This study was undertaken to develop a cascade-type continuous culture system (CCCS) that combines both ICT and biotechnology (BT), for the mass production of microalgae. This system is capable of maintaining the essential culture conditions of pH, temperature, carbon dioxide, and illuminance control, which are key parameters for the growth of microalgae, and is economical for producing microalgae regardless of the season or location. It has the added advantage of providing stable and high productivity. In the current study, this system was applied to culture microalgae for 71 days, with subsequent analysis of the experimental data. The initial O.D. of the culture measured from incubator 1 was 0.006. On the 71st day of culture, the O.D.s obtained were 0.399 (incubator 1), 0.961 (incubator 2), 0.795 (incubator 3), and 0.438 (incubator 4), thereby confirming the establishment of continuous culture. Thus, we present a smart-farm based on ISMC (in-situ monitoring and control) for a mass culture method. We believe that this developed technology is suitable for commercialization, and has the potential to be applied to hydroponics-based cultivation of microalgae and cultivation of high-value-added medicinal plants as well as other plants used in functional foods, cosmetics, and medical materials.
For the photoproduction of molecular hydrogen by photosynthetic bacteria in outdoor conditions, we constructed automatically controlled semi-continuous culture system. When the amount of hydrogen gas produced can be measured by a gas meter with a pulse generator, the same amount of substrate consumed for hydrogen production could be supplied by micro pump related with timers. Using the apparatus, we examined hydrogen production with immobilized cells of Rhodopseudomonas sPhaeroides B6 in outdoor conditions. In spite of severe fluctuation of weather and illumination, the culture was maintained under good control with regard to hydrogen productivity. It was possible to automate the semi-continuous outdoor culture of photosynthetic bacteria for hydrogen production.
A quantitative physiological approach has been employed to estimate the metabolic parameters such as specific uptake rates of nutrients and specific production rate in continuous culture of Pseudomonas elodea for gellan gum production. The estimated values of metabolic parameters are used for process improvement. During the exponential growth phase, the specific growth rate was 0.16hr-1 in batch culture. The gellan gum concentration increased up to 0.7g dry weight/100g broth and the apparent viscosity of the culture broth was about 4,500 cp.(72hrs culture). The ratio of specific uptake rate of carbon to that of nitrogen were found to be optimum at about 3.0mg-carbon/mg-nitro-gen. With the improved medium, the maximum gellan production rate, 0.6g dry weight/1/hr, was obtained at D=0.14 hr-1. The shear stresses of culture broth were fairly well correlated with shear rates by using Casson equation and at highly viscous culture broth, oxygen transfer coefficient was greatly reduced.

This study was carried out to compare the growth and fatty acids composition of the rotifer (Brachionus rotundiformis) cultured in high density by the various enrichments and culture methods. The rotifer fed on condensed freshwater Chlorella was enriched with $\omega$-yeast, Algamac, Super Selco and marine Chlorella. In another culture method, the rotifer was cultured with enrichment supplements for 6 hours after feeding with condensed freshwater Chlorella supplement for 18 hour. The rotifer fed with condensed marine Chlorella for 24 hours without fieshwater Chlorella was used as a control group. Culture tanks (5 $\ell$ working volume) was immersed in a water bath ($28^{\circ}C$). The density of rotifer and dissolved oxygen level in water was stable in control group of rotifer cultured with condensed marine Chlorella for 24 hours and the n-3 HUFA content of rotifer was the highest among the rotifer culture methods. However, the density of rotifer and dissolved oxygen level in the groups of rotifers enriched with $\omega$-yeast, Algamac and Super Selco by methods were drastically decreased, The n-3 HUFA contents of rotifers enriched by Super Selco were higher than those of rotifer enriched by either $\omega$-yeast or Algamac in both methods. The results from this experiment indicated that supplementation of condensed marine Chlorella for 24 hour by the semi-continuous culture was effective for the improvement of the nutritional value of rotifer and it could provide the stable growth condition for rotifer culture in high density.
The growth and fermentation profiles of Clostridium acetobutylicum KCTC 1037 were examined in batch and continuous modes with pH variation and phosphate limitation. Clostridium acetobutylicum KCTC 10 37 grew better at pH 4.5 than at pH 5.5 or 6.5. Acetate and butyrate were produced at pH 5.5, whereas culture at pH 4.5 produced acetone and butanol. Solvent production was increased by the phosphate limitation in a batch culture, but in a phosphate-limited continuous culture for 400 hours steady-state solventogenesis was not observed. The induction and maintenance of solventogenesis presumably require not only acidic condition or phosphate limitation but also favourable bioenergetic condition.

Sprial adaptation technique of conditioned media has been applied to cultivate human cell line which can not survive in a serum free mdium without adding any growth factors in basal medium Doubling time and scu-PA production from serum free adapted cells were 5 days and 890 (IU/mL), respectively in a T-flask, whose values were not much lower than the productivity of 1100(IU/mL) from 5% serum containing medium. It was required to use conditioned media for attaching cells on microcarriers when cells were inoculated into a spinner vessel. Then, cells could continuously grow in serum free medium with having specific growth rate of 0.106 (1/day) and specific scu-PA production rate of $1.58{\times}10_{-5}$(IU/cell/day) in batch cultivation.
A modified amidolytic assay and a fibrin plate method were used to accurately measure the concentration of single chain urokinase type plasminogen activator (scu-PA) and two-chain urokinase type plasminogen activator (tc-PA) in the spent media. $1.65{\times}10^6$(viable cells/ml) of maximum cell density and 1670(IU/ml) of scu-PA concentration were obtained in 1% serum containing medium. The overall conversion ratio from scu-PA to tc-PA was less than 10%. In the results of batch cultivation in a spinner vessel, $4.43{\times}10^6(total cells/ml)$ of maximum cell density and 1560(IU/ml) of scu-PA concentration was observed. The maximum scu-PA concentration and specific scu-PA Productivity were obtained in 1760(IU/ml) and $3.13{\times}10^{-4}(IU/cell)$, respectively, from perfusion cultivation. The conveysion ratios from batch, fed-batch and perfusion cultivations were less than 12%, which means that about 90% of scu-PA secreted from the cells can be maintained during the cultivations.
