조구등이 Glucose Oxidase로 손상된 대뇌신경세포에 미치는 효과 (Effect of Ramulus et uncus uncariae on Glucose Oxidase-Induced Toxicity in Cultured Cerebral Neurons)

  • To examine the cytotoxic effect of glucose oxidase(GO) in cultured mouse cerebral neurons, cytotoxicity was measured by MTT assay after cultured nerve cells were incubated for 3 hours in the media containing 1 ~ 60mU/ml concentrations of GO. In addition, the neuroprotective effect of Ramulus et uncus uncariae(REUU) was determined by MTT assay in these cultrures. Cell viability was remarkably decreased in a dose- and time-dependent manner after cultured mouse cerebral neurons were exposed to 30mU/ml GO for 3 hours. In the neuroprotective effect of REUU on GO-induced toxicity, REUU blocked the GO-mediated neurotoxicity in these cultures. From above the results, it suggests that GO is toxic in cultured mouse cerebral neurons and selective herb extract such as REUU is effective in prevetion of the neurotoxicity induced by GO.

  • The present study examined the effects of water extract of Hwangruynhaedoktang plus Yukmijihwangtang add Chogudeung (HYC) on the blood pressure and renal function in spontaneously hypertensive rats (SHR). The HYC water extract blocked increase of systolic blood pressure (SBP) in SHR. The urinary volume (UV), sodium (UNaV), potassium (UkV), and chloride (UclV) excretions were significantly increased in SHR administered with high dose of water extract of HYC, but urinary osmolality was not changed. The creatinine clearance (Ccr), which is index of glomerular filtration rate (GFR), was increased by administration of HYC, while solute-free water reabsorption (TcH2O) was not changed. Taken together, these results suggest that the depressor effects of HYC, at least in part, caused by diuretic effect due to increase of glomerular filtration rate.

천마구등음가미방(天麻鉤藤飮加味方)이 고혈압에 미치는 영향 (Effects of Chunmagudeungeumgamibang on Hypertension)

  • Chunmagudeungeumgamibang(CGE) has been used for many years as a therapeutic agent for acute stage of cerebrovascular disease and hypertension in oriental medicine. But the effect of CGE on hypertension and vascular system is not well-known. This study was done to investigate the effects of CGE on hypertension. The results were obtained as follow : CGE showed a safety in cytotoxicity and toxicity of liver. CGE showed scavenging activity on DPPH free radical. CGE showed the inhibitory effect on ROS and ACE. CGE significantly decreased the blood pressure and pulse in DOCA-salt hypertensive rat. CGE significantly decreased the levels of aldosterone in DOCA-salt hypertensive rat. CGE significantly decreased the levels of dopamine, epinephrine in DOCA-salt hypertensive rat. CGE significantly decreased the levels of potassium and chloride in DOCA-salt hypertensive rat. CGE significantly increased the levels of calcium in DOCA-salt hypertensive rat. These results suggest that CGE might be effective in treatment and prevention of hypertension

중추신경계 손상 회복에 미치는 대한 조구등의 영향 (The Effect of the Uncariae Ramulus et Uncus on the Regeneration Following CNS Injury)

  • Objective : Following central nervous system(CNS) injury, inhibitory influences at the site of axonal damage occur. Glial cells become reactive and form a glial scar, gliosis. Also myelin debris such as MAG inhibits axonal regeneration. Astrocyte-rich gliosis relates with up-regulation of GFAP and CD81, and eventually becomes physical and mechanical barrier to axonal regeneration. MAG is one of several endogenous axon regeneration inhibitors that limit recovery from CNS injury and disease. It was reported that molecules that block such inhibitors enhanced axon regeneration and functional recovery. Recently it was reported that treatment with anti-CD81 antibodies enhanced functional recovery in the rat with spinal cord injury. So in this current study, the author investigated the effect of the water extract of Uncariae Ramulus et Uncus on the regulation of CD81, GFAP and MAG that increase when gliosis occurs. Methods : MTT assay was performed to examine cell viability, and cell-based ELISA, western blot and PCR were used to detect the expression of CD81, GFAP and MAG. Then also immunohistochemistry was performed to confirm in vivo. Results : Water extract of Uncariae Ramulus et Uncus showed relatively high cell viability at the concentration of 0.05%, 0.1% and 0.5%. The expression of CD81, GFAP and MAG in astrocytes was decreased after the administration of Uncariae Ramulus et Uncus water extract. These results was confirmed in the brain sections following cortical stab injury by immunohistochemistry. Conclusion : The authors observed that Uncariae Ramulus et Uncus significantly down-regulates the expression of CD81, GFAP and MAG. These results suggest that Uncariae Ramulus et Uncus can be a candidate to regenerate CNS injury.

