Four species(25%) of Viperidae(Agkistrodon brevicaudus, Agkistrodon ussuriensis, Agkistrodon saxatilis) and Cloubridae(Rhabdophis tigrenus tigrenus) were Korean poisonous snake. Copulation season of these species was from July to August. Reproduction mode of genus Agkistrodon species was ovoviviparous but Rhabdophis tigrinus tigrinus was the other pattern of oviparous. Optimal movement temperature range was from 20$\circ $C to 29$\circ $C(March~September). Wjen atmosphere temperature was below 10$\circ $C, at that time they hibernate at the ground, rock bottom, stone wall and embankment around the end of a field. The venom of these snakes consist mainly Hematoxin, Cytolysin, Neurotoxin and Cardiotoxin of poisonous liquids. These material injection to animal cause systemic syndrome such as Dizziness(25.7%), Vomitting(23.1%), Fever(22%), Visual trouble(18%), Headace(17.7%), Dyspnoea(17.6%) and bring about other local syndrome such as Discoloration(54.2%), Bleeding(20.2%), Bullae(10.7%) and Skin ulcer(!0.8%). The annual distribution was appeared to decrease 1972 after 1992 and average snakebiting patients was 25.6 per year, but practically total estimated snakebiting was 2,700 per year. The seasonal distribution was most frequent in August(25%), and mortality was 1.8%(26 per 1,430). The sex ratio was 2:1 and according to age distribution, it was most prevalent at one's fifties(19%). The most frequent place where the accident happened was the field(48.2%) and most predilection site of the body for victim were hand(47.8%) and foot(39.5%), Commonly bite snake were Agkistrodon ussuriensis(27.1%), Agkistrodon brevicaudus(22.6%) and Agkistrodon saxatilis(9.6%) but 40.7% of species could not be identified. Treatment of antivenin patient was 75.9% (1,068/1,407).

Polychlorinated biphenyls(PCBs) are large scale industrial chemicals which are using in diverse applications. The goal of this study was to determine if exposure to 2,2',5,5'-tetrachlorobiphenyl (PCB 52) leads to an increase in the production of active oxidants, and subsequently promotes apoptosis of neuronal SK-N-MC cells. Reactive oxygen species (ROS) formation was examined in SK-N-MC cells after treatment of PCB 52 by concentrations and incubation times, respectively. It showed that the rate of ROS production in the cells was increased in a does-dependent manner to 45 min, followed by a return towards control levels after 120 min treatment. We also examined the association of PCB-induced apoptosis with the modulation of biomakers of oxidative damage to lipids (malondialdehyde [MDA]) in SK-N-MC cells. Increased MDA was observed in a dose-dependent manner in groups treated with 10, 15, and 20 figJ me of PCB 52 for 24 h. After treatment of PCB 52, the cells did not show any significant change in the rate of Cu/Zn-superoxide dismutase (Cu/Zn-SOD) activity. Whereas, the cells had a two-fold greater rate of change in catalase activity at 20 ㎍/㎖ of PCB 52 for 24 h when compared to control group. Korean Ginseng is one of the most important crude drugs which has been used as a traditional Oriental medicine. We next investigated protective effect of extracts of ginseng on cytotoxicity induced by PCB 52 in SK-N-MC cells. Pretreatment of SK-N-MC cells with 25-200 μg/ml of ginseng were reduced cell death in a dose-dependent manner in PCB 52-treated cells. To examine the sensitivity of beta-catenin to ginseng, the protective effect of a range of ginseng concentrations was examined in SK-N-MC cells treated with PCB 52. The result demonstrated that ginseng efficiently blocked PCB 52 inducible beta-catenin proteolysis in a concentration dependent manner. The ROS formation was also measured in the presences of extract of ginseng and superoxide dismutase (inhibitor of oxygen free radical production). The both SOD (400 U/ml) and ginseng (200 μg/ml) significantly inhibited RDS generation in PCB 52-treated group.
