Ethanol Extract from Cnidium monnieri (L.) Cusson Induces G1 Cell Cycle Arrest by Regulating Akt/GSK-3β/p53 Signaling Pathways in AGS Gastric Cancer Cells (AGS 위암세포에서 Akt/GSK-3β/p53 신호경로 조절을 통한 벌사상자 에탄올 추출물의 G1 Cell Cycle Arrest 유도 효과)

  • Cnidium monnieri (L.) Cusson is distributed in China and Korea, and the fruit of C. monnieri is used as traditional Chinese medicine to treat carbuncle and pain in female genitalia. In this study, we examined the anti-proliferation and cell cycle arrest effects of ethanol extracts from C. monnieri (CME) in AGS gastric cancer cells. Our results show that CME suppressed cell proliferation and induced release of lactate dehydrogenase (LDH) in AGS cells by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assay and LDH assay. Cell morphology was altered by CME in a dose-dependent manner. In order to identify the cell cycle arrest effects of CME, we investigated cell cycle analysis after CME treatment. In our results, CME induced cell cycle arrest at G1 phase. Protein kinase B (Akt) plays a major role in cell survival mechanisms such as growth, division, and metastasis. Akt protein regulates various downstream proteins such as glycogen synthase kinase-$3{\beta}$ (GSK-$3{\beta}$) and tumor protein p53 (p53). Expression levels of p-Akt, p-GSK-$3{\beta}$, p53, p21, cyclin E, and cyclin-dependent kinase 2 (CDK2) were determined by Western blot analysis. Protein levels of p-Akt, p-GSK-$3{\beta}$, and cyclin E were reduced while those of p53, p21, and p-CDK2 (T14/Y15) were elevated by CME. Moreover, treatment with CME, LY294002 (phosphoinositide 3-kinase/Akt inhibitor), BIO (GSK-$3{\beta}$ inhibitor), and Pifithrin-${\alpha}$ (p53 inhibitor) showed that cell cycle arrest effects were mediated through regulation of the Akt/GSK-$3{\beta}$/p53 signaling pathway. These results suggest that CME induces cell cycle arrest at G1 phase via the Akt/GSK-$3{\beta}$/p53 signaling pathway in AGS gastric cancer cells.

Anti-inflammatory activities of fermented Rhus verniciflua stem bark extract and its growth inhibitory effect on Helicobacter pylori (발효옻 추출물의 헬리코박터파이로리 생장억제 및 항염증 활성)

  • This study was designed to investigate the beneficial effects of fermented Rhus verniciflua stem bark extract (RVSBE) on the stomach. We evaluated RVSBE for its antimicrobial activity against Helicobacter pylori (H. pylori), along with its ability to reduce the viability of human gastric cancer AGS cells. In addition, its anti-inflammatory effect was examined by evaluating nitric oxide (NO) production, and inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 mRNA expression in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. RVSBE showed antimicrobial activity, as 2.0 mg of the extract produced a clear inhibition zone of 4.0 mm. RVSBE inhibited the growth of AGS cells by 20% at concentrations ranging from 0.25-1.0 mg/mL. Regarding the anti-inflammatory effects of RVSBE, at 0.1-1.0 mg/mL, the extract showed more than 75% inhibition of NO production. In addition, cells treated with 0.25 mg/mL RVSBE showed a 25% decrease in iNOS mRNA levels compared to those in the LPS-treated cells. These results suggest that RVSBE may have significant inhibitory effects on inflammatory mediators, and therefore, may be a potential anti-inflammatory candidate.

Apoptosis and Cell Proliferation in Gastric Adenoma and Adenocarcinoma (위샘종과 위샘암종에서의 세포자멸사와 세포증식)

  • Purpose: Cancer is a genetic disease caused by alterations in key regulators of cell growth and cell turnover, We investigated apoptotic cell death and cell proliferation in gastric adenomas and adenocarcinomas. Materials and Methods: The TdT-mediated dUTP-biotin nick end labelling (TUNEL) method and immunohistochemistry for Ki-67 were peformed, using paraffin-embedded tissues of 41 gastric adenomas and 100 gastric adenocarcinomas. These results were compared with histopathologic parameters. Results: The Ki-67 labelling index was higher in adenocarcinomas than in adenomas and the apoptotic index was higher in adenomas than in adenocarcinomas. There were no significant difference between the apoptotic index/Ki-67 labelling index and clinicopathological parameters. Conclusion: We propose that cell proliferation is more closely associated with gastric adenocarcinomas than apoptosis is, but that neither has any clinical significance as a prognostic factor in gastric adenocarcinomas. Expression Pattern of EphB2 in Gastric Cancer (위암에시 EphB2 단백의 발현 양상)