흰쥐의 일과성 전뇌 허혈 손상에 대한 조구등 약침의 효과 (Effects of Ramulus Uncariae Cum Uncis' Herbal Acupuncture on Transient Forebrain Ischemic Injury in Rats)

  • Objectives : Recently, herbal acupuncture has been developed in the Korea since the earlier 1960' applied to various diseases including the cerebrovascular accident. The main characteristics of herbal acupuncture are a combination the merit of acupuncture and herbal medicine. It was not well known the therapeutic effect and the mechanism of herbal acupuncture on transient forebrain ischemic injury, although it has been used frequently in clinics. Ramulus Uncariae Cum Uncis (釣鉤藤) has been claimed to possess sedative, anti-spasmodic actions, and treat childhood epilepsy in oriental medical preparation. Also, it has been stated that Ramulus Uncariae Cum Uncis was antioxidatvie effect and neuroprotection against glutamateinduced neuronal death. Methods : In this study, effects of Ramulus Uncariae Cum Uncis' herbal acupuncture on the $GV_{20}{\;}or{\;}LR_3$, named Baek-hue or Tae-chung, on neuroprotection after the transient forebrain ischemia were investigated in Sprague-Dawely rats. Expressions of cFos, FosB and BDNF protein in the hippocampus and cortex were observed at 2 hrs and 48 hrs after transient forebrain ischemia by immunohistochemistry and ELISA technique. Results : Expression of cFos protein was increased slightly in the hippocampus and cortex at 2 hrs after transient forebrain ischemia, but FosB protein was increased highly comparing to cFos protein. However, pretreatment with Ramulus Uncariae Cum Uncis' herbal acupuncture on $GV_{20}{\;}or{\;}LR_3$ significantly increased expression of cFos protein and significantly decreased expression of FosB protein compared to control group, respectively. These features were observed in the motor cortex and retrosplenial granular cortex as well as the hippocampus. Especially, cFos expression was more increased at the herbal acupuncture on $GV_{20}{\;}than{\;}LR_3$, but FosB expression was more decreased in $LR_3$ group than $GV_{20}$ group. Also, pretreatment with Ramulus Uncariae Cum Uncis' herbal acupuncture on $GV_{20}{\;}or{\;}LR_3$ significantly increased the expression of BDNF protein in the hippocampus ($254.88{\pm}12{\;}pg/ml{\;}in{\;}GV_{20}$ group, $245.93{\pm}44.4{\;}pg/ml{\;}in{\;}LR_3$ group) and the cortex ($85.81{\pm}3.45{\;}pg/ml{\;}in{\;}GV_{20}$ group, $111.51{\pm}15.79{\;}pg/ml{\;}in{\;}LR_3$, group) compared to the hippocampus ($134.07{\pm}2.96{\;}pg/ml$) and the cortex ($61.16{\pm}4.11{\;}pg/ml$) in control group at 48 hrs after transient forebrain ischemia. Conclusion : These results suggest that pretreatment with Ramulus Uncariae Cum Uncis' herbal acupuncture on $GV_{20}{\;}or{\;}LR_3$ has neuroprotective effect on transient forebrain ischemia and the herbal acupunture on $GV_{20}{\;}or{\;}LR_3$ may be related to antioxidative effect and calcium channel block of Ramulus Uncariae Cum Uncis. Also, it could be mentioned there is specificity of acupoints treating ischemic injury through the difference between the herbal acupuncture of $GV_{20}{\;}and{\;}LR_3$.

알코올성 위염에 대한 조구등(釣鉤藤)의 항염증 효과 (Anti-inflammatory effect of Uncariae Ramulus et Uncus on alcohol-induced gastritis)

  • Objective : Gastritis refers to an inflammatory disease of the gastric mucosa. Alcohol is one of the main aggression factors, causing bleeding and inflammation in the gastric mucosa and it is known to not only increase lipid peroxide levels, but also deplete key antioxidant factors. The purpose of this study was to determine the effect of Uncariae Ramulus et Uncus water extract (URW) in alcohol-induced gastritis. Methods : The total polyphenol and flavonoid contents of URW were confirmed through an in vitro experiment. Also, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) activity were confirmed. For in vivo experiments, mice were divided into 4 groups (n=8). Also, 1 hr after oral administration of each drug, 50% ethanol was orally administered to induce gastritis. Results : As a result of in vitro experiments, URW showed excellent antioxidant activity. In alcohol-induced gastritis, URW alleviated the damage to the gastric mucosa caused by alcohol. Also, URW decreased reactive oxygen species (ROS) and malondialdehyde (MDA) levels in serum and gastric tissues, and significantly decreased the expression of NADPH oxidases in gastric tissues. In addition, it significantly modulated the nuclear factor erythroid-derived 2-related factor 2 (Nrf2) and nuclear factor-𝜅B p65 (NF-𝜅B) pathways as well as significantly increased the expression of anti-inflammatory proteins. Conclusions : These results suggest that URW not only reduces oxidative stress through excellent antioxidant activity but also relieves gastric mucosal inflammation as a regulator of Nrf2 and NF-𝜅B pathways.

