Antioxidant Activity and Fatty Acid Aomposition of Beach Rose (Rosa rugosa) Fruit and Seed from Various Ripening Stages (해당화 열매 과육과 종실의 숙기별 항산화능 및 지방산 조성)

  • 해당화 열매를 숙기 별로 과육의 항산화물질 함량과 씨앗의 지방산 함량을 측정하여 평가하였다. 과육의 1,1-diphenyl-2-picrylhydrazyl (DPPH) 항산화능을 갈산 환산농도로 나타내면 미성숙 과육은 $4.36{\pm}0.03mg/g$, 성숙 초기 과육은 $4.54{\pm}0.37mg/g$, 성숙 중기 과육에는 $4.78{\pm}0.21mg/g$, 성숙 후기에는 $4.30{\pm}0.22mg/g$ 범위로 항산화능을 보여주었다. 항산화능력은 성숙중기에서 높은 함량을 보여주었고 성숙함에 따라 감소함을 보여주었다. 미성숙, 성숙 초기는 유의한 걸로 나오지만 성숙 중기와 성숙 후기은 다른 성숙도와 유의하지 않았다. 총 페놀 함량의 미성숙 과육은 $1.60{\pm}0.03mg/g$, 성숙 초기 과육은 $1.80{\pm}0.08mg/g$, 성숙 중기 과육에는 $2.03{\pm}0.67mg/g$, 성숙 후기에는 $1.53{\pm}0.07mg/g$ 범위로 폴리페놀이 존재하였다. 성숙 초기에 제일 높은 항산화능력과 폴리페놀 함량을 보여주고 있으며 성숙할수록 항산화능력과 총 페놀 함량이 떨어지는 것을 알 수 있다. 씨앗의 지방산 함량은 리올레산, 리놀렌산, 올레산, 팔미트산 순으로 많았다. 리올레산은 미성숙 씨앗은 전체 함량 중 56.46%, 초기 과육 씨앗에는 54.26%, 중기 과육 씨앗에는 51.67%, 후기 씨앗에는 50.75%의 함량을 가지고 있다. 리올렌산은 미성숙 씨앗은 전체 함량 중 18.80, 초기 과육 씨앗에는 17.96%, 중기 과육 씨앗에는 18.90%, 후기 씨앗에는 21.42%의 함량을 가지고 있다. 올레산은 미성숙 씨앗은 전체 함량 중 13.54%, 초기 과육 씨앗에는 20.78%, 중기 과육 씨앗에는 22.44%, 후기 씨앗에는 20.81%의 함량을 가지고 있다. 팔미트산은 미성숙 씨앗은 전체 함량 중 6.80%, 초기 과육 씨앗에는 3.40%, 중기 과육 씨앗에는 3.37%, 후기 씨앗에는 3.32%의 함량을 가지고 있다. 리올레산은 미성숙 시기에 가장 많았고 리올렌산은 후기에 올레산은 중기에, 팔미트산은 초기에 많았다.

  • The civil engineering materials used to stabilize the slopes of new riverbanks have a great impact on the types and growth of vegetation introduced after the completion of construction procedure. Recently, microbial-derived, ${\beta}$-glucan- and xanthan gum-based biopolymers are attracting attention as an ecofriendly strengthening material of riverbanks that can possibly stimulate plant growth. This study aimed to assess ecological effects of biopolymer application on native plants in Korean riverbanks. In particular, since dominant plant species could shape characteristics of an ecosystem, we examined the effects of biopolymer on the dominant plant species in riverbanks. Overall, biopolymer did not affect seed germination rates of testing plant species. In contrast, plants grew more vigorously in the soil mixed with biopolymer compared to those in the control soil. The biomass of Echinochloa crus-galli especially increased around two times more in the biopolymer treatment. Plants produced heavier root biomass and leaves with larger specific leaf area, which possibly contributes to the tolerance of environmental stress like drought. These results suggest that biopolymers treated on river banks are expected to stimulate plant growth and increase stress tolerance of domestic dominant plant species.

