Fluoranthene, a common polycyclicaromatic hydrocarbon (PAH), exhibits phototoxicity which may affect aquatic organisms. The eventual goal of this study is to develop a biomarker of Japanese medaka (Oryzias latipes) used in monitoring hazardous chemicals in the ecosystem. In this study we investigated neural toxicity of fluoranthene in Japanese medaka (Oryzias latipes) along with comparative analysis of corresponding behavioral response. The untreated individuals shooed normal behavioral characteristics (i. e., smooth and linear movements). The treated fish, however, showed stopping and abrupt change of orientation (100 ppb), and severely reduced locomotive activity and enhanced surfacing activity (1,000 ppb). Treatment of the medaka fish with fluoranthene caused a significant suppresson of acetycholine esterase (AChE) activities in the body portion but not in the head portion. When fish were exposed to 1,000 ppb of fluoranthene for 24 hr, the body AChE activities decreased from 126.${\pm}$31.89 (nmoles substrate hydrolyzed per min per mg protein) to 49.51${\pm}$11.99. Expressions of tyrosine hydroxylase (TH) protein in the different organs from both head and body portions were comparatively analyzed using an immunohistochemical technique. Five organs of the medaka fish showing a strong TH protein expression were the olfactory bulb, hypothalamus, optic lobe, pons and myelencephalon regions. This study provides molecular and neurobehavioral bases of a biomonitoring system for toxic chemicals using fish as a model organism.
Methamphetamine (MA) is a major drug of abuse in Korea. Currently preliminary evidence suggests that MA dependence may cause long-term neural damage in human. Repeated exposure to psychostimulants such as methamphetamine results in behavioral sensitization, a paradigm thought to be relevant to drug craving and addiction in human. Sensitization alters neural circuitry involved in normal processes of incentrive, motivation, and reward. However the precise mechanism of this behavioral sensitization has not yet been fully elucidated. Repeated use of high dose MA causes neurotoxicity which is characterized by a long-lasting depletion of striatal dopamine (DA) and tyrosin hydroxylase activity of DA, DA-transporter binding sites in the striatum. The loss of DA transporters correlates with memory problems and lack of motor coordination. DA fuels motivation and pleasure, but it' s also crucial for learning and movement. This selective review provides a summary of studies that assess the neurobiological mechanisms of MA.
Purpose: Delayed neuropsychological sequelae (DNS) commonly occurs after recovery from acute carbon monoxide poisoning. The aim of this article is to identify the factors associated with DNS development. Methods: We retrospectively evaluated patients, admitted to the medical center emergency department from June 2005 to March 2011, who were suffering from acute carbon monoxide (CO) poisoning. We categorized the patients into two groups - those with DNS, and those without DNS. Multiple regression analysis was performed to identify the factors related to manifestation of DNS. Results: Of the total one hundred fifty seven patients (157) recruited for the study, twenty two (22) developed DNS. Longer CO exposure times and lower GCS scores were positively associated with development of DNS symptoms. Conclusion: Our study identified two potential factors which are predictive of DNS development in CO intoxication, however, more studies are needed. Adequate follow-up after hospital discharge to monitor for and accurately identify manifestation of DNS, is also important.
Toluene inhalation increases glutamate level and its receptor in various brain regions. In this study, nitric oxide synthase (NOS) activities were investigated in various rat brain regions using NADPH diaphorase staining method which examined histochemical changes of NOS in the neural cells. Also, in vitro LDH leakage assay and MTT test were performed to investigate the toxic influences of toluene in cultured granule cell of rat cerebellum which was significantly affected with toluene in vivo. Rats were exposed to toluene of 10000 ppm for 3 days. 7 days and 14 days by 20 min $\times$ 2 times a day. NADPH diaphorase staining was processed in the different brain regions after inhalation. NADPH diaphorase staining density was not significantly changed at 3 days inhalation group, but the density decreased in proportion to the duration of toluene inhalation. Over 30% of staining density was decreased at 14 days group which was maximum duration of inhalation in this study. The tendency of staining density decrease was significant in granule cell of cerebellum. Cell death by toluene exposure was observed in cultured cerebellar granule cell. $EC_{50}$ measured with LDH leakage assay and MTT test were 43 mM and 72 mM respectively.