  • Purpose: The EphB2 receptor, a member of the receptor tyrosine kinase family, is a target gene of the Wnt signaling pathway and may achieve a tumor suppressor function through regulation of cell growth and migration. Our aim was to determine whether an altered expression of EphB2 might be associated with gastric cancer development and, if so, to determine to which pathologic parameter it is linked. Materials and Methods: For the construction of the gastric cancer tissue microarray, 83 paraffin-embedded tissues containing gastric cancer areas were cored 3 times and transferred to the recipient master block. The expression patterns of EphB2 were examined on tissue microarray slides by using immunohistochemistry and were compared using pathologic parameters, including histological type, depth of invasion, lymph node metastatsis, and peritoneal dissemination. Results: The EphB2 protein was expressed in the normal gastric mucosal epithelium, especially in the bottom of the mucosa. We found loss of EphB2 expression in 30 (36.1%) of the 83 gastric cancer tissues. Statistically, loss of EphB2 expression was more common in gastric cancer with lymph-node metastasis. There was no significant correlation between EphB2 expression and depth of invasion, histologic type, or peritoneal dissemination. Conclusion: Our findings suggest that loss of EphB2 expression may represent a critical step in gastric carcinogenesis.

Inhibition of Cell Invasion by Ethyl Alcohol Extracts of Hizikia fusiforme in AGS Human Gastric Adenocarcinoma Cells (AGS 인체 위암세포에서 톳 에탄올 추출물에 의한 침윤성 저해)

  • In this study, we investigated the effects of ethyl alcohol extracts of Hizikia fusiforme (EHF) on the correlation between tightening of tight junctions (TJs) and anti-invasive activity in human gastric adenocarcinoma AGS cells. Inhibitory effects of EHF on cell proliferation, motility, and invasiveness were found to be associated with increased tightness of the TJs, which was demonstrated by an increase in transepithelial electrical resistance. Activities of matrix metalloprotease (MMP)-2 and -9 in AGS cells were dose-dependently inhibited by treatment with EHF, and this was also correlated with a decrease in expression of their mRNA and proteins; however, tissue inhibitor of metalloproteinase (TIMP)-1 and -2 mRNA levels were increased. Additionally, immunoblotting results indicated that EHF repressed the levels of claudin proteins (claudin-1, -3, and -4), major components of TJs that play key roles in control and selectivity of paracellular transport. Furthermore, EHF decreased expression of insulin such as growth factor-1 receptor proteins, while concurrently increasing that of thrombospondin-1 and E-cadherin. In conclusion, these results suggest that EHF treatment may inhibit tumor cell motility and invasion, and therefore act as a dietary source to decrease the risk of cancer metastasis.

Inhibitory Effect of Persimmon Leaves on the Mutagenicity in Spore Rec Assay and on the Growth of Human Cancer Cells (감잎의 용매별 추출물의 돌연변이 유발 억제 및 암세포 증식억제 효과)

  • The inhibitory effects of persimmon leaves on th e mutagenicity in spore rec assay and on the growth of HT-29 human colon cancer cells and AZ-521 human gastric cancer cells were studied. Methanol extract of persimmon leaves inhibited the mutagenicity induced fly N-methyl- N'-nitro-N-nitrosoguanidine(MNNG) in spore rec assay. The hexane, chloroform and ethylacetate fraction from the methanol extract exhibited strong antimutagenicity against MNNG in spore rec assay The methanol extract of persimmon leaves also revealed the inhibitory effects on the growth of HT-29 human colon cancer cells and AZ-521 human gastric cancer cells. Among the solvent extracted fraction from the methanol extract, the chloroform fraction was most effective and inhibited the growth of HT-29 and AZ-521 cells by 100 percent.

