The Effect of Ginkgo Biloba Extract on the Fractionsted Radiation Therapy in C3H Mouse Fibrosarcoma (Ginkgo Biloba Extract가 C3H 마우스 섬유육종의 분할 방사선치료에 미치는 영향)

  • Purpose : A ginkgo biloba extract (GBE) has been known as a hypoxic cell radiosensitizer. Its mechanisms of action are increase of the red blood cell deformability, decrease the blood viscosity, and decrease the hypoxic cell fraction in the tumor. The aims of this study were to estimate the effect of GBE on fractionated radiotherapy and to clarify the mechanism of action of the GBE by estimating the blood flow in tumor and normal muscle. Materials and Methods : Fibrosarcoma (FSall) growing in a C3H mouse leg muscle was used as the tumor model. When the tumor size reached 7 mm in diameter, the GBE was given intraperitoneally at 1 and 25 hours prior to irradiation. The tumor growth delay was measured according to the various doses of radiation (3, 6, 9, 12 Gy and 15 Gy) and to the fractionation (single and fractionated irradiation) with and without the GBE injection. The radiation dose to the tumor the response relationships and the enhancement ratio of the GBE were measured. In addition, the blood flow of a normal muscle and a tumor was compared by laser Doppler flowmetry according to the GBE treatment. Results : When the GBE was used with single fraction irradiation with doses ranging from 3 to 12 Gy, GBE increased the tumor growth delay significantly (p<0.05) and the enhancement ratio of the GBE was 1.16. In fractionated irradiation with 3 Gy per day, the relationships between the radiation dose (D) and the tumor growth delay (TGD) were TGD $(days)=0.26{\times}D$ (Gy)+0.13 in the radiation alone group, and the TGD $(days)=0.30{\times}D$ (Gy)+0.13 in the radiation with GBE group. As a result, the enhancement ratio was 1.19 ($95\%$ confidence interval; $1.13\~1.27$). Laser Doppler flowmetry was used to measure the blood flow. The mean blood flow was higher in the muscle (7.78 mL/100 g/min in tumor and the 10.15 mL/100 g/min in muscle, p=0.005) and the low blood flow fraction (less than 2 mL/100 g/min) was higher in the tumor $(0.5\%\;vs.\;5.2\%,\;p=0.005)$. The blood flow was not changed with the GBE in normal muscle, but was increased by $23.5\%$ ( p=0.0004) in the tumor. Conclusion : Based on these results, it can be concluded that the GBE enhanced the radiation effect significantly when used with fractionated radiotherapy as well as with single fraction irradiation. Furthermore, the GBE increased the blood flow of the tumor selectively.

Isomalto-oligosaccharide Production from Maltose by Intact Cells of Aureobasidium pullulaans (Aureobasidium pullulaans를 이용한 Maltose로부터 Isomalto-oligosaccharides의 생산)

  • A new method for the production of isomalto-oligosaccharides from maltose was investigated using intact cells of Aureobasidium pullulaans which had been known to produce fructo-oligosaccharides. The cells showed transglucosylation activity producing isomalto-oligosaccharides at high concentrations of maltose, while they showed a hydrolytic activity at low concentrations of substrate when cultivated at $25^{\circ}C$. The optimum reaction conditions for the isomalto-oligosaccharide production were as follows: substrate concentration, 500g/l maltose; pH, 4.5; temperature, $65^{\circ}C$; cell dosage, 10 unit per gram substrate. Under optimized conditions, the maximum yield of isomalto-oligosaccharides achieved was around 48% (w/w). At the early period of reaction, panose was selectively produced from maltose, and thereafter isomaltotriose was synthesized by utilizing panose as a substrate when maltose consumption was discontinued.

Textural Properties of Gelatinized Model Food system (젤라틴화 된 모형식품의 조직특성)

  • The gelatinized model food system were prepared by combining moisture, starch and protein, and the textural properties of their gels of different temperatures and times of heating were investigated by the use of Instron Universal Testing Machine. The hardness, springiness, cohesiveness, gumminess and chewiness of model foods had a high correlation with solid content and the regression equations between the hardness of model foods and moisture content heated for 20min. at $80{\circ}C$ were as follows; $H(PS_4)=18.6405-3.8201M+0.1959M^2,\;H(P_1S_1)=244.7933-5.692M+0.0332M^2,\;H(P_4S)=693.0292-16.6884M+0.1005M^2$, The correlation coefficients were $0.996^{**},\;0.998^{**}\;and\;0.998^{**}$, respectively. Total correlations between textural parameters and temperature and heating times were different according to model foods. The correlation between textural parameters was proportional to protein foods, but the hardness and cohesiveness of starch foods showed the relationship of inverse proportion. Under low solid content, the parameters of model foods appeared to decrease as protein content increased. Under high solid content, the parameters of protein foods were higher than those of starch foods above some level of protein content. The regression equation between the hardness and protein content heated for 20min. at $80^{\circ}C$ was as follows; Hardness(20%)=5.6858-13.5670P+$9.7758P^2$ and the correlation coefficient was $0.95^{**}$.

