Plant Regeneration from Leaf and Internode Segment Cultures of Boxthorn (Lycium chinense Mill.) (구기자나무의 잎과 마디절편체 배양에 의한 식물체 재생)

  • Callus and shoot formation from medicinal crop, Lycium chinense Mill. cv. 'Cheongyang', as influenced by various media, explant sources and plant growth regulators were investigated. The rate of shoots formation, number of shoots, and fresh weight of shoots were the best on MS medium followed by B$_{5}$, WPH, and SH. Callus induction was more effective in leaf than internode segments, and was the best on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA. Effects of plant growth regulators in shoot formation were more effective in BA than TDA combined with NAA. Shoot formation from callus induced in leaf and internode segments was the best on MS medium containing 0.01 mg/L NAA with 0.2 mg/L BA.

High Frequency Plant Regeneration from the Cultures of Cotyledon Explants of Perilla (Perilla frutescens L.) (들깨의 자엽절편배양을 통한 고효율 식물체 재분화)

  • A reliable and effective tissue culture system was established for Perilla frutescens (perilla) using different cultivar, explant source, and growth regulator composition in medium. MS medium supplemented with 2.0 mg/L BA and 0.1 mg/L NAA was maximum at shoot induction. Cotyledon explants formed more shoots than hypocotyl explants. The frequency of plant regeneration through organogenesis was higher (22.8%) than that (6.1%) of somatic embryogenesis. Five genotypes of perilla were screened for the feasibility of shoot regeneration, cotyledon explant of 'Manbaek' showed the highest shoot induction at a frequency of 27.3% among the tested cultivars.

Rooting and Acclimatization of Shoots Harvested from Bioreactor Culture in Rehmania glutinosa (생체반응기에서 수확한 지황 신초의 발근과 순화)

  • This experiment was carried out to know the effect of media and agar concentrations, aeration and growth regulators on rooting and acclimatization of the shoots harvested from bioreactor culture in Rehmannia glutinosa. Half MS media with 1.2% agar improved rooting and acclimatization of shoots. Shoots were effectively acclimatized and rooted well in case of aeration by using membrane filtered vessels. Shoots acclimatized in vessel with membrane Inter were healthier and had higher ex vitro survival rate than those without membrane Inter on plug tray. Addition of paclobutrazol 0.3-0.4 mg/L, to acclimatization media enhanced shoots growth and root development.

  • The most effective cytokinin for shoot multiplication in vitro of Prunus persica cv. Baekmijosaeng, Okubo, and Yumyeong was 2.0 mg/L BA. As the result of combinational treatment of BA and auxin sources (IAA, IBA and NAA), 2.0 mg/L BA with 1.0 mg/L IAA was the most effective for shoot multiplication of cv. Baekmijosaeng. The most effective auxin source for rooting was IAA and the concentration was 5.0 mg/L and 3.0 mg/L for cv. Baekmijosaeng and Okubo, respectively.

  • To establish the mass proliferation system of Ardisia pusilla DC, the shoot tips of Ardisia pusilla DC were cultured on the MS and half-strength MS medium supplemented with $0{\sim}5.0$ mg/L BA or $0{\sim}0.5$ mg/L thidiazuron(TDZ), respectively. A few multiple shoot formation observed when the shoots were cultured on MS medium containing TDZ. However, the frequency of multiple shoot formation was reached up to 82.4%, when the shoots were cultured on half-strength MS medium supplemented with 0.5 mg/L BA. Also the number of shoot per explant was 7.1. To promote rooting from multiple shoot, newly formed shoots were transferred to half-strength MS medium containing 0.5 mg/L IBA or 0.5 mg/L NAA, respectively. Regenerated plantlets were grown to normal mature plants in soil.

  • 복주머니난(Cypripedium)속 식물은 우리나라에 광릉요강꽃을 비롯하여 털복주머니난, 흰털복주머니란, 복주머니란, 노랑복주머니란등 5종이 분포하는 것으로 알려져 있다. 광릉요강꽃과 털복주머니란 두 종은 환경부에서 지정한 멸종위기 식물 1급과 2급에 지정되어 보호를 받고 있고 나머지 3종은 제도적으로도 보호를 받지 못하는 실정이다. 본 연구에서는 충분히 성숙한 털복주머니난과 복주머니난의 종자에 NaOCl처리를 하여 발아율을 향상시킬수 있었는데 이러한 전처리가 발아에 미치는 원인에 대한 연구를 실시하였다. 털복주머니란의 무균적인 종자발아는 1.0% NaOCl 처리구에서 70% 이상의 종자발아율을 보였으며, POM배지가 MS배지보다 신초분화가 양호했다. GA3와 활성탄(Activated charcoal)의 혼합첨가는 신초증식에 효과적이었다. 신초분화 후 저온처리는 신초의 증식 율을 증가시켰다. 한편 NaOCl 농도(0, 1, 3, 5, 10%)와 NaOCl 처리시간(0, 5, 15, 30, 45, 60분)에 따라서 복주머니란의 종자발아를 확인한 결과, NaOCl 1%를 30분간 처리하였을 때 가장 높은 발아율을 나타냈다. NaOCl을 처리하여 종자의 종피상태를 SEM과 TEM으로 관찰한 결과 NaOCl의 처리는 종피 세포벽의 부분적 해리 및 작은 구멍을 만들게 하였는데 이러한 종피의 물리화학적 변화가 물과 양분의 이동을 원활히 하여 종자의 발아를 촉진하는 것으로 사료된다. 복주머니난의 신초분화에 미치는 casein과 활성탄의 효과를 알아본 결과 casein 200 mg/L와 활성탄 200 mg/L를 혼합 첨가한 실험구에서 가장 높은 신초분화율을 보였다. 토양순화 후 생존률은 극히 저조했으며 30 개체중에 5 개체만이 다음해 어린동아를 싹틔우는 것을 확인 하였다. 본 결과들을 종합하여 보면 멸종위기식물, 특히 털복주머니란과 복주머니란의 조직배양을 통해서 서식지외 보존방안(기내증식)에 관해 가능성을 제시하였다고 생각되어 진다.

