Fertilization and in vitro Development of Bovine Oocytes Following Round Spermatid Injection (원형정자 미세주입에 의한 소 난자의 수정과 체외 배 발달)

  • In this study we determined fertilization processes and developmental ability of porcine oocytes following injection of round spermatid in the presence of artificial activation. Electrical stimulation at 3 h before spermatid injection significantly increased the incidence of normal fertilization as compared to those following injection without stimulation or with stimulation immediately after injection. The incidences of two pronuclear formation and apposition were not different in oocytes between following intracytoplasmic spermatid and spermatid nucleus injection. Indirect immunocytochemistry and laser scanning confocal microscopy study revealed that micro tubules were organized from the oocyte cortex following round spermatid injection, and this seemed to move both male and female pronuclei into the oocyte center. Paternal mitochondria which are introduced with spermatid have been observed up to 4-cell. Our study indicated that either round spermatid or it's nucleus can be used to produce viable bovine embryos by injection into unfertilized oocytes.

  • Germ cell development and cyclic changes in the epithelial cells of the seminal vesicle of the male rapa whelk, Rapana venosa, were investigated by cytological and histological observations. The process of germ cell development can be classified into five stages: (1) spermatogonial, (2) primary spermatocyte, (3) secodary spermatocyte, (4) spermatid, and (5) spermatozoon. In particular, four atypical cells (Type IA, IB, IIA and IIB cells) occur among normal germ cells in the acini during spermatogenesis. Presumably, the atypical cells, which have lysosome-like vacuoles or lysosome-like bodies in the cells, are involved in breakdown and absorption themselves in the acini. However, atypical cells were not found in the epithelial cells of the inner layer of the seminal vesicle. A considerable amount of spermatozoa are transported from the testis towards the the seminal vesicles until late July. The main coupulation period is between June and July. The process of the cyclical changes of the seminal vesicles can be classified into three phases: (1) resting, (2) accumulating, and (3) spent. Yellow granular bodies are involved in resorption or digestion of residual spermatozoa.

  • The aim of this study was to clarify the differentiation of seminiferous epithelial cells and spermiogenesis in the testis of Rana catesbeiana. Spermatogenesis of R. catesbeiana consists of primary spermatogonia, secondary spermatogonia, primary spermatocytes, secondary spermatocytes and spermatids. They were subdivided into eight stages on the basis of the morphological features of the germ cell differentiation. From the spermatocytes except primary spermatogonia to before the spermiation of spermatids were surrounded by spermatocyst. Spermiogenesis of R. catesbeiana can also be divided into three stages on the basis of morphological features of the nucleus and the cytoplasm organelles. Spermatozoon contained a saccular acrosome, a cylindrical and tapered slighty at both ends head, and a tail with only the axoneme.

  • The testis of Neoditrema ransonneti is testicular tubule type, each testicular tubule consists of numerous testicular cysts which contain numerous germ cells showing the same developmental stage. During spermatogenesis, well developed rough endoplasmic reticula and the Golgi complex are observed in the cyst cell. Secretory activity of cyst cell was the highest in the late spermiogenesis. Sperm binding materials of spermatozeugmata are secreted by testicular cyst cell. One spermatozeugmata is produced by a testicular cyst during spermatogenesis. The capsular structure was not found in the spermatozeugmata discharged from male. According to observations under transmission electron microscopy approximately 1,500 to 1,700 of sperm tails were observed in the cross sectioned spermatozeugmata.

  • The ultrastructures of germinal cells of male marsh clam, Corbicujar lena were studied. The mature sperm was primitive type, consisting of head, middle piece and tail. The mature sperm was whip-shaped and its head was divided into two parts; the acrosomal part shaped long hollow cone about $3{\mu}m$ in length and the sperm nuclear part shaped a long stick about $9\;{\mu}m$ in length. The posterior part of the sperm nuclear projected to centriole, The middle piece of the sperm-nuclear had four mitochondria and two centrioles. The sperm tail part had the 9+2 microtubular arrangement known as a typical pattern, During spermiogenesis, chromatin within sperm nuclear became fiberic materials by condensation.

