Polymorphisms and Allele Distribution of Novel Indel Markers in Jeju Black Cattle, Hanwoo and Imported Cattle Breeds (제주흑우, 한우 및 수입 소 품종에서 새로운 indel 마커의 다형성과 대립인자 분포)

  • The aim of this study was to screen the polymorphisms and distribution of each genotype of insertion/ deletion (indel) markers which were found in a preliminary comparative study of bovine genomic sequence databases. Comparative bioinformatic analyses were first performed between the nucleotide sequences of Bovine Genome Project and those of expressed sequence tag (EST) database, and a total of fifty-one species of indel markers were screened. Of these, forty-two indel markers were evaluated, and nine informative indel markers were ultimately selected for population analysis. Nucleotide sequences of each marker were re-sequenced and their polymorphic patterns were typed in six cattle breeds: Holstein, Angus, Charolais, Hereford, and two Korean native cattle breeds (Hanwoo and Jeju Black cattle). Cattle breeds tested in this study showed polymorphic patterns in eight indel markers but not in the Indel-15 marker in Charolais and Holstein. The results of analysis for Jeju Black cattle (JBC) population indicated an observed heterozygosity (Ho) that was highest in HW_G1 (0.600) and the lowest in Indel_29 (0.274). The PIC value was the highest in HW_G4 (0.373) and lowest in Indel_6 (0.305). These polymorphic indel markers will be useful in supplying genetic information for parentage tests and traceability and to develop a molecular breeding system for improvement of animal production in cattle breeds as well as in the JBC population.

  • In this paper, we proposed a new algorithm of the inverse scattering for the reconstruction of unknown dielectric scatterers using the finite-difference time-domain method and the design sensitivity analysis. We introduced the design sensitivity analysis based on the gradient information for the fast convergence of the reconstruction. By introducing the adjoint variable method for the efficient calculation, we derived the adjoint variable equation. As an optimal algorithm, we used the steepest descent method and reconstructed the dielectric targets using the iterative estimation. To verify our algorithm, we will show the numerical examples for the two-dimensional $TM^2$ cases.

  • 강유전성 세라믹스 재료로써는 PZT계열의 세라믹재료가 널리 쓰이고 있다. 이는 우수한 유전 및 압전특성을 가지고 있으나, PbO을 다량 함유하고 있어 $1000^{\circ}C$이상에서 PbO가 급격하게 휘발되는 성질 때문에 조성의 변동이 생겨 재현성이 어려우며 이를 방지하기 위하여 PbO를 과잉 첨가시키기 때문에 PbO휘발로 인한 강한 독성이 인체에 유해하고, 비환경 친화적인 물질로 최근에는 환경문제가 대두됨에 따라 대체 또는 보완 할 수 있는 방안에 검토되고 있다. 본 연구는 그 해결책의 한 방안으로 압전특성이 우수한 $(Pb_{0.94}Sr_{0.06})[(Ni_{1/2}W_{1/2})_{0.02}(Mn_{1/3}Nb_{2/3})_{0.07}(Zr_{0.51}Ti_{0.49})_{0.91}]O_3$계 조성을 설계하고 Glass frit(0~1.1 wt%)를 소량 첨가하여 액상 소결 특성을 부여하고 $1000^{\circ}C$ 이하의 저온에서 소결하여 유전 및 압전 특성을 평가하였다. 실험방법은 일반적인 세라믹스 제조공정으로 24시간 ball milling하고 $850^{\circ}C$에서 2시간 하소 후 Glass frit를 소결조제로 소랑 첨가하여 $1000^{\circ}C$ 이하 온도에서 소결을 진행하여 각 소결온도에 따른 유전 및 압전 특성을 평가하였다. 최종 소결된 시편의 밀도와 수축율을 분석하여 최적의 소결온도를 확립하였으며 XRD분석을 통해 perovskite구조를 확인하고 미세구조확인을 위해 SEM으로 관찰하였다. 압전 특성을 평가하였다.

