A Proposition for the Removal of Algae and Phosphorus from River Water Using Multi-Purpose filtration pond (다목적 여과저류지를 이용한 하천수의 조류와 인 제거방안 제안)

  • In an effort to find a solution to the eutrophication of major Korean rivers, a method to utilize multi-purpose filtration pond was investigated. As literature showed that oyster shell is known to be the most adequate for the removal of dissolved phosphorus in Korean rivers, batch and column experiments were performed using oyster shell as an adsorbent in this study. The results of the batch experiment showed that the removal of dissolved phosphorus from river water through adsorption as a way of preventing algal growth was not practical. The results obtained from the column experiment, however, suggested that oyster shell may be utilized as an adsorbent under limited conditions. Based on the results of the experiments a methodology was proposed to remove algae from river water through the use of multi-purpose filtration pond. This method involves mechanically removing the accumulated algae cake from the surface of the artificial stream in the pond towards the condensing part located at the lower reach of the stream, where particles gather before the final removal. In addition, employment of oyster shell as an adsorbent in the condensing part allows prevention of phosphorus released from the dead algae re-entering the river water.

Isolation of marine algicidal bacteria from surface seawater and sediment samples associated with harmful algal blooms in Korea (유해조류번성 주변의 해수와 침전물에서 살조균의 분리)

  • This study mainly focused on isolation of marine algicidal bacteria associated with phytoplankton blooms and characterization of algicidal activity against harmful algae. Harmful algal blooms (HABs) found naturally in surface waters have caused many environmental problems worldwide. In this study, forty bacterial strains that have capability of inhibiting harmful algal growth were isolated from Masan Bay, Jinhae Bay, Dol Island, Jangmok Bay, and the Tongyeong Sea, Republic of Korea. The bacteria were screened furthermore for the characteristics on algicidal activities against Cochlodinium polykrikoides, Chattonella marina, Skeletonema costatum, Heterosigma akashiwo, Heterocapsa triquetra, Prorocentrum minimum, and Scrippsiella trochoidea. As a result, the algicidal bacteria that were screened from double over layer agar and microscopic counts tests belonged to genera Pseudomonas, Vibrio, Bacillus, Pseudoalteromonas, Ruegeria, Joostella, Marinomonas, Stakelama, Porphyrobacter, and Albirhodobacter. One of the most important HAB species is Co. polykrikoides and the strongest algicidal activity against the dinoflagellate was 94.00% after 6 h treatment with 10% bacterial culture filtrate. In this study, Marinomonas sp. M Jin 1-8, Stakelama sp. ZB Yeonmyeong 1-11 & 1-13, Porphyrobacter sp. M Yeonmyeong 2-22, and Albirhodobacter sp. 6-R Jin 6-1 were found to be as new genera of bacteria having anti-algal activity. These results suggest that these bacteria might play an important role in controlling phytoplankton blooms.

Assessment of the possibility as Cosmetics Materials by Brown algae from Jeju Island Using Supercritical Fluid System (초임계 유체 기술을 이용한 제주산 갈조류의 화장품 소재로서의 가능성 평가)

  • Although various biological extracts derived from the resources from Jeju are currently being developed for cosmetic materials, few studies using Brown algae from Jeju have been conducted. This study identified materials with high antioxidant levels and examined their whitening effects to assess their feasibility as potential cosmetic resources. Brown algae from Jeju (Undaria pinnatifida (Harvey) Suringar, Laminaria japonica, Ecklonia cava, Hizikia fusiforme (Harvey) Okamura (HF), and Sargassum fulvellum (Turner) C. Agardh (SF) were processed using a supercritical fluid system at 50℃ and 400 bar, and their total phenol content, antioxidant activity, tyrosinase inhibition, and primary skin irritation were measured. Of all the samples, only SF using the supercritical fluid system (SFS) displayed concentration-dependent behavior for the total phenol content (p<0.05). Among the samples, the antioxidant activity of SFS was significantly higher (p<0.05) and the one increased at higher concentrations (p<0.05). On the other hand, tyrosinase was inhibited to a greater extent with HF than the others. The primary skin irritation tests showed that none of the samples were skin irritants. This study confirmed the feasibility of using Phaeophyceae from Jeju as potential cosmetic materials, where the samples can be selected and utilized based on the required physicochemical properties.

