Cultivation Condition of Transformant Alcaligenes eutrophus Harboring Cloned phbC Gene for Production of P(3-hydroxybutyrate-3-hydroxyvalernte) Containing High Molar Fraction of 3-Hydroxyvalerate. (P(3-hydroxybutyrate-3-hydroxyvalerate)의 생산을 위한 재조합 phbC 유전자를 형질전환시킨 Alcaligenes eutrophus의 배양조건 검토)

  • The cultivation conditions of transformant Alcaligenes eutrophus AER5 harboring cloned phbC gene for mass production of poly (3-hydroxybutyrate-3-hydroxyvalerate)[P(3HB-3HV)] containing high molar fraction of 3-hydroxyvalerate (3-HV) were investigated. In two-stage batch cultivation, transformant accumulated P(3HB-3HV) containing 52.2 mol% of 3HV compared to 30 mol% of parent strain A. eutrophus H16. The increased 3-HV molar fraction was due to the amplified activity of PHB synthase participating in condensation of 3-HB and 3-HV. To increase efficiency of P(3HB-3HV) accumulation, fructose was added along with precursor compound valerate, and total cell mass and P(3HB-3HV) concentrations remarkably increased, but not 3-HV molar fraction. The effect of magnesium ion showed that P(3HB-3HV) concentration and 3-HV molar fraction were significantly increased upto 6.1 g/L and 71.3 mol% at 0.01 g/L of MgSO$_4$, respectively. The efficiency of several pH adjuster, NaOH, NaOH and (NH$_4$)$_2$SO$_4$, and NH$_4$OH, on total cell mass, p(3HB-3HV) concentration, and 3-HV molar fraction was also compared. To overcome the disadvantage of two-stage cultivation, one-stage intermittent fed-batch cultivation was attempted, such a way 10.0 g/L of fructose was supplied for cell growth at initial 36 hr and then 10.0 g/L of valerate and 5.0 g/L of fructose were applied to induce the accumulation of P(3HB-3HV), consequently, 10.4 g/L of P(3HB-3HV) with 38 mol% of 3-HV fraction could be obtained after 72 hr. These results can be used for elucidating cultivation strategy for mass production of P(3HB-3HV) containing high 3-HV molar fraction using transformant A. eutrophus AER5 harboring cloned phbC gene.

The Dietary Effect of Royal Jelly Supplementation on Epidermal Levels of Filaggrin and Free Amino Acids during Menopause in Rats (폐경기 노화 유도 전후의 로얄제리 식이공급이 백서 표피의 필라그린과 유리아미노산 함량 및 관련 대사 효소의 단백질 발현 변화에 미치는 영향)

  • Epidermal hydration is mainly maintained by natural moisturizing factors (NMFs). Of these various NMFs, free amino acids (AAs) are major constituents generated by filaggrin degradation. The reduction of these AAs has been reported in aging skin induced during menopause. In this study, we examined whether the dietary supplementation of royal jelly (RJ) during the pre- and post-menopausal period alters epidermal levels of filaggrins, free AAs, and peptidylarginine deiminase-3 (PAD-3) (an enzyme involved in filaggrin degradation processes). Sprague Dawley rats were divided into five groups: groups fed a control diet for 12 weeks, in which an ovariectomy (OVX) or sham operation (SHAM) were underwent at week 4; groups fed a diet with 1% RJ harvested in different area of Korea (RJ1 and RJ2); and a group fed a diet with isoflavone (IF), the typical functional food for menopause prevention, for 4 weeks before and 8 weeks after an ovariectomy operation. In the epidermis of group OVX, total filaggrins (including profilaggrin and filaggrin) were reduced; these levels in groups RJ1 and IF were similar or less than in group OVX. However, total AAs, which showed no apparent difference between groups SHAM and OVX, were highly increased in groups RJ1 and IF. Specifically, aspartate (Asp) and proline (Pro), the major AAs in functioning NMF, were highly increased in group RJ1. Although total filaggrins, profilaggrin, filaggrin and PAD3 increased, total AAs (including Asp and Pro) in group RJ2 were modest or less than in group RJ1. The PAD3 alteration was not apparent among the four other groups. Taken together, we demonstrate that the diet supplementation of RJ1 enhanced filaggrin degradation (but not through the increased protein expression of PAD3), and increased total AAs, Asp and Pro. RJ1 could be a dietary supplementation for preventing the skin aging induced during menopause.

