Comparison on phosphate solubilization ability of Pantoea rodasil and Burkholderia stabilis isolated from button mushroom media (양송이배지로부터 분리한 Pantoea rodasil 와 Burkholderia stabilis의 인산가용화능 비교)

  • This study investigated the synergistic effect of single inoculation and co-inoculation of phosphate-solubilizing bacteria (PSB) Pantoea rodasil LH-1and Burkholderia stabilis HS-7. Phosphate-solubilizing abilities of these strains were assessed by measuring phosphorus content in culture media that were singly inoculated or co-inoculated with these strains for 7 days. The co-inoculation of P. rodasil LH-1and B. stabilis HS-7 was found to release the highest content of soluble phosphorus ($783.41{\mu}g\;mL^{-1}$) into the medium, followed by single inoculation of B. stabilis HS-7B ($743.90{\mu}g\;mL^{-1}$) and P. rodasil LH-1 ($736.59{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production, and glucose consumption were also observed in the medium inoculated with both the strains, compared with that in the medium inoculated with the strain alone. Results of a plant growth promotion bioassay showed 7.7% and 15.5% higher leaf and root growth, respectively, in romaine lettuce co-inoculated with P. rodasil LH-1and B. stabilis HS-7 than those inoculated with the strain alone. However, no significant difference was observed between single inoculation and co-inoculation of these strains with respect to phosphorus release and plant growth. Although the results of the present study did remarkdly not show the synergistic effect of phosphate solubilization by co-inoculation of the PSB strains examined, these results indicate that treatment with PSB exerts a beneficial effect on crop growth.

Phosphate solubilizing effect by two paraburkholderia bacteria Isolated from button mushroom medium (양송이배지로부터 분리한 두 Paraburkholderia 속 세균에 의한 인산가용화 효과)

  • The present study was conducted to investigate the synergistic effects caused by single and co-inoculation of the phosphate solubilizing bacteria (PSB), Paraburkholderia phenazinium YH3 and Paraburkholderia metrosideri YH4. Phosphate solubilization was assessed by measuring the phosphorus contents for 7 days in a single and co-inoculation medium. Co-inoculation of the two strains was found to release the highest content of soluble phosphorus ($1,250{\mu}g\;mL^{-1}$) into the medium, followed by the single inoculation of P. metrosideri YH4 ($1196.59{\mu}g\;mL^{-1}$) and P. phenazinium YH3 ($994.34{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production and glucose consumption was also observed in the co-inoculation medium of the two strains. A plant growth promotion bioassay revealed that co-inoculation with the two strains enhanced the growth of romaine lettuce more than single inoculation with either of the two strains (28.5% for leaf and 16.6% for root). Although there was no significant difference between single and co-inoculation of bacterial strains in terms of phosphorous release and plant growth, the synergistic effects of co-inoculation with PSB could be beneficial for crop growth.

  • The results of series studies on the ratio of supplements, out-door composting and out-door fermentation induced by using the rice straw as a main substrates at the cultivation of Agaricus bisporus, and the cultivation of Pleurotus ostreatus using the rice straw bundles on its compost are as follows; When rice straw for cultivation of A. bisporus was used as the main substrates in synthetic compost as a carbon source, yields were remarkably high. Fermentation was more rapid than that of barley straw or wheat straw and the total nitrogen content was high in rice straw compost. Use of barley straw compost for cultivation of A. bisporus was shown of low yield compared with rice straw, but when a 50% barley straw and 50% rice straw mixture was used, the yield was almost the same as that using only rice straw. The total organic nitrogen on the compost were shown the positive relation to the yield of A. bisporus, but the ammonium nitrogen negative relation to the mycelial growth and yield of A. bisporus. When rice straw was used as the main substrate for compost media, urea was the most suitable source of nitrogen. Poor results were obtained with calcium cyanamide and ammonium sulfate. When urea was applied three separate times, nitrogen loss during composting was decreased and the total nitrogen content of compost was increased. The supplementation of organic nutrient activated compost fermentation and increased yield of A. bisporus. The best sources of organic nutrients selected were as follows: perilla meal, sesame meal, wheat bran and poultry manure, etc. Soybean meal, tobacco powder and glutamic acid fermentation byproducts which were industrial wastes, could be substituted for perilla meal, sesame meal and wheat bran as organic nutrient sources for compost media. During out door composing of rice straw for cultivation of A. bisporus, using of tuner, composter and tunnel system increased up to 13% of its yield, and also cut down 34% of production Cost. The cultivation of P. ostreatus and utilizing of rice straw and wheat straw was established and its yield was high on the rice straw pots. When the substrates 'Rice straw' was heated by steam at $60^{\circ}C$ for 6 hr. mycelial growth of P. ostreatus was moderately rapid and its yield was high.