분자단계에서 천연소재 유래 다당체는 면역 세포 표면(예, 대식세포)에 있는 한가지 또는 여러가지 수용체(${\beta}$-glucan 수용체, mannose 수용체, complement 수용체 3, Toll-like 수용체, scavenger 수용체)와 면역 반응을 시작하기 위하여 결합할 수 있고, 그 후에 신호전달을 위하여 cytokine과 second messenger($Ca^{2+}$, cAMP, cGMP, NO 등등)들을 생성한다. 그 결과로 선천면역과 적응면역을 유도하여 다양한 질환에 영향을 미친다. 추가적으로 다당체들의 면역조절은 신경내분비 체계의 조절과도 연관되어 있다. 최근 수많은 천연소재유래 다당체의 생리활성에 관한 연구가 진행되고 있다. 다양한 연구를 통해 정확한 약효, 낮은 독성과 부작용을 가진 일단의 다당체가 발견되어 현재 임상에서 암 치료에도 사용되고 있다. 이러한 사실들은 면역증진 다당체들이 항 종양치료를 위한 새로운 치료제로서 큰 시장을 개척할 수 있다는 가능성을 보여주고 있다. 이러한 사실에도 불구하고 현재 다당체에 대한 연구는 다음과 같은 취약점을 내포하고 있다. 첫째 다당체의 생리활성에 대한 정해진 비교기준이 없다. 특별히 임상 환경에서는 이러한 연구들이 중국, 일본, 한국과 일부 다른 동양국가들에 국한되어 있어 유럽이나 미국에서의 연구 자료가 결여되어있다. 둘째 현재 연구에 사용되고 있는 대다수의 천연소재 유래 다당체는 순수한 다당체가 아닌 천연 그대로의 것을 사용하고 있어 소재로부터 유래하는 다른 활성성분에 의해 쉽게 영향을 받을 수 있어 순수 다당체가 면역체계에 어떠한 역할을 수행하는지 진단하는데 어려움이 있다. 셋째 면역조절 경로에서 다당체의 정확한 신호전달 경로가 아직 정확하게 밝혀지지 않았고, 또한 동물과 임상실험에서 다당체가 작용하는 수용체 및 신호전달 경로에 대한 상반되는 결과가 발표되고 있기도 하다. 따라서 다당체의 면역조절활성에 대한 연구에 있어 다음과 같은 점들에 주목해야 할 필요가 있다. 첫째 다당체의 구조와 활성간의 상관관계를 밝혀야 할 것이다. 특히 다당체의 세부 구조 및 활성 부위를 밝혀낸다면 분자수준의 면역조절 기작을 명확히 하는데 도움을 줄 수 있을 것이다. 이러한 정보를 확보할 수 있다면 다당체의 구조를 변형시켜 활성을 증진시킬 수 있을 것이며, 또한 활성이 증가된 다당체를 합성하는 일도 가능해 질 것으로 예상된다. 마지막으로 ${\beta}$-glucan과 lentinan과 같은 정제된 다당체의 연구를 지속적으로 수행하여 효과적인 동물실험 및 임상실험 protocol을 확보하고 연구결과들에 대한 database를 구축해야 할 필요가 있다. 이러한 노력들이 성공적으로 수행된다면 천연소재유래 다당체들이 가까운 장래에 암 치료를 포함한 다양한 질병에 적용 가능한 새로운 면역조절제로 사용될 수 있을 것이다.

Died Gardenia jasminoides fruit is used as a dye in the food and clothes industries in Asia. The present study investigated the anti-inflammatory effects of aqueous extract of G. jasminoides fruits (GJ) in BV-2 microglial cells. GJ inhibited lipopolysaccharide-induced nitric oxide (NO) secretion, inducible nitric oxide synthase (iNOS) expression, and reactive oxygen species production, without affecting cell viability. Furthermore, GJ increased the expression of heme oxygenase-1 (HO-1) in a dose-dependent manner. Moreover, the inhibitory effect of GJ on iNOS expression was abrogated by small interfering RNA-mediated knock-down of HO-1. In addition, GJ induced nuclear translocation of nuclear factor E2-related factor 2 (Nrf2), a transcription factor that regulates HO-1 expression. GJ-mediated expression of HO-1 was suppressed by LY294002, a phosphoinositide 3-kinase (PI-3K) inhibitor, and SB203580, a p38 kinase inhibitor, but not by the extracellular signal-regulated kinase (ERK) inhibitor PD98059 or c-Jun N-terminal kinase (JNK) inhibitor SP600125. GJ also enhanced the phosphorylation of Akt and p38. These results suggest that GJ suppresses the production of NO, a pro-inflammatory mediator, by inducing HO-1 expression via PI-3K/Akt/p38 signaling. These findings illustrate a novel molecular mechanism by which extract from G. jasminoides fruits inhibits neuroinflammation.

The antioxidant activity of commercial sweet persimmon wine concentrate (SPWC) was evaluated by determining the total phenol content (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, reducing power (RP), and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities. TPC in the SPWC was $9.29{\pm}0.11\;mg$ gallic acid equivalents (GAE)/g, which corresponds to 31.59 mg GAE/100 mL of the wine. The IC50 for the DPPH radical scavenging activity, RP, and ABTS radical scavenging activity of SPWC were 2.96, 1.44, and 0.48 mg/mL, respectively. The neuroprotective effect of SPWC against glutamate-induced neurotoxicity in N18-RE-105 cells was investigated. Treatment of N18-RE-105 cells with various SPWC concentrations under glutamate resulted in the induction of a protective effect in a dose-dependent manner, as determined by the MTT reduction assay. These results suggest that SPWC exhibits considerable antioxidant and neuroprotective activity.
