Ensemble Projection of Climate Suitability for Alfalfa (Medicago Sativa L.) in Hamkyongbukdo (함경북도 내 미래 알팔파 재배의 기후적합도 앙상블 전망)

  • It would be advantageous to grow legume forage crops in order to increase the productivity and sustainability of sloped croplands in Hamkyongbukdo. In particular, the identification of potential cultivation areas for alfalfa in the given region could aid decision-making on policies and management related to forage crop production in the future. This study aimed to analyze the climate suitability of alfalfa in Hamkyongbukdo under current and future climate conditions using the Fuzzy Union model. The climate suitability predicted by the Fuzzy Union model was compared with the actual alfalfa cultivation area in the northern United States. Climate data obtained from 11 global climate models were used as input data for calculation of climate suitability in the study region to examine the uncertainty of projections under future climate conditions. The area where the climate suitability index was greater than a threshold value (22.6) explained about 44% of the variation in actual alfalfa cultivation areas by state in the northern United States. The climatic suitability of alfalfa was projected to decrease in most areas of Hamkyongbukdo under future climate scenarios. The climatic suitability in Onseong and Gyeongwon County was analyzed to be over 88 in the current climate conditions. However, it was projected to decrease by about 66% in the given areas by the 2090s. Our study illustrated that the impact of climate change on suitable cultivation areas was highly variable when different climate data were used as inputs to the Fuzzy Union model. Still, the ensemble of the climate suitability projections for alfalfa was projected to decrease considerably due to summer depression in Hamkyongbukdo. It would be advantageous to predict suitable cultivation areas by adding soil conditions or to predict the climate suitability of other leguminous crops such as hairy vetch, which merits further studies.

Assessment of Allelopathic Potential of Some Weed Species on Alfalfa(Medicago sativa L.) Germination and Early Seedling Growth (알팔파 발아와 초기생육에 대한 잡초종의 Allelopathic 잠재성 평가)

  • Greenhouse and laboratory studies were conducted to investigate allelopathic potential of some weed species on alfalfa(Medicago sativa L.) germination and seedling growth. In the comparison between top(leaves+stems) and root extracts, top extract exhibited greater allelopathic effects on alfalfa germination than that of root. The various weed species extract differently responded to alfalfa test species, WL-320, in terms of allelopathic effect. Top and root aqueous extracts of lambsquarter(Chenopodium album L.), giant foxtail(Setaria faberii Herrm.), redroot pigweed (Amaranthus retroflexus L.), velvetleaf(Abutilon theophrasti Medic.), crabgrass(Digitaria sanguinalis L.), canada thistle(Cirsium arvense L.) and prostrate knotweed(Polygonium aviculare L.) significantly inhibited germination, seedling length, weight, vigor, and rate of germination of alfalfa. The regression slopes of various top extracts showed that velvetleaf(b=3.69) extracts were the most inhibitory, while large crabgrass(b=2.39) extracts had the least allelopathic effect on alfalfa germination. Germination, seedling length and weight of alfalfa were inversely proportional to the concentration of dried velvetleaf extracts. Also, more of the toxic effects were observed from the dried extracts compared to the fresh extracts. Residue of velvetleaf inhibited significantly alfalfa emergence and survival percentage compared to the control. The emergence and survival percentage of alfalfa were 44%, 57% at 1.0% residue treatment, respectively. When weed residues were mixed with silica sand with incubation time, velvetleaf residue most inhibited alfalfa growth. The degree of inhibition increased as incubation time increased. An incubation for 72h caused the greatest inhibition of alfalfa growth. These results demonstrate the different allelopathic activity of weed species extracts on alfalfa and suggest that weed may affect alfalfa growth and development through the inhibitory effects of allelochemicals present in weed tissue.

