Effects of Pre-sowing Seed Soaking and Planting Depth on Dormancy Breaking and Seedling Emergence of Pearl Millet (Pennisetum americanum(L.) Leeke) (진주조의 파종전 침종 및 파종심도가 휴면타파와 출아에 미치는 영향)

  • One of the most important cultural techniques of pearl millet (Pennisetum americanum (L.) Leeke) is to encourage rapid and uniform emergence of seedlings to establish good stand and to let them grow well. Thus the objectives of the study were to investigate the effects of pre-sowing seed soaking and planting depth on dormancy breaking, germination and emergence of the seedlings, and to estimate the optimum planting season of pearl millet in Suwon, Korea. The seeds with dormancy germinated 99 to 100 percent when soaked in the H$_2$O$_2$ 1% solution for 24 hours and rinsed with pure water, but germinated only 38% and 83% when soaked in pure water for 24 hours just after harvest and drying, and one month later from the harvest time, respectively. The seeds of Australia inbred line did not germinate at the constant 10$^{\circ}C$, but germinated at the constant 11$^{\circ}C$. It also was possible to estimate the optimum planting season by applying minimum temperature 11$^{\circ}C$ for germination. The minimum air temperature reached from late April in Suwon, Korea in regular years but fluctuated from late April to early May in 1986 and 1987. Thus, the safe planting season was mid-May for rapid and uniform germination of pearl millet seed. The optimum depth of planting was 2∼4cm under the optimum soil moisture condition, and 4 to 6 cm under the drier soil moisture condition. Subcoleoptile internode(mesocotyle) length increased according to increased depth of planting. Seedling crown placement also became deeper due to deeper planting of the seeds. The subcoleoptile internode length and seedling crown depth were positively correlated with actual planting depth, indicating that deeper planting would be not good for appropriate adventitious root and tiller development.

Distribution of Vascular Plants in Youngjongdo and Yongyudo (영종도와 용유도 일대의 식물상)

  • Vascular plant surveys in Iss. Youngjongdo and Yongyudo, Incheon were conducted 6 times from May to October 2009 and July 2011. The vascular plants inventoried during the surveys were 644 taxa in total; 11 formas, 73 varieties, 4 subspecies, 556 species, 352 genera and 98 families. Among them, 250 taxa were newly found from the survey areas, including Cnidium japonicum Miq., Argusia sibirica (L.) Dandy, Breea setosa (Willd.) Kitam., Dryopteris erythrosora (D. C. Eaton) Kuntze and Dryopteris nipponensis Koidzumi. In the case of dormancy form, hemicryptophytes were most abundant by 37.1%. Six taxa of Korean endemic plants and nine Korean rare plants were also newly recognized along with 45 taxa of notable plants of regional flora announced by the Ministry of Environment. The naturalized plants were 95 taxa, and the Urbanization Index of the survey areas was 29.6%.

Modification of the commercial silkworm eggs adequate for Bluemoon0silkworm transgenesis (누에 형질전환에 적합한 실용품종 누에알의 제조)

  • Silkworm transgenesis scientists have done some genetic modification work on multivoltine silkworms, but that type of silkworms is less commercial feasible. They are easy to manipulate, because they breed all year round. But the commercial silkworm variety must undergo hydrochloric acid treatment at a high temperature to be artificially hatched. Hydrochloric acid penetrates through the holes in the silkworm eggs, fatally damaging their reproduction. So it had been thought that altering the properties of the commercial silkworm variety would be very difficult. So we have tried to make from diapause to non-diapause eggs using diapauses varieties, 'Backokjam' and 'Jam 124'. At present, our group has establishing the conditions for non-diapause eggs. Oviposited eggs after 40 ~ 60 hours were incubated for 24 hours at $15{\sim}20^{\circ}C$ with dark condition. Non-diapause eggs were completely induced. The hatching rate, molting rate and pupation rate of non-diapause 'Jam 124' and 'Backokjam' eggs showed no differences compared to diapause eggs. When transgenic silkworm using the non-diapause eggs, the hatching rate showed that non-diapause eggs induced from diapause were 40 ~ 70%, diapause eggs treated with artificial incubation were 10 ~ 30%, and polyvoltine strains, HM eggs were 30 ~ 50%. Therefore, we suggest that modification techniques of the commercial silkworm eggs adequate for silkworm transgenesis can be used to develop transgenic silkworms more easily.

A Study on the Germination of Apple Rootstock Seed, Malus sieboldii (사과 대목 종자의 발아에 관한 연구)

  • The seeds of Malus sieboldii were collected in late fall of 1988 and stored in cold stratification condition for 15 weeks. Growth regulators such as $GA_3$, BA and ABA were treated to the seed to know germination capacity and seedling growth. The results were as follows. 1) Higher germination was achieved in $GA_3$ and BA plots, But ABA treatment showed lower germination percentage and physiological dwarf and also induced to the secondary dormancy condition in partly. 2) Average length of time for germination was delayed over 2 days in ABA plot when comparing with the other plots. 3) Average length of plumule and radicle also was inhibited by ABA treatment. This means lower seedling vigor. 4) Fresh and dry weight of the seeding were higher in BA plot while ABA plot was lower and undesirable seedling was produced. From above mentioned results, it is believed that ABA treatment to the seed induced the secondary dormancy and physiological dwarf pattern. Therefore, utilization of BA can be produced the normal seedling, the authors believed.

