Estimation of Physico-Chemical Characteristics of Domestic Aroma Rice and Foreign Aroma Rice (국내 육성 및 도입 향미자원의 이화학적 특성 평가)

  • This study was conducted to provide the fundamental data based on the analysis of phsyco-chemical characteristics of domestic aroma rice and foreign aroma rice for breeding of aroma rice. The average amylose content of domestic aroma rice and widely cultured domestic traditional rice were 18.1% and 19.0% respectively. The Indica and the Japonica types were 25.6% and 25.0%. In the domestic rice of Tongil and Japonica type, the average protein content were 7.6% and 6.4% respectively. And the average protein content of foreign Indica and Japonica type were 7.6% and 7.0% respectively. And the average protein content of widely cultured domestic traditional rice was 5.6%. The Japonica type of rice shown a low protein content compared with Indica type of rice, and the foreign Indica types of rice exhibited a wide range of protein contents. The average alkali digestive value (ADV) of Tongil type of the domestic aroma rice was about 5.0 and the value of Japonica type was about 6.0. The average ADV of foreign Indica and Japonica type were 4.3 and 5.1 respectively. Also the average ADV of widely cultured domestic aroma rice was about 6.0. The foreign aroma rice were distributed a variable range of ADV value and lower than the both domestic aroma rice and widely cultivated domestic rice. In the whole aspect of the amylogram, the highest viscosity, the lowest viscosity and the last viscosity of the total resources, in domestic aroma rice were clearly lower with the next order, Japonica type of foreign aroma rice, Indica type of foreign aroma rice, Japonica type of domestic aroma rice, widely cultured domestic traditional rice, and Tongil type of domestic aroma rice. It shown the difference aspect of amylograms according to the eco-type of the domestic and foreign aroma and the general rice, and it was distinguishable in difference of the aspect of the amylogram of the endosperm of rice as non-glutinous rice, waxy rice, and middle-waxy rice.

  • Volatile flavour components rates from aromatic rices were analyzed by Electronic nose systems. In functional group, polar compounds and aldehyde compounds showed much of volatile flavour components than apolar compounds, sulphur compounds and aminated compounds. The profiles of volatile flavour components rates were markedly differents of sen-sing times, amylose content.

  • Gelatinization of Japonica(30 varieties) and J/Indica(22 varieties) rice flours in 0.2N NaOH was examined. J/Indica rice flours were easily gelatinized and showed higher viscosities at all reference points than Japonica counterparts. Viscosity development rate was not correlated with protein or amylose content. Only J/Indica rice varieties showed positive correlation between hydration rate and viscosity development rate.

  • Interrelationship between. certain physicochemical properties of milled rice was investigated. Grain hardness and milling time were correlated with grain dimension. Equilibrium moisture content of rice upon soaking at room temperature showed a high negative correlation with the amylose content. The contents of protein, fat and ash had no relationship with hardness, milling time and equilibrium moisture content.

Analysis of Grain Quality Properties in Korea-bred Japonica Rice Cultivars (우리나라 자포니카 벼 품종의 식미관련 미질특성 분석)

  • This study was conducted to make clustering analysis based on major physicochemical characteristics related to palatability of cooked rice. 89 Korea-bred japonica rice cultivars could be largely classified into two groups, that is, Dongjinbyeo and Ilpumbyeo groups. The Ilpumbyeo group was divided into two subgroups; Ilpumbyeo and Chucheongbyeo groups. The two major rice groups showed significant difference in viscogram properties of rice flour. Ilpumbyeo group revealed slightly higher estimates of viscogram traits as compared with Dongiinbyeo group in average. Early-maturing rice group showed slighly lower estimates of taste meter and higher protein content compared with medium or medium late maturing ones. Also, early and medium-maturing groups exhibited slightly higher estimates of peak, hot and breakdown viscosities but lower estimates of consistenency and setback viscosities compared with medium-late-maturing one. The rice cultivars developed in 2000's revealed slightly higher estimates of peak, hot, cool and consistency viscosities compared with those in $1980's{\sim}1990's$. The grain quality properties significantly associated with the esimates of Toyo taste meter were protein and amylose contents and hot viscosity. The lower protein content and hot viscosity and the higher amylose content, the higher estimates of the taster meter. The protein content was highly negatively correlated with amylose content of milled rice. The important quality components contributed to multiple regression formula for estimating the Toyo taster meter values were protein content, alkali digestion value, and hot viscosity. The fittness of this formula was about 49% along with the coefficients of determination.

