The chemical and gelling properties of porphyran from Porphra yezoensis collected from Buan and Wido in Korea at different months of the year were studied. Crude porphyran was prepared by hot-water extraction and further purified by cetylpyridinium chloride precipitation, Crude porphyran and porphyran were modified by alkali treatment to eliminate sulfate. The yields of alkali-modified crude porphyran (AMCP) and porphyran(AMP) were between $4.9\~10.9\%$ and $4.0\~6.7\%$ of the dried algae weight and were maximum in February for Buan and January for Wido, respectively. Gel strength of AMCP were highest in February ($790g/cm^2$) for Buan and January ($740 g/cm^2$) for Wido. Alkali modification increased 3,6-anhydro galactose content and the molar ratio of galactose and 3,6-anhydrogalactose of AMCP and AMP showed 1 : 0,8$\~$1.1. GLC and FT-IR measurement of AMP showed that most of sulfate residues were combined to C-6 of galactose, Thus, results of this study suggest that crude porphyran extracted from Porphra yezoensis produces an agar of a reasonably good quality after alkali treatment.
Domestic seaweeds Gracilaria verrucosa collected from coastal areas of Namhae, Wando and Yeosoo, Korea were subjected to the examination of yield and quality of agar prepared according to various pretreatment conditions. In alkali treatment at high temperature, higher alkali concentration for one-hour period at $90^{\circ}C$ gave rise to higher yield of agar. In acid treatment, higher yield was obtained by higher acid concentration and longer treating period. Alkali treatment at room temperature gave rise to a slightly decreased yield at higher alkali concentration and longer treating time. Total nitrogen and crude ash of agar samples were greatly decreased by pretreatments. Jelly strength, gelation ability, gelation point and viscosity of agar samples tended to increase as the alkali-treating condition of seaweed became stronger. It was shown that sulfur content of agar had a high negative correlation with jelly strength of its gel. Various alkali treatments of seaweed at room temperature showed no marked difference in agar quality and did not exhibit any good effect comparable to alkali treatment at high temperature.
In this study, various influent sludge pre-treatment methods and the internal recirculation of thickened sludge from effluents using a liquid/solid separation unit were adopted to investigate their effects on the sludge digestion and methane production in a combined mesophilic anaerobic and thermophilic aerobic sludge digestion process. A lab-scale combined sludge digestion process was operated during 5 phases using different feed sludge pre-treatment strategies. In phase 1, the feed sludge was pre-treated with a thermal-alkaline method. In contrast, in phases 2, 3 and 4, the internal recirculation of thickened sludge from the effluent and thermal-alkaline, thermal, and alkaline pre-treatment (7 days) were applied to the combined process. In phase 5, the raw sludge without any pre-treatment was used to the combined process. With the feed sludge pre-treatment and internal recirculation, the experimental results indicated that the volatile suspended solid (VSS) removal was drastically increased from phases 1 to 4. Also, the methane production rate with the thermal-alkaline pre-treatment and internal recirculation was significantly improved, showing an increment to 285 mL/L/day in phase 2. Meanwhile, the VSS removal and methane production in phase 5 were greatly decreased when the raw sludge without any pre-treatment was applied to the combined process. Considering all together, it was concluded that the combined process with the thickened sludge recirculation and thermal-alkaline pre-treatment can be successfully employed for the highly efficient sewage sludge reduction and methane gas production.
In a natural fiber-reinforced composite material, many studies have been devoted to improving the interfacial adhesion between natural fiber and polymer matrix and the composite properties through various fiber surface modifications. In the present study, waste wool-reinforced polypropylene matrix composites were fabricated by compression molding and their mechanical and impact properties were characterized. As a result, the tensile and flexural properties and the impact strength of waste wool/polypropylene composites strongly depended on the treatment medium, alkali treatment with sodium hydroxide (NaOH) and silane treatment with 3-glycidylpropylsilane(GPS). The composite with waste wool by silane treatment exhibited higher mechanical properties and impact resistance than that by alkali treatment. The fracture surfaces of the composites support qualitatively the increased properties, showing the improved interfacial bonding between the waste wool and the polypropylene matrix.