Effects of Temperature and Photoperiod on the Growth of Tatary Buckwheat(Fagopyrum tataricum) (온도 및 일장처리가 쓴 메밀(Fagopyrum tataricum)의 생육에 미치는 영향)

  • This study was carried out to investigate the effects of temperature and photoperiod on the growth of Fagopyrum tataricum. It showed a tendency to promote the germination rate and plant height of tartary buckwheat under high temperature. Plant height and number of leaves were promoted in the order of 25 > 20 > 15 > 3$0^{\circ}C$ under the different temperature after 30 days of the planting. In the 10, 12, 14hr photoperiods at 2$0^{\circ}C$, plant height, number of leaves and dry weight were increased as much as photoperiod became long except by 16hr. Rutin contents was not regular both common and tartary buckwheat under the photoperiods. The rutin content in leaves was higher than stem. Compared to yield of tartary and common buckwheat, plant height, number of leaves, number of branch and stem diameter of tartary buckwheat increased more than common buckwheat. Especially, number of leaves was increased about 2.5 times more than common buckwheat. And there was a difference in about 2.1 times in grain weight and about 5.4 times in number of grains. Rutin content was 1469.8mg/l00g in grain of tartary buckwheat. It was about 60 times higher than 22mg/100g in grain of common buckwheat.

Mid-late Maturing Male Sterile Line of Onion (Allium cepa L.) 'Wonye 30001' (양파 중생계 웅성불임 중간모본 '원예 30001')

  • A new intermediate parent 'Wonye 30001' (Allium cepa L.) was developed by Bioenergy Crop Research Center, NICS in 2009. As a male sterile line, 'Wonye 30001' could be used for hybrid seed production by crossing with pollen parent. The first cross was conducted in 2000. Male sterile plants were selected from 'Ginque' breeding lines and fertile plants were selected from YG-1-1. The male sterile line 'Wonye 30001' has circular bulb and bulb weight of 260 g. As mid-late maturing type, lodging date is around May 20. Plant height and stem diameter are 57 cm and 15.9 mm, respectively. In seed harvesting characteristics, number of flower stalks and the length are 6 and 135cm, respectively. Flowering date of 'Wonye' is May 23 and is completely male sterile. 'Wonye 30001' is a promising male sterile line for hybrid bulb onion seed production.

Morphological and Progeny Variations in Somaclonal Mutants of 'Ilpum' (Oryza sativa L.) ('일품'벼 체세포변이체의 표현형과 후대변이)

  • A total of 424 plants was regenerated from the seed-culture of a rice cultivar, 'Ilpum'. The regenerated plants were grown in a greenhouse. The 297 plants with high fertility were selected among 424 plants. The harvested seeds from each plant were planted to each line at experiment field in 2008 and 2009. The each line was evaluated for the agronomic and morphological traits, also. The 64 lines (21.5%) showed significant differences in agronomic and morphological traits from donor cultivar 'Ilpum' among 297 lines. The heading date different from donor cultivar 'Ilpum' showed highest frequency in 297 lines, and accounts for 9.1% (29 lines). The phenotype of opaque endosperm and rolling leaf account for 1.7% and 1.3% in 297 lines, respectively. The genetic segregation was observed in dwarf/semi-dwarf, rolling leaf and opaque endosperm at $S_1$ generation, but not in $S_2$ generation. These results suggest that the mutant derived from a tissue-culture will be one of the promising genetic resources, due to its wide variation and high frequency of mutation, comparatively.

Breeding of a Red Rose Cultivar, 'Noble Red' with Good Color and Shape (화형과 화색이 우수한 적색 스탠다드 장미 '노블레드' 육성)

  • A new standard rose cultivar 'Noble Red' was bred by the cross between the pink standard cultivar 'Silba 87' and the red standard cultivar 'Bravo' at the National Institute of Horticultural and Herbal Science. The cross was made in 1999 and 'Noble Red' was finally selected in 2003 after investigating the characteristics such as flower Color, Fragrance, stem length, stem diameter, weight, and number of leaves for three years from 2001 to 2003. 'Noble Red', a red standard cultivar grows vigorously and has good flower shape. The major characteristics of this cultivar are $125stems/m^2/year$ in yield, 71.9 cm in length of cut flower, 6.0cm in flower diameter, 30.8 in petal number, and 9.2 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cultivar, 'Rote Rose'.

