Lychnis wilfordii (Regel) Maxim. is one of the wetland specialists mainly distributed in peatlands at high latitudes. In Korea, it is isolated in two regions. This study investigated habitats, growth traits, and self-compatibility of L. wilfordii and assessed the internal dynamics of its population persistence. Its population has remained stable in the Yongneup Wetland Protected Area (YWPA). There was a clear difference in vegetation environment between YWPA and the distribution area in Daegwallyeong (DWL), Pyeongchang-gun. It has self-compatibility while pollinators facilitate its seed production. It produces a large number of hibernacles and bears the maximum number of branches and fruits in soil with rich organic contents. However, it grows and bears fruits even under the condition of low organic contents. In YWPA, L. wilfordii is not distributed in high moor but widely distributed in low moor where tussocks by Carex thunbergii var. appendiculata are developed. It is mainly distributed on the top of tussocks also. Therefore, it is judged that the formation, growth, and extinction of tussocks by C. thunbergii var. appendiculata is closely related to the establishment, growth, and extinction of plants distributed in this space. It is assessed that the current YWPA has well-developed tussocks in which L. wilfordii is widely distributed, and extinction and re-establishment progress well. Accordingly, the L. wilfordii population is expected to be sustainable in the long term given if its current ecological process is maintained well.

Homoharringtonine isolated from the plumyew tree (Cephalotaxus koreana) is currently considered as one of the most promising chemotherapeutic agents for anti-cancer. Variation of homoharringonine contents from eight natural populations of Korean native plumyew trees was determined by HPLC (high performance liquid chromatography), and compared with the variation among populations, the variation of the different parts(needle, root, stem and seed), and the variation according to their ages and growth features. Homoharringonine contents among populations were significantly different with Mt. South Dukyu ($1,048{\mu}g/g$) having the highest and Mt. Obong having the lowest. The analyses of plant pans showed that the homoharringtonine contents were highest in plant needles ($874{\mu}g/g$), and followed by roots, stems and seeds. Homoharringtonine contents of the plumyew trees decreased about 30% in increment with their ages and growth.
Salt cress (Thellungiella halophila or Thellungiella parvula), species closely related to Arabidopsis thaliana, represents an extremophile adapted to harsh saline environments. To isolate salt-tolerance genes from this species, we constructed a cDNA library from roots and leaves of salt cress plants treated with 200 mM NaCl. This cDNA library was subsequently shuttled into the destination binary vector [driven by the cauliflower mosaic virus (CaMV) 35S promoter] designed for plant transformation and expression via recombination- assisted cloning. In total, 305,400 pools of transgenic BASTA-resistant lines were generated in Arabidopsis using either T. halophila or T. parvula cDNA libraries. These were used for functional screening of genes involved in salt tolerance. Among these pools, 168,500 pools were used for primary screening to date from which 7,157 lines showed apparent salt tolerant-phenotypes in the initial screen. A secondary screen has now identified 165 salt tolerant transgenic lines using 1,551 (10.6%) lines that emerged in the first screen. The prevalent phenotype in these lines includes accelerated seed germination often accompanied by faster root growth compared to WT Arabidopsis under salt stress condition. In addition, other lines showed non-typical development of stems and flowers compared to WT Arabidopsis. Based on the close relationship of the tolerant species to the target species we suggest this approach as an appropriate method for the large-scale identification of salt tolerance genes from salt cress.
Sex ratios, and patterns of tree size and growth variation, resource allocation, spatial and age class distribution between the sexes were investigated in natural populations of the sexual trees, R. trichocarpa and R. sylvestris, and the mainly asexual, clonal tree, R. javanica of the dioecious Rhus (Anacardiaceae) distributed in Korea. Sex ratios for three species exhibited a significant degree of female bias, but among the populations, sex ratios were seen to vary quite widely. The measurement of tree size and annual increment of male trees in R. trichocarpa and R. sylvestris were significantly higher than those of female trees, but not significantly different in R. javanica. In all of the species, flowering branch number per individual and inflorescence number per branch of males outnumbered those of females. Branch number per individual, rachis(leaf) number per branch and rachis(leaf) number per inflorescence were more in females than in males. These results were considered as reproductive efforts to increase the pollen supply in males and the fruit production in females. Spatial distribution analysis in two different populations of R. trichocarpa indicated that males and females were randomly distributed in space, but seedlings were clumped around parental trees. Analysis of age class distributions between the sexual reproduction trees, R. triclaocarpa and R. sylvestris, and the asexual, clonal tree, R. javanica showed a different distribution in frequencies of males and females in each age class. These results showed that sexual and asexual reproduction tree species had almost different preference of habitats, and different sex ratio and annual growth.

