  • Vanilloid receptor TRPV1 (known as capsaicin channel, transient receptor potential vanilloid 1) is known to be a key protein in the pain signal transduction. However, the proteins controlling the activity of the channel are not much known yet. Recently mouse Rab11-FIP3 (Rab11-family interaction protein 3) was found and reported to interact with rat TRPV1. Rab11 has been shown to play a key role in a variety of cellular processes including plasma membrane recycling, phagocytosis, and transport of secretory proteins from the trans-Golgi network. Therefore, Rab11-FIP3 was proposed to be involved in the membrane trafficking of TRPV1. In this study, the unreported rat Rab11-FIP3 was yet cloned in order to show the specific interaction of the TRPV1 and Rab11-FIP3 in the same species of rat and to examine the membrane trafficking of TRPV1. The result showed that rat Rab11-FIP3 is expected to have 489 amino acids and showed 80% identity with that of human and over 90% identity with that of mouse. Rab11-FIP3 was found to be expressed in heart, brain, kidney, testis using northern and western blot analyses. We also found that rat Rab11-FIP3 was colocalized with rat TRPV1 but not with TRPV2 of same family in the rat brain by using immunohistochemistry showing that two proteins interact specifically, suggesting the role of Rab11-FIP3 in the membrane trafficking.

  • With the development of genomics, wearable device and IT/NT, a vast amount of bio-medical data are generated recently. Also, healthcare industries based on big-data are booming and big-data technology based on bio-medical data is rising rapidly as a core technology for improving the national health and aged society. A pathway is the biological deep knowledge that represents the relations of dynamics and interaction among proteins, genes and cells by a network. A pathway is wildly being used as an important part of a bio-medical big-data analysis. However, a pathway analysis requires a lot of time and effort because a pathway is very diverse and high volume. Also, multidimensional analysis systems for various pathways are nonexistent even now. In this paper, we proposed a pathway analysis system that collects user interest pathways from KEGG pathway database that supports the most widely used pathways, constructs a network based on a hierarchy structure of pathways and analyzes the relations of dynamics and interaction among pathways by clustering and selecting core pathways from the network. Finally, to verify the superiority of our pathway analysis system, we evaluate the performance of our system in various experiments.

  • This research deals with an estimation method for kinetic reaction model. The kinetic reaction model is a model to explain spread or changing process based on interaction between species on the Biochemical area. This model can be applied to a model for disease spreading as well as a model for system Biology. In the search, we assumed that the spread of species is stochastic and we construct the reaction model based on stochastic movement. We utilized Gillespie algorithm in order to construct likelihood function. We introduced a Bayesian estimation method using Markov chain Monte Carlo methods that produces more stable results. We applied the Bayesian estimation method to the Lotka-Volterra model and gene transcription model and had more stable estimation results.

  • As development in technology of bioinformatics recently makes it possible to operate micro-level experiments, we can observe the expression pattern of total genome through on chip and analyze the interactions of thousands of genes at the same time. Thus, DNA microarray technology presents the new directions of understandings for complex organisms. Therefore, it is required how to analyze the enormous gene information obtained through this technology effectively. In this thesis, We used sample data of bioinformatics core group in harvard university. It designed and implemented system that evaluate accuracy after dividing in class of two using Bayesian algorithm, ASA, of feature extraction method through normalization process, reducing or removing of noise that occupy by various factor in microarray experiment. It was represented accuracy of 98.23% after Lowess normalization.