Identification of Alga-lytic Bacterium AK-07 and Its Enzyme Activities Associated with Degradability of Cyanobacterium Anabaena cylindrica (Anabaena cylindrica 분해세균 AK-07의 동정과 분해 관련 효소활성 조사)

  • To investigate bacteria with algal Iytic activities against Anabaena cylindrica when water blooming occurs and to study enzyme profiles of alga-Iytic bacteria, various bacterial strains were isolated from surface waters and sediments in eutrophic lakes or reservoirs in Korea. Abacterial strain AK-07 was characterized and identified as Acinetobacter johnsonii based on its16S rDNA base sequence. When AK-07 was co-cultivated with A. cylindrica, bacterial cells propagated to $8\;{\times}\;10^8$ cfu $ml^{-1}$ and Iyses algal cells. However, culture filtrates of AK-07 did not exhibit algal Iytic activities. That suggesting the enzymes on the surfaces of the bacterium might be effective algal Iytic agents to cause Iyses of cells. Acinetobacter johnsonii AK-07 exhibited high degradation activities against A. cylindrica, and formed alginase, caseinase, lipase, fucodian hydrolase, and laminarinase. Moreover, glycosidases for example ${\beta}$-galatosidase, ${\beta}$-glucosidase, ${\beta}$-glucosaminidase, and ${\beta}$-xylosidase, which hydrolyzed ${\beta}$-0-glycosidic bonds, were found in cell-free extracts of A. johnsonii AK-07. Other glycosidase such as ${\alpha}$-galctosidases, ${\alpha}$-N-Ac-galctosidases, ${\alpha}$-mannosidases, and ${\alpha}$- L-fuco-sidases, which cleavage ${\alpha}$-0-glycosidic bondsare not detected. In the results, enzyme systemsof A. johnsonii AK-07 were very complex to do-grade cell walls of cyanobacteria. The polysaccharides or peptidoglycans of A. cylindrica maybe hydrolyzed and metabolized to a range of easily utilizable monosaccharides or other low molecular weight organic substances by strain AK-07 of A. johnsonii.

Effects of Nonylphenol on the Population Growth of Algae, Heterotrophic Nanoflagellate and Zooplankton (내분비장애물질 Nonylphenol이 미세조류, 종속영양편모충, 동물플랑크톤의 개체군 성장에 미치는 영향)

  • Nonylphenol (NP) has been well known as a major substance of surfactant and/or estrogenic environmental hormone. We tested toxic effects of nonylphenol on the population growth and development of aquatic organism such as algae (Microcystis aeruginosa), heterotrophic nanoflagellate (Diphylleia rotans), micro- (Brachionus calyciflorus) and macro-zooplankton (Daphnia magna) among eutrophic water food-web constituents. Dosage of NP treatment were 4 to 5 grades, according to each organism's tolerance based on pre-experiments; algae (0.01, 0.05, 0.10, 1.00 mg $L^{-1}$) Diphylleia rotans (0.5, 1,2. 5,6, 10 ${\mu}g\;L^{-1})$, Brachionus calyciflorus (0.1, 0.5, 1, 2.5, 5 ${\mu}g\;L^{-1}$), and Daphnia magna (0.5, 1, 5, 10, 50 ${\mu}g\;L^{-1}$), respectively. Toxic effects were measured by the changes of biomass of each organism after NP treatment. All experiments were triplication. As suggested, the higher concentration of NP treatment, the stronger inhibited the population growth of all organisms tested. In view of toxicity, a variety of concentration of NP showed a significant growth inhibition to organism; algae to 0.05 $mg\;L^{-1}$, D. rotans and B. calyciflorus to 1.0 ${\mu}g\;L^{-1}$, and D. magna to 5.0 ${\mu}g\;L^{-1}$, respectively. The $EC_{50}$ of each organism to the nonylphenol are as follows; 3. calyciflorus (2.49 ${\mu}g\;L^{-1}$), D. rotans (3.49 ${\mu}g\;L^{-1}$), D. magna (7.61 ${\mu}g\;L^{-1})$, and M. aeruginosa (47 ${\mu}g\;L^{-1})$. NP toxic effects on the development of zooplankton like egg production showed some differences in treatment concentration between Brachionus calyciflorus ${0.1{\sim}1NP{\mu}g\;L^{-1})$ and Daphnia magna $(0.5{\sim}5NP\;{\mu}g\;L^{-1})$. These results suggest that a strong growth inhibition of predator or grazer by the nonylphenol can stimulate the algal growth, or can play important role in evoking the nuisance algal bloom in eutrophic water with enough nutrients.

