Development and Application of Rubric for Assessing Nature of Technology in the Context of Socioscientific Issues (과학기술관련 사회쟁점에 대한 의사결정에서 나타나는 NOT 이해 수준의 평가를 위한 루브릭 개발 및 적용)

  • Current science education aims to guide students as future responsible citizens to make informed decisions on socioscientific issues (SSI). In the authors' previous study, it was found that conceptions of nature of technology (NOT) were explicitly represented in various contexts of SSI with differentiated levels of understanding, and cases of the informed NOT understanding included the key features of well-reasoned SSI decision-making. Therefore, enhancing NOT understanding could be one of the elements to leverage students' informed SSI decision-making. In this study, we developed a rubric to assess NOT understanding in the context of SSI and applied it to evaluate the impact of SSI instruction. Participants were 58 college students who took an SSI course for 6 weeks. Before and after the SSI course, they were asked to write decision-making essays on the Golden Rice issue (a type of genetically modified food). As a result of analyzing the pre- and post decision-making essay using the rubric, it was found that NOT understanding was improved after the SSI course; in addition, the salient patterns of NOT changes were analyzed in detail in order to gauge the influence of the SSI classes. Implications for science education were discussed.

Properties and Structures of Bi2O3-B2O3-ZnO Glasses for Application in Plasma Display Panels Rib (PDP Rib용 Bi2O3-B2O3-ZnO계 유리의 물성과 구조)

  • This study, compared with data of PbO-base glass system is a part of new glass composition design with Bi-base composition for PDP Rib. As $Bi_2O_3-B_2O_3-ZnO$ glass composition including Bi, which have similar density value and work facility to PbO, properties of softening point, thermal expansion coefficient, chemical durability, dielectric constant, and structural changing by XPS were investigated. $Bi_2O_3-B_2O_3-ZnO$ glass system, added 50∼80 wt% $Bi_2O_3$ widely, were presented 400∼480$^{\circ}C$ softening temperature, $68{\sim}72{\times}10^{-7}/^{\circ}C$ thermal expansion coefficient and 13∼25 dielectric constant. These results were showed similar physical properties with Pb-base glass system of same composition content, application possibility as starting composition of rib material was identified through micro-control of components and physical properties. The bonding energy of $O_{1s}$ as the $Bi_2O_3$ content decreasing was increased and full width at half-maximum (FWHM) was decreased, which is caused by non-bridging oxygen increasing.

  • The brown planthopper, Nilaparvata lugens, is among the most serious insect pests of rice. It is widely distributed in Asia, Australia and Pacific islands. An earlier mitochondrial DNA study revealed that there exist significant genetic differences between populations north and south of the Red River Delta region in Vietnam. However the mitochondrial DNA was not sufficiently variable to examine the sources of immigration. For a more detailed analysis of geographic population structure of N. lugens, we developed microsatellite markers. Thirty-seven putative microsatellite loci were isolated using a magnetic biotin method, and five primer pairs designed from the flanking regions of sequenced microsatellite clones were labeled with fluorescent. Of these five primer sets, two have proven to be useful across all the samples we used in this study. We used variation at these two microsatellite loci to test the hypothesis that N. lugens biotypes (1, 2, and 3) sampled from laboratory selection constituted distinct genetic units. Allele frequency differences among the three major biotype categories were not significantly different at one locus (27035). However, the other (7314) did show differences among the major three biotypes. The methods we describe here will be useful for studying population structure of crop pest and for tracking the patterns of migratory pest like the rice planthoppers.