Cultivars or genetic resources with a black seed coat and green cotyledons are rich in lutein, which can promote eye health, and anthocyanin, known for its numerous health benefits. However, mature seeds also contain P34, 7S α' subunit, and lectin proteins, which are allergenic and degrade quality. Here, we report the breeding of a new soybean line with a black seed coat, green cotyledon, and free of P34, 7S α' subunit, and lectin proteins. A total of 157 F2 seeds with black seed coats and green cotyledons were selected by crossing a female parent with a brown seed coat, green cotyledon, and lacking the 7S α' subunit and lectin proteins with a male parent with a black seed coat, green cotyledon, and lacking the P34 and lectin proteins. The P34 and 7S α' subunit proteins were consistent with a ratio of 9:3:3:1, indicating that they are independent of each other. From 14 F2 seeds that were recessive (cgy1cgy1p34p34) for both proteins, one individual F2 plant (F3 seeds) with the desired traits-black seed coat, green cotyledon, and lacking P34, 7S α' subunit, and lectin proteins- was finally selected. The triple null genotype (absence for P34, 7S α' subunit, and lectin proteins) was confirmed in random F3 seeds. The selected line has a black seed coat and green cotyledons, and when sown on June 14 in the greenhouse, the maturity date was approximately October 3, the height was about 68 cm, and the 100-seed weight was about 26.5 g.
Genetic differentiation of populations is resulted from the environmental and the genetic effects, and the interactions between them. Whereas, the major factors influencing to the genetic differentiation within populations are the gene flow induced by seed or pollen dispersial, the microsite heterogeneity, and the density-dependent distribution of individuals. For the purpose of studying spatial genetic structure and the distribution pattern of Korean pines(Pinus koraiensis), we set up one $100{\times}100m$ plot at a Korean pine stand in Quercus mongolica community on Mt. Jumbong in Korea. To estimate the coefficient of spatial autocorrelation as Moran's index and an analogue, simple block distance, isozyme markers were analyzed in 325 Korean pines. For 11 polymorphic loci observed in 9 enzyme systems, the average percentage of polymorphic loci, the observed and expected heterozygocity were 72.2% 0.200, and 0.251, respectively. It was revealed the excess of homozygotes was observed in the plot, which suggests that here may be more number of consanguineous trees than expected. On the basis of isozyme genotypes observed in this study, 325 trees were classified into 147 groups in which the maximum number of trees for one group was 34. From the distance class of 24-32m, the genetic heterogeneity began to increase. The variation of simple block distance against the growth performance by tree height and diameter also showed the same trend at 24~32m class. According to high fixation index(F=0.204), the spatial genetic structure within a stand, the analysis of the growth performance, and the distribution patterns of identical genotypes, we inferred that the genetic structure of a Korean pine stand in Mt. Jumbong has been maintained rather density-dependent mechanism than the gene flow, such as the pollen dispersial or the heavy input of seeds following the forest gaps. The genetic patchy size was determined between 24~32m, which suggests that the selection of individuals for the ex situ conservation of Korean pine in Mt. Jumbong may be desirable to be made with the spatial distance over 37 meters between trees.
Soybean [Glycine max (L.)] is a major source of protein for human and animal feed. Inter- and intra-genotype variation of soybean protein has been investigated by soybean researchers. However, limited sample amount of soybean single seed there is no report that investigated intra-plant variation of soybean protein within soybean plant. Recently a non-destructive NIR (near-infrared reflectance) spectroscopy using single seed grain to analyze seed protein was developed. The objectives of this study were to understand variation of seed protein content within plant and to determine the amount of minimum sample size which can represent protein content for a soybean plant. Frequency distribution of protein content within plant showed normal distribution. There was an intra-cultivar variation for protein content in soybean cultivar Seonnogkong. Difference of protein content among single plants of Seonnokong was recognized at 5% level. Seeds in lower position on plant stem tended to accumulate more protein than in higher position. There was significant difference for protein content between sample size 5 seeds and sample size of more than 5 seeds (10, 20, 30, 40, and 50 seeds) at a soybean plant with 57 seeds however no difference was recognized among sample size (5, 10, 20, and 30 seeds) at a soybean plant with 33 seeds. Around 20% seeds of soybean from single plant needed to determine the protein content to represent protein content of single soybean plant. This study is the first one to report evidence of intra-plant variation for proteincontent which detected by non-destructive NIR spectroscopy using single seed grain in soybean.

