A new bacterial disease of sesame(Sesamum indicum) was observed on field-grown plants in Suwon, Hongchun and Yeonchun in 2000. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Isolations made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Two bacterial strains were purified and used for farther tests. Pathogenicity of strains was confirmed on 3-week-old sesame plants sprayed with bacterial suspensions containing $10^{8}cfu/ml$ of phosphate buffered saline. The Biolog and fatty acid analyses of the two strains(SL3451 and SL3476) 1mm sesame leaf blight showed that they could be identified as ft campestris pv. sesami because of their high similarity to the tester strain(X. campestris pv. sesami LMG865) with a match probability of $100\%$. The bacterium grew well between 18 and 36$^{\circ}C$, but optimum temperature was $27^{\circ}C$ on LB broth. This is the first report of bacterial blight of sesame in Korea. Symptoms of bacterial blight of sesame are difficult to differentiated with those of bacterial leaf spot caused by Pseudomonas syringae pv. sesami.
Powdery mildew of pepper is one of the most devastating diseases which is occurring all the year under greenhouse condition. In this study, control efficacy against powdery mildew was evaluated by mixed culture solutions of two chitinolytic bacteria, Lysobacter enzymogenenes strain C-3 and Chrornobacterium sp. strain C-61, cultivated in the chitin-supplemented medium. In all experiments, white powder on the reverse side of pepper leaves perfectly disappeared 3 days after application of mixed culture solutions. However, periods required for formation of new white powder on the same sites after application (control-lasting period) were largely differed according to environmental conditions. In particular, the control-lasting period was much longer when sprayed on 6 PM than 9 AM and especially, on rainy days than sunny days. This indicates that control efficacy of culture solution may be largely affected by environmental conditions after application. The undiluted culture solution resulted in a perfect control with control value more than 95% by application of 5-day-intervals under severely diseased field and 7-day-intervals under disease-started field. A ten-fold diluted product also showed control value more than 81% by application of the same method. These results suggest that this culture solution can be practically used to control powdery mildew disease in pepper plants.
Gymnophclloides seoi is a human intestinal trematode prevalent on southwestern islands in Korea. In the present study, we investigated whether G. seoi metacercariae can grow and develop into adults by in vitro cultivation. The metacercariae were obtained from naturally infected oysters, and cultured in uitro for 5 days under three conditions; $37^{\circ}C/5%{\;}CO_2,{\;}41^{\circ}C/8%{\;}CO_2,{\;}or{\;}41^{\circ}C/15%{\;}CO_2$, in NCTC 109 complete media containing 20% FBS and 1% antibiotics-antimycotics. The degree of worm growth and development was compared with that grown in uiuo of C3H mice. The length of the worms cultivated in uitro was $200-300{\;}\mu\textrm{m}$, not significantly different from metacercariae, whereas the length of the worms recovered from C3H mice was significantly larger, $300-400{\;}\mu\textrm{m}$. The worms produced eggs when grown in C3H mice or cultured in vitro for 2 days under $41^{\circ}C/8%{\;}CO_2{\;}or{\;}41^{\circ}C/5%{\;}CO_2$, but not when cultured under 37$^{\circ}C/5%{\;}CO_2$. Among the in vitro conditions, $41^{\circ}C/15%{\;}CO_2$ was best for egg Production, although the number of eggs was about half of worms obtained from C3H mice. In conclusion, in vitro cultivation of G. semi metacercariae into egg-pioducing adults was partially successful under culture conditions of $41^{\circ}C/5%{\;}CO_2{\;}or{\;}41^{\circ}C/8%{\;}CO_2$.