Objectives : The amyloid $\beta$-protein ($A\beta$) is the principal component of the senile plaques characteristic of Alzheimer's disease (AD) and elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors including antioxidants and proteoglycans modify $A{\beta}toxicity$. In this study, we have investigated the protective effects of water- and organic solvent-extracts of Hemerocallis fulva root fractions pre-extracted with methanol on $A\beta$-induced oxidative cell death in cultured rat pheochromocytoma (PC12) cells. Methods : For this study, we used MTT reduction assay for detection of protective effects of water- and organic solvent-extracts of Hemerocallis fulva root fractions pre-extracted with methanol on $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells. We also used cell-based $\beta$-secretase assay system to investigate the inhibitory effect of water- and organic solvent-extracts of Hemerocallis fulva root on $\beta$-secretase activity. Results : We previously reported that methanol extracts of Hemerocallis fulva root strongly attenuated cytotoxicity induced by the three $A\beta$ fragments ($A{\beta}_{25-35},\;A{\beta}_{1-42}\;A{\beta}_{1-43}$) to both SK-N-MC and PC12 cells. In the present study, we found that butanol-, ethylacetate-, chloroform-, and water-extracts of Hemerocallis fulva root fractions pre-extracted with methanol had strong protective effects against $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells and inhibitory potency to $\beta$-secretase activity. Conclusion : These results suggest that butanol-, ethylacetate-, chloroform-, and water-extracts of Hemerocallis fulva root fractions pre-extracted with methanol may contain the protective component(s) against $A\beta$-induced cell death in PC12 cells as well as inhibitory component(s) to $\beta$-secretase activity.

Objective : Growth factor receptors on the tumor cells are known to be expressed highly allowing the tumor cells to bind growth factors to stimulate cellular division. Immunotoxin therapy is one of the novel approaches to the primary malignant brain tumor, and expression of cell-surface receptor is essential for the immunotoxin to have specific anti-tumor activity. Despite promising cytotoxic activity of immunotoxin, tumor responses are not curative on clinical trials, and additional studies are needed regarding various factors influencing the efficacy of the immunotoxin. The purpose of this study is to detect the expression of various growth factor receptors on brain tumor cell lines which are going to be used in these studies. Materials and Methods : The authors detected transferrin receptor(TR), insulin-like growth factor-1 receptor(IGF-1R), and interleukin-4 receptor(IL-4R) on medulloblastoma cell line(Daoy) and glioblastoma cell lines(U373 MG and T98 G) by flow cytometric analysis. Results : TR was expressed on Daoy, U373 MG, and T98 G. IGF-1R was expressed on Daoy and U373 MG, but not on T98 G. IL-4R was expressed on all cell lines tested. Conclusion : The transferrin and interleukin-4 receptors might be good targets for immunotoxin therapy. The results should be considered in additional in vitro and in vivo studies regarding immunotoxin and in establishing the proper treatment model of the immunotoxin therapy including selection of the adequate immunotoxin.
Damage assessment based on in situ surveys for oil spills in marine environment is limited by fundamental difficulties as well as tremendous expenses. Except for intertidal zones, the damage is not preserved well. Also such surveys are usually confined to adult organisms. To overcome these limitations a computer model, NRDAM/CME, was developed in the case of USA (Reed et al., 1989), where an acute toxicity data base was used to assess indirect damages through food webs and loss due to recruitment as well as adult losses. In the present study damage assessment of natural biological resources for hypothetical oil spills is attempted using a computer model for hypothetical spills of Bunker C and heavy crude oil. In the model, the logical structure of NRDAM/CME was adopted, and biomass and productivity database were compiled for the Korean waters. The results showed that the damage increased in a nonlinear fashion as the spill amount increased. The magnitude of the damage depended upon the chemical properties of oil viscosity and solubility in particular, which implies that usage of oil dispersant might increase the damage by dispersing oil. The results also indicate that long term damage due to recruitment loss could be greater than short term damage.
Subcutaneous pannicultis-like T cell lymphoma is a rare cutaneous T cell lymphoma. It presents with multiple subcutaneous nodules or plaques involving the extremities or trunk, and with constitutional symptoms that include fever, malaise, fatigue, myalgia, chills and weight loss. Histologically, the lesions of this disease are reminiscent of panniculitis and are composed of a mixture of small and large atypical lymphoid cells infiltrating between adipocytes. The optimal treatment for this disease is undefined and prognosis of this disease is poor, even when treated with multiagent chemotherapy regimens considered optimal for agressive lymphoma of other types. Poor prognosis factors include clinical features such as anemia, leukocytopenia, hepatosplenomegaly, lymphadenopathy and coagulopathy, which are suggestive of hemophagocytosis. Much of the mortality of this disease is due not to disseminated lymphoma with organ failure, but rather to complications of the cytopenias associated with the hemophagocytic syndrome. We report a case of subcutaneous panniculitis-like T cell lymphoma in a 12 year-old boy who presented with initial complaints of fever and multiple subcutaneous nodules, and briefly review the related literature.
A randomized, double blind, placebo-controlled human intervention study involving 62 healthy volunteers was performed to investigate the effects of Keumsa sangwhang (Phellinus linteus) mushroom extracts (KPLE) on natural killer (NK) cell activity in peripheral blood. The volunteers were randomly distributed into two groups, one receiving KPLE (3.3 g/day) and the other a placebo by oral administration for 8 weeks. In this study, the number of NK cells did not increase with KPLE administration, however the cytotoxic activity of NK cells against the Jurkat leukemia cell line increased significantly. This result suggests that administration of KPLE induces cell-mediated immunity by increasing NK cell activity in humans.
