Chayacterization of Bacillus polyfermenticus SCD as a Probiotic. (Bacillus polyfermenticus SCD의 생균제로서의 특성)

  • Bacillus polyfermenticus SCD which is commonly called as Bisroot strain is being used for functional foods through the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine in humans. The cells of B. polyfermenticus SCD were treated for 4h in artificial gastric juice (pH 2.0,3.0) and bile acid. Final viability of the strain in artificial gastric Juice (pH 2.0, 3.0) is reached to 62.8% and 81.2% respectively B. polyfermenticus SCD is resistant to antibiotics such as streptomycin, rifampicin, nystatin and ampicilin. B. polyfermenticus SCD is well known supplies the nutrients by synthesizing vitamin $B_1$, $B_2$, C and K. B. polyfermenticus SCD produces various digestive enzymes and the enzymes enable to completely digest diets in our body. Above all, $\alpha$-amylase and pretense activities are very higher than B. subtilis KCTC 1020, about two fold and twenty five fold respectively. B. polyfermenticus SCD is very stable during long-term storage period in phosphate buffers of wide-range pH, solutions of various concentrations of sodium chloride, 5% glucose solution and water.

Digestibility of Infant Food and its Nutritional Meaning (유아식품(乳兒食品)의 소화율(消化率)과 영양적(營養的) 의의(意義))

  • Digestibilities of flour of white wheat, barley, potato, sweet potato, are $5{\sim}11$ percent (3hrs.) in unripe starch state and in gelatinization, digestibilities are up to $20{\sim}30$ percent (3hrs.). And in two states, digestibilities of flour of potato and sweet potato are lower than barley and white wheat flour. If it is not gelatinization, digestibilities of biscuit and cracker are much lower than popped cake (29%). In gelatinization, digestibilities of biscuit and cracker $(19{\sim}29%,\;3hrs.)$ are similar to gelatinized flour of materials. Digestibilities of bread and castern is $21{\sim}32$ percent, it is similar to gelatinized flour of materials. And bread is higher than castera. Digestibility of on the marketing weaning food (II) (15%, 3hrs.) is much lower than food (I) (24.1%, 3hrs.) When the date of manufacturing passed $1{\sim}2$ months, digestibility of some cookies have a tendency to low. When mothers choice the infant weaning food, they consider metritional value (42%) and digestibility (27%). Also when they make weaning food at home, they generally use flour of rice (41%) and white wheat (23%). And they think, the rate of digestion is 49% in the rice flour and white wheat (23%). It was sort of cooked of rice (16%) and weaning food on the marketing(13%), which accepts to good digest. Also it was sort of biscuits (44%) which comparatively high choice snack food.

Food Quality of Muffin with Germinated Brown Rice Soaked in Mycelial Culture Broth of Phellinus linteus (상황버섯균사체 배양액 침지 발아현미를 첨가한 머핀의 식품학적 특성)

  • The purpose of this study was to examine the qualities of optimized muffins with germinated brown rice soaked in mycelial culture broth of Phellinuslinteus (GBRP) using response surface methodology. Firstly, general compositions of optimized muffins with GBRP were higher than that of control and total sugar contents were similar. However, the total free amino acid and constitutional amino acid contents except for GABA were lower than those of control. Starch hydrolysis in control was higher than in optimized muffins with GBRP, whereas protein digestibility and protein efficiency ratio were not. The weights of optimized muffins with GBRP were higher than that of control (p<0.01), whereas height (p<0.01) and pH (p<0.001) were similar. The hardness (p<0.05) and chewiness (p<0.05) of optimized muffins with GBRP were higher compared to control; adhesiveness, springiness, and gumminess were similar, but cohesiveness (p<0.01) was not. The flavor (p<0.05) and taste (p<0.01) of optimized muffins with GBRP were higher than those of control; appearance, texture and overall acceptability were similar, but color (p<0.05) was not. The total polyphenol contents (p<0.01), DPPH radical scavenging activity (p<0.01), and superoxide dismutase-like activity (p<0.05) of optimized muffins with GBRP were higher than those of control, but nitrite scavenging activity was similar.

