Comparison of Void Content between Cyldrical Concrete Specimen and Concrete Core Specimen Using ASTM C 642 Test Procedure (ASTM C 642 시험방법을 이용한 구조체 코어공시체와 원주형 공시체의 공극률 비교 평가)

  • Recently, construction accidents have occurred due to illegal water addition and insufficient quality control at domestic construction sites. In this study, the void content test method proposed in ASTM C 642 was used to provide a reference guideline for evaluation on the quality control status of cast-in-place structural concrete. For this purpose, simulated structural concrete for coring purpose was prepared in addition to the concrete cylindrical specimens with the same formulation, and the changes in compressive strength, elastic modulus, and void content related to coring were evaluated. According to experimental results, the compressive strength and modulus of elasticity were reduced by coring, which was associated with the generation of microcracks during coring. With respect to void content, the difference in void content between the cylindrical specimen and the cored specimen was up to 1.69%. If this value is used as a correction factor, it is possible to estimate the real void content of the cast-in-place structural concrete. By comparing this with the void content obtained from cylindrical concrete specimens, it is possible to evaluate the quality control status and amount of illegal water addition on the structural concrete.

Effect of Dietary Acid Hydrolysates of Cocoon on Performance, Egg Quality, Blood Immune Substance and Serum Cholesterol in Laying Hens (산란계 사료 내 누에고치 산가수분해물 첨가 시 산란율, 계란 품질, 혈중 면역 물질 및 혈청 콜레스테롤 함량에 미치는 영향)

  • The study was conducted to evaluate the effects of dietary acid hydrolysates of cocoon on laying performance, egg quality, blood level of immune substance and cholesterol in laying hens. The animals used in the experiment were a total of 240 Hy-Line Brown laying hens (41 weeks old). Dietary treatments included 1) CON (basal diet), 2) SP1 (basal diet + acid hydrolysates of cocoon 0.05%), 3) SP2 (basal diet + acid hydrolysates of cocoon 0.1%) and 4) SP3(basal diet + acid hydrolysates of cocoon 0.2%) twelve laying hens were allotted to a block (pen) with five replicated. Through the whole period of experiment, egg production, egg weight, egg shell breaking strength and egg shell thickness were no significant difference. However, acid hydrolysates of cocoon treatments (SP1, SP2, SP3) significantly increase their egg yolk height and Haugh unit compared to control (P<0.05). IgG and WBC (white blood cell) contents in blood were higher in SP1, SP2, SP3 treatment compared to control (P<0.05). However, albumin and total protein content in blood were no significant difference. Phospholipid, HDL-cholesterol, LDL-cholesterol and total cholesterol contents in serum were higher in SP1, SP2, SP3 treatment compared to control (P<0.05). In conclusion, supplement dietary acid hydrolysates of cocoon improved egg yolk height, Haugh unit, and blood level of phospholipid, HDL-cholesterol, LDL-cholesterol, total cholesterol in laying hen.

Effects of Dietary Hydrolyzed Yeast on Egg Production and Egg Quality during Late Phase of Laying Hens (산란후기 사료 내 가수분해 효모의 첨가 급여가 생산성과 계란 품질에 미치는 영향)

  • An experiment was conducted to investigate the effects of varying levels of hydrolyzed yeast on egg production and egg quality in aged laying randomly allotted to three dietary treatments such that egg production was similar in each treatment (6 replicates of 10 birds each). The layers were fed diets containing 0, 0.1, or 0.2% hydrolyzed yeast for eight weeks. No significant difference was observed in egg production during the first half of the experiment. Egg production and daily egg mass in groups fed diets containing hydrolyzed yeast were significantly higher (P<0.05) than those of the control groups during the second half of the experiment. Egg weight was not affected by the dietary treatment. Eggshell strength and thickness in groups fed diets containing hydrolyzed yeast were significantly higher than those of the control groups during the overall experimental period (P<0.05). Although no significant differences were observed in the Haugh units, yolk color in the group fed diets containing 0.1% hydrolyzed yeast was significantly higher than that in the control group (P<0.05). The mammillary layer thickness increased in a linear manner and significantly following treatment with dietary hydrolyzed yeast (P<0.05). Antibody titer against avian influenza virus in the group fed diets containing 0.2% hydrolyzed yeast was significantly higher (P<0.05) than that in the control group. In conclusion, dietary hydrolyzed yeast improved egg production and eggshell quality of laying hens in the late stages of production.

Application of the Equivalent Point Method for Estimation of Kinetic Parameters (Kinetic Parameters 결정을 위한 Equivalent Point Method의 이용)

  • A method for application of the equivalent time and temperature point for estimating kinetic parameters was proposed. The feasibility of the method was demonstrated with both theoretical and empirical kinetic data. The theoretical kinetic data were obtained from the arbitrarily chosen time-temperature data for three chemical reactions whose kinetics are well established. The experimental kinetic data were obtained for the acid (0.0005 N HCl) catalyzed hydrolysis of sucrose(2%). The activation energy and the frequency factor determined by the proposed method were $104.74{\pm}1.87kJ/mol\;and\;5.62{\times}10^{14}\;hr^{-1}$ respectively and the results agreed well with those obtained by a different method of kinetic parameter estimation, i.e. the linearly increasing temperature method.