  • 에너지 해외의존도 97%, 무역의존도 70%인대한민국. 우리는하루220만배럴, 연간8억배럴이상을소비한다. 부존자원이 극도로 빈약한 우리나라가 나아가야 할 길은자명하다. 에너지 자원을 거의수입해 의존하다보니 이를고부가화 해 내수에 돌리면서 최우선으로 수출을 통해 온나라가 먹고 살고 있다해도 과언이아니다. 해외 에너지자원을 안정적으로 확보하는 것이 바로 국가경쟁력의 근원이 되는것은 어제오늘의 일이 아니다. 고유가 파고를 헤쳐나가야 한다는 절박한현실에서안정적인 에너지자원확보를 위한 정부의 해외자원분야 정책 및 민관전략이 올들어 의욕적으로 전개되고 있다. 우선정부는 자원개발 업무를 강화하기 위해 현재1과1팀의 자원개발조직을 보강하 고유전개발팀을 신설하는 등 모두3개팀으로 늘린다. 나아가 해외 자원확보에주력 산업 동반진출도 착착진행하고 있다. 국내 자원 개발 기업과 에너지관련기업, 해외진출기관등 14곳으로 구성된 ‘에너지산업해외진출협의회’도 최근 출범시켰다. 유전을탐사하거나 개발하는데 개인들도간접투자할수있는 ‘유전개발펀드’도 7월에 첫 선을 보인다. 약2000억원규모의이 시범펀드 도입 등을 통해 자원개발을 가속화,2013년까지원유-가스자주개발률을18%(원유15%, 가스30%)까지 끌어 올릴 계획이며 여기에16조원 신규투자가 소요 될 것으로보고있다. 이같은 일련의 공격적인 움직임은 단순히 돈을 주고 자원을 사오는 기존방식을 앞으로는과감히탈피,자원부국이 원하는 산업인프라등을 수주해제 공한뒤 그 대가로자원을 확보하는 총체적 자원확보전략에 무게를두겠다는 의지를 담고 있어 고무적이다. 여기에 다원유 등 수입원자재 가격이 급등세를 보이고있는 가운데 우리나라의 해외 자원개발사업이 활기를 띠고있다.석유공사는 민간기업들과 함께 지난해에만 예멘2곳 등 전세계 5개국9곳의 석유 및 천연가스 광구계약을체결, 올들어 최근참여사들과 함께 주주간계약 서명식을 갖고본격적인 사업을 추진 중이다. 광업진흥공사역시 SK네트웍스와제휴, 최근 전체 매장량이650만t으로 추정되는 중국 후난성의 아연광 탐사권을 획득, 합작 사설립계약을 체결한 것으로 알려지고 있다. 민간기업들도 해외자원분야진출이 활발하다. 기업들의 해외 자원개발을 위한 투자규모도 늘어 올해38개 해외 자원개발기업이 32개국 211개 사업에 모두31억달러를 투자할 계획인것으로 나타났다. 이에본지는 올해 들어서도 고유가의 지속으로 속증하고 있는에너지수입동향과 해외 자원 개발의 현실과 문제점, 개선방향, 나아가 최근 안전적인 자원확보를 위한 동향을 진단해본다.

Diversity and Phylogenetic Analysis of Fluorescent Pseudomonads Isolated from Soil-Root System of Red Pepper in Greenhouse (비닐하우스 고추재배지의 토양과 근계로부터 분리된 형광성 Pseudomonads의 계통 분류 및 다양성)

  • Among the fluorescent pseudomonad isolates from soil- root system of red pepper in Chinju, Kyunsangnam-Do, the phylogenetic analysis for 35 isolates were conducted. The partial 16S ribosomal DNA sequences were used as taxonomic key for phylogenetic analyses, and these sequences were enabled to identification of the fluorescent pseudomonad isolates on the species level. The 17 isolates among them were classified into Pseudomonas putida group, and consisted of the strains isolated mainly from soil. This group were subdivided into 4 subgroups (I, II, III, and IV). The subgroup I and IV were unique ones which were relatively remotely related with subgroup II and III including the type strain of P. putida. The 15 isolates among 35 isolates were grouped along with the type strain of Pseudomonas fluorescens, and 3 isolate were characterized as intermediates of P. fluorescens and Pseudomonas chlororaphis. Most of strain isolateds from the rhizosphere soil and rhizoplane of red pepper were identified as P. fluorescens and closely related with each other. In this study, root of red pepper was supposed to be colonized by a specific strain or strains of P. fluorescens.