An Overview for Molecular Markers in Plants (식물에서 분자 마커의 동향)

  • A molecular marker is a molecule contained within a sample taken from an organism or other matter. The development of molecular techniques for genetic analysis has led to a great contribution to our knowledge of plant genetics and our understanding of the structure and behavior of various genomes in plants. Recently, functional molecular markers have been developed to detect the presence of major genes from the analysis of pedigreed data in absence of molecular information. DNA markers have developed into many systems based on different polymorphism-detecting techniques or methods such as RFLP, AFLP, RAPD, SSR, SNP, etc. A new class of very useful DNA markers called genic molecular markers utilizing the ever-increasing archives of gene sequence information being accumulated under the EST sequencing projects on a large number of plant species. Functional markers are derived from polymorphic sequences, and are more likely to be involved in phenotypic trait variation. Based on this conceptual framework, the marker systems discussed below are all (gene)-targeted markers, which have the potential to become functional. These markers being part of the cDNA/EST-sequences, are expected to represent the functional component of the genome i.e., gene(s), in contrast to all other random DNA based markers that are developed/generated from the anonymous genomic DNA sequences/domains irrespective of their genic content/information. Especially I sited Poczai et al’ reviews, advances in plant gene-targeted and functional markers. Their reviews may be some useful information to study molecular markers in plants.

Molecular Cloning and Expression in Escherichia coli of a Rabbit Globin Gene (유전공학적 방법에 의한 토끼 글로빈 유전자의 재조합과 대장균에서의 발현)

  • The structural gene of rabbit hemoglobin was cloned into Pst I site of pBR322 in E. coli. The complementary DNA (cDNA) was synthesized from rabbit globin mRNA with avian myeloblastosis viral reverse transcriptase, and then RNA was destroyed at pH 11. The double stranded cDNA was synthesized with both Klenow fragment of E. coli DNA polymerase I and reverse transcriptase and then the hairpin loop was opened with Sl nuclease. Double stranded cDNA was subsequently tailed with dCTP and annealed to dGMP-tailed vector DNA. After transformation and initial screening of appropriate clones by plasmid size, the cloned colonies were identified by in situ colony hybridization using by plasmid size, the cloned colonies were identified by in situ colony hybridization using $[^32P]$-labeled cDNA probes and characterized the inserts with restriction endonucleases. The expression of cloned globin gene was investigated by standard radioimmunoassay using rat anti-rabbit Hb serum as primary antibody and goat antirat IgG serum as secondary antibody. The result suggested that the chimeric proteins (the part of $\\beta$-lactamase from the vector pBR322 and globin from rabbit) were supposedly produced in E. coli and the product had the antigenic determinant of rabbit hemoglobin.

Development of Chloroplast Genome-based Insertion/Deletion Markers in the Genus Broussonetia (닥나무 속 식물의 엽록체 유전체 기반 InDel 마커의 개발)

  • Several members of the genus Broussonetia are woody plants with high-quality cellulose fibers and are used to make a traditional type of Korean paper known as Hanji. Three of these species, Broussonetia kazinoki, Broussonetia monoica, and Broussonetia papyrifera, are found in the Korean Peninsula. Because it is challenging to distinguish different Broussonetia species based on morphology alone, we have developed a set of insertion/deletion (InDel) markers for genetic identification of these species. From twenty-two Broussonetia samples collected throughout Korea, we selected six for next-generation sequencing analysis. InDel marker candidates were identified by comparing this sequence information with the B. kazinoki chloroplast genome sequence. The marker candidates were used to screen the genomes of the twenty-two Broussonetia plants, and five useful chloroplast-based InDel markers were identified. Detailed genotyping using these five markers showed that the twenty-two plants of the genus Broussonetia could be clustered into five groups, verifying that the markers developed here can be used for breeding, identification, and analysis of species in the genus Broussonetia.

Analysis of Genetic Variation in the Small Subunit Ribosomal RNA Gene of Euplotes Ciliates for Developing Species Diagnostic Molecular Marker (종 식별 분자 마커 개발을 위한 섬모충류 Euplotes의 small subunit ribosomal RNA 변이성 분석)

  • To verify which loop regions of 18S rRNA gene are suitable as species-specific genetic markers in ciliates, we analyzed the genetic variation of 18S rRNA gene among 9 Euplotes species (Hypotrichia : Ciliophora). In our result, no inter-specific variation was detected from V1, V3 and V5 regions, and the length of V7 and V8 are 44 bp and 79 bp, respectively, which are too short to make genetic marker. In contrast, V2 and V4 may be good candidate segments of species-specific diagnostic molecular markers because these two regions are most variable ($1.75{\sim}20.61%$) and showed good inter-specific phylogeny. Furthermore, the sequences of V2 and V4 are 123 bp and 306 bp, respectively in length which are enough to make species-specific marker.