  • In the present study, we synthesized mesoporous silica hollow spheres with different wall thickness using polystyrene (PS) spheres as a structure template, tetraethoxysilane (TEOS) as a silica source, cetyltrimethylammonium bromide (CTAB) as a template. Particle size and dispersion of PS spheres were strongly depended on the concentration of surfactant in the aqueous solutions. The size of PS spheres was increased with decreasing concentration of surfactants. Dispersion of PS particle was improved when the surfactant concentration was lower than 0.5 g of surfactant.

  • The product specificity of Bacillus ${\alpha}-amylase$ on raw rice starch has teen studied by using HPLC and scanning electron microscopy (SEM). Analysis of starch degradation products digested by ${\alpha}-amylase$ showed considerable differences between raw and gelatinized rice. The hydrolysis of raw rice starch resulted in formation of more glucose and maltose than those of gelatinized starch. SEM revealed characteristic enzyme degradation patterns. Hollow curvatures were observed in gelatinized starch, indicating the substrate is hydrolyzed in the interior of the starch chain by Bacillus ${\alpha}-amylase$. In contrast, raw starch were hydrolyzed from the end of the substrate, resulting in pinholes over the surface of the starch granules.

Production and Characterization of α-Galactosidases from Two Bacillus licheniformis Isolates (Bacillus licheniformis 분리균 2종의 α-Galactosidase 생산성과 효소특성)

  • Two bacterial strains, Bacillus licheniformis YB-1413 and YB-1414, producing extracellular α-galactosidase, were obtained from homemade Doenjang. On the basis of their biochemical properties, 16S rRNA sequences and random amplified polymorphic DNA patterns by polymerase chain reaction, they were found to be somewhat different from one another. α-Galactosidase productivities of the two isolates were increased by wheat bran, but drastically decreased by melibiose, raffinose and sucrose which were used as carbon sources. The enzyme productivities were increased by yeast extract as a nitrogen source with maximum levels of 1.87 U/ml for YB-1413 and 1.69 U/ml for YB-1414, respectively. The enzymes of both isolates exhibited maximum activity for hydrolysis of para-nitrophenyl-α-D-galactopyranoside (pNP-αGal) under reaction conditions of pH 6.0 and 45℃. Their hydrolyzing activities for pNP-αGal were drastically decreased by the addition of low concentrations of ribose and galactose. They were capable of hydrolyzing completely α-1,6 linked galactosyl residue in melibiose, raffinose and stachyose, which are known to be anti-nutritional factors in products of soybean and legume. In relation to the latter, the isolates YB-1413 and YB-1414 have potential applicability in improving soybean-fermented foods and the nutritional value of soybean feed.

The Korean Stock Market Surveillance System : Changes in Volatility Before and After Surveillance Designation (한국의 감리종목 제도 : 감리지정 전.후의 변동성 비교)

  • The Korean Stock Market Surveillance System is desinged to control the volatility of stocks by drawing investor's attention and suppressing disguised demand, when stocks run up so rapidly in short period of time. Yet the Surveillance System has not been under empirical examination about its role and evolved in line with the Price Limit System. This study looks at the security returns under surveillance designation for 1995 -2001 period. The results indicate that the volatility of stocks has not been affected after surveillance designation. The constraints against the disguised demand, however, seems to limit the security returns rather than volatilities. These findings raises a question about the role of The Korean Stock Market Surveillance System for the control of volatility. The Surveillance System needs to be examined thoroughly about its role, function, and its conditions. Otherwise, the shareholders with less information could be placed at a disadvantage. This paper suggests that the system should be amended in an effort to make the volatility of stocks under control.

Production of ${\beta}-Galactosidase$ with High Transgalactosylation Activity by Bacillus sp. A4442 Mutant (변이주 Bacillus sp. A4442에 의한 갈락토스 전이활성이 높은 ${\beta}-Galactosidase$의 생산)

  • In an attempt to improve the productivity of ${\beta}-galactosidase$ from Bacillus sp. A1, which was isolated from soil and has remarkably higher transgalactosylation activity than lactose hydrolysis activity, a chemical mutation procedure using N-methyl-N'-nitro-N-nitrosoguanidine followed by selection was conducted. The final selection, designated as Bacillus sp. A4442, turned out to show a substantially increased enzyme productivity. Catabolite repression by glucose and lactose requirement as an inducer for the enzyme biosynthesis, which were shown in the parent strain, was markedly diminished; instead it was found out that galactose acts as another inducer. Because pH of medium, one of the most important factors for cell growth as well as enzyme production, is closely related with the sugar concentration during culture, it was kept in the optimum range of $6.5{\sim}7.5$; for this the initial glucose concentration was adjusted to be 0.5% which was thereafter maintained by the controlled pumping-in of lactose using the pH-stat technique. By doing so, we were able to increase the productivity of ${\beta}-galactosidase$ with high transgalactosylation activity up to $44\;unit/m{\ell}-broth$.
