Effects of Senenium and Vitamin E Administration on the Semen Characteristics, Blood Chemical Values and Hormone in Hanwoo Sires I. Effects of Selenium, Vitamin E and rBST Administration on the Semen Characteristics in Hanwoo Sires (Selenium 과 Vitamin E 투여가 한우 종모우의 정액성상, 혈액성분 및 호르몬 변화에 미치는 효과 I.   • The objectives of this study were to investigate 1) the effects of Selenium(Se), Vitamin E (Vit. E) or recombinant Bovine Somatotropin(rBST) administration on fresh and frozen/thawed semen characteristics and 2) the effect of taurine on frozen/thawed semen characteristics in Hanwoo sires Hanwoo sires were randomly assigned to five groups (1. control, 2. rBST, 0.09mg/kg body weight (BW), 3. Vito E 1,500IU/kg BW, 4. Se 0.l mg/kg BW, 5. Vit. E 1,500IU plus Se 0.1 mg/kg BW). The administration of Se, Vit. E and rBST for each experimental group were given 6 times at 15 days interval by intramuscular injection. The administration of Se, Vit. E or rBST in Hanwoo sires didn't affect semen volume and pH values, but sperm viability was significantly increased comparing to the control group. Also, frozen/thawed semen analysis showed that the sperm viability increased, but any other effects were not found in total sperm :lumber, motility and abnormality among treatments. The addition of taurine in semen freezing extender had a beneficial effects on frozen/thawecl semen characteristics in all groups. The administrations of rBST, Vit. E and Se did not affect the sperm capacitation and acrosome reaction, either the ratio of F pattern(uncapacitated and acrosome intact sperm) or AR pattern(capacitated and acrosome-reacted sperm), but the ratio of B patten(capacitated and acrosome intact sperm) of treatment groups was significantly higher than that of control group, These results indicated that the viability, motility and quality of semen in Hanwoo sires were slightly increased by the injection of rBST, Vit. E and Se, and the addition of taurine in semen freezing extender were also increased the semen characteristics after thawing.

Oil Fluorescence Spectrum Analysis for the Design of Fluorimeter (형광 광도계 설계인자 도출을 위한 기름의 형광 스펙트럼 분석)

  • To evaluate the degree of contamination caused by oil spill accident in the sea, the in-situ sensors which are based on the scientific method are needed in the real site. The sensors which are based on the fluorescence detection theory can provide the useful data, such as the concentration of oil. However these kinds of sensors commonly are composed of the ultraviolet (UV) light source such as UV mercury lamp, the multiple excitation/emission filters and the optical sensor which is mainly photomultiplier tube (PMT) type. Therefore, the size of the total sensing platform is large not suitable to be handled in the oil spill field and also the total price of it is extremely expensive. To overcome these drawbacks, we designed the fluorimeter for the oil spill detection which has compact size and cost effectiveness. Before the detail design process, we conducted the experiments to measure the excitation and emission spectrum of oils using five different kinds of crude oils and three different kinds of processed oils. And the fluorescence spectrometer were used to analyze the excitation and emission spectrum of oil samples. We have compared the spectrum results and drawn the each common spectrum regions of excitation and emission. In the experiments, we can see that the average gap between maximum excitation and emission peak wavelengths is near 50 nm for the every case. In the experiment which were fixed by the excitation wavelength of 365 nm and 405 nm, we can find out that the intensity of emission was weaker than that of 280 nm and 325 nm. So, if the light sources having the wavelength of 365 nm or 405 nm are used in the design process of fluorimeter, the optical sensor needs to have the sensitivity which can cover the weak light intensity. Through the results which were derived by the experiment, we can define the important factors which can be useful to select the effective wavelengths of light source, photo detector and filters.