Carbon monoxide (CO) intoxication is a leading cause of severe neuropsychological impairments. Peripheral nerve injury has rarely been reported. Following are brief statements describing the motor peripheral neuropathy involved bilateral lower extremities of a patient who recovered following acute carbon monoxide poisoning. After inhalation of smoke from a fire, a 60-year-old woman experienced bilateral leg weakness without edema or injury. Neurological examination showed diplegia and deep tendon areflexia in lower limbs. There was no sensory deficit in lower extremities, and no cognitive disturbances were detected. Creatine kinase was normal. Electroneuromyogram patterns were compatible with the diagnosis of bilateral axonal injury. Clinical course after normobaric oxygen and rehabilitation therapy was marked by complete recovery of neurological disorders. Peripheral neuropathy is an unusual complication of CO intoxication. Motor peripheral neuropathy involvement of bilateral lower extremities is exceptional. Various mechanisms have been implicated, including nerve compression secondary to rhabdomyolysis, nerve ischemia due to hypoxia, and direct nerve toxicity of carbon monoxide. Prognosis is commonly excellent without sequelae. Emergency physicians should understand the possible-neurologic presentations of CO intoxication and make a proper decision regarding treatment.
BASTA is an herbicide which contains glufosinate ammonium as a main component with an anionic surfactant, polyoxyethylene alkylether sulfate, and nonselectively inhibits glutamine synthetase. It became a wildly used herbicide in Korea and its intoxication is now increasing. A 42-year old woman ingested about 300ml of BASTA in a suicide attempt. She showed unconsciousness and respiratory distress in the beginning, and later developed multiple generalized convulsions, low blood pressure, fever and diabetes insipidus. Although she became alert 12 days after the ingestion, she showed retrograde amnesia for a period of about recent 10 years. A neuropsychological test on day 22 revealed frontal lobe dysfunction, visual memory disturbance and slight decrease in visuospatial function. All these neurological abnormalities that might occur due to glufosinate ammonium were almost improved in the follow-up test performed a month later.

Stupp등(1973)에 의하면, Neomycin, polymyxin-G, gentamycin, erythromycin, tetracycline penicillin중, 다만 penicillin만이 독작용이 없다고 알려져 있다. Ototoxic antibiotics는 round window막을 투과하여 내이, perilymph에 들어가고 또다시 Reissner's막을 통하여 Endolymph에 이르게 된다고 보고있다. 주병변은 Stria vascularis의 degeneration, Sensory epithelia의 degeneration 및 Ganglion cell의 degeneration을 이르킨다. 63세 여자의 양측천공성 만성 중이염 환자가 가정에서 중이강내에 Terramycin분말 한 Capsule을 양이에 나누어 넣고, 심한 Sensorineural Deafness를 초래, 6개월이 지난 현재, 병변의 호전을 보지못하고 있는 일증례를 문헌적고찰과 함께 보고하는 바이다.
산화적 스트레스로부터 참다래 과실 추출물의 신경세포 보호효과에 미치는 영향을 알아보기 위하여 신경세포주인 PC12 세포를 이용하여 참다래 과실추출물의 전처리가 산화적 손상으로부터 유발되는 신경세포사멸을 억제할 수 있는지 조사하였다. t-BHP에 의해 유도된 신경세포손상으로부터 세포사멸을 억제하여 세포생존도를 증가시켰으며 세포사멸로부터 형성되는 핵의 농축현상과 단편화가 현저히 감소함을 확인 할 수 있었다. 그리고 Bcl-2 단백의 발현 증가, Bax 단백의 발현 감소, caspase-3의 활성, PARP 분해 단백(85KDa)감소, ERK, p38 활성을 감소시켰다. 따라서 참다래 과실의 추출물은 신경세포증식효과를 통해 신경세포손상으로부터 유발되는 다양한 퇴행성 뇌질환의 예방에 도움이 될 것으로 나타났다.