A Study on Prediction of Asian Dusts Using the WRF-Chem Model in 2010 in the Korean Peninsula (WRF-Chem 모델을 이용한 2010년 한반도의 황사 예측에 관한 연구)

  • The WRF-Chem model was applied to simulate the Asian dust event affecting the Korean Peninsula from 11 to 13 November 2010. GOCART dust emission schemes, RADM2 chemical mechanism, and MADE/SORGAM aerosol scheme were adopted within the WRF-Chem model to predict dust aerosol concentrations. The results in the model simulations were identified by comparing with the weather maps, satellite images, monitoring data of $PM_{10}$ concentration, and LIDAR images. The model results showed a good agreement with the long-range transport from the dust source area such as Northeastern China and Mongolia to the Korean Peninsula. Comparison of the time series of $PM_{10}$ concentration measured at Backnungdo showed that the correlation coefficient was 0.736, and the root mean square error was $192.73{\mu}g/m^3$. The spatial distribution of $PM_{10}$ concentration using the WRF-Chem model was similar to that of the $PM_{2.5}$ which were about a half of $PM_{10}$. Also, they were much alike in those of the UM-ADAM model simulated by the Korean Meteorological Administration. Meanwhile, the spatial distributions of $PM_{10}$ concentrations during the Asian dust events had relevance to those of both the wind speed of u component ($ms^{-1}$) and the PBL height (m). We performed a regressive analysis between $PM_{10}$ concentrations and two meteorological variables (u component and PBL) in the strong dust event in autumn (CASE 1, on 11 to 23 March 2010) and the weak dust event in spring (CASE 2, on 19 to 20 March 2011), respectively.

Soil Salinity and Vegetation Distribution at Four Tidal Reclamation Project Areas (4개 간척 지구에 분포하는 식생과 토양 염류농도)

  • This research was conducted to present reference data to be used as newly reclaimed tidal land management. We investigated vegetation succession at 4 reclaimed/reclaiming project areas and discussed relationship with soil and vegetation trhrough investigation and analysis soil chemical characteristics at 2 areas. 14 families 58 kinds were investigated. Vegetation were variou at Dea-Ho conservation polt and Seok-Mun National Industrial Area which are maintaining naturally. Vegetation were simple at Hong-Bo and Dongjin and MinKyong river areas which effected sea water. Common species that were investigated at 9 sites were Suaeda asparagoides, Aster tripolium, Phragmites australis, Suaeda maritima, Suaeda japonica, Carex scabrifolis. As soil desalinization progressing, soil classified at first saline-soidc soil, the nest saline soil and then normal soil. Chenopodiaceae revealed at about 30 dS/m of soil ECe and existed to 10 dS/m of soil ECe. At about 20 dS/m of soil ECe. Aster tripolium, Calamagrostis epigeios, and Sonchus brachyotus revealed and then non-halophytes and common plants at inland revealed at low soil ECe of about 10 dS/m. However it was not to progress vegetation sucdession and soil desalinization at the same time, owing to input of seeds or plants ect from out-ecosystem. So for promotion of vegetation at newly reclaimed tidal land, we proposed that it was very effective to plant artificially halophytes or suitable species through soil test.

Charaterization of Biomass Production and Wastewater Treatability by High-Lipid Algal Species under Municial Wastewater Condition (실제 하수조건에서 고지질 함량 조류자원의 생체생성과 하수처리 특성 분석)

  • Wastewater treatment using algal communities and biodiesel production from wastewater-cultivated algal biomass is a promising green growth technology. In literature, there are many studies providing information on algal species producing high content of lipid. However, very little is known about adaptability and wastewater treatability of such high-lipid algal species. In this study, we attempted to characterize algal biomass production and wastewater treatability of high-lipid algal species under municipal wastewater condition. For this, four known high-lipid algal strains including Chlorella vulgaris AG 10032, Ankistrodesmus gracilis SAG 278-2, Scenedesmus quadricauda, and Botryococcus braunii UTEX 572 were individually inoculated into municipal wastewater where its indigenuous algal populations were removed prior to the inoculation, and the algae-inoculated wastewater was incubated in the presence of light source (80${\mu}E$) for 9 days in laboratory batch reactors. During the incubations, algal biomass production (dry weight) and the removals of dissolved organics (COD), nitrogen and phosphorous were measured in laboratory batch reactors. According to algal growth results, C. vulgaris, A. gracilis and S. quadricauda exhibited faster growth than indigenuous wastewater algal populations while B. braunii did not. The wastewater-growing strains exhibited efficient removals of total-N, ${NH_4}^+$-N, Total-P and ${PO_4}^{3-}$-P which satisfy the Korea water quality standards for effluent from municipal wastewater treatment plants. A. gracilis and S. quadricauda exhibited efficient and stable treatability of COD but C. vulgaris showed unstable treatability. Taken together with the results, A. gracilis and S. quadricauda were found to be suitable species for biomass production and wastewater treatment under municipal wastewater condition.