Processing Conditions for Protein Enriched Jeung-Pyun (Korean Fermented Rice Cake) (전통증편의 단백질보강에 관한 연구)

  • To improve the food quality of traditional Jeung-pyun(Korean fermenfed rice cake), effect of mixed ingredients on the quality of protein enriched product was studied. Changes were evaluated in chemical and textural properties of protein enriched Jeung-pyun altered by fermentation time, temperature, and steaming time. The maximum volume wxpansion was noted in dough mixed with rice flour(100 g), soy flour(25 g), sucrose(20 g), Tak-ju(50 ml) and water(10 ml), and then fermented at $35^{\circ}C$ for 3 hours. Soy protein isolate(SPI) enriched rice dough had a maximum expanded volume when 15g(w/w) of SPI, 20g(w/w) of sucrose, 60ml(v/w) of Tak-ju and 50ml(v/w) of water added into 100g rice flour, and then fermented at $30^{\circ}C$ for 2 hours. Steaming after fermentation made the smaller volume of bulk $45^{\circ}C$-50% of the initial volume be showed just after fermentation. Protein enriched Jeung-pyun prepared under the maximal volume endowing conditions showed the best protein quality(protein digestibility and protein efficiency ratio) and starch structure which was susceptible towards enzyme reaction. The improved starch quality of protein enriched Jeung-pyun could be confirmed by reducing power, gelatinization degree and amylose content. Jeung-pyun riched with soy flour at 20% level or with SPI at 5% showed the best overall quality by sensory and textural property but it had a problem in browning.

Physicochemical Properties of Rice Flour of Different Cultivars using Wet and Dry Milling Processes (제분방법에 따른 품종별 쌀가루의 이화학적 특성 및 소화율)

  • Rice flours from five rice (Oryza sativa L.) varieties with different amylose content were prepared by both wet and dry milling processes. The moisture content of wet-milled rice flours (WMR) was approximately three-times higher than that of dry-milled rice flours (DMR). Water absorption index (WAI), water solubility index (WSI), and swelling power (SP) increased in proportion to temperature. The WAI, WSI and SP values of DMR were higher than those of WMR. Baeokchal (BOC), which is a waxy rice cultivar, had a significantly high WSI value. Pasting properties of DMR, except for the BOC cultivar, resulted in an increase in peak, trough, final, and setback viscosities. The levels of resistant starch in four cultivars, except for Dodamssal (DDS), were under 1%, irrespective of the milling process, whereas the resistant starch contents of DMR and WMR in DDS were 9.18% and 6.27%, respectively. In vitro digestibility of WMR was higher than that of DMR, and the estimated glycemic index of the rice flour varieties ranged from 57.6 to 81.3. Damaged starch content of WMR was less than that of DMR; in addition, a negative correlation was observed between the amylose and damaged starch contents of WMR. These results suggest that the properties of rice flour vary depending on the milling method and flour variety, and could be a reference for selecting the appropriate processing method.

Replacement of Imported Timothy Hay with Domestic Italian Ryegrass Silage in a Horse Feedstuff (말에서 국내산 이탈리안 라이그라스 사일리지의 수입산 티모시 건초 대체 효과)

  • This study was conducted to investigate the possibility of replacing imported Timothy hay (TH) with domestic Italian ryegrass silage (IRGS) as a horse feed considering feed quality, nutrient digestibility and feed price. Two experimental diets (TH and IRGS) were fed to six-headed Thoroughbred (body weight, 475.7±33.3kg) of the Korea Racing Authority of Wondang Stud Farm. The 3 head animals were assigned to Control group (TH) and Treatment group (IRGS), respectively. The nutrient digestibility was determined by the total collection method. IRGS is enough for using as a horse feed because its Relative feed value(RFV) was higher than TH and its fermentation quality is suitable for horses. Although no difference was observed in nutrient digestibility, Total digestible nutrients(TDN), and Digestible energy(DE) between Control and Treatment group (p>0.05), the fact that price of IRGS was much lower (53.7~62.4%) than that of TH indicates IRGS has competitive advantage over TH as a horse forage feed. The present study indicates that IRGS can be fully replaced with TH due to its superior economic value even though the similarity of its nutrient digestibility, TDN, and DE to TH.

Effects of mushroom composition on the quality characteristics of extruded meat analog (버섯 첨가가 압출성형 대체육의 품질 특성에 미치는 영향)

  • This study was conducted to investigate the effects of mushroom composition (0, 4, 8, and 12%) on the quality characteristics of an extruded meat analog. The meat analog blend was isolated soy protein, wheat gluten, and corn starch (50:40:10). The extrusion condition was set to 55% feed moisture, 170℃ barrel temperature, and 150 screw speed by high moisture extrusion using a twin-screw extruder equipped with a cooling die. The integrity index, hardness, cohesiveness, springiness, chewiness, and cutting strength of the meat analog increased with the increasing mushroom content, while its water holding capacity and nitrogen solubility index (NSI) decreased. The protein digestibility decreased with the increasing mushroom content, while the DPPH radical scavenging activity significantly increased. In conclusion, the incorporation of mushrooms into the investigated meat analog enhanced its texture and antioxidant level.