'Jopung', A New Forage Winter Oat Cultivar for the Southern Regions of Korea (남부지역 적응 조숙 다수 조사료용 월동귀리 신품종 '조풍')

  • 'Jopung' (Avena sativa L.), a winter oat for forage use, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, NICS, RDA in 2009. It was derived from an original cross between the $F_1$ hybrid of 'Early80' and 'PA202-210-S' and 'Walken' in 1999. Subsequent generations followed by the cross were handled in bulk and pedigree selection programs at Suwon and Yeoncheon, respectively. A line, 'SO99006-B-YB-29-Y1', was selected for earliness and good agronomic characteristics and designated as a line name of 'Gwiri65'. The line 'Gwiri65' was subsequently evaluated for earliness and forage yield in four different locations, Yesan, Iksan, Kimjae, and Jeju, from 2007 to 2009 and finally named as 'Jopung'. Over 3 years, the average forage dry matter yield of cultivar 'Jopung' harvested at milk-ripe stage was $17.3MT\;ha^{-1}$, compared with $13.6MT\;ha^{-1}$ of check cultivar 'Swan'. The silage quality of 'Jopung' was superior to the check cultivar 'Swan' in crude protein content, and was similar to the check cultivar in percent total digestible nutrients (TDN). Cultivar 'Jopung' is suitable primarily for winter planting in the southern regions of Korean peninsula.

Breeding of a White Rose Cultivar, 'Snow Ball' with Good Color and Good Vase Life (절화수명이 긴 백색 스탠다드 장미 '스노우 볼' 육성)

'Snow Ball', a white standard cultivar grows vigorously and has good flower shape. The major characteristics of this cultivar are $90.3stems/m^2/year$ in yield, 57.8 cm in length of cut flower, 6.1 cm in flower diameter, 85.7 in petal number, and 15.0 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer's preference of this cultivar is relatively higher than that of control cultivar, 'Tineke'.

The effects of Stigma Removal and Auxin Treatment on Fruit Shape and Quality in Strawberry (딸기의 주두 제거와 옥신 처리가 딸기의 과형 및 품질에 미치는 영향)

  • The effects of stigma removal from floral organ and treatment of auxin on those regions were evaluated on the fruit growth and quality in strawberry (Fragaria ananassa Duch.). The removal of stigma reduced the fruit weight, fruit length, and fruit width, while fruit growth declined as the number of removed stigma increased. The removal of stigma also resulted in reduced fruit firmness, sugar content, and total anthocyanin content, and decreased value of Hunter a for red color and increased value of Hunter b for yellow. The number of seeds deceased as the regions of removed stigma increased. The treatment of auxin on the region of removed stigma improved fruit size, while it reduced fruit firmness, sugar content, and anthocyanin content. The delay in fruit ripening was observed from both stigma removal and auxin treatment. The incidence of malformed fruits increased as the regions with removed stigma increased. The treatment of auxin also promoted fruit malformation.

Influence of Abiotic Treatments on Isoflavone Accumulation in Soybean Seeds during Germination (물리적 요인이 콩 발아 중 isoflavone 함량에 미치는 영향)

  • Isoflavones, a group of secondary metabolites present in plants and especially abundant in soybeans, are garnering increasing interest because of their medicinal properties. Numerous environmental factors influence isoflavone accumulation in plants. Successful attempts to increase isoflavone contents in soybean seeds have not been widely reported. In this report, we examined an effective method for producing a high amount of isoflavones during soybean seed germination, by treating the seed with 4 abiotic factors. Soybean seeds were exposed to ultrasound (50 Hz) and heat ($40^{\circ}C$) for 30minutes, then germinated for 5 days in darkness. Another sample was subjected to water deficit during germination in darkness, and the other sample was subjected to light conditions during the germination period. The results showed that the ultrasound treatment increased isoflavone content during soybean germination. Three days after germination, the ultrasound-treated seeds presented the highest isoflavone content (6,042.1 ug/g dry weight) compared to the other treatments, which was also 1.3-fold increases over the content of the raw seed's before germination. With respect to each type of isoflavone group, aglycone (partially genistein) content was markedly higher, whereas glucoside was lower than that of the untreated samples. In other experiment, total isoflavone content after heat, water deficit, and light treatments was 5,600.0, 4,740.1, and 5,631.4 ug/g, respectively. These present work suggests that ultrasound treatment is a novel approach for improving the production of isoflavones in soybean seeds during germination.