The Naturally Occurring Levels of Nitrate and Nitrite in Livestock Products (축산물 중 천연유래 질산염 및 아질산염 함유량 조사)

  • This study was done in order to investigate the naturally occurring levels of nitrate and nitrite in livestock products. Total samples of 458 consisting of meats (n = 223), processed meat products (n = 51), raw milks (n = 30), processed milk products (n = 142), eggs (n = 5) and processed egg products (n = 7) were analyzed for contents of nitrate and nitrite by ion chromatography (IC). That methods showed good results in terms of linearity, limit of detection (LOD), limit of quantitation (LOQ), recovery, reproducibility and uncertainty. Nitrate and nitrite were detected in 167 and 40 samples, respectively. The nitrate levels (mg/kg) were not detected (ND)~40.23 for modified milks, ND~37.97 for sauce meats, ND~32.40 for process cheeses, ND~31.50 for processed egg products, ND~27.73 for dry milks, ND~24.76 for sausages, ND~22.45 for bacons, ND~21.55 for natural cheeses, ND~20.82 for hams and fermented milks, ND~13.57 for eggs, ND~12.77 for butters, ND~9.31 for milks and ND~3.88 for meats while the nitrite levels (mg/kg) were ND~17.35 for processed egg products and ND~1.92 for meats. In conclusion, the result of this study of nitrate and nitrite in livestock products could be used as one of scientific base datum to determine whether they are naturally occurring or not, including ingredients and their percentage, manufacturing processes, other papers relating to naturally occurring levels of them, and so on.

Chemical components and hepato-protective effect of Lentinula edodes fermented by lactic acid bacteria (표고 유산균 발효물의 성분 및 간기능 보호 효과)

  • This study was conducted to improve the useful components and biological activity of Lentinula edodes fermented by lactic acid bacteria (LAB). Three LAB strains (Lactobacillus brevis KCCM 11904, L. plantarum KCCM 354469, and L. fermentum KCCM 12116) were inoculated and used for L. edodes hot water extract (10%, 20%, 30%) fermentation. LAB fermentation of L. edodes hot water extracts decreased pH and thus were more acidic than non-fermented L. edodes hot water extract. β-glucan and ergothioneine contents were increased by L. edodes in a concentration-dependent manner. The ergothioneine and β-glucan contents were highest in fermented with 30% L. edodes hot water extract fermented by L. plantarum and L. brevis (40.48 mg/100 g and 13.94%, respectively). The hepato-protective effect of fermented L. edodes hot water extracts by the three LAB were tested using Sprague-Dawley rat primary hepatocytes. In primary hepatocytes obtained following liver injury induced by acetaminophen, fermented L. edodes hot water extracts by the three LAB showed protective effects, as evident by reduction of the aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase liver markers. The collective results indicate that the fermented L. edodes hot water extracts obtained using LAB are potentially valuable in preventing or treating liver disease.

Development of Potassium Impregnated Carbon Absorbents for Indoor CO2 Adsorption (K계열 함침 탄소계 흡착제의 실내 저농도 이산화탄소 흡착성능 강화)

  • Relatively high indoor CO2 concentration (>1,000 ppm) has a negative impact on human health. In this work, indoor CO2 adsorbent was developed by impregnating KOH or K2CO3 on commercial activated carbon, named as KOH/AC and K2CO3/AC. Commercial activated carbon (AC) showed relatively high BET surface area (929 m2/g) whereas KOH/AC and K2CO3/AC presented lower BET surface area of 13.6 m2/g and 289 m2/g. Two experimental methods of TGA (2,000 ppmCO2, weight basis) and chamber test (initial concentration: 2,000 ppmCO2, CO2 IR analyzer) were used to investigate the adsorption capacity. KOH/AC and K2CO3/AC exhibited similar adsorption capacities (145~150 mgCO2/g), higher than K2CO3/Al+Si supports adsorbent (84.1 mgCO2/gsample). Similarly, chamber test also showed similar trend. Both KOH/AC and K2CO3/AC represented higher adsorption capacities (KOH/AC: 93.5 mgCO2/g K2CO3/AC: 94.5 mgCO2/gsample) K2CO3/Al+Si supports. This is due to the KOH or K2CO3 impregnation increased alkaline active sites (chemical adsorption), which is beneficial for CO2 adsorption. In addition, the regeneration test results showed both K-based adsorbents pose a good regeneration and reusability. Finally, the current study suggested that both KOH/AC and K2CO3/AC have a great potential to be used as CO2 adsorbent for indoor CO2 adsorption.
