Separation of Lipase Using Reverse Micelles in Spray Column (Spray Column에서 역미셀을 이용한 Lipase의 분리)

  • Lipase was separated using reverse mlcelles in a spray column. The 50 mM AOT-Isooctane solution was used as reverse micellar solution for the extraction of lipase (crude containing 25% Protein). Ionic strength was controlled by KCl(0.1M KCl for extraction, 0.5M KCl for back exlractlon). Acetate buffer and phosphate buffer were used for control of pH. The efficiencies of extraction and stripping were 30% and 50%. An increase of circulation did not change the efficiency of extraction in forward extraction. The optimum flow rate was around 0.10ml/sec.

  • 북한 농업용수 수요량의 파악은 앞으로 다가올 농업 수자원 분야의 과제에 대처하고 통일 후의 수자원 개발 계획을 수립하기 위한 가장 기본적인 요소이다. 그러나 농업용수 수요량 파악을 위한 북한에의 접근은 불가능한 것이 사실이며, 위성영상을 이용한 원격탐사 기법의 활용은 이러한 문제를 해결할 수 있는 가장 효과적인 방법이다. 이에 본 연구에서는 먼저 시계열 MODIS 영상을 이용하여 북한 전 지역을 대상으로 논의 분포를 추출한 후, MODIS 영상의 해상도에 의해 발생하게 되는 논 추출의 불확실성이 지형학적 인자에 따라 어떻게 나타나는지 살펴보고 이를 적용하여 논 추출 결과를 보완하였다. 또한 북한 지역에 적용 가능한 농업용수 수요량 산정 방법을 선정하고 시계열 MODIS 영상을 이용하여 추출된 논 지역을 대상으로 북한 전역의 연간 농업용수 수요량을 추정하였다. 본 연구에서 정립된 시계열 MODIS 영상을 이용한 논 추출 방법 및 북한의 농업용수 수요량 산정 방법은 향후 북한의 농업용수 관련 연구에 대한 중요한 기초 자료가 될 수 있을 것으로 기대된다.

Orostachys japonicus Hexane Fraction Attenuates Pro-inflammatory Cytokines in LPS-activated Macrophage Cells by Suppression of AP-1 and IRF3 Transcription Factors (LPS로 유도된 대식세포에 대한 와송 핵산추출물의 AP-1과 IRF3 전사인자의 억제에 의한 전염증성 사이토카인의 감소 효과)

  • Orostachys japonicus (O. japonicus) is known as a medicinal plant for the treatment of various symptoms. This study investigated the anti-inflammatory effect of the hexane fraction from O. japonicus (OJH) on the LPS-stimulated response in RAW 264.7 macrophage cells. This study was conducted to confirm the effect of cell cytotoxicity and production of reactive oxygen species (ROS) in OJH-treated macrophage cells. Additionally, pro-inflammatory cytokines and transcription factors were determined using RT-PCR and western blotting assay. OJH showed no change in lactate dehydrogenase (LDH) levels and exhibited reduced ROS levels in LPS-induced inflammatory cells. Moreover, OJH significantly suppressed the mRNA levels of proinflammatory cytokines, including IL-1β, IL-2, IL-6, TNF-α, and IP-10. Furthermore, OJH effectively inhibited the protein levels of AP-1 (p-c-Jun and p-c-Fos) and p-IRF3 in a dose-dependent manner. In conclusion, our results demonstrate that OJH exhibits strong anti-inflammatory activities via regulation of inflammatory factors.

  • Cultural heritages emit volatile organic compounds(VOCs) during degradation as chemical, biological factors. These VOCs in the atmosphere are degradation factors for another materials. But non-destructive organic analysis methods are uncommon, and have difficulties for field application. In this study, solid phase micro-extraction(SPME) and gas chromatography( GC) are carried out for analysis of VOCs which are emitted from leather, and prove SPME field holder's efficiency. I analize leather archival objects at The National Archives of Korea by cross-checking GC and SPME methods. It could be confirmed that the compounds are (E)-2-nonenal, butyl hydroxy toluene emitted itself from leather.

Development of a Screening Method for Deforestation Area Prediction using Probability Model (확률모델을 이용한 산림전용지역의 스크리닝방법 개발)

  • This paper discusses the prediction of deforestation areas using probability models from forest census database, Geographic information system (GIS) database and the land cover database. The land cover data was analyzed using remotely-sensed (RS) data of the Landsat TM data from 1989 to 2001. Over the analysis period of 12 years, the deforestation area was about 40ha. Most of the deforestation areas were attributable to road construction and residential development activities. About 80% of the deforestation areas for residential development were found within 100m of the road network. More than 20% of the deforestation areas for forest road construction were within 100m of the road network. Geographic factors and vegetation change detection (VCD) factors were used in probability models to construct deforestation occurrence map. We examined the size effect of area partition as training area and validation area for the probability models. The Bayes model provided a better deforestation prediction rate than that of the regression model.