Enzymatic Characterization of a Thermostable 4-α-Glucanotransferase from Thermotoga neapolitana (Thermotoga neapolitana 유래 내열성 4-알파-글루칸전이효소의 효소적 특성)

  • The gene encoding 4-$\alpha$-glucanotransferase (mgtA) from Thermotoga neapolitana was cloned and expressed in Escherichia coli in order to investigate whether this enzyme was capable of producing cycloamylose for industrial applications. MgtA was purified to homogeneity by HiTrap Q HP and Sephacryl S-200 HR column chromatographies. The size of the enzyme as determined by SDS-PAGE was about 52 kDa, which was in good agreement with its deduced molecular mass of 51.9 kDa. The optimal temperature and pH for the activity of the 4-$\alpha$-glucanotransferase was found to be $85^{\circ}C$ and 6.5, respectively. The enzyme hydrolyzed the 1,4-$\alpha$-glucosidic bonds in oligomeric 1,4-$\alpha$-glucans and transferred oligosaccharides (maltotriose being the shortest one) to acceptor maltodextrins. However, the enzymes had no activity against pullulan, glycogen, and other di- or trioligosaccharides with rare types of $\alpha$-bond. MgtA is distinguished from 4-$\alpha$-glucanotransferase from Thermotoga maritima in that it can convert maltotriose into maltooligosaccharides. The treatment of glucoamylase after the reaction of MgtA with maltotriose, maltotetraose, maltopentaose, or maltohexaose as sole substrate revealed that MgtA yielded linear maltooligosaccharides instead of cycloamylose.

Variability of Quality Related Characters in the Recombinant Inbred Lines from Milyang 23 and Gihobyeo (밀양23호와 기호벼 교잡 재조합자식계통(RILs)의 품질관련 특성 변이)

  • The rice quality related traits including physico-chemical traits were evaluated with one hundred sixty-four MG RILs derived from the cross between Milyang 23, Indica/Japonica hybrid type, and Gihobyeo, Japonica type. The variation distribution of all traits examined approximately fit normal distribution and transgressive segregants over parents were observed for all traits. The occurrence of such transgression could be associated with the interactions of complementary QTL alleles from two parents. However in this study, it could not be concluded that our results of segregation were due to either complementary gene effects or overdominance of a major gene. These factors should be verified by further studies. Correlations between traits were evaluated by regressing phenotypic values of one trait on those of another traits. There are highly significant correlation between grain thickness with grain width, white core and white belly. But between white belly and alkali digestion value showed highly negative significant correlation. Contents of protein showed highly negative correlation with amylose and Mg/K ratio but showed highly correlation with K and Fat contents. Hardness of cooked rice showed highly correlation with adhesiveness, elasticity, gumminess, chewiness.

  • The effect of N-fertilization on yield, milling characteristics and quality of Samcheonbyeo, an early maturing rice variety, was investigated in 2002 to 2004. The study was carried out in the southern alpine area of Un-bong Sub-Station, Honam Agricultural Research Institute. Higher nitrogen levels increased number of panicle and grain per unit area, but lowed ripening rate and decreased 1,000-grain weight. Higher nitrogen levels of up to 90 kg/ha also increased head rice yield. A nitrogen level of 90 kg/ha was found optimal for increasing ripening rate and head rice yield, and rice palatability.

Physicochemical Properties of Domestic Millet Starches (국내산 조전분의 이화학적 특성)

  • Physicochemical properties of domestic millet starches were determined. Amylose contents of nonwaxy and waxy millet starches were 28.0 and 8.0%, respectively. The water binding capacity of nonwaxy millet starch was slightly higher than that of waxy millet starch. There was no recognizable difference on granule size between nonwaxy and waxy millet starch. Abrupt increases in swelling power over $80^{\circ}C$ of gelatinization temperature were characteristic features of millet starches. The initiation of increase in light penetration was started slightly earlier in case of waxy millet starch. Maximum viscosities of $6{\sim}7%$ nonwaxy and waxy millet starch were 300-460 and 800-1080 B.U., respectively. The conspicuous break-down was noticed in waxy millet starch.

Effect of Cold Water Irrigation on the Growth, Yield and Grain Quality in rice (냉수관개가 벼 생육, 수량 및 미질에 미치는 영향)

  • This experiment was carried out at Chuncheon substation, Crop Experiment Station to investigate the effect of cold water irrigation on the growth and grain yield of rice in 1990. Irrigation of cold water (17$^{\circ}C$) delayed 10 to 19 days of heading, and reduced about 17 to 31% of plant height in terms of culm and panicle length. Number of panicle per plant was decreased and / or increased by cold water irrigation. Cold water irrigation reduced spikelet number per panicle and percent of filled grain. Grain yield was increased but, straw weight was decreased according to water temperature gradient from inlet to outlet. There was varietal differences in head rice recovery by cold water treatment. In general the dark brown and / or black colored kernel increased at cold water inlet but decreased percent of green kernel. Alkali digestible value increased and amylose content decreased at cold water inlet.