We investigated the delay of kimchi fermentation by the addition of plant extracts. Fifteen plant extracts were screened for inhibitory activity aginst Lactobacillus plantarum by using an agar well diffusion assay, and determined the minimal inhibitory concentration (MIC) and minimal lethal concentration (MLC) were determined. The lowest MIC for grapefruit seed extract (GFSE; 0.0313 mg/mL) was determined, followed by Caesalpinia sappan L. extract (CSLE; 0.25 mg/mL), and oregano essential oil (OREO; 1.0 mg/mL). GFSE, CSLE, and OREO were individually added to kimchi, and incubated the samples at 10 for up to 20 days. Results showed that the addition of GFSE (0.3 and 0.5%), CSLE (0.1, 0.3, and 0.5%), or OREO (0.5 and 1.0%) led to a significant increase in the pH of kimchi, and also a significant reduction in the numbers of lactic acid bacteria. Taken together, the addition of natural antimicrobial agents can delay kimchi fermentation.
The variation of needle, cone and seed characteristics and of allelic frequency in isoenzyme, ADH, ME and PGI, according to different resin duct index in Pinus densiflora, Pinus thunbergii and their hybrids was analyzed. The results obtained were as follows : 1. With increase of number of resin ducts, morphological characteristics such as needle length, needle sheath length, cone size, seed size, seed wing size, 1000 seeds weight, etc. tended to he increased, while number of stomata row in needle to be decreased. 2. As the results of discriminant analysis for the morphological characteristics of needle, cone and seed, most individuals are generally coincided with number of resin duct mostly in Pinus densiflora were not. 3. According to the canonical discriminant function obtained from the morphological characteristics in Pinus densiflora, Pinus thunbergii and their hybrids including introgressive hybrid, the resin duct index, 1000 seeds weight, cone size and neelde sheath length characterized fairly their species. 4. With increase of resin duct index, hybrid index tended to be higher. The results obtained from the discriminant analysis and the hybrid index were nearly same each other. 5. With increase of number of resin duct, the allelic frequencies for isoenzyme, ADH-$B_2$, ME-$A_2$ and PGI-$B_1$. $B_2$ tended to increase but those of ADH-$B_3$, ME-$A_4$, and PGI-$B_3$, to decrease. It is thought that this increase of frequency for the former four isoenzymes was resulted in introgressive gene flow from Pinus thunbergii to Pines densiflora and accordingly the frequency of latter three isoenzymes tended to decrease.
The germination responses of Echinochloa crus-galli (L.) Beauv. seeds to temperature were examined under the various thermal conditions. While almost all the seeds tested did not germinate immediately after collection, almost of the seeds which were stored for 7, 10, and 17 months showed very high germination percentages (85-95%) at their own constant temperatures between $16^{\circ}C\;and\;40^{\circ}C$. The total thermal time which was required for germination(10-70%) of Echinochloa crus-galli seeds ranged from 539Kh(degree Kelvin X hour) to 1,279Kh in accordance with the distribution function of thermal time, $F({\Theta})=1-[3D^{-3}({\Theta}-m+D)+1]^{-1/2}$, where m is 935Kh and D is 555Kh. Moist chilling treatment at $2^{\circ}C$ for 20 days increased the final germination percentage as well as the germination rate. In the increasing temperature(IT1 regime, E. crus-galli seeds started to germinate at $12^{\circ}C$. and showed greater germination rate with increasing temperatures, with the final germination percentage of 80%. On the other hand, in the decreasing temperature(DT1 regime, the seeds began to germinate at $12^{\circ}C$(10% germination) with the final germination percentage of 20%. An induced dormancy occurred at $4^{\circ}C$ in the DT regime.