조구등이 MCAO 모델 흰쥐에서 gliosis 억제에 미치는 영향 (The Effect of the Water Extract of Uncariae Ramulus et Uncus on Gliosis in the Middle Cerebral Artery Occlusion(MCAO) Rats)

  • Objectives : In condition of brain infarction, irreversible axon damage occurs in central nerve system(CNS), because gliosis becomes physical and mechanical barrier to axonal regeneration. Reactive gliosis induced by ischemic injury such as middle cerebral artery occlusion is involved with up-regulation of GFAP and CD81. The current study is to examine the effect of the Uncariae Ramulus et Uncus on CD81 and GFAP expression in the rat brain following middle cerebral artery occlusion. Methods : In order to study ischemic injuries on brain, infarction was induced by middle cerebral artery occlusion(MCAO) using insertion of a single nylon thread, through the internal carotid artery, into a middle cerebral artery. Cresyl violet staining, cerebral infarction size measurement, immunohistochemistry and microscopic examination were used to detect the expression of CD81 and GFAP and the effect on the infarct size and pyramidal cell death in the brain of the rat with cerebral infarction induced by MCAO. Results : The following results were obtained 1. Measuring the size of cerebral infartion induced by MCAO in the rat after injection of Uncariae Ramulus et Uncus showed the size was decreased. 2. Intravenous injection of Uncariae Ramulus et Uncus showed pyramidal cell death protection in the hippocampus in the MCAO rat. 3. Water extract injection of Uncariae Ramulus et Uncus decreased GFAP expression significantly in the MCAO rat. 4. Uncariae Ramulus et Uncus water extract decreased CD81 expression in the MCAO rat. 5. The administration of water extract of Uncariae Ramulus et Uncus induced up-regulation of c-Fos expression significantly compared with MCAO. 6. The admistration of water extract of Uncariae Ramulus et Uncus increased ERK expression significantly compared with MCAO. Conclusion : We observed that Uncariae Ramulus et Uncus could suppress the reactive gliosis, which disturbs the axonal regeneration in the brain of the rat with cerebral infaction after MCAO by controlling the expression of CD81 and GFAP. The effect may be modulated by the up-regulation of c-Fos and ERK. These results suggest that Uncariae Ramulus et Uncus can be a candidate to regenerate CNS injury.

아마란스(Amaranth) 종실의 가공에 따른 비스킷 제품에의 적용 (Application to the Biscuits Manufacture of Processed Amaranth Seeds)

  • 아마란스를 충분한 양의 물에 침지한 후 물기를 제거하고 수분함량이 30~50중량%일 때 가열솥에서 이취가 없어지고 고소한 냄새가 나며 색이 더 노랗게 변하면서 입자가 puffing될 때까지 볶아준 다음 비스킷에 첨가하여 관능성과 상품성을 향상시켰다. 가수량에 따른 팽창용적 측정 결과 1.3배에서 1.6배 가수량에서 가장 큰 부피의 팽창이 있었고, 가공 방법에 따른 호화도 측정 결과 취반의 경우는 가수량이 증가함에 따라 호화도가 증가하였고, 볶은 아마란스의 호화도가 64.10%로 쪄서 말린 아마란스의 호화도 57.59%보다 높은 것을 알 수 있었다. 또한 찐 후 말린 아마란스와 생 아마란스는 경도($\times$$10^{5}$dyn/$\textrm{cm}^2$)가 각각 16,197과 13,601로 단단한 것으로 측정되었고 볶은 아마란스가 1,580으로 가장 무른 것으로 측정되었다. 생 아마란스와 볶은 아마란스, 찐 후의 아마란스를 첨가하여 비스킷을 제조한 결과 생 아마란스는 이취와 이질감이 문제가 되었고 찐 후 말린 아마란스는 점성은 제거 할 수있었으나 색이 검게 변화하는 것을 개선할 수 없었으며 볶은 아마란스의 경우 비스킷 첨가시 이취나 이질감이 거의 없었고 흰색으로 변화함에 따라 비스킷의 색을 더욱 밝게 해주는 효과도 있었다. 또한 비스킷에 첨가될 볶은 아바란스의 함랑은 5%일 때 가장 적절한 미감을 보이는 것으로 여겨진다. 볶은 아마란스를 5% 첨가한 비스킷과 볶은 아마란스 자체의 시간에 따른 산패도 변화를 측정하였을 때 볶은 아마란스 자체의 산가와 과산화물가는 어느 정도 높고 서서히 산패가 진행되지만, 이를 5% 함유한 비스켓의 산가와 과산화 물가는 매우 낮게 나타나 실험결과로 미루어 제품의 품질은 약 6개월 정도의 유통기한에서는 매우 안정한 것으로 추정되었다.다.of NAA/Cr ratios of lentiform nucleus between the symptomatic and the nonsymptomatic side, the present $^1$H MRS study shows a significant neuronal laterality in Parkinson's disease with unilateral symptom. CS 가스 흡입이 흰쥐의 폐포막내 Laminin-1에 미치는 영향 (Expressions of Laminin-1 in Lung Alveolar Septa after CS gas Exposure in Rats)