Expression of tissue-type plasminogen activator and its derivative proteins in transgenic alfalfa plants (조직형 플라스미노겐 액티베이터와 관련 변이 단백질들을 발현하는 알팔파 형질전환체)

  • Tissue-type plasminogen activator (t-PA) is a thrombolytic agent important in fibirn clot lysis. T-PA causes fibirn-specific plasminogen activation. Six binary vectors harboring t-PA and its derivative genes were cloned and expressed in transgenic alfalfa plants. The insertion of the t-PA and its derivative genes in genomic DNA of alfalfa plants was confirmed by PCR. The presence of the t-PA and its derivative transcripts in total RNAs of the transgenic alfalfa leaves was verified by RT-PCR. ELISA experiments demonstrated that the highest level of recombinant t-PA expression was $75.1{\mu}g$/ total soluble protein (mg) in alfalfa plants. The amount of recombinant t-PA and its derivative proteins in transgenic plants was estimated to range from 9.7 to $39.5{\mu}g$/ total soluble proteins (mg). Western blot analysis of the transformed alfalfa leaves revealed bands of approximately 68-kDa recombinant t-PA and its derivative proteins. The fibrinolysis of recombinant t-PA and its derivative proteins was confirmed by a fibrin plate assay (range from 3.2 to 8.1 cm). The results presented provide information for the development of an additional production of recombinant human proteins having pharmaceutical applications using transgenic plants.

Susceptibility of the Alfalfa Weevil, Hypera postica (Coleoptera: Curculionidae) to Korean Entomopathogenic Nematodes in Laboratory Assays (한국산 곤충병원성 선충에 대한 알팔파바구미의 감수성 실내 검정)

  • Laboratory tests were conducted to determine the susceptibility of the alfalfa weevil, Hypera postica to Korean entomopathogenic nematodes, Steinernema carpocapsae GSN1 strain (ScG), S. glaseri Dongrae strain (SgD), Heterorhabditis bacteriophora Hamyang strain (HbH), and Heterorhabditis sp. Gyeongsan strain (HeG) at the petri dish assay. The larval mortality of H. postica was significantly different depending on nematode species and treatment concentration. SgD and HeG strain were more effective against H. postica larva than ScG and HbH strain. When SgD and HeG strain were treated with the rate of >20 infective juveniles (ijs) per alfalfa weevil larva, mortality was ca. 77.5-100% at the late instars of H. postica in 3 days. The number of established nematode was significantly different depending on nematode species, whereas number of progeny was not significantly different. The mean number of established us of SgD strain in a host was the highest at 80 ijs by 30.2. The highest progeny number of HeG strain was 2,671.5 with 80 ijs. All nematode strains were not parasitic entirely to H. postica adults. These results show that alfalfa weevil late larva is highly susceptible to Korean entomopathogenic nematodes in the laboratory.

Bioassay of Allelopathy Substance Related Injury by Successive Cropping in Alfalfa(Medicago sativa L.) (알팔파 연작장해에 관여하는 타감작용 물질의 탐색 및 생물검정)

  • This experiments were conducted to determine the effect of allelopathy and autotoxicity in alfalfa. Among several alfalfa cultivars, the substances exudated from seven cultivars including Medicago sativa, c.v. Luna, Sparta, Magnum, Husky, Milkmaker, Challenger and Anchor inhibited the germination and seedling growth of radish, ranging from 60 to 80%. When allelopathic substances were exudated from the soil of the alfalfa-cultivated field, the increased levels of substance retarded the germination rate and seedling growth of radish and alfalfa. The inhibition rate was about 80%, compared with that of control. Thus, this study indicates that the allelopathy and autotoxicity substances of alfalfa influenced the germination and seedling growth in alfalfa itself and radish. When allelopathic substances were analysed by using gas chromatograph, several kinds of phenol compounds were detected as follows; Salicylic acid, hydroxybenzoic acid, vanillic acid, syringic acid, coumaric acid, and ferulic acid.