Inhibitory Effects of Ginger and Processed (Beopje) Ginger Extracts on HCl-ethanol Induced Gastritis in Rats (염산-에탄올에 의해 유발된 흰쥐 위염에 대한 생강 및 법제생강 추출물의 억제작용)

  • This study examined the protective effects of ginger and processed (Beopje) ginger extracts on gastritis induced by HCl-ethanol in SD-rats. Beopje (or Poje) is a branch of herbal medicines processed using a Korean traditional method to achieve specific pharmacological effects. Gastric lesions were induced in the rats by a treatment of 1 mL of HCl-ethanol (60% ethanol+150 mM HCl). The rats were divided into seven groups: Normal (1 mL of saline without HCl-ethanol treated group), Control (HCl-ethanol treated group), GL (35 mg/kg of ginger treated group), BGL (35 mg/kg of Beopje ginger treated group), GH (350 mg/kg of ginger treated group), BGH (350 mg/kg of Beopje ginger treated group) and Cimetidine (80 mg/kg of cimetidine treated group). The gastric injury inhibition rate was 40.2% and 64.9% in GL and BGL and 68.4% and 99.6% in GH and BGH respectively, showing significantly lower rates than the control (p<0.05). The level of gastric juice secretion decreased significantly in all ginger administered groups. The pH of the gastric juices of BGH increased and the acidity of BGH and cimetidine decreased significantly (p<0.05) compared to the other groups. Beopje ginger had stronger inhibitory effects on gastritis than ginger without the Beopje process. The protective effect on gastritis by the ginger and Beopje ginger extracts increased in a dose-dependent manner (p<0.05). These results suggest that ginger has inhibitory effects on HCl-ethanol induced gastritis in rats that can be improved through the Beopje process.

Quality Comparison of Commercial Cider Vinegars by Their Acidity Levels (시판 사과식초의 산도에 따른 품질특성 비교)

  • The quality of commercial cider vinegars was compared according to their acidity levels (low, general, double strength and triple strength). The pH, reducing sugar content and brown color intensity decreased, while the total acidity increased with the increasing acidity levels, which may be resulted from difference in formulation and manufacturing procedures. The free sugars were mainly composed of fructose and glucose, which were the highest in low acidity vinegar, followed by double strength, general, and triple strength acidity vinegars. Acetic acids and malic acids were identified as the major organic acids. The citric acid concentration was the highest in triple strength vinegars. The contents of total phenolics and flavonoids were the highest in low acidity vinegars, which indicated their dependence on the apple juice content. DPPH and ABTS radical scavenging abilities were the highest in low acidity vinegars, that showed high amounts of antioxidants.

The Clinical Efficacy of the Low-dose FSH Regimen for Intrauterine Insemination (인공수정 시술시 저용량 FSH(Low-dose FSH) 용법의 임상적 효용성에 관한 연구)

  • Objective: This study is to investigate the clinical efficacy of low-dose FSH regimen, comparing with clomiphene citrate and human menopausal gonadotropin (CC/hMG) regimen. Methods: Retrospective study of the ovulatory factor infertility 39 patients who had been treated by intrauterine insemination (IUI). The 31 cycles of 21 patients were stimulated by CC/hMG regimen, the 22 cycles of 18 patients were stimulated by low-dose FSH regimen. We compared the rate of clinical pregnancy, multiple pregnancy and ovarian hyperstimulation syndrome (OHSS) of both group. Results: The rate of clinical pregnancy of the CC/hMG group was 25.7% per cycle, and that of the low-dose FSH group was 54.5% per cycle. The low-dose FSH group showed a higher rate of clinical pregnancy per cycle than CC/hMG group (p=0.028). However, no differences was found statistically in the rate of multiple pregnancy and OHSS between CC/hMG group (22.2%, 5.7%) and low-dose FSH group (33.3%, 13.6%). Conclusion: This study showed that the low-dose FSH regimen is superior to CC/hMG regimen in getting clinical pregnancy, but dose not reduce the ovulation induction complications.

Adsorption and Degradation of Alkylbenzenesulfonate by Soils (Alkylbenzenesulfonate의 토양(土壤)에 의한 흡착(吸着)과 분해(分解))

  • A laboratory experiment was conducted to investigate the effects of pH, organic matters and anion on the adsorption and degradation of surfactant by different soils; Anmi series (limestone region), Gangseo series (alluvial soil). For this study, Alkyl Benzene Sulfonate (ABS ; Sodium Dodecylbenzenesulfonate) was used as a surfactant. The results were as follows: 1. Adsorption of ABS by soils was correlated positively with the equilibrium concentration of ABS in a soil suspension. (Anmi seris : r=0.9855, Gangseo series : r=0.9931). 2. Adsorption rate of ABS by soils was about 70% of the treated concentration ($600{\mu}g$ ABS/g soil) in a range of pH 4 to pH 5, and about 20% for pH 8. 3. Addition of electrolytes increased ABS adsorption by soils in a soil suspension; the higher concentration, the higher adsorption. But the influence among electrolytes was not significant. 4. Adsorption of ABS by soils was not affected by soil organic matter content in this experiment. 5. Degradation rate of ABS in a soil suspension was about 85% at $30^{\circ}C$, and about 10 to 15% at $10^{\circ}C$. Addition of sewage accelerated the degradation rate regardless of temperature and reached about 85% in a week.