Effect of Testicular Histopathology on Pregnancy Outcomes in Non-Obstructive Azoospermia (비폐쇄성 무정자증 환자에서 고환의 조직병리학적 진단에 따른 체외수정시술 결과의 비교)

  • Objective: To evaluate outcomes of patients with non-obstructive azoospermia (NOA) undergoing the testicular sperm extraction (TESE) combined with intracytoplasmic sperm injection (ICSI) with different histopathologic subgroups. Method: A total of 122 embryo-transferred TESE/ICSI cycles were compared among NOA subgroups; Germ-cell aplasia (GA, 40 cycles), Maturation arrest (MA, 32 cycles) and severe hypospermatogenesis (S-HS, 50 cycles). Obstructive azoospermia (OA, 667 cycles) patients were served as a control. TESE/ICSI outcomes such as fertilization rate (FR), clinical pregnancy rate (CPR) and live birth rate (LBR) were evaluated. Results: The 2PN FR of embryo-transferred TESE/ICSI cycle was 58.1% in GA, 42.2% in MA and 48.0% in S-HS, which was significantly lower than that of OA (72.9 %, p<0.001). For ICSI-spermatozoa cycles, there were no significant differences in CPR (22.6%, 29.4% and 26.1%) and LBR (16.1%, 29.4% and 19.6%) among NOA subgroups. The CPR of ICSI-spermatid cycles was 0.0%, 9.1% and 0.0% without a live birth. For ICSI-spermatocyte cycles, no clinical pregnancies occurred in any group. Conclusion: There was no significant difference in the FR of embryo-transferred TESE/ICSI cycles among NOA subgroups. The FR among all NOA subgroups was significantly lower than that of OA. Testicular histopathology in NOA did not affect successful pregnancy if spermatozoa extraction from the testis is successful and embryo transfer is possible.

Separation of Highly Motile Bovine Spermatozoa (고활력 우정자의 분리)

  • 소의 번식효율을 증가시키기 위한 기초실험으로서 6%, 10% 및 20%의 bovine serum albumin 을 사용한 선인들의 방법과 tyrode액을 사용하여 시도한 저자의 방법으로 소의 원정액, 희석정액 및 일반시판 냉동정액으로부터 고활력정자를 분리수집하여 정자의 각종 성상과 광학현미경적 형태를 비교관찰한 결과는 다음과 같다. 1. 원정액으로부터 bovine serum albumin을 사용하여 분리한 정자는 대조군에 비하여 운동성, 운동성 정자수, 정상정자율 및 전진운동성이 현저히 높았고 정자회수율은 6%일 때 가장 높았다. 2. 원정액으로부터 bovine serum albumin을 사용하여 고활력정자를 분리한 후 냉동한 정액의 정자운동성, 정상정자율 및 전진운동성은 대조군에 비하여 현저히 높았고, 이러한 현상은 bovine serum albumin의 농도가 20%일 때에 가장 현저하게 나타났다. 3. Bovine serum albumin을 사용하여 분리한 고활력정자의 냉동전 및 냉동후의 광학현미경적인 기형율은 대조군에 비하여 현저히 낮았다. 4. Bovine serum albumin을 사용하여 분리한 고활력정자는 전자현미경으로 세포막의 확장 및 공포 형성, acrosome의 확장과 density loss의 변형율이 대조군에 비하여 낮았다. 5. 일반시판 냉동정액으로부터의 고활력정자의 분리는 bovine serum albumin을 사용할 때는 어려웠으나, tyrode액을 사용한 이 실험에서는 가능하였다. 6. 원정액, 희석정액 및 냉동정액의 고활력정자회수율은 tyrode 액을 이용하여 80분간 정치하였을때 현저히 높았다. 7. Tyrode 액을 이용하여 원정액, 희석정액 및 냉동정액으로부터 분리된 고활력정자의 운동성, 전진운동성 및 정상정자율은 대조군에 비하여 현저히 높았다. 8. Tyrode 액을 이용하여 원정액, 희석정액 및 냉동정액으로부터 분리한 고활력정자의 광학현미경적 기형율은 대조군에 비하여 현저히 낮았다.

  • Study on spermatid and spermatozoon of a Korean endemic freshwater fish, Microphysogobio yaluensis, has been carried out by transmission electron microscope. Spermiogenesis involves conspicuous modifications intercelluar movement such as centrioles and mitochondria migration, nuclear depression and structural changes for instance chroatin condensation, excess of cytoplasm. The mature spermatozoa are similar to those of other cyprinids as follows: a spherical nucleus with a shallow nuclear fossa, a short midpiece containing several mitochondria and encircling the basal body of the flagellum. However there are some differences in the orientation of the centrioles,the number of the mitochondria and distribution of vesicles from other cyprinids. It could be concluded that the M. yaluensis spermiogenesis belongs to type 2, and its mature spermatozoon is charaterized by a unique feature which may provide a useful systematic character.
