Pellet and straw methods in canine semen freezing are compared with respect to motility, viability and acrosome demage of sperm during each of the two major processing steps, to prior-freezing and to frozen-thawing. Senen was extended with a tris-buffered egg yolk contained 4% glycero1 Pellet freezing in the hole of dry ice and straw freezing on the surface of liquid nitrogen were carried out, respectively. The frozen semen 10 days after storage in liquid nitrogen container. wao thawed. In the comparison of two freezing methods, the straw freezing method with 42.7% in motility. 49.2% in viability and 0.186 acrosome score after thawing seems to be superior to the pellet freezing method with 31.2%, 34.5% and 0.314%, respectively. Sperm motility of processing step to frozen-thawing against decrease rate 12.67% to Prior freezing appeared of 33.84% and 49.37% in straw and pellet freezing and increase of 0.02 in acrsomal score to prior freezing appeared of 0.08 and 0.21 in straw and pellet freezing method to frozen-thawing
It would be one of the most important tests that determination of semen in crime scene as a matter of significant evidences. Recently, it has been developed for the identification of semen in forensic specimens which was used simply, easily and reproductively. In this study, Prostate-Specific-Antigen (PSA) Rapid Test kit was evaluated for the forensic identification of semen and compared with one step semen inspection forensic rapid test kit. The sensitivity and specificity of the rapid PSA kit were examined in addition to the stability of PSA. The positive band of rapid PSA kit shown even with 1,000,000-fold diluted semen, which was at least 100 timed higher than qualitative one step semen inspection forensic rapid test kit. PSA was detected in urine from normal male adult, however, it was not detected in urine from young boys and female body fluids. It was shown that PSA was very stable to resist boiling for 20 minutes and the effect of bacteria. In crime scene investigation, rapid PSA kit is expected to help to identify semen easily in the evidences.

The Korean Construction Industry significantly contributed to the Korean economic development not only by preparing the domestic infrastructure but also by successfully conducting various international projects. However, major incidents such as the collapse of Sung-Su Bridge and Sam-Pung Department Store along with stagnation of Korean Economy have raised questions about underlying systematic problems of the Korean Construction Industry. As a solution to resurrect the Korean Construction Industry, the Construction Management (CM) system introduced in late 1990's and increasingly utilized as an innovative delivery system. Despite of the fast growth of CM, the performance of CM has not been up to the hype and low CM fee has been identified as one of the main reasons of unsatisfactory CM performance. Therefore, this research attempted to propose ways of improving 'CM Fee Guidelines' published by Korean Ministry of Construction and Transportation by conducting a benchmarking study on Global Standards of estimating CM Fee. International organizations benchmarked in this research include CMAA, ASCE, DOL, DOE, etc. Various investigation and analysis revealed that Korean 'CM Fee Guidelines' need to significantly modified comparing to Global Standards. This research also tried to prepare recommendations to improve the CM deliver system focusing on the 'Method of CM Fee Estimation' and Selection of a CM firm as an owner's agent.
This study was conducted to investigate the effect of infusion of sterile boar and their semen as pregnancy antigen factor for improvement of production in gilts. We observed 160 gilts between 7 to 8 months of age that were divided into 4 treatment groups vie. A group-epidimetomized boar, B group-no sperm semen infusion, C group-vasectomized boar, and D group-control in a completely randomized design. The number of total pigs born(NT) of A, B, C, and D groups were 10.05, 10.44, 11.63, and 9.97 pigs, respectively(P<0.05). And the NT of C g.cup was the highest among treatment. The number of live pigs born(NB) was similar to NT The NB of C group (10.70) was higher than that of A(9.12) and control(9.11)(P<0.05). However, there was not significant between B and C groups. The progesterone concentration of C group was the highest compare to other group at the 6th day after breeding. However, the progesterone concentration of C was lower than other groups after 8th day. There were not significant among cortisol of A, B, C and D groups. According of the results of this study, the infusion with vasectomized boar and semen at estrus before gestation can be improved reproductive efficiency because of more litter size in gilts.
This study was conducted to evaluate the feasibility of in situ soil flushing for TPH-contaminated soil remediation with column test. The soil texture of the soil was sand and the initial TPH concentration was $9,369mg\; kg^{-1}$. 0.1% Tween-80 was selected as surfactant solution. And the acrylic and the glass syringe columns were used as reactors. In the acrylic column test, 35% of the initial TPH was removed in 1 PV of flushing and approximately 40% in 5 PV and finally 7 PV showed about 60%. The glass column test showed 3 ~ 12% higher removal efficiency than that of acrylic test until 5 PV of flushing. However, there was no difference in TPH removal efficiency when 7 PV of surfactant was finally flushed. Both of alum only and alum+polymer mixed surfactants showed also the best coagulation efficiency in $150mg\;L^{-1}$ of concentraion. When Tween 80 was newly dissolved in 0.1% to the recovered solution after the coagulation treatment, the removal efficiency was increased from 32.0% to 41.0% in comparison to the new 0.1% Tween 80 solution without reuse by coagulation treatment.
The totipotential spermatogonial stem cells of adult testis which give rise to mature sperm cells is well-known to incorporate foreign DNA as well as those of somatic cells. Also, the integration rates of foreign DNA after haploid stages are generally known to decrease and /or is simply bound foreign DNA into the sperm plasma membrane. To overcome these problems, liposome and DNA complexes were used to determine how direct injection of these complexes into testis were integrated into sperm genome and resulted in transgenic offspring. To study this purpose, cation liposome was gently mixed with WAF/hGH DNA (1 : 2) and the complexes were injected into testis. At 10, 20, 40, 60, and 80 days after direct injection into testis, mature sperm cells were recovered by using artificial virgin method from two goats and each semen except a part of semen used for DNA analysis such as PCR or Southern blotting was cryopreserved for the artificial insemination. The results obtained in this study are summarized as follows. 1. By PCR, the presence of exogenous DNA was confirmed up to 80 days after injection with liposome/DNA complexes. The highest integration rates were obtained at day 40 after direct injection. This results suggested that spermatogonial stem cells were integrated exogenous DNA into their genome. 2. Among 23 Korean Native Goats which were artificially inseminated, 4 goats resulted in pregnancy and produced 7 young goats. 3. Two young goats were confirmed as a transgenic by PCR and Southern blot analysis. Therefore, our results suggested that testis-mediated gene transfer can be used as a feasible tools for the production of transgenic livestock.