  • A genetic study on the panicle characters in Oryza sativa was carried out by means of a 5 x 5 diallel cross. The five parental varieties were Raekyung, Yeongnamjosaeng, Nongbaek, Yushin and Honenwase. All characters were correlated positively each other, except number of kernels per primary branch. The number of secondary branches per primary branch.was the most effective factor in determining the number of kernels per panicle, the next being the number of primary branches per panicle. Regression analyses of the data of Vr/Wr indicated the presence of non-allelic gene interactions for all characters. Overdominant characters were the number of kernels per panicle, the number of primary branches per panicle, the number of secondary branches per primary branch, the number of kernels per primary branch and sum of kernels on all the tertiary branches per panicle, suggesting that the characters were more influenced by dominant effect than additive effect. However, the number of kernels per secondary branch was partially dominant where the genetic variation was due more to additive effect than to dominance effect. But after omitting the parent which had non-allelic interaction gene, the characters; the number of kernels per panicle, the number of secondary branches per primary branch, and the number of kernels per secondary branch, were partially dominant. Narrow sense heritabilities(h$^2$ N) in number of kernels per panicle and number of secondary branches were high and moderate, respectively, but those of the rest were lower. Mean squares of GCA and SCA of all characters, except SCA of the number of kernels per secondary branch, were highly significant. Effects of GCA were larger than SCA effects in all characters. Raekyung, Yushin and Nongbaek had highly positive GCA, and the best positive SCA was observed in crosses of Nongbaek x Tongillines (Raekyung, Yushin, and Yeongnamjosaeng) in all characters.

  • To information on the inheriance of heading date of rice under the short-day condition, the $F_2$ seeds obtained from all possible combinations of the diallel crosses between five leading rice varieties: Nongbaek, Tongil, Palgueng, Mangyeong and Gimmaze were grown under the short-day condition and natural condition. The days to heading were investigated and analysed for genetic components. The results obtained are summarized as follows. 1. Under the natural condition, the durations to heading of all combinations excluding Nong-baek x Palgueng and Nongbaek x Mangyeong were longer than those of their parents. mean. Whereas, under the short-day condition, the duration to heading of Mangyeong x Gimmaze combination was equal to that of their parents mean, and those of all other combinations were significantly shorter than those of their parents mean. 2. Under the natural condition, the non-allelic gene interactions were significant in days to heading, but under the short-day condition, the non-allelic gene interactions were not significant, and inheritance of days to heading revealed partial dominace in which additive effects were greater than dominant effects. 3. Tongil and Gimmaze were located on dominant zone under the natural day-length, and Nongbaek and Mangyeong were under the short-day condition. 4. Under the natual condition, dominant effects were greater than additive effects, and both heritabilities of the broad and the narrow senses were significantly low. Under the short-day condition, additive effects were significantly higher than dominant effects, and both heritabilities of the broad and the narrow senses were high.

  • We performed a molecular marker-based analysis of quantitative trait loci (QTLs) for traits that determine the quality of the appearance of grains, using 120 doubled-haploid (DH) lines developed by another culture from the F1 cross between ‘Cheongcheong’ (Oryza sativa L. ssp. Indica) and ‘Nagdong’ (Oryza sativa L. ssp. Japonica). The traits studied included length, width, and thickness of the grains, as well as length-to-width ratio and 1,000-grain weight. The objective of this study was to determine the genetic control of these traits in order to formulate a strategy for improving the appearance of this hybrid. Within the DH population, five traits exhibited wide variation, with mean values occurring within the range of the two parents. Three QTLs were identified for grain length on chromosomes 2, 5, and 7. Three QTLs were mapped for grain width on chromosome 2: qGW2-1, qGW2-2, and qGW2-3. Six chromosomes were identified for the grain length-to-width ratio; four of these were on chromosome 2, whereas the other two were on chromosomes 7 and 12. One QTL influencing 1,000-grain weight was identified and located on chromosome 8. The results presented in the present study should facilitate rice-breeding, especially for improved hybrid-rice quality.