The growth responses of Chinese Cabbage, lettuce, and spinach were evaluated to investigate the feasibility of carbonized rice-hulls as a sanitary cultivation media instead of soil. The carbonized rice-hulls were pretreated with several inorganic acid such as nitric, sulfuric, hydrochloric, and phosphoric acid for neutralization. The neutralization of the carbonated material using nitric, sulfuric, and phosphoric acid did not alter the growth responses of these vegetables compared to those of control plants grown on the normal cultivation soil. Especially, the nitric acid pretreatment resulted in the better growth of these plants than the mixture of soil and compost. This is probably due to the additional supply of the nitrate-nitrogens. However the growth responses of lettuce and spinach on the carbonated materials neutralized with hydrochloric acid were different and appeared to be having symptom which is analogous to the chloride toxicity during the late stage of growing. Although no difference was observed in chemical composition, the contents of protein-nitrogen were higher in the plants grown on the carbonated material compared to those of plant grown on the mixture of soil and compost. Based on our preliminary results, the carbonized and neutralized rice-hulls are very useful bed material for the sanitary cultivation under the controlled environmental condition.
Saccharomyces cerevisiae KNU5377 is a constitutively thermotolerant, fermentative strain at high temperatures over 4$0^{\circ}C$. The exposure to 0.5 M NaCl caused S. cerevisiae KNU5377 to be lost its constitutive thermotolerance. Furthermore, the NaCl adaptation beyond 0.3 M during the overnight culture forced the strain-specific fermentation ability of S. cerevisiae KNU5377 to be disappeared. However, these phenomena did not occur in the reference, Saccharomyces cerevisiae ATCC24858. As a result, this adaptation led both strains to show the closely similar thermotolerance level and alcohol fermentation ability, implying the NaCl adaptation eliminated its strain-specific characteristics of S. cerevisiae KNU5377 Therefore it indicated that the superior intrinsic characteristics of S. cerevisiae KNU5377 must be related to the NaCl adaptation. On the other hand, the heat adaptation elevated alcohol productivity for both strains, but surprisingly did it for KNU5377 at the rate of two times higher than the reference's one; this suggests that KNU5377 possesses more efficient system enough to cause the difference. Consequently, these characteristics of S. cerevisiae KNU5377 must be interesting targets for further study to understand on how KNU5377 could acquire the constitutive thermotolerance and the outstanding fermentative capacity at high temperatures.

The seasonal variation in the proximate composition, pH, and glycogen contents of oysters Crassostrea gigas collected in Geoje and Jaran Bays on the southern coast of Korea was studied between March 2012 and February 2013. In the Geoje Bay oysters, the moisture content was 77.49-81.50 g/100 g, lipids ranged between 1.22 and 2.47 g/100 g, proteins between 9.46 and 13.11 g/100 g, and ash between 1.88 and 2.58 g/100 g. In the Jaran Bay oysters, the moisture content was 74.22-82.05 g/100 g, lipids comprised 1.32-2.37 g/100 g, proteins 9.19-13.35 g/100 g, and ash 1.96-2.45 g/100 g. The moisture content was highest in October and January in Geoje and Jaran Bay, respectively, and tended to increase from July until September. The highest protein levels occurred in August in both bays, which coincided with the timing of oocyte maturation, and then decreased at the beginning of total spawning. The highest lipid levels occurred in April in Geoje Bay, and February in Jaran Bay. The glycogen content was 0.40-2.28 g/100 g in Geoje Bay, and 0.61-3.53 g/100 g in Jaran Bay, and was highest in February and decreased from March onwards. The lowest glycogen content occurred in September and then increased from October onwards. The pH ranged between 6.29 and 6.48, and 6.32 and 6.59, for Geoje and Jaran Bay, respectively, and was highest in February.