Effect of Sarcodon aspratus Extract on Expression of Cell Cycle-Associated Proteins in HepG2 Cells (HepG2세포에서 향버섯 추출물이 세포주기 조절단백질에 미치는 영향)

  • We investigated the effect of Sarcodon aspratus extract on expression of cell cycle regulators. Methanol extract of Sarcodon aspratus showed a growth suppression on HepG2. As shown by western blot analysis, the expressions of cyclin A and Dl known as cell cycle regulators were decreased after treatment of Sarcodon aspratus extract. On the other hand, the expression of cyclin Bl was increased in the presence of Sarcodon aspratus extract. Furthermore, the expression of p53, a tumor supressor gene, and p27, a cell cycle dependent protein kinase inhibitor, were increased, whereas the expression of PCNA was decreased. In conclusion, our study suggests that growth inhibitory effect of Sardodon aspratus methanol extract on HepG2 is induced by cell cycle arrest in the Gl phase caused by decrease in cyclin A, Dl expressions and increases in p53, p27 expression.

The Analysis of Whitening Effects on Extracts from Ginkgo (Ginkgo biloba L.) Seeds (은행나무 종자 추출물의 미백효능 분석)

  • Ginkgo (Ginkgo biloba L.) seeds, called 'Baekqwa', were extracted from 70% ethanol to investigate the whitening effect and to confirm the application potential as a cosmetic material. Ginkgo seed ethanol extracts (GBE) were treated with B16F10 melanoma cells, and melanin contents and tyrosinase, which is the main enzyme concerning the synthesis process of melanin, inhibitory activity were confirmed. As a result, there were inhibited in a concentration-dependent manner, and GBE also significantly reduced the protein expression and mRNA levels of tyrosinase, tyrosinase-related protein-1, -2 (TRP-1, -2), and their upstream transcription factor, microphthalmia-associated transcription factor (MITF). These results suggest that GBE could be used as an effective whitening agent that has an inhibitory effect on melanin production by regulating the expression and degradation of MITF in melanocytes.

Anti-obesity effects of an enzymatic extract of mandarin (Citrus unshiu) peel in 3T3-L1 adipocytes (감귤피 효소적 추출물의 지방세포에서의 항비만 효과)

  • Mandarin peel (MP) is a by-product of the processing of citrus juice or other products. This study aimed to investigate the potential anti-obesity effect of an enzymatic extract of MP on the inhibition of adipogenesis in 3T3-L1 adipocytes. The enzymatic extract (MPCE) was prepared using the commercial food-grade carbohydrase Celluclast. Lipid accumulation and triglyceride levels were significantly lower in MPCE-treated cells than in untreated cells. In addition, MPCE treatment reduced the protein expression levels of peroxisome proliferator-activated receptor-γ, CCAAT/enhancer-binding protein α, sterol regulatory element-binding protein 1, and fatty acid-binding protein 4. These results suggest that MPCE inhibits adipogenesis by downregulating the expression levels of adipogenesis-related proteins. Therefore, the current findings demonstrate that MPCE possesses potent anti-obesity properties and could be a potential ingredient in functional food industries.

Anti-inflammatory effects of ethanolic mulberry extract on the murine macrophage cell line, RAW 264.7 (RAW 264.7 큰포식세포에서 상백피 및 상지 에탄올 추출물의 항염증 활성 비교)

  • The aim of this study was to compare the anti-inflammatory effects of ethanol extracts of root peel and spear of mulberry (RME and SME, respectively) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Both extracts significantly inhibited the production of reactive oxygen species (ROS), nitric oxide (NO), and interleukin-6 (IL-6). However, prostaglandin $E_2$ ($PGE_2$) levels decreased in LPS-stimulated RAW 264.7 cells treated with SME. Additionally, the extracts reduced inducible NO synthase (iNOS) expression and cyclooxygenase-2 (COX-2) in mRNA levels. Although ROS production was lower in the RME-treated cells than in the SME-treated cells, the levels of other inflammatory parameters, including IL-6 and $PGE_2$, and mRNA levels of iNOS and COX-2 reduced more in the SME-treated cells. These results indicate that SME showed higher anti-inflammatory activities than RME. Therefore, SME can be used as a functional food ingredient to enhance health.

Enhancement of the Anti-inflammatory Activities of Aralia continentalis Kitagawa Extracts Fermented by Lactobacillus plantarum (추출용액에 따른 유산균 발효 땅두릅의 항염증 효과)

  • We investigated the anti-inflammatory activities of various organic solvent extracts with and without Lactobacillus plantarum fermentation of Aralia continentalis Kitagawa which has hypotensive effects in addition to excitatory effects on the central nervous system. It has been used to treat arthritis, colds, neuralgia, rheumatism, and itchy skin. Our extracts were tested for their anti-inflammatory potential on NO production and the expression of inflammatory factors in lipopolysaccharide-stimulated RAW264.7 macrophages. Extracts with and without L. plantarum fermentation were prepared using water, ethanol, hexane, ethyl acetate, and butanol. The RAW264.7 cells were tested for toxicity and the anti-inflammatory activity of each extract was determined at a concentration with no toxicity to the cells. The extracts used in this study significantly inhibited both the production of NO and the mRNA expression of COX-2 and iNOS, the major inflammatory factors. The production of inflammation-related cytokines $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ was also significantly reduced. These results suggest that the extracts involving fermentation by L. plantarum can inhibit cytokines by controlling the expression of inflammatory cytokine genes. It is considered that the water, ethanol, and butanol extracts after fermentation with L. plantarum could be useful as functional natural materials with anti-inflammatory effects.