Effects of BA Treatment and Cut Planting in Root Cuttings of Lacquer Tree (Rhus verniciflua Stokes) (옻나무 근삽시 BA 처리방법과 삽식법의 효과)

  • Effects of BA treatment and cut planting methods on shooting and seedling characteristics were investigated to increase seedling products by cuttings of lacquer tree. Dipping on 1,000mg/L BA solution was better than dripping with same solution at 5 weeks after cut planting. As shooting rate was increased suddenly, it was 85% at 10 weeks after cut planting. Among 3 dripping treatments, interval was not effective on shooting rate. Dripping per 2 days with 100 ㎕ of 1,000 mg/L BA solution was most effective on seedling height and leaf number and than dipping was more effective than dripping per 5 and 7 days. Branch developed on near part from dripped scion. Shooting of horizontal planting was earlier about a week and rate was increased suddenly from 3 to 6 weeks, it was highest with 96% than others at 10 weeks after planting. Seedling of horizontal cut planting was tallest about 40cm and branch developed lower than vertical and oblique cut planting. In case of horizontal cut planting, 2-3 shoots were formed on different part of scion and than those were could divided to seedling, so horizontal cut planting was most effective to propagation of seedling than any others.

In Vitro Shoot Tip Culture of Pear 'Niitaka' as Related to Tree Vigor Sampling Time and Plant Growth Regulators (배 '신고'의 경정배양에 있어서 모수의 수세, 경정의 채취시기 및 생장조절제의 영향)

  • In vitro shoot tip culture technique was established in pear (Pyrus pyrifolia 'Niitaka') as related to tree vigor, sampling time, and plant growth regulators and sucrose supplemented to medium. Shoot tips excised in June from the tree having medium-vigor developed good shoots. BA (1.0 and 2.0 mg/L) without NAA produced shoots suitable for proliferation, and NAA supplemented to medium resulted in poor shoot growth and excessive callus formation. BA of 2.0 mg/L combined with 0.01 mg/L NAA provided shoots suitable for rooting and sucrose of 30 g/L was recommended for proliferation medium. A fourth strength MS medium supplemented with 0.1 mg/L NAA produced plantlets in good quality of root number and root length.

Effect of in vitro Culture Condition and Lines on Growth Pattern of Lateral Bud from Nodal Cutting of Phalaenopsis Flower Stalk (팔레높시스 기내 화경 배양조건 및 계통이 액아의 발육형태에 미치는 영향)

  • This study was carried out to investigate the effects of in vitro culture condition and among lines on growth pattern of lateral buds from nodal cuttings of Phalaenopsis flower stalks. The ratio of bud growing into shoot from nodal cuttings of flower stalks were 90.9% and 54.4% on MS and hyponex medium, respectively. The number of buds grown vegetatively were increased remarkably on the MS medium containing 5 mg/L BA. The rate of buds grown vegetatively was higher in basal and middle parts than in upper part of flower stalks. The flower stalk sections cultured at 25~28$^{\circ}C$ showed the highest ratio of vegetative growth. Medium contamination was decreased by pretreatment of etiolation to the flower stalk. However, the pretreatment did not show specific effects on shoot development and reduction of phenolic compound. Average shoot number which was formed from flower stalk segments in 27 of 30 accessions were 3.17, while high number of shoots were obtained from Phal. 3020 and Phal. 3039. The growth pattern of lateral buds in F$_1$hybrids was similar to that of their parents.

Effect of silver nitrate(AgNO3) and polyamines on shoot organogenesis and plant regeneration of Lycopersicon esculentum cultivar, Micro-Tom (질산은과 polyamines이 미니토마토, Micro-Tom 신초 기관발생과 식물체 재분화에 미치는 영향)

  • The study was carried out to establish an efficient protocol for shoot organogenesis and plant regeneration from stem explant cultures of Lycopersicon esculentum cv. Micro-Tom. The regenerated shoots obtained from stem explant cultures on solid MS medium containing the different concentrations of BAP. The highest number of shoots (5.3) per explant and shoot growth (0.7 cm) was obtained on MS medium containing 4.0 mg/L BA. The additions of AgNO$_3$ and putrescine substantially improved the shoot regeneration frequency, at the optimal concentration of 7 mg/L and 50 mg/L respectively. The regenerated shoots (about 1 cm) were normal and could be easily rooted with 0.1 mg/L IBA treatment. The rooted plants were hardened and transferred to vermiculite with a 92% survival rate where they grew normally.