The Integrational Operation Method for the Modeling of the Pan Evaporation and the Alfalfa Reference Evapotranspiration (증발접시 증발량과 알팔파 기준증발산량의 모형화를 위한 통합운영방법)

  • The goal of this research is to develop and apply the integrational operation method (IOM) for the modeling of the monthly pan evaporation (PE) and the alfalfa reference evapotranspiration ($ET_r$). Since the observed data of the alfalfa $ET_r$ using lysimeter have not been measured for a long time in Republic of Korea, Penman-Monteith (PM) method is used to estimate the observed alfalfa $ET_r$. The IOM consists of the application of the stochastic and neural networks models, respectively. The stochastic model is applied to generate the training dataset for the monthly PE and the alfalfa $ET_r$, and the neural networks models are applied to calculate the observed test dataset reasonably. Among the considered six training patterns, 1,000/PARMA(1,1)/GRNNM-GA training pattern can evaluate the suggested climatic variables very well and also construct the reliable data for the monthly PE and the alfalfa $ET_r$. Uncertainty analysis is used to eliminate the climatic variables of input nodes from 1,000/PARMA(1,1)/GRNNM-GA training pattern. The sensitive and insensitive climatic variables are chosen from the uncertainty analysis of the input nodes. Finally, it can be to model the monthly PE and the alfalfa $ET_r$ simultaneously with the least cost and endeavor using the IOM.

Effect of Alfalfa Saponin on the Serum Cholesterol Level in Rats (알팔파 사포닌이 흰쥐 혈청 콜레스테롤 농도에 미치는 영향)

  • Alfalfa saponin was isolated from the roots of alfalfa(Medicage sativa L.) and incoporated at the level of 0.2% and 0.4% cholesterol-free diets and cholesterol enriched-diets. Wistar male rats weighing approximately 80g were fed with for 21 days. The serum lipids and fecal excretion lipids of the rats were determined and the result obtained are as follows. 1. Alfalfa saponin contained 36.4% medicagenic acid. 2. The feeding of alfalfa saponin increased cholesterol excretion but not bile acid excretion. 3. Alfalfa saponin showed hypocholesterolemic activity at 0.4% level in both the diets with and without cholesterol but the effect was not observed at 0.2% level in a diet. 4. It was indicated that the hypocholesterolemic effect of alfalfa saponin was associated with the decreased absorption of cholesterol from alimentary canal.

Mineralization of organic materials and plant uptake in upland condition (밭 토양 조건에서 시비용 유기물질의 무기화와 식물체 흡수)

  • 화학비료를 이용할 수 없는 유기농업에서의 양분관리를 위해서 녹비, 퇴비, 유박 등을 주 원료로 하는 유기질비료 등 다양한 유기물이 농경지에 투입되고 있다. 그러나 다양한 성분 및 탄소/질소비로 이루어진 유기물은 토양 중에서 분해되는 속도가 다르고, 토양수분함량, 통기성, 온도 등과 같은 토양조건에 따라서도 분해속도가 크게 차이가 난다. 본 실험에서는 유기농업에서 시비원으로 자주 이용하는 퇴비, 유박비료, 녹비조건의 알팔파 등을 이용하여 무기화율 및 양분이용률을 화학비료와 비교하였다. 농경지 투입시 유기물은 질소양분양(21kg N/10a)을 동일하게 투입하였으며 화학비료는 기비(전체량의 1/3)만을 투입하였는데 알팔파, 유박비료의 경우 노지조건에서 토양처리 후 1달 내에 가장 높은 무기화율을 보였으며 수분조건이 제한된 무기화통내에서의 무기화도 2개월 내에 대부분 이루어지는 것으로 나타났다. 수딘그라스를 2개월 재배한 결과 화학비료 질소 양분이용율은 70%, 탄질비가 낮은 유박 및 알팔파는 40%내외, 탄질비가 높고 분해가 어려운 가축분 왕겨퇴비는 10%에 불과했다. 즉 화학비료 대비 유기물 양분(질소)의 비효화 율은 알팔파는 60%, 유박비료는 54%, 퇴비는 14% 였다. 또한 화학비료(100%) 대비 인산 이용률(유박: 296%, 알팔파: 660%, 퇴비: 36%로, 인산의 이용율이 높은 것은 유기물로 투입된 인산의 량이 화학비료보다 낮아 상대적으로 유기물질에 의한 작물의 인산이용율이 높고, 화학비료는 토양중 고정화가 작물생육 초기에 일어나는데 비하여 유기물질은 서서히 분해되면서 작물에 흡수되어 인산이용율이 높은 것으로 추정된다. 수단그라스 1차수확 후 포장침수로 인한 생육불량으로 전 생육 과정을 통한 양분흡수율은 측정할 수 없었다.