To improve osseointegration at the boneto-implant interface, several studies have been carried out to modify titanium surface. Variations in surface texture or microtopography may affect the cellular response to an implant. Osteoblast-like cells attach more readily to a rougher titanium surface, and synthesis of extracellular matrix and subsequent mineralization were found to be enhanced on rough or porous coated titanium. However, regarding the effect of roughened surface by physical and mechanical methods, most studies carried out on the reactions of cells to micrometric topography, little work has been performed on the reaction of cells to nanotopography. The purpose of this study was to examme the response of osteoblast-like cell cultured on blasted surfaces and alkali treated surfaces, and to evaluate the influence of surface texture or submicro-scaled surface topography on the cell attachment, cell proliferation and the gene expression of osteoblastic phenotype using ROS 17/2.8 cell lines. In scanning electron micrographs, the blasted, alkali treated and machined surfaces demonstrated microscopic differences in the surface topography. The specimens of alkali treatment had a submicro-scaled porous sur-face with pore size about 200 nm. The blasted surfaces showed irregularities in morphology with small(<10 ${\mu}m$) depression and indentation among flatter-appearing areas of various sizes. Based on profilometry, the blasted surfaces was significantly rougher than the machined and the alkali treated surfaces (p$TiO_2$) were observed on alkali treated surfaces, whereas not observed on machined and blasted surfaces. The attachment morphology of cells according to time was observed by the scanning electron microscope. After 1 hour incubation, the cells were in the process of adhesion and spreading on the prepared surfaces. After 3 hours, the cells on all prepared surfaces were further spreaded and flattened, however on the blasted and alkali treated surfaces, the cells exhibited slightly irregular shapes and some gaps or spaces were seen. After 24 hours incubation, most cells of the all groups had a flattened and polygonal shape, but the cells were more spreaded on the machined surfaces than the blasted and alkali treated surfaces. The MTT assay indicated the increase on machined, alkali treated and blasted surfaces according to time, and the alkali treated and blasted surfaces showed significantly increased in optical density comparing with machined surfaces at 1 day (p<0.01). Gene expression study showed that mRNA expression level of ${\alpha}\;1(I)$ collagen, alkaline phosphatase and osteopontin of the osteoblast-like cells showed a tendency to be higher on blasted and alkali treated surfaces than on the machined surfaces, although no siginificant difference in the mRNA expression level of ${\alpha}\;1(I)$ collagen, alkaline phosphatase and osteopontin was observed among all groups. In conclusion, we suggest that submicroscaled surfaces on osteoblast-like cell response do not over-ride the one of the surface with micro-scaled topography produced by blasting method, although the microscaled and submicro-scaled surfaces can accelerate osteogenic cell attachment and function compared with the machined surfaces.
This study is to compare characteristics of saccharification reactions applying to enzymatic hydrolysis of pretreated fallen leaves for bio-ethanol production. It experimented pretreatment of acid, alkaline in the chemical. This experiment includes preteatment of acid and alkaline in chemical, soaking, shaking and autoclaving method, which were applied to biomass. In result, the glucose production from alkaline-NaOH method was 263 mg glucose/ g biomass comparing with them of acid-HCl method. Thus, alkaline-NaOH method is superior than the acid-HCl method for chemical preteatment of fallen leaves. Also, when various chemical treatments were compared, they were all. Based on the results of this study, we found that leaves, one of biomass, are possible in pretreatment and enzymatic hydrolysis process, and they are likely to affect bio-ethanol production in the future.

The slow degradation rate of sewage sludge in anaerobic digesters is due to the rate limiting step of sludge hydrolysis. Therefore, the pre-treatment process had been carried out using acidic(pH 1.5, 3, 4, 5) and alkaline(pH9, 10, 13), thermal(50, 100, 150, $200^{\circ}C$) and ultrasonic treatment(400W, 20kHz, 15, 20, 25, 30, 35, 40, 50, 60min). In the best conditions of each treatment, the SCOD ratio(%) of treated/untreared samples were increased 102% in acid(pH5), 986% in alkali(pH13), 959% in thermal($200^{\circ}C$) and 1123% in ultrasonic(35min) treatment. As the result, the ultrasonic treatment was most effective, followed by alkali, thermal, acidic treatment. In the effects of total gas productivity, the thermal($200^{\circ}C$) pretreatment was the highest, followed by thermal($150^{\circ}C$), ultrasonic(90min), alkaline(pH9) and ultrasonic(50min).
This study was conducted to improve circulation system and the storage of garlic against the spoiling microorganisms during the storage. To fulfill this objective, the isolation and identification of spoiling microorganisms from horticultural products were performed at the first step. Antimicrobical activity of electrolyzed alkaline water (EAW) was tested against the spoiling microorganisms and MIC(minimun inhibitory concentration) was determined. As a result, it showed the formidable MIC value at near 2.0%(v/v) against the spoiling microorganisms.
This study was conducted to determine the inactivation effect of electrolyzed water and organic acids either alone or in combination on L. monocytogenes or natural microflora on lettuce. Acidic electrolyzed water completely inactivated L. monocytogenes in broth system within 60 sec, but alkalin electrolyzed water caused approximate 1.7 log CFU/g reduction. However, acidic electrolyzed water reduced only 2.5 log CFU/g of L. monocytogenes on lettuce, and similar antimicrobial effect was observed with alkalin electrolyzed water. In the meantime, acidic and alkaline electrolyzed water caused approximately 2 log CFU/g reduction compared to control, whereas both electrolyzed water combined with $1\%$ organic acids ranged from 2.6 to 3.7 log CFU/g reduction. Among the organic acids, both electrolyzed water combined with $1\%$ citric acid showed the strongest synergistic antimicrobial effect to reduce L. monocytogenes on lettuce as well as total counts, yeast and molds. When antimicrobials, alone or in combination were treated into L. monocytogenes inoculated lettuce at $5^{\circ}C\;and\;15^{\circ}C$ for designed periods, the combined alkalin electrolyzed water with $1\%$ citric acid showed the greatest potential to inhibit growth of the bacteria. According to Scanning Electron Microscopy(SEM), the treatment of electrolyzed alkali water in combination with $1\%$ citric acid highly reduced the growth of the L. monocytogenes compared to single treatment and resulted in causing the destruction of cell membrane.