  • 배 경 : Laminin은 생체조직의 세포외기질에 함유되어 있는 당단백질로 특히 기저막내에 특이적으로 분포되어 있다. 저자들은 군중집회 해산용으로 혹은 군사용으로 널리 사용되고 있는 CS 가스(ortho-chlorobenzylidine malononitrile)가 폭로되었을 때 흰쥐의 폐조직내 laminin에 미치는 영향을 밝히고자 하였다. 대상 및 방법 : 본 실험에 사용한 실험동물은 청정동물실에서 사육한 생후 21일령의 흰쥐 (Sprague-Dawley계) 35마리를 사용하였다. 실험동물은 대조군에 5마리, 실험군에 30마리를 배정하였다. CS 가스에 폭로 후 흰쥐를 12시간, 1일, 2일, 3일, 5일 및 7일 경과후 희생시켜 폐장을 절취하여 면역조직염색 및 면역도금법을 시행하였다. 결 과 : 상기와 같이 조직면역반응과 염색을 시행한 후 조직을 관찰하여 다음과 같은 결과를 얻었다. CS 가스 투여후 2일 경과군의 흰쥐 폐포막에는 림파구, 단핵구등이 침투되기 시작하여 3일 경과군에서는 강도의 염증조직이 관찰되었다. 대조군과 CS 가스 투여 12시간군의 폐장의 제2형폐포세포내에 금과립은 층상체주위와 외형질에서 소수 관찰되었다. 결 론 : 이상과 같은 결과로 흰쥐에 CS 가스를 계속 3일간 투여했을 경우 폐포에서 염증조 직이 형성되고 이때 기저막내 laminin의 분포는 일시 감소되나 염증조직의 소멸과 함께 다시 증가되는 것을 보아 laminin이 조직재형성에의 역할이 흰쥐 폐포에서도 일어나고 있음을 알 수 있었고, 면역금과립방법을 이용한 전자현미경 검사에서 제2형폐포세포내 laminin의 분포도 증명할 수 있었다.

고품질 복합내병성 조생종 벼 신품종 "주남조생" (A New Early Maturing Rice Cultivar "Junamjosaeng" with Multiple Disease Resistance and High Grain Quality Traits)

  • "주남조생"은 소득작물 전 후작에 적응하는 벼를 육성하고자, '98/'99년 동계에 밥맛이 좋은 것으로 알려진 일본의 고시히까리에 밀양165호(주남벼)를 2회 여교배하여 육성한 조생종 단기성 품종으로 주요특성과 수량성을 요약하면 다음과 같다. 1. 주남조생의 영남 평야지의 소득작물 후작 만기재배에서 8월 26일로 "금오벼"보다 6일 빠른 조생종 품종으로 주당수수는 12개로 금오벼보다 2개 많은 편이고, 수당립수는 약간 적은편이며 현미천립중은 금오벼보다 약간 무거운 편이다. 2. 불시출수는 금오벼보다 덜 되는 편이지만 수발아는 금오벼보다 잘되는 편이다. 유묘기 내냉성은 금오벼와 비슷하며, 성숙기 임실율은 78%로 금오벼보다 높은 편이다. 3. 잎도열병 저항성반응은 금오벼와 비슷한 수준이며, 흰잎 마름병 레이스 $K_1$, $K_2$$K_3$에 저항성이며, 바이러스병은 줄무늬잎마름병에만 저항성이고, 벼멸구등 해충에는 감수성이다. 4. 쌀알은 맑고 심복백은 없다. 아밀로스 함량은 19.8%이고 단백질함량은 6.7%로 금오벼와 같고 밥맛은 금오벼보다 약간 양호하며, 도정수율은 금오벼와 약간 높은편이다. 5. 쌀수량은 '04~'06년 3개년간 실시한 지역적응시험에서 4.43 MT/ha로 금오벼와 비슷하다.