Standard Estimating System, which can be used for estimating a construction cost, has been pointed out having some problems such as non-reflecting a variety of construction environments and site conditions, non-adapting new technologies and methods since it was established in 1970. For solving these problems, the Korean Institute of Construction Technology(KICT) has chosen organizations for amending Standard Estimating System. However they have had many mistakes in the process of amending works because of non-establishing amending process and absence of the depreciation measurement method. This research derived the necessity to improve the amending process of the Standard Estimating System, and presented the detailed amending process and the performing method in the each process. Furthermore, this research proposed the depreciation measurement method available for the construction industry by means of analyzing researches that performed in the manufacturing industry and the measurement method for depreciation of general accounts.
Purpose: This study is to contribute to the development of monitoring technology through the increase of confidence in construction monitoring by deriving the analysis of construction monitoring cost and improvement measures of railway tunnel construction in Seoul. Method: It presents the status on design and contract of construction monitoring cost, status on application construction monitoring cost and its analysis, analysis on safety management cost and quality management cost, expansion of application of the price calculation standard for monitoring management services to improve this, and monitoring for direct order of ordering organization. Results: If the monitoring management service that was meanwhile ordered as included in the construction work is performed by the directly selected company of ordering organization through the preliminary screening for bidding qualification, then the improvement of monitoring quality and the accurate monitoring data can be secured. Conclusion: For the price calculation standard for monitoring management service, the application of actual cost addition method under the Engineering Promotion Act and the calculation standard of monitoring management cost for standard estimation for ground survey should be extended through the direct order of ordering organization, not the method to be included in the net construction cost where it is performed by a subcontractor via contractor.
This experiment was conducted to investigate the effects of artificial insemination(AI) frequency on reproductive performance of sows. A total of 48 F1 sows(Yorkshire×Landrace) were allocated to 1 of 4 treatments using completely randomized design(CRD). Four experimental treatments were AI frequency from one to four times(AI1, AI2, AI3, AI4) respectively. Estrus detection was done at approximately 09:00 and 21:00 daily by applying back pressure to females with the presence of a mature boar and the weaning to estrus interval(WEI) of all sows were 5~6 day. Sows detected in estrus were mated at 12 hour after and mating interval was 12 hour by treatments. This experiment demonstrated that the lowest farrowing rate was observed AI3 treatment. Frequency of AI did not influence on reproductive performance when WEI was 5-6 day. No significant differences were observed on litter size, born alive and litter birth weight. Consequently, decreased AI frequency did not have any detrimental effect on reproductive performance when estrus detection was adequate. Decreased AI frequency could reduce cost of production of pigs when sows showed normal reproductive performance.

Recovery of low level nitric acid and sulfuric acid ions, which were contained in wastewater of a wet scrubber for de-NOx and de-SOx from ship engines, was attempted as fertilizing materials. This study utilized a selective extraction method using four organic solvents to precipitate the solid salts of ammonium nitrate and ammonium sulfate. The IR analysis showed almost same composition of the extracted ammonium salts with a commercial product, and recovery rate of nitrogen and sulfuric ions was 89% and 80% respectively. It was found that the selectivity and solubility consequently could be the crucial factors to recover the low level ions from the waste scrubbing water.
