Bioactive Components from Red Tide Plankton, Cochlodinium polykrikoides (Cochlodinium polykrikoides 적조 조체의 생리활성 성분)

  • Large amounts (300 grams) of natural red tide plankton, Cochlodinium polykrikoides, was collected at the Yokji island coastal waters, Kyeongnam, Korea, on October, 1993. Using the bioassay systems, bioactive materials were screened from methanol extracts of C. polykrikoides. Live C. polykrykoides was toxic to fishes, however, the water soluble and chloroform soluble fraction of their methanol extracts did not shown ichthyotoxicity (5 mg/ml), and toxicity to mice (50 mg, i.p,). These fractions did not show any peaks corresponding to paralytic shellfish toxins or diarryhetic shellfish toxins on the fluorometric HPIC chromatograms. Neither fractions did not show antibiotic activities by paper disk (10 mg/disc) test and chloroform soluble fraction showed only $20\%$ growth inhibition activity on the Lymphoid P-388 at the concentration of $180\;{\mu}g/ml$. Hemolytic activity was detected by both fractions. Fatty acid analysis by GC, GC/MS and proton NMR showed that the chloroform soluble fraction composed of $25.3\%$ of DHA (dorosahexaenoic acid) and $15.3\%$ of IPA (eicosapentaenoic acid) as the hemolytic components.

Retrospective Clinical Analysis of Treatment for Primary Warm Autoimmune Hemolytic Anemia (원발성 온난자가면역용혈빈혈의 치료에 대한 후향적 분석)

  • Background: Primary warm autoimmune hemolytic anemia (AIHA) is a relatively rare hematologic disorder resulting from autoantibody production against red blood cells. There has been very few studies about primary warm AIHA in South Korea because of its low incidence. We retrospectively analyzed the treatment outcome of primary warm AIHA. Method: We reviewed retrospectively the medical records of 9 primary warm AIHA patients from December 2002 to January 2015. We analyzed the causes and clinical characteristics of primary warm AIHA patients. We retrospectively analyzed the clinical data in electronic medical records for 9 Korean patients with AIHA patients who were diagnosed during the period from December 2002 to January 2015 at the Regional University Hospital in Korea. The study protocol was approved by the Institutional Review Board (IRB #2015-08-007, Chosun University Hospital IRB). Results: The mean age was 52 years (range 27~78), the mean hemoglobin level was 5.0 g/dL (range 2.5~6.4 g/dL). All patients received steroids at therapeutic dosages (corticosteroid 1 mg/Kg) as first line treatment. Eight of them showed complete response (5/8, 62.5%) and partial response (3/8, 37.5%), one patient required second-line treatment with rituximab. Two patients who responded first line treatment were relapsed at 86 weeks and 24 weeks after response, respectively. Only one patient of them was retreated with corticosteroid because of anemic symptoms. Conclusion: This study indicates that oral corticosteroid is an effective therapy for primary warm AIHA.

  • In-vitro flow characteristics downstream of a polyurethane artificial heart valve and a Bjork-Shiley Monostrut mechanical valve have been comparatively investigated in pulsatile flow using particle image velocimetry (PIV). With a triggering system and a time-delayed circuit the velocity distributions on the two perpendicular measurement planes downstream of the valves are evaluated at any given instant in conjunction with the opening behaviors of valve leaflets during a cardiac cycle. The regions of stasis and high shear stress can be found simultaneously by examining the entire view of the instantaneous velocity and Reynolds shear stress fields. It is known that high shear stress regions exist at the interface between strong axial jet flows along the wall and vortical flows in the central area distal to the valves. In addition. there are large stagnation or recirculation regions in the vicinity of the valve leaflet, where thrombus formation can be induced by accumulation of blood elements damaged in the high shear stress zones. A correlation between the unsteady flow patterns downstream of the valve and the corresponding opening postures of the polyurethane valve membrane gives useful data necessary for improved design of the frame structure and leaflet geometry of the polyurethane valve.

  • The capacity of tobacco infusions of Korean cigarettes to activate the complement pathway was investigated in vitro by hemolytic assay. Complement activity was studied using samples of normal rabbit serum incubated at $37^{\circ}C$ with tobacco infusions. In all mixtures containing tobacco infusion A, B, C, D, E, F, G, H and I at a concentration of 0.1 : 1, total hemolytic complement showed a progressive decrease of 55.92%, 48.62%, 42.89%, 49.12%, 42.26%, 52.26%, 71.96%, 60.90%, and 61.14% respectively, after 60min of incubation. This study suggests that all tobacco infusions of Korean cigarettes are activators of complement.

A Case of Gilbert's Syndrome with Severe Neonatal Hyperbilirubinemia

  • Gilbert's syndrome is caused by a reduction in the activity of uridine diphosphate glucuronosyltransferase (UGT) and induces chronic, non-hemolytic unconjugated hyperbilirubinemia. It has been suggested that 3-10% of the population has Gilbert's syndrome. Commonly, Gilbert's syndrome causes mild symptoms. However, a case of Gilbert's syndrome with severe neonatal hyperbilirubinemia is presented here. The patient developed jaundice three days after birth. Five days after birth, the patient's total serum bilirubin level was 34 mg/dL. The patient received intensive phototherapy and was given oral phenobarbital. Hemolytic hyperbilirubinemia was excluded on the basis of laboratory tests. Heterozygote polymorphisms of the promoter region (-3279T>G) and exon 1 (211G>A) were found in UGT1A1 gene. After discharge, the patient did not require any further treatment. This is the first case of proven Gilbert's syndrome with severe neonatal hyperbilirubinemia in Korea.