Effect of Solvent Extracts from Sargassum hemiphyllum on Inhibition of Growth of Human Cancer Cell Lines and Antioxidant Activity (짝잎모자반(Sargassum hemiphyllum)의 암세포주 증식 억제 및 항산화 효과)

  • This study was carried out to determine the inhibitory effects of solvent extracts from Sargassum hemiphyllum on growth of cancer cell lines (AGS human gastric adenocarcinoma and HT-29 human colon cancer cells) and production of lipid peroxides. Inhibitory effects of acetone with methylene chloride extract from S. hemiphyllum on the growth of AGS and HT-29 cancer cells were increased as dose dependent patterns (p<0.05). The methanol extract was more effective on inhibition of growth of AGS. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of cancer cells (p<0.05) and the inhibitory effect was stronger in HT-29. In DCFH-DA (dichlorodihydrofluorescin diacetate) assay, acetone with methylene chloride and methanol extracts showed a stronger inhibitory effect on the production of cellular lipid peroxides (p<0.05) compared with the butanol and hexane fractions. These results indicate that the consumption of S. hemiphyllum may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Investigation and evaluation of appearance ratio using wildbird intrusion prevention device in domestic fowl farms (국내 가금 농장에서 야생조류 침입 억제 장치를 이용한 야생조류 출현율 조사 및 평가)

  • The epidemiological investigation of outbreak in korea confirmed that the inflow of avian influenza (AI) is related to the migration of migratory birds. In this study, avian repellents instrument were implemented and developed using the visual effects of lasers in accordance with the situation of small domestic fowl farms, and monitoring cameras were installed around each instrument to investigate the frequency of wild birds appearing and evaluate the performance of the instrument. Observation showed that the appreance ratio was reduced by 95%, and no significant reduction in the intrusion prevention effect by adaptation was observed on all fowl farms. In conclusion, it is expected that the outbreak of wild bird-borne infectious diseases such as avian influenza will be decreased if the device is installed on domestic fowl farms.

  • The ouput power of wave energy system in buoy is determined according to the inner diameter of oscillating water column and flow resistance. The increase of adhered shellfish inside the water column leads to decrease the inner diameter of wave energy converter. Influx loss of seawater reduces the efficiency of output power in the wave generation system. In this paper, the test result of AFS characteristic is described for preventing the deposition with shellfish and etc. The current of anode is controlled by buck converter, and the control algorithm developed for AFS in buoy. The experimental results is shown excellent preventing capapbility of AFS in buoy.