Analysis of Melanocortin Receptor 1 (MC1R) Genotype in Korean Brindle Cattle and Korean Cattle with Dark Muzzle (칡소와 비경흑색 한우의 Melanocortin Receptor 1 (MC1R) 유전자형 분석)

  • PCR-RFLP analysis was carried out to investigate the genotype of Melanocortin receptor 1 (MC1R) gene in Korean Brindle Cattle and Korean Cattle with dark muzzle, which are coat color and muzzle pigmentation variants of Korean Cattle, respectively. Allelic variants of MC1R in cattle were analyzed by digestion with BsrFⅠ, AciⅠ. Among six genotypes, $E^D/E^D,\;E^D/E^+,\;E^D/e,\;E^+/E^+,\;E^+$/e and e/e, detected in cattle, only two genotypes, $E^+/E^+\;and\;E^+$/e, were observed in Korean Brindle Cattle, probably reflecting the necessary of $E^+$ allele for the expression of black brindle coat color. As in Korean Cattle with light muzzle, the $E^+$/e and e/e genotypes were detected in Korean Cattle with dark muzzle. The $E^+$ and e alleles frequencies in two populations of Korean Cattle with dark muzzle and with light muzzle were 0.37, 0.63 and 0,11, 0.89, respectively. Although the frequency of $E^+$ allele in Korean Cattle with dark muzzle was higher than in Korean Cattle with light muzzle, the $E^+$ allele was not completely associated with dark muzzle pigmentation. The results of this experiment indicate that the difference of MC1R genotype and frequency may be useful for fixation of coat color in Korean Cattle as well as Korean Brindle Cattle.

  • Background: We conducted a case-control study to evaluate the potential association between SERPINA1 genotypes ($M1_{Val}$, $M1_{Ala}$, S, and Z) and the risk COPD. Methods: The study population consisted of 93 patients with COPD and 112 healthy controls. The polymerase chain reaction and restriction fragment length polymorphism for detecting the SERPINA1 variants. Results: The M2 allele of the SERPINA1 gene was significantly associated with the risk of COPD in Koreans. The effect of the M2 allele on the risk of COPD was more pronounced in the subgroup<64 years. Conclusion: These results suggest that SERPINA1 polymorphisms may contribute to a genetic predisposition for COPD. However, additional studies with larger sample sizes are required to confirm our findings.

Variation in Leaf Characteristics among Hybrids of Jeoktanshim Hibiscus syriacus L (적단심계 무궁화 품종간 교배차대묘의 엽 특성 변이)

  • This study was performed to provide basic information of the development and breeding of new cultivars of Hibiscus syriacus L., which have more beautiful and diverse flowers. Morphological characteristics of the leaves and genetic variation of the leaves of two Jeoktanshim-line cultivars, Bulsae and Pyungsung, were crossed each other. The result of the cross between Bulsae and Pyungsung are as follows: Mean leaf length and width were 6.09cm and 3.36cm, respectively. Mean petiole length was 1.08cm, and mean shoulder width and incision depth were 1.50cm and 0.84cm, respectively. Mean leaf base angle was $98.56^{\circ}$. Leaf index and petiole length were the most important components of leaf characteristics. Two groups were clustered in distance level 5.4. The result of the cross between Pyungsung and Bulsae are as follows: Mean leaf length and width were 5.25cm and 3.10cm, respectively. Mean petiole length was 0.87cm, and mean shoulder width and incision depth were 1.50cm and 0.76cm, respectively. Mean leaf base angle was $107.87^{\circ}$, which was the widest among ten cross combinations. Two groups were clustered with seven leaf characteristics in distance level 4.2. Group I included two small groups, and group II included four small groups.