Nut Characteristics of Chestnut Variety Cultivated in Suncheon (순천지방에 재배되고 있는 밤나무 품종의 과실 특성)

  • This study was carried out to provide the basic information for variety improvement by means of nut characteristics of chestnut variety cultivated in Suncheon area. While Tsukuba has the largest number of burr with 125.5, it showed significant difference among chestnut variety. Average number of nuts per burr ranged from 1.3 in Kunumi to 2.4 in Pyeonggi, which showed significant difference among chestnut variety. Average nut weight varied between 17.5 g in Isseumo to 28.1 g in Kunumi. Nut yield ranged form 1,000.4 g in Sangrim to 5,479.5 g in Tsukuba, which showed significant difference among chestnut variety. Changbanggamyul had the highest rate of healthy nut, Kunumi, Tanzawa, Tsukuba, Daebo, Pyeonggi, and Sinyipyeong had the rate of healthy nut more than 80%. The rate of split pericarp showed significant difference among chestnut variety as ranged from 0.0% in Changbanggamyul to 26.1% in Gwangdeok.

Gene expression profile of the early embryonic gene of the silkworm, Bombyx mori (누에 수정란 초기발현유전자 데이터베이스 구축)

  • This study was aimed for development of a useful genes that has a transcript expressional specificity in the early embryonic stage of the silkworm, Bombyx mori. We constructed and analyzed a full-length cDNA library from silkworm's eggs which after a lapse of 2 ~ 6 hours post oviposit. A total 960 clones were randomly selected, and the 5' ends of the inserts were sequenced to generate 652 expressed sequence tags(EST). 334 unique ESTs were generated after the assembly of 652 ESTs. The annotation of 334 unique ESTs by BLAST search revealed that 156(47%) of the sequences represented known genes, whereas 178(53%) of the sequences has no matches in the database. Of the 156 known genes, the most abundant genes were heat shock protein hsp20.8 gene(12 times) and ubiqutin-like protein gene(11 times). The functional groups of these ESTs with matches in the database were constructed according to their putative molecular functions. Among thirteen functional categories, the largest groups were protein synthesis(9.6%) and cellular organization( 8.1%). Further defined studies on molecular functions and biological roles of their promoters will give us wellfined information and its application.

Applying the basic knowledge about regulation of pigmentation towards development of strategies for cutaneous hypopigmentation

  • The extensive variation in human cutaneous pigmentation is mainly due to differences in the rate of melanin synthesis by epidermal melanocytes, the relative amounts of eumelanin and pheomelanin synthesized, and the manner and rate of transfer of melanosomes from melanocytes to keratinocytes. Pigmentation is a complex trait that is regulated genetically and environmentally. One gene that has been receiving a lot of attention is the gene for the melanocortin 1 receptor The extensive polymorphism of this gene in human populations suggests its significance in the diversity of pigmentation. Exposure to solar ultraviolet radiation (UV) results in increased synthesis of a variety of growth factors, cytokines and hormones, and in modulation of their receptors in the epidermis. Knowledge about the regulation of pigmentation has led to strategies for clinical treatment of hyperpigmented skin lesions. Three main strategies are: 1) the use of chemicals that interfere with the melanin synthetic pathway, 2) the design of peptides or peptide-mimetics based on the structure of hormones that regulate eumelanin synthesis, and 3) the use of agents that reduce melanosome transfer from melanocytes to keratinocytes. All three strategies are expected to induce hypopigmentation, by inhibiting total melanin synthesis, eumelanin production, or the epidermal melanin unit, respectively.