Hydrogen Production from Tofu Manufacturing Wastewater by Heat-treated Anaerobic Microflora from the Concentrated Sewage Sludge (농축 하수오니 유래 열처리 혐기세균 복합체를 이용한 두부제조 폐수로부터 수소 생산)

  • 합성 및 두부 제조 폐수로부터 혐기 세균 복합체를 이용하여 수소를 생산하였다. 수소생산 혐기 세균 복합체는 하수처리장 농축 소화조에서 발생하는 슬러지를 $90^{\circ}C$에서 20분간 열처리하여 얻었다. 혐기 세균 복합체는 $37^{\circ}C$ 회분식 운전조건에서 1% (w/v) 포도당 함유 PYG (peptone-yeast extract-glucose) 배지로부터 1.15 L-$H_2$/g-균체건조량의 수소를 생산할 수 있었고, 이때 주요 유기산으로 15 mM acetate와 32 mM butyrate가 생성되었다. 같은 발효조건에서 1.4% 전분과 0.07% 환원당을 포함하는 두부 제조 폐수로부터 1.76 L $H_2$/L-두부제조폐수의 수소를 발생하였다. 이와 같은 결과로 부터 포도당과 두부 제조 폐수로부터 혐기세균 복합체에 의한 수소생산 효율은 각각 1.9과 0.9 mol $H_2$/mol 포도당을 나타내었다. 반연속운전(HRT, 12 시간)시 합성폐수를 이용하여 60일 이상 안정적으로 수소를 생산할 수 있었고, 이 때 혐기 세균 복합체는 1.3-2.0 L $H_2$/L-배양액을 발생하였다. PCR-DGGE(polymer chain reaction-denaturing gradient gel electrophoresis) 분석결과, 반응기 내 세균 복합체의 주요 미생물은 Clostridium 종이었다. 본 연구는 적절한 열처리를 통해 혐기 소화조 슬러지로부터 고활성 수소생산 세균 복합체를 얻을 수 있으며, 이들 세균 복합체를 이용하여 합성 및 두부제조 폐수로부터 효율적인 수소생산이 가능하다는 것을 나타내고 있다.

  • The objectives of this study was to determine whether a new physically modified cornstarch by ultra-fine- or nanoscale pulverizer to reduce particle size offers better bioactive function than native cornstarch in weanling Sprague-Dawley rats. Male weaning Sprague-Dawley rats were fed diets containing native cornstarch (NAC), ultra fine pulverized cornstarch (UFC) or nano-scale pulverized cornstarch (NSC) for 4 weeks. In vitro rate of starch hydrolysis, growth performance, organ weight, intestine length intestinal proliferation and the fermentation by Bifidobacterium of rat cecum were evaluated. The diet with reduced particle size (UFC or NSC) significantly increased body weight gain and organ weight. Feed efficiency was increased in NSC fed rats and was not affected in UFC fed rats. Intestinal proliferation was decreased in NSC group. Reduction of particle size also increased cecal short chain fatty acid concentration and the growth and acidifying activity of Bifidobacterium. It is concluded that a reduction of particle size of starch granules by physically modification may increase growing performance and gut function.

Changes in Retrogradation Characteristics of Mungbean Starch Gels during Storage (저장에 따른 녹두전분 겔의 노화 특성 변화)

  • This study was carried out to investigate the effects of storage temperature(5$\^{C}$ and 25$\^{C}$) and time(1, 24, 48 and 72hours) on the retrogradation characteristics of mungbean starch gels by using differential scanning calorimeter, rapid visco-analyzer, $\beta$-amylase method, X-ray diffractometer and scanning electron microscope. Endothermic peaks of gels stored at 5$\^{C}$ were shifted to the lower temperature than those stored at 25$\^{C}$ and endothermic enthalpy of gels stored at 5$\^{C}$ was greater than that stored at 25$\^{C}$. DSC thermograms showed that the retrogradation rate of gels stored at 5$\^{C}$ was higher than that shored at 25$\^{C}$. The pasting properties of the gels such as peak viscosity and final viscosity increased with increasing storage time except the gels stored at 25$\^{C}$ for 72 hours and this trend was more apparent at the storage temperature of 5$\^{C}$. The digestibility measured by $\beta$-amylase method decreased with increasing storage time and this trend was more apparent at the storage temperature of 5$\^{C}$. X-ray diffraction patterns of the gels stored at 5$\^{C}$ showed more distinct peak than those stored at 25$\^{C}$, but no difference was detected by the storage time. The microstucture of all the gels showed a well developed three dimensional network The network structure of the gels stored at 25$\^{C}$ formed a thick matrix after 72 hours and those stored at 5$\^{C}$ from the early stage of storage.