Detection of Pathogenic Yersinia Enterocolitica in Drinking Water and Vegetables by Mutiplex-PCR (Multiplex-PCR에 의한 먹는샘물 및 야채류로부터의 병원성 Yersinia enterocolitica의 신속검출)

  • The study was conducted to develope a rapid method for the detection of Yersinia enterocolitica in spring water and vegetables via multiplex polymerase chain reaction (PCR) technique using ail, yst, uirF and subgenus-specific Y16S primers. Specificity and sensitivity of multiplex PCR and application of best primers for the detection of Y. enterocolitica from spring water and vegetables were investigeted. Y. enterocolitica ATCC 27729 strains gave 356 bP and 200 bp (Y16S) and 134 bp (yst) bands. but Y. enterocolitica ATCC 9610 and ATCC 23715 strains gave 200 bp and 134 bp bands.In the meanwhile, non-pathogenic Yersinia species, such as Y. frederikseni, Y. inter-media, Y. kristenseni and Y. pseudotuberculosis gave only single 200 bp band, and other bacteria including Escherichia coli O157:H7 ATCC 25392, Shigella dysenteri. Staphylococcu aureus ATCC 25923 and Listeria mo-nocytogenes ATCC 19111 did not show any bands. Among primers, yst and Y16S primer showed the best sensitivity. Seven CFU/mL Y. enterocolitica cells could be detected with yst and Y16S primers and the sensitivity was significantly improved by the further 2nd PCR after 38 cycles of first PCR amplication. Spring water, cabbage and mushroom were inoculated with Y. enterocolitica to determine the sensitivity of multiplex-PCR for the rapid detection of Y. enterocolitica. Multiplex-PCR assay could detect 7 or 70 cells in spring water and vegetables using whole cell lysate with repeating PCR amplication.

Seasonal Variation of Mineral Nutrients in Korean Common Fruits and Vegetables (한국인 상용 과일과 채소의 계절별 무기질 함량 변화)

  • The purpose of this study was to investigate the seasonal variation in the mineral contents of fruits and vegetables available all the year -round. We analyzed Na, K, Mg, Ca, P, Fe Cr, Mn, Cu, Zn, Se, and Al concentrations by ICP-mass in 4 fruits and 17 vegetables items which are frequently consumed in Koreans. For Na content, fall apples showed the highest levels among 4 seasons, but fall tangerines contained the lowest amount of Na among the four seasons. Among the vegetables, the spinach, carrots, and lettuce contained relatively high amounts of Na. The K contents of cucumbers, cabbage, and zucchini were higher in the fall than in the other seasons. For Ca and Fe contents, spring strawberries, fall pears, and fall cabbage had the highest levels. The Cr contents of the apples were higher in the fall and winter than those were in the spring and summer. The strawberries contained their highest amounts of Cr, Mn, and Cu in the spring. The above results showed there are seasonal differences in the mineral contents of fruits and vegetables. Additionally, the fruits and vegetables tended to contain more minerals in the season they were harvested. Therefore, it is recommended to consume those fruits and vegetables during their harvest season.
