effects of it since the early stage of medical history. It is commonly used for lifestyle related diseases including stroke in recent days thanks to the increasing number of manufacturers. However, products from pharmaceutical companies as well as food companies are not classified as Bambusae Caulis in Liquamen but Bamboo vinegar. This study aims to discern disparate manufacturing process. Methods : In this study, original texts were searched to corroborate the correct method to produce Bambusae Caulis in Liquamen. It is essential to inspect the original texts thoroughly in the course of modernizing traditional knowledge. Result : Discrimination between the two substances starts from manufacturing process; Bambusae Caulis in Liquamen is made under the temperature of less then $170^{\circ}C$, containing mild fragrance and sweet taste, and the Bamboo vinegar over $170^{\circ}C$ upto $400^{\circ}C$, carrying strong smoky scent and acidness. Although some researches show conforming clinical actions of both products, still there is a possibility that closer clinical study would reveal their discrepancy. Conclusion : Current use of Bambusae vinegar needs speculation. Authenticity in proper manufacturing process can be attained through passed-on medical texts.

Effects of Mixed Extracts with Acanthopanax chiisanensis and Bambusae Caulis in Liquamen on the Blood Sugar of Diabetic mice induced with Streptozotocin (오가피와 죽력 배합약물이 Streptozotocin으로 유발된 당뇨 생쥐에 미치는 영향)

  • This study was carried out to investigate the optimal mixed extract with Acanthopanax Chiisanensis Nakai in order to strengthen anti-diabetic effects on the hyperglycemia induced by streptozotocin in mice. The original Bambusae Caulis in Liquamen filtered and refined. The effects of Acanthopanax Chiisanensis Nakai with Bambusae Caulis in Liquamen were administered to mice for 4weeks and its anti-diabetic effect examined. Mice used in this experiment were divided into two groups and distrilled water(control), Acanthopanax Chiisanensis Nakai mixed with refined Bambusae Caulis in Liquamen(AC+BCL.D) were given orally for 28days respectively. And then, experimental groups were observed in terms of blood sugar, creatinine, BUN and GPT. The amount of glucose was significantly decreased compared with the control group(P < 0.01). The amount of creatinine was decreased compared with the control group(P < 0.05). The amount of GPT did not show any differences between two groups.

Effect of Bambusae Caulis in Liquamen manufactured by Different Production Process and Silk Worm Powder on Blood Sugar in db/db Mice (생산공법차이에 따른 죽력과 누에가루를 배합한 약물이 db/db mouse의 혈당강하에 미치는 영향)

  • This study was carried out to understand the effects of Bambusae Caulis in Liquamen manufactured by different production process mixed with Silk Worm Powder on blood sugar in the db/db mice. Refined Bambusae Caulis in Liquamen D(L-BCL.D and H-BCL.D) manufactured by low or high temperature production process and original Bambusae Caulis in Liquamen producted by Hanlim pham Co. (BCL) and Silk Worm Powder were used. The effects of L-BCL.D+Silk Worm Powder, H-BCL.D+Silk Worm Powdr and BCl+SWP were observed in terms of blood sugar, creatinine, BUN, GPT in db/db mice. The amount of glucose was significantly decreased (P < 0.01) in the expremental groups compared with the control. The amount of Creatinine did not show any differences among four groups. The amount of blood urea nitrogen observed significant decrease in the case of BCL group. The amount of GPT did not show any differences among four groups.

Property Comparison of Purified Bambusae Caulis in Liqamen extracted at low temperature (정제방법에 따른 저온추출 죽력의 특성비교)

  • Jookrhyuk (Bambusae Caulis in Liqamen) was extracted at low temperature (100℃ - 160℃) and purified by adsorption and distillation and their physicochemical properties were compared. In case of colour among all Jookrhyuk samples the L value appeared to be higher in the purified Jookrhyuk,and the b value appeared to be higher in crude Jookrhyuk. Purified Jookrhyuk showed much lower level of total sugar and tar contents than crude Jookrhyuk. A considerable reduction in concentration of methanol and phenolic compounds was achieved by purification process, whereas the content of acetic acid was increased. Therefore it was suggested that adsoption and distillation could be used as one of effective purification methods of various Joockrhyuk samples.

  • Jookrhyuk (Bambusae Caulis in Liqamen) is one of the most important ingredients for treating Diabetes mellitus. Various aspects of quality of Jookrhyuk produced in Korea depending on carbonization process were compared. The pH Value was the highest in Jinyoungsangsa Jookrhyuk(pH 3.17) among the three different Jookrhyuk samples. Damyangsan Jookrhyuk that contains high level of total sugars(8.0 Brix, %) show incresed level of tar contents, it exhibited positive realationship between total sugar and tar contents. Concentration of methanol and phenolic compounds which are related to toxicity were the highest in Damyangsan Jookrhyuk(1.34㎎/㎖, 3.84㎎/㎖ respectively). The harmful heavy elements were not found in the evaluated Jookrhyuk samples. In coclusion, it was suggested that Bambusae Caulis in Liqamen should be further puryfied for using as main oral medicines for treating Diabetes mellitus.

  • The purpose of this research was to investigate effects of Bambusae Caulis in Liquamen(BCL) on T-lymphocytes and peritoneal macrophages in mice. The apoptosis and subpopulation of T-lymphocytes were tested using a flow cytometer. The phagocytic activity of mouse peritoneal macrophage was tested using a luminometer. Nitric oxide production was tested using a Griess reagents. BCL induced T-lymphocytes apoptosis. BCL increased $T_H$ cells population and decreased $T_C$ cells population of T-lymphocyte, but did not affect splenocytes subpopulation. BCL increased nitric oxide production and phagocytic activity of peritoneal macrophage in mice. These results suggest that BCL regulates the immune system in consequence of an increase in helper T cell population and macrophages activation.

  • This study was carried out to understand the effects of Bambusae Caulis in Liquamen on blood sugar in the db/db mice. Refined Bambusae Caulis in Liquamen C. D(BCL.C. D)manufactured by high temperature production process and Bambusae Caulis in Liquamen(H-BCL) manufactured & distributed by HANLIM PHARM.COM., LTD were used. The Bambusae Caulis in Liquamen extracted from bamboo charooal manufacturing process was filtered and refined. The effects of Bambusae Caulis in Liquamen were administered orally to mice for 6weeks and its anti-diabetic effect examined. The effects of BCL.C. D and H-BCL were observed in terms of blood sugar. creatinine. BUN and GPT in db/db mice. The results were as follows : The amount of glucose was slightly decreased (P<0.05) in the B CL.C-treated groups compared with the control. The amount of glucose was significantly decreased (P<0.01) in the BCL.D and H-BCL-treated groups compared with the control. The amount of creatinine did not show any differences among four groups. The amount of blood urea nitrogen did not show any differences in the case of BCL.C-treated groups. but observed significant decrease in the case of BCL.D and H-BCL-treated groups. The amount of GPT did not show any differences in the case of BCL.D-treated groups. but observed significant increase in the case of BCL.C and H-BCL-treated groups.