연구배경 : 만성 폐쇄성 폐질환 환자에서의 다발성 말초신경병변은 상지보다는 하지, 운동 신경 보다는 감각 신경을 주로 침범하며, 병리학적으로 주로 수초탈락을 보인다는 것은 많은 연구에서 일치하는 소견을 보이나, 그 발생 빈도는 보고자마다 서로 다른 결과를 보고하였으며, 정확한 원인인자에 대해서는 확실하게 알려져 있지 않다. 저자들은 만성 폐쇄성 폐질환 환자에서 임상신경학적 검사와 전기 신경학적 검사를 시행하여 다발성 말초신경병변의 발생빈도와 침범부위 및 원인 인자를 찾고자 하였다. 방 법 : 만성 폐쇄성 폐질환의 여러 증상을 보이면서 폐기능 검사상 FEV1과 FEV1/FVC이 70% 이하이고 provocation test 상 가역반응을 보이지 않으며 어떠한 대사성 질환도 지니지 않으며, 신경에 독성을 야기하는 약물 복용의 과거력이 없는 44명을 대상으로 하여 임상 소견으로 팔 다리 저림, 신체검사상 심부건 반사감소, 진동 및 위치감각 저하, 내적근 위축을 조사하였고, 전기 신경학적 검사로서 운동신경과 감각신경의 전도검사 및 침근전도를 시행하여 다발성 말초신경병변을 진단하였다. 각 환자는 연령, 흡연력, 동맥혈 개스 분석, 폐기능검사 소견을 조사하였다. 결 과 : 44명의 대상 환자 중 22명이 전기 신경학적 검사상 진단되어 50%의 발생빈도를 보였으며, 임상 신경학적 검사상 20%인 9명이 양성을 보였다. 전기 신경학적 검사상 양성을 보인 22명 중 임상 신경학적 검사상 양성을 보인 군은 9명이었으며, 13명은 음성을 보여 무증상의 잠재성 다발성 말초신경병변이 많았다. 전기 신경학적 검사상 상지보다는 하지, 운동신경보다 감각신경 이상이 많았으며, 신경 손상 변성이 수초탈락보다 많았다. 유발 인자에 대한 검사로서 연령, 흡연력, 동맥혈 개스 분석, 폐기능 검사 의미 있는 인자는 찾을 수 없었다. 결 론 : 만성 폐쇄성 폐질환 환자에서 다발성 말초신경병변이 비교적 높은 발생빈도를 보여, 이에 대한 조기 발견에 관심을 가져야 하며, 향후 대단위 검사로서 보다 정확한 유병율과 함께 원인과 조직 병리학적인 조사가 더 이루어져야 되리라 여겨진다.
인간이나 환경에 해로운 영향을 주는 지속성 유기오염물질의 독성을 평가하기 위해서 염분과 온도에 대해 내성이 강한 Acartia 종들과 Artemia을 대상으로 실험하였다. 지속성 유기오염물질인 PAHs와 TBT에 대한 요각류의 독성을 평가하기 위해 3가지의 실험을 실시하였다. 1) 광양만에서 주로 나타나는 5가지 PAHs(anthracene, benzo〔a〕pyrene, fluoranthene, phenanthrene, pyrene)에 대한 A. omorii의 48h-LC$_{50}$ 을 구하였다. 2) Artemia를 이용하여 온도에 따른 benzo〔a〕pyrene과 TBT의 독성의 변화를 측정하였다. 3) PhMs중에서 독성이 강한 benzo〔a〕pyrene에 노출된 먹이를 섭취한 A. erythruea와 A. omorii의 난 생산, 부화율, 고형화된 배설물 양의 변화를 측정하였다. A. omorii에 대한 5가지 PAHs 중에서 fluoranthene (48 h-LC$_{50}$ 19.20 $\mu\textrm{g}$ L$_{-1}$)과 benzo〔a〕pyrene (48h-LC$_{50}$ 29.89$\mu\textrm{g}$ L$_{-1}$)의 독성이 강하게 나타났다. 온도실험에서는 동일한 유해물질을 가지고 실험을 하더라도 온도의 변화에 따라 급격한 독성의 차이가 나타날 수 있고, 유해물질 간에도 온도에 따라서 나타나는 독성에 대한 특성이 다르게 나타났다. 특히 15$^{\circ}C$에서는 TBT (9.982 $\mu\textrm{g}$ L$^{-1}$)의 독성이 benzo〔a〕pyrene (1,173 $\mu\textrm{g}$ L$^{-1}$)의 독성보다 약 100배 정도 더 독성이 강하게 나타났다. Benzo〔a〕pyrene에 노출된 먹이는 요각류의 난 생산, 부화율, 고형화된 배설물 양의 변화에 영향을 주었다. 특히 benzo〔a〕pyrene의 농도 증가는 요각류의 생산력 변동에 영향을 미칠 수 있는 것으로 사료된다. 본 연구는 요각류를 이용하여 해양생태계에 유입되는 지속성 유기오염물질에 대한 위험성을 알리는 지표생물로서 사용할 수 있을 것으로 판단된다.