  • Background : No association between schizophrenia and dopamine D4 receptor polymorphisms have been reported. Despite these results, it is premature to exclude the association. It has been suggested that the susceptibility to develop schizophrenia could result from variation at a number loci which may interact or coact with each other. Therefore, we investigated a possible association of combinations of exon III 48bp polymorphism[D4E3] and exon I 12bp polymorphism of the DRD4 gene [D4E1] with schizophrenia. Methods : 207 unrelated Korean schizophrenic patients and 191 healthy controls were recruited. DRD4 genotype was established using the polymerase chain reaction. Statistical analysis consisted of ${\chi}^2$ tests for Hardy-Weinberg proportions and genotypic and allelic frequencies in the patients and control groups. Results : There were no statistically significant differences in the each polymorphisms between schizophrenics and controls. And all genotype frequencies were within Hardy-Weinberg expectations. When the combinations of the polymorphism in schizophrenia and controls were compared, however, there were significant differences at $A1A2^*2/4$ in the distributions of the combinations of D4E1 and D4E3(p<0.01). Conclusions : These findings suggest that the certain combination of D4E1 and D4E3($A1A2^*2/4$) has the protective role to a susceptibility for schizophrenia.

  • Objectives: This study was conducted to investigate whether joint effects between family allergy history and environmental tobacco smoke(ETS) by parents were associated with pediatric asthma and wheezing. Methods: The study objects of this study were 2301 element school students and their parents in an urban-rural areas of Gyeonggi-do. Pediatric asthma and wheezing were identified by measures of International Study of Asthma and Allergies in Childhood (ISAAC) questionnaires. We investigated history of parental allergy, ETS, and other socioeconomic status of both parent. Data were analyzed using logistic regression methods. Results: After adjusting other variables, children with maternal asthma history were more likely to be reported life time wheezing (OR: 3.79 95%CI:2.43-5.90), recent wheezing (OR:4.09 95%CI:2.28-7.38), and diagnostic asthma (OR:2.61 95%CI: 1.44-4.75). Paternal asthma history increasing risk of life time wheezing (OR 2.01 95%CI:1.19-3.38) and recent wheezing (OR:2.38 95%CI:1.24-4.56). Joint effect between parental allergy history and ETS significantly effected on child's life time wheezing and recent wheezing. The risks of life time wheezing (OR:2.47 95%CI:1.64-3.717) and recent wheezing (OR: 2.51 95%CI:1.34-4.69) were significantly higher than others without both factors. The risk of recent wheezing of children with maternal recent smoking and parental allergy history (OR:4.83 95%CI:1.89-12.33) was higher than their counterpart. Conclusions: The result of this study implies that children with family allergy history and passive smoking are more likely to be get asthma and wheezing than children with family allergy history and non-passive smoking. This study provide the object information to increase the efficiency of non-smoking campaign and education for decreasing pediatric asthma risk.

  • This study aimed to investigate the effects of beak trimming and crossbreeding-combinations on the productive performance and stress response levels of Korean native chickens. The study divided 248 individuals from six crossbreeding-combinations into two groups: one underwent beak trimming, and the other did not. The survival rate, body weight, egg production rate, egg quality, feather damage score, HSP-70 gene expression level, H/L ratio, and intracellular DNA damage rate were measured and analyzed. The results showed that the beak-trimmed group had significantly higher survival rates and hen-housed egg production compared to the non-beak-trimmed group (P<0.05). Feather damage and DNA damage rates were significantly lower in the beak-trimmed group (P<0.05). On the other hand, there were no significant differences between the two groups in adult body weight, hen-day egg production, egg quality, HSP-70 gene expression level, and H/L ratio. Among the crossbreeding-combinations, there were significant differences in survival rate, body weight, feather damage score, egg quality, and DNA damage rate (P<0.05), while egg production rate, HSP-70 gene expression level, and H/L ratio showed no significant differences. There was an interaction between beak trimming and crossbreeding-combinations in some traits. In conclusion, beak trimming in Korean native chickens has a positive impact on productive performance, and in terms of stress response, beak trimming may not act as a stress factor or may even reduce stress after the growing period. Furthermore, there were differences in productive performance and stress response levels among crossbreeding-combinations, but the effects of beak trimming were similar across these combinations.
