Acid Production by Lactic Acid Bacteria in Soy Milk Treated by Microbial Pretense or Papain and Preparation of Soy Yogurt (미생물 Protease 또는 Papain으로 처리된 두유에서 젖산균의 산생함과 대두요구르트의 제조)

  • The soy milk prepared from soy protein concentrate was treated with microbial protease or papain. Growth and acid production by Lactobacillus acidophilus in soy milk containing partially hydrolyzed proteins were investigated. Sensory evaluation of yogurt beverage prepared from protease treated soy milk was also performed. Protease treatment of soy milk enhanced acid production by lactic acid bacteria, particularly in case of microbial pretense and simultaneous treatment by two types of protease showed synergistic effect. pH and number of viable cells were not affected markedly by pretense treatment. Microbial pretense treatment up to 15 minutes or papain treatment up to 45 minutes enhanced acid production, but further treatment up to three hours did not affect the acidity markedly. rho sensory evaluation showed that overall acceptability and taste of soy yogurt beverage were slightly improved when soy milk was treated with microbial pretense of 0.2% or papain of 0.2%. The amount of non-protein nitrogen considerably increased by pretense treatment of 15 minutes and it increased gradually by further treatment up to three hours.

  • The soy milk prepared from soy protein concentrate was treated with pretense of various concentrations. Growth and acid production by Lactobacillus acidophilus or L. bulgaricus in soy milk containing partially hydrolyzed protein were investigated. Sensory evaluation of soy yogurt beverages prepared from pretense treated soy milk was also performed. The treatment of soy milk with pretense markedly enhanced acid production by lactic acid bacteria, particularly by L. acidophilus, whereas pH and number of viable cells were not .affected by pretense treatment. Protease treatment of 15 minutes greatly enhanced acid production by lactic acid bacteria, but further treatment up to three hours did not affect the acidity markedly. The sensory evaluation showed that overall acceptability of soy yogurt beverages was slightly improved when soy milk was treated with pretense of 0.1%. The amount of non-protein nitrogen was considerably increased by pretense treatment of 15 minutes and it reached the maximum value by treatment of two hours.

  • Jeungpyun has a unique sponge-like texture and sour taste imparted by the lactic acid and alcohol that are produced by the addition of Takju(turbid rice wine) as a starter. Its consumption, however. has been decreased due to the long preparation time, the difficulties in quality control and the offensive odor derived from the Takju. The present study was carried out in order to shorten the preparation time and to improve the quality of Jeungpyun. To achieve the objectives an appropriate commercial lactic acid starter was selected and a cofermentation system with yeast was developed. A starter containing Lactococcus lactis, Lactococcus cremoris and Lactococcus diacetylactis was selected based on the acid production rate and the quality of the produced sour taste. It took 3 hr for the lactic acid fermentation of rice slurry. The optimum addition levels of the lactic acid starter and yeast were 0.45% and 0.60%, respectively. The lactic acid fermented rice slurry was mixed with the rice slurry separately fermented for 2 hr by yeast, and cofermented for another 1 hr before steaming. Jeungpyun Prepared by the developed method was superior in quality to that Prepared by conventional method using Takju. The developed method reduced the preparation time more than 50% compared with the conventional method.

Characteristics of Fermented Fruit and Vegetable Mixed Broth Using by Bacteriocin-producing Lactic Acid Bacteria and Yeast (박테리오신 생성 젖산균과 효모를 이용한 과채발효액의 특성)

  • A mixed culture of bacteriocin-producing lactic acid bacteria and yeast was used to prepare unique fermented fruit and vegetable beverage which contains bacteriocin. Fruit and vegetable broth fermented by Lactococcus lactis 11454 showed inhibitory activity against foodborne pathogens such as Listeria monocytogens and Staphylococcus aureus, and Streptococcus mutans which is a major causative agent for dental caries. Bacteriocin was detected at the end of fermentation of fruit and vegetables and its antimicrobial activity was stable for 5 weeks during storage at $4^{\circ}C$. These results suggest that the presence of bacteriocin in beverages during fermentation and storage will provide a safeguard against foodborne pathogens and spoilage bacteria. To improve flavor of the fermented broth, post-fermentation of the fermented broth was carried out at $4^{\circ}C$ by using yeast isolated from raw material mixture. Total acidity of the post-fermented broth was slightly decreased, but significant increase in the concentration of succinic acid was observed in the post-fermented broth. It was also observed that bitter and disagreeable taste compounds such as phthalates were decreased, and mild acidic and fruity flavor esters and alcohols were increased by the post-fermentation of the fermented broth with yeast.