Safety of the genus Enterococcus and the development of food fermentation starters in Korea: Current status and future steps (Enterococcus 속 박테리아의 안전성과 식품발효용 종균 개발의 방향성)

  • Bacteria of the genus Enterococcus are of importance in food fermentations as well as their use as probiotics in humans and livestock. However, they are also important nosocomial pathogens that cause infections. Some strains are resistant to multiple antibiotics and possess virulence factors. The role of Enterococcus species in disease has raised issues on their safety for use in foods or as probiotics. First, this review summarized the positive and negative traits of Enterococcus spp. to illustrate the controversial nature of this bacterial genus and discussed the current genomic approaches can eliminate pathogenic strains. Then, this review examined the current status of starter development for traditional food fermentations and the regulation on the approval of novel food microorganisms in Korea to point out problems in the regulation. Based on the conclusions from the studies on Enterococcus spp., we suggested the direction of safety assessment of novel food microorganisms in Korea.

A Case of Mycoplasma haemofelis Infection in a Korean Domestic Shorthair Cat (한국 토종 단모종 고양이에서 Mycoplasma Haemofelis 감염 증례)

  • A six-months-old male Korean domestic shorthair cat was presented with fever, tachypnea, anorexia, and weight loss and admitted to Lee Seung Jin Animal Medical Center. During the routine physical examination, clinical signs such as mild dehydration and jaundice in the sclera were present. The complete blood count (CBC) and serum chemistry result showed anemia, thrombocytopenia, neutrophilia, and hyperbilirubinemia. Radiography revealed hepatomegaly and splenomegaly. Blood smear and microscopic examination showed severe hemolysis and anisocytosis. We sent the blood sample to the Neodin Veterinary Laboratory for PCR analysis to conduct a test to find out Ehlichia, feline hemoplasmas (haemobartonella), feline leukemia virus (FeLV), feline immunodeficiency virus (FIV) and anaplasma infection. According to PCR examination, the blood of this cat was positive for feline hemoplasmas (Mycoplasma haemofelis), but negative for other pathogens. The patient was prescribed doxycycline for 4 weeks and prednisolone for 1 week. The free of feline hemoplasmas infection was confirmed by PCR recheck after six months.

Antitumor Components of the Cultured Mycelia of Lepiota procera (갓버섯의 항균 분석)

  • To find antitumor components in the cultured mycelia of Lepiota procera, the proteinpolysaccharide obtained from the mycelia was subjected to DEAE - Sephadex A-50 column chromatography and Sepharose-4B gel filtration. Of the fractions, the purified Fraction C1 was named lepiotan and examined for antitumor activity against the solid form of sarcoma 180 in ICR mice. The inhibition ratio of lepiotan was 64% at the dose of 10 mg/kg/day for 10days. The chemical analysis of lepiotan showed 60% polysaccharide and 21 % protein. The polysaccharide moiety was found to be a heteromannoglucan which consisted of 46.3% glucose, 40.2% mannose and 11.0% fucose. When the antitumor component, Fraction A, was examined for immunopotentiation activity, it was found to increase the number of plaques in hemolytic plaque assay and to restore the immunity in the tumor-bearing mice up to 89.7% of the normal level. Also the antitumor acitivity was suppressed by the treatment with carrageenan, an antimacrophage agent. These results indicate that the antitumor activity was exerted through immunopotentiation, but not through direct cytotoxicity against the tumor.

  • 1. The total 1,204 of human sera from January to December, 1976 of the out-patients in St. Mary's Hospital were tested one by one. The total 57.6% of the 1,204 cases were confirmed as a hemagglutination inhibition antibodies, and 58.18% out of 574 male cases and 57.14% of 630 female cases were confirmed to have antibodies, respectively. 2. The monthly distribution of hemagglutination inhibition antibody was shown to be 91.2% out of 34 cases in October, 1976 but other months it was low. 3. In the group whose age was from 51 to 60, it was 64.6% and this shows that the group was markedly higher than others.

  • A hemolysin producing strain was isolated from Kum rivet estuary located in west part of Korea. In the process of identification the isolated strain was similar to V. mimicus but did not show characteristics of known Vibrio species; therefore, the strain was designated as Vibrio sp. D9 ( V. kumkang) tentatively and further identification study was carried out by comparing its bacteriological characteristics, Morphologically Vibrio sp. D9 was a typical straight roe with a polar flagellium. Among known Vibrio species no identical strains were found when using automatic bacteria identification system ($MicioLog^(TM)$system, release 4.0, Biolog Inc., USA) which evaluated the ability of metabolizing 95 kinds of carbon and nitrogen sources. Vibrio sp. D9 showed 18 and 13 different responses as compared to V. mimicus and V. cholerae, respectively. Clear hemolysis zones were observed with the strain against human and sheep blood agar plate, Hemolytic toxicity was confirmed by strong vascular permeability and fatal toxicity against mouse was also observed. Thus the strain was a pathogenic vibrio. Growth conditions for Vibrio sp. D9 were salinity of $0{\~}5.0{\%}$, pH of $6.4{\~}9.8$, temperature of $15{\~}41^{\circ}C$, respectively.
