Evaluation of Insecticidal and Antifeeding Activities of Eco-friendly Organic Insecticides Against Agricultural Insect Pests (농업해충에 대한 친환경유기농자재들의 살충력 및 섭식저해력 평가)

  • Insecticidal and antifeeding activities of 29 commercialized eco-friendly organic products for managing plant diseases and insect pests against Plutella xylostella larvae, Spodoptera exigua larvae, Frankliniella occidentalis adults, and Myzus persicae adults were tested using spraying and leaf dipping bioassays under laboratory conditions. Products containing 60% Sophora extract (EOIS) and mixtures (EOISm) with Sophora extract, Stemona japonica extract, Melia azedarach extract, and Nepeta cataria extract as well as mixtures (EOISc) with Sophora extract, Chenopodium ambrosioides extract, and Melia azedarach extract as active ingredients showed strong insecticidal activity at recommended concentration against P. xylostella larvae. At half concentration, their insecticidal activities were decreased under 50%. The EOIS gave good insecticidal activity against S. exigua larvae and also showed 85% and 95% insecticidal activity at 24 and 48 hours after treatment to F. occidentalis adults, respectively. For M. persicae adults, EOISm and mixtures (EOIR) containing rape seed extract, neem extract, and castar oil produced 93% and 68% insecticidal activity, but their activities did not be increased at double concentration. EOISm only showed 100% contact toxicity against M. persicae adults exposed to dipping leaves. Interestingly, the insecticidal activity of EOIR and EOICi (citronella oil and derris extract) against M. persicae adults was increased with exposed time and concentration. In addition, EOICe (cedar oil), EOIS, EOISm, EOISc, EOIM (microorganism), EOIR, EOIPe (plant extract), and EOIT (tea tree extract) gave strong antifeeding activity against S. exigua and P. xylostella larvae. EOIB, EOIBs, EOIM, EOICi, and EOIMc showed above 70% antifeeding activity to the lepidopteran larvae. These results indicate that mixtures containing 2 to 3 plant extracts with Sophora extract show good activities against insect pests, although the difference of insecticidal and antifeeding activities was produced depending on both a tested insect species and an active ingredient or concentration.

Development of molecular markers for varietal identification of Brassica juncea on the basis of the polymorphic sequence of ITS regions and MITE families (갓 (Brassica juncea) 품종구분을 위한 ITS 영역 및 MITE Family 정보를 이용한 분자표지 개발)

  • Brassica juncea (2n = 4x = 36, AABB genome, 1,068 Mb) is a U's triangle species and an amphidiploid derivative of B. rapa and B. nigra. Fifteen varieties were used to study the ITS (internal transcribed spacer) regions of ribosomal DNA and MITEs (miniature inverted-repeat transposable elements) with a view of developing specific molecular markers. ITSs and MITEs are an excellent resource for developing DNA markers for genomics and evolutionary studies because most of them are stably inherited and present in high copy numbers. The ITS (ITS1 and ITS2) sequence was compared with the consensus sequence of B. rapa and B. nigra. Variation in ITS1 created two separate groups among 15 varieties, with 10 varieties in one group and 5 in the other. Phylogenetic analysis revealed two major clusters for those 10 and 5 varieties. Among the 160 different MITE primers used to evaluate the selected 15 varieties of B. juncea, 70 were related to the Stowaway, 79 to the Tourist, 6 to the hAT, and 5 to the Mutator super-families of MITEs. Of 160 markers examined, 32 were found to be polymorphic when fifteen different varieties of B. juncea were evaluated. The variety 'Blackgat' was different from the other mustard varieties with respect to both phenotype and genotype. The diversity of 47 additional accessions could be verified using eight selected molecular markers derived from MITE family sequences. The polymorphic markers identified in this study can be used for varietal classification, variety protection, and other breeding purposes.

Monitoring of Residual Pesticides in Commercial Agricultural Products in Korea (국내 유통 농산물의 농약 잔류실태 모니터링)

  • We tested for residual pesticide levels in agricultural products purchased from 5 provinces within the middle region of Korea during 2006. A Total of 488 samples of 23 different types of agricultural products were analyzed by GC/MS, GC-NPD, and LC/MS/MS. We used multi-analysis methods to analyze for 231 different pesticide types; a single residual pesticides were detected in 92 samples (18.9%), of which 9 samples (1.9%) exceeded the Korea Maximum Residue Limits (MRLs). we detected pesticide residue in more than 50% of the pepper leaf and welsh onion samples. For the welsh onions, 16 among the 30 analyzed samples contained pesticide residue, and 4 samples exceeded the Korea MRLs. Among the 234 kinds of pesticides we tested for, 42 were detected, and 21 of them were detected more than twice. Six pesticide residue types, including cypermethrin, iprodione, fludioxonil, ethoprophos, flutolanil, and lufenuron, exceeded the MRLs. No residual pesticides were detected in 396 of the samples (81.1%), and the residual pesticide levels in 83 samples (17.0%) were lower than the Korea MRLs, indicating that 98.1% of the samples were relatively safe.

Study on Status of Nutritional Supply by Lunch-box in High School (고등학생(高等學生)의 도시락에 의한 영양섭취상태(營養攝取狀態)에 관(關)한 조사연구(調査硏究))

  • This study was projected to get basic data which can provide a basis for future direction in nutritional education, and also to find the way how to improve the nutritional supply by evaluating the current nutritional intake of average high school students through the survey study of their daily packed lunch. Five hundred twenty seven students from two boys high school and two girls high school including one general and one vocational school respectively were chosen as random sampling technique. Four hundred forty nine among the 527 students had brought lunch. The contents of lunch box were weighed and converted into nutritional values according to the food composition table and compared with recommended dietary allowances. The results compared and classified by sex, School and housewives' educational level were as follows: 1. The nutritional supply in the lunch box was 671 Cal of energy and 22.3 gm of protein for male students which were respectively 55.9% and 74.2% of the dietary recommendations. On the other side female student's lunch boxes were found to contain 495 Cal of energy and 21.3gm of protein which are respectively 61.8% and 80% of the dietary prescriptions. Excluding niacin, all vitamins and minerals were found to be short. 2. Calorie intake in the vocational high school was found to be higher than in the general high school but lower in protein intake especially significant difference (P<0.01) in animal protein. 3. From the nutritional point of view the educational backgrouud of the housewives was not found to have any influence in the way of preparing the lunch boxes. 4. Nutrients of lunch box were heavily inclined to grain rather than to side dishes.

  • The pool size of plasma glucose, turnover rate and other concerned items for glucose metabolism in normal laying hen were investigated by a single-injection method using $U-C^{14}-glucose$. The 11.6 nCi of pure dose was injected to a hen normally fed through the wing vein. The glucose concentration in plasma sample taken at 5 minutes after injections was 214mgper 100ml. From the plottings of logarithmic standard specific activities of plasma taken from 5 to 120 minutes against the time after injection and from the regresion analysis, metabolic states were determined. The pool size was 1.07g, turnover rate was 0.024 per minute, turnover time was 41 minutes, utilization rate was 26mg/min. (0.83 g/hr/kg B.W. 3/4) and glucose space(extracellular fluid volume) was 25.3 per cent of body weight. The values obtained from. 10-50 minutes samples were similar to those described above, which we from 5-120 minutes samples.