Burcucumber (Sicyos angulatus) is an annual invasive plant species originated from North America. This species grows by twining around tree trunks, and blocks other plants from photosynthesis. Thus, it has caused the destruction of the ecosystem and biological diversity by threatening native plants. This study was performed to find out the effect of different soil water content (20%, 13%, 10%, 3%) and various shading degrees (0%, 60%, 80%) on the growth and photosynthesis-related activity of burcucumber. In the responses of burcucumber to soil water content, plant height (PH), leaf length (LL), leaf width (LW) and photosynthetic efficiency (PE) were lower at 20% water content than 10% reflecting that burcucumber plant grow well in the less dry soil and shows poor growth under wet soil condition. In shading experiment, PH, LL, LW and PE were lower at 80% than 60% shading and in general, the growth characteristics were lowered as the shading intensity increased. The improved growth of burcucumber under highly or moderately shaded condition implies that the plant can grow well without growth retardation and can be adapted to shading condition with other tall plant species including tree. Further study on the combination effects of above factors should be conducted in future for effective ecological control of burcucumber.

This study was conducted to investigate the climatic factors affecting bud phenology of Pinus densiflora provenances. Data were collected from Jungseon, Chungju and Jeju plantations which were parts of the 11 provenance trials established by Korea Forest Research Institute in 1996. The 36 provenances were included in this trial ($33^{\circ}30^{\prime}{\sim}38^{\circ}08^{\prime}$ in latitude and $126^{\circ}30^{\prime}{\sim}129^{\circ}20^{\prime}$ in longitude). The bud swelling date and bud burst date of the provenances were investigated from March to May in 2004 in two-day interval. The four geographic factors and fifteen climatic factors of the test sites and provenances were considered in this study. Canonical correlation analysis was conducted to examine the major factors affecting the bud phenology. Our results suggested that the major factors affecting the timing of bud swelling and burst are the differences in latitude, longitude, extremely low temperature (during December-February), extremely high temperature (during November-February) and annual mean growing days between test plantation and provenance. The provenances with lower winter temperature than that of plantation showed the faster bud swelling and bud burst. Based on these results, the implication on the seed transfer of P. densiflora was discussed.
From January 1997 to May, the red marbled rockfish, Sebastiscus tertius (Barsukov et Chen) was reared in the laboratory, and observed the morphological characteristies of egg and development of larvae and juveniles. The egg is spherical in shape colorless and transparent, measuring 0.72~0.80mm in diameter (mean 0.75mm, n=50). The naturally bearing larvae were 3.79~3.97mm in total length with 7~8+15~16=22~24 myomeres, and mouth and anus were open. Melanophores were present on the yolk of the larva and ventral margins of the tail. The 1~2 days after bearing larvae measured 3.90~4.13mm in total length, and transformed to postlarval stage, and yolk sac were completely resorbed. On the lower jaw melanophores were present. Total length of the larvae was 4.23~4.60mm in total length (11 days after bearing), number of elongated pectoral fin rays was 9~10, and melanophores distributed on the top of head, around the intestine tube, ventral margins of the tail and on the pectoral fin. In 21 days after bearing, the larva were 5.83~7.10mm in total length, and the caudal notochord flex $45^{\circ}$. Individuals of 9.80~12.36mm in total length (33 days after bearing) are regarded to have reached to the juvenile stage with all the fins were developed.
Protoplasts were isolated from cotyledons of germinating Pinus densiflora S. et Z. seeds. The seeds were germinated for nine days, and excised embryonic cotyledons about 3-4mm long were incubated with Cellulase Onozuka R-10 and Macerozyme. After 8 hours of incubation, large number of viable protoplasts were isolated. Isolated protoplasts were cultured in a medium containing basal salts of $B_5$ medium, vitamines, amino acids, organic acid, sugars, and growth hormones. The first evidence of protoplast budding was observed after twelve hours in culture, and it suggested that high potential of the embryonic cotyledons for rapid cell division affected the early budding, rather than effect of culture medium was shown in twelve hours. The three- to four-cell stage was reached after three to four days of culture. Most cell divisions were achieved by additional buddings rather than equal binary cell division. No further cell division was observed beyond the four-cell stage. Protoplasts isolated from fully expanded cotyledons (germinated for 17 to 24 days) seldom initiated or failed to initiate cell division.