Changes in the Conrent of Chlorophylls and Their Derivatives in Brined Korean Cabbages Added with Ingredients during Sorage (부재료의 첨가에 따른 절임배추의 숙성 중 Chlorophyll 및 그 유도체의 함량변화)

  • The brine Korea cabbage (BKC) with various ingredients was stored at 5$^{\circ}C$ and 15$^{\circ}C$ for 13 days to examine the changes in pH, total acidity, ascorbic, and cholrophylls. Decrease in pH and increase in total acidity in the BKC stored at 15$^{\circ}C$ were greater than in the BKC stored at 5$^{\circ}C$, indicating these changes are closely related to the storage temperature. The effect of ingredints was various; garlic, red pepper powder, and fermented anchovy juice accelerated the decrease in pH and increase in total acidity; mustard powder and leaf mustard suppressed their changes and freen onion and ginger had no effect. At both temperatures, ascorbic acid was remained at high level in the BKC with leaf mustard, but it was maintained at lowest level in the BKC with ferented anchovy juice. Degradation od chlorophylls was slow in the BKC with leaf mustard, which maintained the high level os ascorbic acid during storage. Meanwhilr the degradation of chlorophylls or production of pheophytin and pheophorbide was cinsistent with the production of acid. These results suggset that degradation of chlorophylls in a BKC was positively correlated with ascorbic acid content and the acid produced during storage.

광미/광폐석 처리를 위한 고형화 공정 실증 실험

  • 본 연구에서는 폐광산 주변에 산재되어 있는 광미/광폐석을 처리하기 위하여 고형화 실증 실험을 수행하였다. 고형화 공정에서 흔히 사용하는 포틀랜드 시멘트와 MSG-E, MSG-N을 고화제로 사용하였으며 현장 광미 및 광폐석을 대상으로 고화체를 양생하고 고화체의 압축강도 및 중금속 용출 정도를 측정하였다. 고화체의 물리/화학적 특성을 비교하기 위해 광미/고화제 비율, 배합수/고화제 비율 그리고 고화체 양생기간을 실험인자로 설정하였다. 실험 결과 광미/고화제의 비율 1:1 만을 고려하더라도 중금속 용출의 급격한 감소가 이루어지는 것을 확인할 수 있었으며 광미/고화제의 비율을 3:1 이하로 유지시키는 경우, 고화체의 압축강도가 현행 폐기물 관리법(20조 관련)에서 규정하고 있는 차단형 매립시설 내부막의 압축강도 기준인 $0.21kgf/mm^2$ 보다 높은 것으로 나타났다. 다양한 pH를 갖는 수용액에 대하여 시간에 따른 고화체의 중금속 용출률을 측정한 결과, 수용액의 pH가 1과 13인 강산/강염기 용액에서 일부 중금속의 용출 농도가 지하수 생활용수 기준치를 초과하였으나, pH와 3 - 11인 경우에는 중금속 용출률이 급격히 감소하여 모두 기준치 이하를 나타내었다. 또한, pH가 1과 13인 수용액의 경우에도 고화체와 반응하는 시간이 증가할수록 고화체의 buffering 효과에 의해 수용액의 pH가 감소하였다. 이러한 결과는 현장에서 접촉수의 pH가 강산이나 강염기라 하여도, 고화체의 buffering 효과에 의해 시간이 지남에 따라 수용액의 pH가 낮아져 고화체로부터의 중금속 용출은 매우 감소할 것임을 의미한다.ss of an active application defined using the model. Physicochemical Properties of a Biopolymer Flocculant Produced from Bacillus subtilis PUL-A (Bacillus subtilis PUL-A로부터 생산된 Biopolymer 응집제의 물리화학적 특성)

  • Soybean milk cake (SMC) was used for the solid-state fermentation by Bacillus subtilis PUL-A isolated from soybean milk cake. In the presence of 5% glutamate the maximum production of biopolymer (59.9 g/kg) was performed by fermentation at $42^{\circ}C$ for 24 hr. The recovered biopolymer was consisted of 87% $\gamma$-polyglutamic acid with molecular weight of $1.3{\times}10^6$ dalton and other biopolymer. The biopolymer solution showed the great decrease in consistency below pH 6.0, regardless of the molecular weight of PGA. Biopolymer solution has a typical pseudoplastic flow behavior and yield stress. The consistency of biopolymer solution was greatly decreased by increasing heating time and temperature in acidic condition compared to the alkaline condition. In kaolin clay suspension, the flocculating activity of biopolymer was the highest value with 15 mg/L biopolymer and 4.5 mM $CaCl_2$, but decreased greatly with $FeCl_3$. The flocculating activity of biopolymer was maximum at pH5, but decreased drastically by heating at $60{\sim}100^{\circ}C$. In particular, biopolymer with native PGA showed the efficient flocculating activity compared to that of modified biopolymer containing low molecular weight of PGA.