  • Seed dormancy and germination responses to light and gases were determined for Echinochloa colona (L.) Link. E. colons seeds did not require a period of after-ripening for breaking dormancy. Water movement occurred readily across the seed coat. Repeated cycles of hydration and dehydration reduced viability and thence germination. Water imbibition for 24 h increased seed moisture by 21%; seeds returned to their original weight after drying at room temperature for 13 h. Removal of seed-coats increased germination in the dark. Light stimulated germination. Germination at a daylight intensity of 51.9 $Wm^{-2}$ or less was significantly reduced. Germination of seeds which were exposed to light for 1 h each day was significantly less than that of seeds exposed for longer than 2 h a day. Seeds subjected to blue light had delayed and decreased germination compared to seeds exposed to red light. Ethylene or carbon dioxide exogenenously added in the presence of light stimulated germination. The addition of the two gases together had a synergistic effect. In the dark, however, the two gases did not increase germination.

Establishment of the Successive Rearing Method of Cabbage Butterfly, Pieris rapae L. in a Room Condition (배추흰나비의 실내 계대사육법 확립)

  • Cabbage butterfly, Pieris rapae was reared in a room to establish a year-round rearing system. The eggs oviposited by the parent fed on host plant showed 89.2% of hatchability and hatched in 3.9 days after oviposition. The larval period was 18.1 days under high temperature, long day condition ($25^{\circ}C$, 16L : 8D), showing 97.8% pupal ratio and emergence rate. However, under low temperature, short day condition ($21^{\circ}C$, 10L : 14D) the larval period extended to 23.6 days and the pupal ratio was 70%. All of those pupae went into diapause. The oviposition preference experiment on different hosts (Chinese cabbage, cabbage, tulip and kale) showed that hot-water extract was preferred over methanol extracts or squeezed raw juices. The host preference showed that Chinese cabbage was less preferred than the other three. The artificial ovipositing kit was constructed for the oviposition in a room. The 48-hours old eggs could be stored for 7 days at$ 5^{\circ}C$ and showed 70% of hatchability. Non-diapausing pupae could be stored for 30 days at 5 to $15^{\circ}C$, showing 85% of emergence rate. However, the pupae stored at $5^{\circ}C$ showed longer storage period and higher emergence rate. The systematic successive rearing method of cabbage butterfly in a room was completed, based on the above experiments.

Biochemical Properties of Haemolymph Carboxylesterase in Diapausing Pupae of Helicoverpa assulta (Guenee) (담배나방의 휴면 용 혈림프 Carboxylesterase의 생화학적 특성)

  • Haemolyph carboxylesterases induced in diapausing pupae of Helicoverpa assulta Guenee were investigated. Increase in the activity of the electrophoresed isozyme bands were observed during the diapausing pupae. The isozymatic composition exhibited remarkable alterations represented as disappearance and induction of some isozyme bandsp which were identified as carboxylesterase (CE) on the basis of their specificities to inhibitors. Much higher activity of the induced CE was shown in reaction with $\beta$-naphthyl acetate ($\beta$-Na) than $\alpha$-naphthyl butyrate ($\alpha$-Nb), representing the high regioselectivity to $\beta$-naphthyl group. Optimal temperature for the enzyme activity was different to the substrates used 37$^{\circ}C$ in $\beta$-Na and 4$0^{\circ}C$ in $\alpha$-Nb, respectively. However, the optimal pH for the enzyme activity was the same as 7.5 regardless of the substrates used, and relatively high thermostability of the CE was demonstrated by showing the denaturation at high temperature (50~55$^{\circ}C$).

Emergence Characteristics of Weedy Rice Collected in South Korea (국내 수집 잡초성벼의 출아특성 분석)

  • In order to provide basic data for efficient control of weedy rice, we analyzed emergence rate of 100 weedy rice genetic resources collected in South Korea according to growth temperature and soil burial depth. Analyzing of emergence rate using low dormancy weedy rice genetic resources, emergence was increased at 12 and $14^{\circ}C$ of mean temperature condition. When calculating the effective accumulated temperature above $10^{\circ}C$, emergence rate increased 0.6% and 0.5% in 1 cm and 3 cm of soil depth, respectively according to increase of $1^{\circ}C$. However it reduced as 0.28% in 5 cm of soil depth condition. Analyzing of emergence rate using high dormancy weedy rice, red rice showed 55%, 46%, 41% of emergence rate in 1 cm, 3 cm and 5 cm of soil depth condition, respectively. However those of white rice decreased as 10%, 8% and 5.7% in 1 cm, 3 cm and 5 cm of soil depth condition, respectively. Relative emergence, assuming the final emergence rate as 100%, reached 80%, 90% and 100% when effective accumulated temperature was $83-112^{\circ}C$, $100-123^{\circ}C$, $148-160^{\circ}C$ respectively.

Differences in Emergence and Growth of an Exotic Weed Quamoclit coccinea Moench under Different Environment Conditions (환경조건에 따른 외래잡초 둥근잎유홍초의 출현과 생육 차이)

  • This study was carried out to investigate the differences in germination and growth of Quamoclit coccinea Moench under various temperatures, seeding depths, and levels of shading and soil moisture for effective weed management. Seed dormancy of Q. coccinea Moench was over 1 year; best results were obtained when seeds were soaked in sulfuric acid for 15 minutes in order to break the dormancy. Germination rates of Q. coccinea Moench ranged from 69 to 73% at $25-35^{\circ}C$ and 26% at $15^{\circ}C$. The germination rates ranged from 70-84% at 2, 3, 5, 7, and 8 cm of seeding depths. In addition, the germination rates were 7% and 13% at 12 cm and 15 cm of seeding depths, respectively, and showed normal growth at the both seeding depths. Q. coccinea Moench showed a high germination rate regardless of shading levels, but shoot fresh weight varied depending on the level of shading as follows: 20%>no shading=shading 35%>shading 50%>shading 75%=shading 90%. Q. coccinea Moench did not germinate when soil had a saturation rate of either 30% or 100%. However, 60-83% of seeds germinated with optimal growth when soil had saturation rates of 60% and 80%.