A Comparative Analysis of the Illumina Truseq Synthetic Long-read Haplotyping Sequencing Platform versus the 10X Genomics Chromium Genome Sequencing Platform for Haplotype Phasing and the Identification of Single-nucleotide variants (SNVs) in Hanwoo (Korean Native Cattle) (일루미나에서 제작된 TSLRH (Truseq Synthetic Long-Read Haplotyping)와 10X Genomics에서 제작된 The Chromium Genome 시퀀싱 플랫폼을 이용하여 생산된 한우(한국 재래 소)의 반수체형 페이징 및 단일염기서열변이 비교 분석)

  • In Hanwoo cattle (Korean native cattle), there is a scarcity of comparative analysis papers using highdepth sequencing and haplotype phasing, particularly a comparative analysis of the Truseq Synthetic Long-Read Haplotyping sequencing platform serviced by Illumina (TSLRH) versus the Chromium Genome Sequencing platform serviced by 10X Genomics (10XG). DNA was extracted from the sperm of a Hanwoo breeding bull (ID: TN1505D2184/27214) provided by Hanwoo research canter and used for the generation of sequence data from both the sequencing platforms. We then identified SNVs using an appropriate analysis pipeline tailored for each platform. The TSLRH and 10XG platforms generated a total of 355,208,304 and 1,632,772,004 reads, respectively, corresponding to a Q30 (%) of 89.04% and 88.60%, respectively, of which 351,992,768(99.09%) and 1,526,641,824(93.50%) were successfully mapped. For the TSLRH and 10XG platforms, the mean depth of the sequencing was 13.04X and 74.3X, the longest phase block was 1,982,706 bp and 1,480,081 bp, the N50 phase block was 57,637 bp and 114,394 bp, the total number of SNVs identified was 4,534,989 and 8,496,813, and the total phased rate was 72.29% and 87.67%, respectively. Moreover, for each chromosome, we identified unique and common SNVs using both sequencing platforms. The number of SNVs was directly proportional to the length of the chromosome. Based on our results, we recommend the use of the 10XG platform for haplotype phasing and SNV identification, as it generated a longer N50 phase block, in addition to a higher mean depth, total number of reads, total number of SNVs, and phase rate, than the TSLRH platform.

Coat Color Patterns and Genotypes of Extension and Agouti in Hanwoo and Jeju Black Cattle (제주흑우와 한우에서 Extension, Agouti 유전자형과 모색 출현 양상)

  • To understand the relationship between coat color inheritance patterns and genotypes of Extension (E) and Agouti (A) loci in cattle, the genotypes for melanocortin-1 receptor (MC1R) and agouti signaling protein (ASIP) were analyzed in Hanwoo, Jeju black cattle (JBC), and their crossbred progeny. Three MC1R alleles ($E^D$, $E^+$, and e) were found in the black-colored JBC population. JBC had no recessive homozygotes (e/e), but this genotype was predominant in the Hanwoo breed. However, MC1R $E^+$/e Hanwoo did not produce a black coat color as they appeared either as brown or solid red. For ASIP, three genotypes (A/A, A/$A^{Br}$, and $A^{Br}/A^{Br}$) were determined by insertion/deletion of an L1-BT element in Hanwoo. The ASIP $A^{Br}$ allele was rarely observed, and no ASIP $A^{Br}/A^{Br}$ homozygotes were detected in the JBC population. Cattle carrying ASIP $A^{Br}$ did not show any agouti-like brindle pigmentation patterns in either breed or their progeny. The coat colors of the crossbred progeny were discriminated by two colors, yellowish-brown versus dark-brown or black, and their coat colors were directly related to the genotypes of the Extension locus, yellowish-brown (e/e) and dark-brown or black ($E^+$/e), but not to the Agouti locus. ASIP genotypes probably did not affect coat color development in the Hanwoo or crossbred progeny. Our results suggest that the ASIP genotypes do not play key roles in coat color variation, but the MC1R genotypes do direct the phenotypes of Hanwoo, JBC, and their progeny.