Isolation and Characteristics of a Homofermentative lactic Acid Bacterium (호모발효 젖산군의 분리 및 특성)

  • This study was targeted to isolate and characterize a bacterium producing lactic acid in a large amount. Lactic acid bacteria of about fifty strains were isolated from kimchi, a Korean traditional fermented vegetable food. Strain KH-1 of them was most effective in the lactic acid production and showed 99% homology with Lactobacillus casei from analysis of 16S rRNA sequencing. The conversion ratio of lactic acid from glucose by 1. casei KH-1 was 98% in anaerobic condition, and the lactic acid was composed as racemic mixture of D(-)-and L(+)-lactic acid, 7% and 93%, respectively. This result indicated that L. casei KH-1 was a homofermentative bacterium mainly producing L(+)-lactic acid. The strain KH-1 used glucose as a preferential substrate but not utilized lactose. In investigation of more inexpensive nitrogen source for cultivation of strain KH-1 using industrial MRS medium, when yeast extract and corn steep liquor were used at the ratio of 1 to 1, the molar yield of lactic acid produced per mole of glucose(Yp/s) was 1.09.

Properties Changes of Radish Variety Group of Altari Dongchimi Innoculated with Leuconostoc citreum IH22 during Fermentation (알타리 무 동치미 제조시 젖산균 첨가가 숙성중 품질에 미치는 영향)

  • This study examined the chemical and sensory characteristics of Altari Dongchimi with inoculation of Leuconostoc citreum IH22. Dongchimi was stored at room temperature for the first day and at 4$^{\circ}C$ from 2nd to 28th day. The pH was somewhat lower in Altari Dongchimi when first inoculated with Leuconostoc citreum IH22 than non -treated Dongchimi. As fermentation proceeded, titratable acidity of Altari Dongchimi significantly increased. The patterns of microflcral changes in both the inoculated and the control were similar during fermentation: the total number of bacteria increased at the beginning, but rapidly decreased right after the optimum ripening point. The hardness and fracturability of both Dongchimi's decreased gradually during fermentation. The evaluation of the sensory qualities showed that the sour taste and fresh taste resulted in similar scores in both Dongchimi's but overall acceptability of the control was higher than that of inoculated one with Leuconostoc citreum IH22.

Antibacterial Activity of the Bamboo(Pseudosasa japonica Makino) Leaves Extracts on Lactic Acid Bacteria Related to Dongchimi (동치미 젖산균에 대한 대나무(이대)잎 추출물의 항균활성)

  • Antibacterial activities of the bamboo(Pseudosasa japonin Makino) leaves extract on lactic acid bacteria related to Dongchimi fermentation were investigated. The bamboo leaves were extracted with 70% ethanol, fractionated it with 4 solvents which have a different polarity. Total viable count was very similar to those obtained by lactic acid bacteria during fermentation of Dongchimi. Its maximum counts were (equation omitted) 10$^{7}$ CFU/m1 at 8 days of fermention. Lactic acid bacteria isolated during Dongchimi fermentation were identified as Leuconostoc species(BK-2, BK-07, BK-22), Lactobaillus species (BK-05, BK-08) and Streptococcus species(BK-15) 70% ethanol extract of the bamboo leaves showed antibacterial activity against strain BK-02, BK-07, BK-lS and BK-22. Especially, strain BK-22 was more effective to the extracted substance. Among the each fractions, 70% ethanol and diethyl ether had the strongest antibacterial activity against strain BK-22 and BK-07, respectively. The two fractions also showed similar activities against all of the test strains.

Effects of Psychrotrophic Lactic Acid Bacterial Starter on Kimchi Fermentation (저온성 젖산균 스타터가 김치발효에 미치는 영향)

  • Five strains of psychrotrophic lactic acid bacteria which had been isolated from kimchi were inoculated as kimchi starters, and then their effects were investigated during fermentation at $8^{\circ}C$. The initial numbers of Gram negatives and coliforms in no-starter-inoculated kimchi were $4.0{\times}10^{6}CFU/ml\;and\;3.2{\times}10^4CFU/ml$ respectively, while these of lactic acid bacteria were only $1.6{\times}10^3CFU/ml$. Starter-inoculated kimchi showed sharp decreases in Gram negatives and coliforms from initial stage, but no-starter-inoculated kimchi showed slow increases in those bacteria in the early stage, and sharp decreases after 10 days. It took 10 days in no-starter-inoculated kimchi, $4{\sim}6$ days in Leuconostoc-inoculated kimchi and 2 days in Lactobacillus-inoculated kimchi for lactic acid bacteria to reach stationary phase of growth. No-starter-inoculated kimchi required 10 days to reach optimum ripened state, while all starter-inoculated kimchi samples required 4 days. The inoculations of all these starters did not cause over acidifications. Remarkable softening occurred in Leu. dextranicum-inoculated kimchi in 6 days, but did not in the rest. Except the Leu. paramesenteroides-inoculated kimchi, the qualities of Leuconostoc-inoculated kimchi were as good as those of no-starter-inoculated kimchi, but those of Lactobacillus-inoculated kimchi were not so good.