Elucidation of Anti-tumor Initiator and Promoter Derived from Seaweed-3 : Anti-tumor Promoters of Ecklonia stolonifera Extracts (해조류 중의 anti-tumor initiator 및 promoter의 해석-3 : 곰피 추출물중의 발암 promotion억제 인자)

  • To elucidate anti-tumor promoter from seaweed, the anti-tumor promoting activity of Ecklonia stolonifera, Undaria pinnatifida and Laminaria japonica extracts were determined by Epstein-Barr virus (EBV)-early antigen (EA) induction caused by a tumor promoter, teleocidin B-4. The methanol extracts of seaweed were subsequently fractionated with diethyl ether, distilled water, chloroform and ethyl acetate. Among the solvent fractions tested, chloroform and ethyl acetate fraction of E. stolonifera showed a high anti-tumor promoting activity at the levels of 88.0 and $85.9\%$ by the addition of 20 ${\mu}g/m{\ell}$, respectively. To characterize anti-tumor promoters from solvent fractions of E. stolonifera, the effects of phenols, chlorophyll derivatives and carotenoids on the anti-tumor promoting activity were investigated. Phenols, such as bromophenol and phloroglucinol showed anti-tumor promoting activity of $57\~66\%$ at 20 ${\mu}g/m{\ell}$. Pigments, such as chlorophylls and carotenoids exerted high anti-tumor promoting activities. Chlorophyll a and pheophorbide a exhibited the activity of $77.4\%$ and $66.6\%$ at 5${\mu}M/m{\ell}$, respectively. The active compounds of carotenoids were tentatively identified as lutein and $\alpha-cryptoxanthin$ from the profiles of visible spectra and R_f value of their authentic compounds, and showed anti-tumor promoting activities of $76.9\%$ and $84.4\%$ at dose of 20 ${\mu}g/m{\ell}$, respectively.

Basic Studies on the Development of Diet for the Treatment of Obesity II. Comparison of the Inhibitory Effect of Algae and Crude Drug Components on Obesity (비만치료식 개발을 위한 기초연구 2. 조류와 생약성분의 비만억제작용 비교)

  • This study was designed to compare the inhibitory effects of algae components such as alginic acid and chlorella extract with the red ginseng saponin and garlic components such as alliin and scordinin on obesity. These components supplemented to basic diet were fed ad libitum to Sprague Dawley rats for 4 weeks, and the inhibitory effects on obesity were evaluated by measuring body weight gain and feed efficiency, the contents of glucose and cholesterol in serum, lipase activity in serum, lipid contents in liver and adipose tissue around epididymis and ovarium, and Lee index, etc. Among the inhibitory effects of these components on obesity, Na-alginate($71\%$) was the most effective (p<0.01), and the others were effective (p<0.001) in the order of scordinin ($76\%$) = chlorella ($76\%$)> alliin ($86\%$) for body weight gain, but Na-alginate ($68\%$) was the most effective, followed by alliin ($79\%$) for feed efficiency (p<0.001). In the inhibitory effects on lipid accumultion in liver and adipose tissue, Na-alginate ($84\%$) among these components was the most effective (p<0.001) in liver, but Na-alginate ($54\%$) was the most effective (p<0.001), followed by alliin ($64\%$), and followed by scordinin ($66\%$) in adipose tissues around epididymis and ovarium (p<0.001). The inhibitory effect on obesity evaluated by Lee index was observed in Na-alginate ($91\%$) only (0.001). The decreasing effects of lipid content in liver by these components were effective in the order of alliin ($40\%$)> saponin ($58\%$)> Na-alginate($62\%$)> chlorella ($64\%$)> scordinin ($74\%$) (p<0.001). G lucose contents in serum were significantly decreased in cases of Na-alginate ($67\%$), saponin ($73\%$), scordinin ($74\%$) and alliin ($83\%$), and cholesterol contents in serum were significantly decreased in cases of Na-alginate ($82\%$), chlorella ($86\%$) and saponin ($89\%$) (p<0.001). The increasing effect of lipase activity in serum was effective in cases of Na-alginate ($109\%$) and saponin ($104\%$), while garlic components such as alliin and scordinin, and chlorella were decreased. It is proved that the inhibitory effect of alginic acid which is the major component of brown algae on obesity was more effective than those of ginseng and garlic components such as saponin, alliin and scordinin.