Anti-proliferative Properties of p-Coumaric Acid in SNU-16 Gastric Cancer Cells (SNU-16 위암 세포주에서 p-coumaric acid의 세포성장 억제 효과)

  • The ubiquitous plant metabolite p-coumaric acid (p-CA) has antioxidant and anti-inflammatory properties, but its anti-cancer activity has not been established in gastric cancer cell lines. In this study, we investigated the effects of p-CA on the proliferation and transcriptome profile of SNU16 gastric cancer cells. Treatment with p-CA induced apoptosis of the SNU-16 cells by regulating the expression of pro-apoptotic and anti-apoptotic proteins, such as Bcl-2, poly (ADP-ribose) polymerase (PARP), Bax, procaspase-3, and cleaved-caspase-3. The genes differentially expressed in response to p-CA treatment of the SNU-16 cells were identified by RNA sequencing analysis. Genes regulated by p-CA were involved mainly in the inflammatory response, apoptotic processes, cell cycle, and immune response. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that the phosphatidylinositol-3-kinase-Akt and cancer signaling pathways were altered by p-CA. Protein-protein interaction (PPI) network analysis also revealed that p-CA treatment was correlated with differential expression of genes associated with the inflammatory response and cancer. Collectively, these results suggest that p-CA has potential utility in gastric cancer prevention.

고분자량 Chitooligosaccharides의 in vitro 항종양성

  • Chitosan에 Ttichoderma viride 유래의 cellulase를 처리하여 효소분해함으로써 고분자량 chitooligosaccharides를 얻었다. 생산된 올리고당을 HPLC로 분석한 결과 octamer 이상의 것이 전체 올리고당 중 49.3%에 달하는 비교적 고분자 화합물로 구성된 chitooligosaccharides의 혼합물이었다. MTT검색법에 의하여 고분자량 chitooligosaccha- rides의 정상세포주와 암세포주에 대한 세포독성을 실험하였다. 정상세포주인 아프리카 초록원숭이의 신장세포에 대한 $IC_{50}$/(50% 저해농도)값은 1,107.95$\mu$g/ml로 정상세포에 대한 독성은 거의 없었다. 폐암세포주인 A549, 방광암세포주인 J82, 대장암세포주인 SNU-C4, 위암세포인 SNU-1, 유방암세포주인 ZR75-1에 대한 $IC_{50}$/값은 각각 421.06$\mu$g/ml, 417.99$\mu$g/ml, 445.54$\mu$g/ml, 380,65$\mu$g/ml, 460.49$\mu$g/ml로 in vitro 종양세포 증식억제 활성을 나타내었다.

김치에서 분리한 유산균과 starter를 첨가한 김치의 항암효과

  • 본 연구는 김치에서 분리한 유산균과 starter를 첨가하여 제조한 김치의 항암효과가 in vitro 상에서 어떻게 나타나는지 알기위해 실시하였다. 유산균의 균체 자체로는 모든 암세포에 있어서 세포독성을 거의 나타내지 못하였으며, 김치액의 경우는 평균적으로 $30{\sim}40%$정도의 세포독성을 보여주었다. 김치액인 K2가 K1에 비하여 다소 높은 세포독성을 나타내어 첨가된 starter에 따라 약간의 차이를 보여주었다. 유산균의 파쇄액의 경우에는 모든 암세포에서 50%이상의 세포독성을 보였고 대장암 세포인 WiDr과 위암세포인 MKN-45에서 $70{\sim}80%$ 정도의 세포독성으로 암세포 성장 저해에 상당히 효과적으로 관찰되었다. 유산균의 종류에 따른 세포독성을 비교해보면, 혼합 균체 파쇄액인 E3가 단일 균체 파쇄액보다 전반적으로 약간 높은 경향을 보여주었다.

Inhibitory Effect of Green-Yellow Vegetables on the Mutagenicity in Salmonella Assay System and on the Growth of AZ-521 Human Gastric Cancer Cells (녹황색 채소류의 돌연변이유발 억제 및 AZ-521 위암세포의 성장 저해효과)

  • The antimutagenic effect of green-ye1low vegetables on the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and N-nitrosodimethylamine (NDMA) in Salmonella assay system and also their inhibitory effect on AZ-521 human gastric cancer cells were studied. Twenty-four items from twenty six kinds of vegetables(92%) revealed antimutagenic activity toward MNNG (p< 0.0l, 0.05). Perilla leaf, Korean cabbage, cauliflower, lettuce, mustard leaf, water dropwort, small water dropwort, carrot and burdock inhibited the mutagenicity more than 80%. The methanol extracts of the vegetables also showed the antimutagenic activity toward NDMA (p< 0.01, 0.05). Especially, perilla leaf, kale, soybean sprout and onion inhibited more than 80% of the NDMA induced mutagenicity in S. typhimurium TA100. Small water dropwort and perilla leaf exhibited the strong inhibitory effect (97~100%) on the growth of the AZ-521 human gastric cancer cells. Soybean sprout, water dropwort, broccoli, crown daisy, green red pepper, red pepper leaves, spinach, cabbage and sweet potato also inhibited growth of the cancer cells (p < 0.001~0.05).
